Understanding the transect survey for abundance of anemone fishes and other living organisms is important to asses reef associated fish diversity in North Bay island. The percentage distribution of 10 different substratum from the disturbed, semi-disturbed and undisturbed areas was recorded during the survey in North Bay islands during November 2009 to April 2010. The survey observations reveal that the fishes were the dominant groups followed by mollusks, lobsters and octopus. There are 5 different anemone fishes were collected during the transect survey and their distribution is more in undisturbed area. We are standardizing the different mass culture techniques for production of phytoplankton and zooplankton for the nutritional source for the anemone fish larvae. Monitoring the water quality parameters and culture the phytoplankton and zooplankton used in different culture media with 2 adjustment studies like with and without salinity adjustment. The results of this experiment indicate that zooplankton was rich in protein and fat content and it will be used as high nutritional source for feeding fish larvae.

Penelitian ini bertujuan untuk mengetahui pengaruh ketinggian air yang berbeda terhadap pertumbuhan dan kelangsungan hidup larva ikan botia (Chromobotia macracanthus, Bleeker). Hasil penelitian menunjukkan bahwa sintasan atau kelangsungan hidup tertinggi terdapat pada perlakuan B dengan ketinggian air 10 cm yaitu sebesar 98.88%, sedangkan kelangsungan hidup terendah terdapat pada perlakuan A dengan ketinggian air 5 cm yaitu sebesar 97.99%. Pertambahan bobot nilai tertinggi terjadi pada perlakuan A dengan ketinggian air 5 cm dengan nilai bobot rata–rata 0.02252 gr dan pertambahan bobot rata–rata terendah terjadi pada perlakuan C dengan ketinggian air 15 cm yaitu sebesar 0.01132 gr. Pertambahan panjang rata–rata tertinggi terjadi pada perlakuan A yaitu sebesar 0.42 cm dan yang terendah terjadi pada perlakuan B yaitu sebesar 0.29 cm. Parameter kualitas air selama penelitian masih dalam kisaran optimal dimana suhu 24,3–28,9oC, pH 6,5–7,0, DO 6,84–7,69 ppm, NH3 0,00–0,03 ppm, NO2 0,00–0,07 ppm. Analisis statistik dengan uji F diperoleh bahwa ketinggian air yang berbeda dalam pemeliharaan larva ikan botia (C. macracanthus, Bleeker) berbeda sangat nyata (P>0.01) terhadap pertambahan bobot dengan nilai Fhitung (21.00) > Ftabel (10.92), dan berbeda sangat nyata terhadap pertambahan panjang dengan nilai Fhitung (23.56) > Ftabel (10.92), sedangkan untuk sintasan atau kelangsungan hidup tidak berbeda nyata antar perlakuan. Hasil uji BNT menunjukkan, setiap perlakuan memberikan pengaruh terhadap pertumbuhan dan tidak berpengaruh terhadap kelangsungan hidup.The study aims to determine the effect of different water levels on the growth and survival of fish larvae Botia (Chromobotia macracanthus, Bleeker). The results showed that the survival rate was highest in treatment B with the water level was 10 cm which was equal to 98.88 %, whereas the lowest survival rate was found in treatment A with the water level was 5 cm which was equal to 97.99 %. The highest value of weight gain occurred in treatment A with average value in weight was 0.02252 g and the average weight gain was lowest in the treatment of C that reached 0.01132 g. The highest growth of length was occured in treatment A that was equal to 0.42 cm and the lowest occurred in treatment B that was 0.29 cm. Water quality parameters during the study were in the tolerance range where the optimum temperature were 24,3-28,9 oC , pH 6.5 to 7.0, DO 6.84 to 7.69 ppm, NH3 0.00 to 0.03 ppm , NO2 0.00 to 0.07 ppm . Statistical analysis by F test showed that the different water levels in the larval rearing of fish Botia (C. macracanthus, Bleeker) was significantly different ( P > 0.01) in the weight gain with the value of Fcount ( 21.00 ) > Ftable ( 10.92 ), and it was significantly different against the length with the value of F count ( 23:56 ) > F table ( 10.92), while for the survival rate showed has not significantly different between treatments. The LSD test showed that all the treatment were effected the growth rate, but not for survival rate.

First culture media for chydoridae used horse manure dried in the sun with a composition of 10 grams and 50 grams of soil dissolved in 1 L of water for 5 days, then filtered and diluted three times. Laboratory-scale cultures were carried out in a 500 ml volume jar containing horse manure culture media; then in the process fed with bread yeast and rice bran (0.05 gr yeast and rice bran/500 ml) every 3 days. Mass culture uses 12 jar containers with a volume of 3 L.The administration of chydoridae to two-day old koi fish larvae was carried out with the following treatments: treatment A, which was ornamental fish fed with boiled egg yolk in ad libitum; treatment B is ornamental fish fed with 5 individual chydoridae/ml and treatment C, which is not given any food with three replications. The design of the response in this study was to see the survival rate of ornamental fish larvae (%).The results of measurements of water quality during the study showed that the temperature during the study was 27°C, while the pH was 7. From the statistical analysis it was found that administration of chydoridae had a significant effect on the survival of two-day-old koi fish larvae which were kept for 14 days.Keywords: chydoridae, life feed, ornamental fish larvae, growthABSTRAKMedia kultur awal chydoridae menggunakan kotoran kuda yang dikeringkan di bawah sinar matahari dengan komposisi 10 gram kotoran kuda serta 50 gram tanah yang dilarutkan dalam 1 L air selama 5 hari, kemudian dilakukan penyaringan dan diencerkan sebanyak tiga kali.  Kultur skala laboratorium dilakukan dalam wadah toples dengan volume 500 ml berisi media kultur kotoran kuda; selanjutnya dalam proses kultur chydoridae diberi makan ragi roti dan dedak (0,05 gr ragi dan dedak /500 ml) setiap 3 hari sekali. Kultur massal menggunakan wadah toples sebanyak 12 buah dengan volume 3 L dengan mengikuti metode pada kultur skala laboratorium.Pemberian chydoridae kepada larva ikan koi yang berumur dua hari dilakukan dengan beberapa perlakuan sebagai berikut : perlakuan A yaitu  ikan hias diberi makan kuning telur rebus secara ad libitum; perlakuan B yaitu  ikan hias diberi makan chydoridae 5 individu/ml dan perlakuan C yaitu  ikan hias tidak diberi makan apapun dengan tiga ulangan.  Rancangan respons dalam penelitian ini adalah melihat laju kelangsungan hidup larva ikan hias (%).Hasil pengukuran kualitas air selama penelitian menunjukkan, suhu selama penelitian adalah 27 °C, sedangkan pH adalah 7.  Dari hasil analisis statistik diperoleh bahwa pemberian chydoridae sangat memberi pengaruh nyata kepada kelangsungan hidup larva ikan koi berumur dua hari yang dipelihara selama 14 hari.Kata kunci: chydoridae, pakan alami, larva ikan hias, pertumbuhan

The rotifer Brachionus rotundiformis (total body length 240.59±10.24 μm, lorica length 175.28±9.18 μm, and lorica width 124.28±7.76μm) is commonly used as starter food in the larval rearing of marine fish. But, larvae of some marine tropical fish species required starter food with body size smaller than B. rotundiformis. The present study was aimed to isolate minute rotifers from nature and to assess the possibility of culturing these rotifers. Sampling of rotifers was conducted in an estuary of Mangket (Kema-Minut), using plankton net (mesh size 40 µm). A trial of culturing the rotifers was conducted at salinities of 10, 20 and 30 ppt by using a microalga, Nannochloropsis oculata. A species of rotifer identified as Colurella sp. (family Lepadellidae) was successfully isolated from the sampling location. Body size of Colurella sp. was extremely small (Total length 123.22±5.45 μm, lorica length 95.96±3.81 μm, and lorica width 53.57±3.11 μm), which were smaller than Brachionus rotundiformis SS-type as a conventional starter food for marine fish larvae.  Results of culturing the minute rotifer Colurella sp. showed that the species grew well at salinities of 10, 20 and 30 ppt with no significant difference among treatments (ANOVA, p>0.05), indicating a potential use of minute rotifer Colurellasp. as starter food for marine fish larvae. Rotifera Branchionus rotundiformis (ukuran tubuh: panjang total 240,59±10,24 μm, panjang lorika 175,28±9,18 μm, dan lebar lorika 124,28±7,76μm) sering digunakan sebagai pakan awal pemeliharaan larva ikan laut. Namun, larva beberapa spesis ikan laut tropis membutuhkan pakan awal berukuran tubuh lebih kecil dari Branchionus rotundiformis. Penelitian ini bertujuan untuk mendapatkan minute rotifer dari alam (berukuran tubuh lebih kecil dari B. rotundiformis) dan menguji kemungkinan pemeliharaannya. Sampling rotifer dilakukan di perairan estuari Desa Mangket (Kema-Minut), menggunakan plankton net (ukuran mata jaring 40 µm). Uji coba pemeliharaan dilakukan pada salinitas (10, 20, dan 30 ppt) dengan menggunakan Nannochloropsis oculata. Satu spesies minute rotifer yang teridentifikasi sebagai Colurella sp. (family Lepadellidae) berhasil diisolasi dari lokasi sampling. Colurella sp. memiliki ukuran tubuh sangat kecil (panjang total [PT] 123,22±5,45 µm, panjang lorika [PL] 95,96±3,81 µm, dan lebar lorik [LL] 53,57±3,11 µm) yang mana lebih kecil dari Branchionus rotundiformis tipe-SS sebagai pakan awal larva ikan laut. Hasil uji coba pemeliharaan minute rotifer Colurella sp. menunjukkan bahwa spesis ini dapat tumbuh pada salinitas 10, 20, dan 30 ppt dengan perbedaan kepadatan populasi yang tidak signifikan antar perlakuan (Uji ANOVA, p > 0.05) mengindikasikan potensi pemanfaatan minute rotifer Colurella sp. sebagai pakan awal larva ikan laut.

pH is one of the important water quality parameters in aquaculture. This study aimed to observe the growth performance and survival rate of common carp (Cyprinus carpio) Rajadanu strain reared in culture media with different pH levels. Fish (length: 3.60 ± 0.18 cm; weight: 1.68 ± 0.18 g) were stocked in nine aquariums (40 cm x 40 cm x 30 cm) for 10 days with a stocking density of 25 fish per aquarium. The treatments used were (A) pH 4-5, (B) pH 5-6, and (C) pH 6-7, with three replications. A commercial feed containing 28% protein was given daily and as much as 3% of the total biomass. The observed data were analyzed using variance analysis (ANOVA), followed by Duncan test. The results showed that the pH treatments did not affect the growth (length and weight) and survival rate of common carp Rajadanu strain (P>0.05). The best growth was achieved by the fish group reared in water with a pH range of 6-7 (length = 0.38 cm; weight = 0.17 g). The highest survival was attained by the fish group reared in water with a pH range of 5-6 (90.66%).

Abstract One type of phytoplankton that are used in marine fish hatchery activities namely Nannochloropsis oculata. The purpose of study case is to know patterns growth of N. oculata in culture scale laboratory, intermediate, and bulk. Study was held in Situbondo Brackishwater Aquaculture Center of East Java in January-February 2011. Study case methods used is descriptive method. N. oculata culture techniques performed by multilevel methods, beginning with the isolation of phytoplankton in the media for/liquid, culture in test tubes 10 ml and elenmeyer 50-100 ml, culture on the bottle 100-1000 ml, culture in karboy of 20 liters, intermediate scale culture 100 - 1000 liters and a mass-scale culture with a volume of more than 1000 liters. Laboratory-scale use Walne for fertilizer. Intermediate-scale use of fertilizers FeCl3 1-2 ppm, 10 ppm EDTA, Na2HPO4 10-15 ppm, 100-150 ppm KNO3, and Tracemetals/vitmix 5 ml/m3, mass-scale culture use of fertilizers FeCl3 1 ppm, 5 ppm EDTA, TSP 20 ppm, ZA 40 ppm and 50 ppm urea. Water quality measurement results obtained by laboratory scale temperature is 23 - 25°C, intermediates and mass scale is 30°C, pH in the culture laboratory scale and intermediate is 7.7 to 7.8, mass culture pH is 7.9, salinity laboratory scale 29-30 ppt, intermediates and mass scale 30 ppt. During mass culture of N. oculata showed that the growth pattern in accordance with the general pattern of phytoplankton growth. Mass-scale culture produce 1504 x 104 cells/ml for six days.

Serum in embryo culture medium may be a potential cause of abnormal accumulation of lipid droplets, which is correlated to a higher sensitivity to cryopreservation. Moreover, serum may introduce pathogens. With the aim of developing a serum-free culture medium, we first (Experiment 1) investigated the effect of adding ITS (5 μg/mL insulin, 5 μg/mL transferrin, 5 ng/mL selenium) as a serum substitute in SOF medium on embryos cultured in large groups (20 embryos per culture drop of 20 μL) and we then (Experiment 2) analyzed the effect of adding BSA. In this second experiment, our serum-free culture media were also tested on embryos cultured in small numbers (5 embryos per drop of 20 μL) in order to mimic ovum pickup (OPU) conditions. Embryos were obtained from slaughterhouse oocytes, matured in vitro for 24 h in a serum-free enriched 199 medium (Donnay et al. 2004 Reprod. Fertil. Dev. 16, 274) containing ITS, and fertilized for 18 h. In experiment 1, embryos were cultured in SOF (Holm et al. 1999 Theriogenology 52, 683–700) supplemented with 0.1 mg/mL polyvinylpyrrolidane (PVP) without (SOF) or with ITS (SOF-ITS), or with 5% FCS (SOF-FCS). Cavitation occurred earlier in presence of serum (Table). Adding ITS to SOF increased blastocyst rates at Day 7 and Day 8 post-insemination (p.i.) and also the hatching rate. In experiment 2, embryos were cultured in SOF-FCS, SOF-ITS, or SOF-ITS supplemented with 4 mg/mL fatty acid free BSA (SOF-ITS-BSA). Within each condition, no differences were observed for blastocyst and hatching rates between embryos cultured in large or in small groups. Adding BSA to SOF-ITS increased blastocyst rate at Day 6 p.i. and also the hatching rate. At Days 7 and 8 p.i., blastocyst rates were higher in SOF-FCS than in SOF-ITS and tended to be higher than in SOF-ITS-BSA, especially for embryos cultured in small groups. Cell numbers of the resulting embryos were unaffected. These results indicate that: (1) ITS as supplement to SOF medium promotes embryo development in vitro. (2) BSA as protein supplement to SOF-ITS medium accelerates blastulation and improves hatching rate. (3) SOF-ITS and SOF-ITS-BSA are two serum-free culture media that can sustain development of embryos, also when cultured in small number, even though SOF-FCS tended to afford better rates of development. Further studies will include evaluation of other quality parameters including resistance to cryopreservation. The increase of different types of cell cultures, which can be used for the in vitro studies of physiological and/or pathological processes, has introduced the need to improve culture techniques through the use of materials and culture media that promote growth, recreating a cellular micro-environment that can be asserted in in vivo condition. The standard methods for the functionalization of supports used for cell cultures are based on the use of synthetic or natural biopolymers, which generally have high costs, such as poly-lysine and polyornithine. The aim of this work is to demonstrate the alternative use of the polysaccharide chitosan as adhesion factor and structural component for 2D/3D neuronal cultures. Thanks to its versatility, it could be easily functionalized for the fabrication of personalized of in vitro models

Abstract Folliculogenesis is a complex process that requires integration of autocrine, paracrine, and endocrine factors together with tightly regulated interactions between granulosa cells and oocytes for the growth and survival of healthy follicles. Culture of ovarian follicles is a powerful approach for investigating folliculogenesis and oogenesis in a tightly controlled environment. This method has not only enabled unprecedented insight into the fundamental biology of follicle development but also has far-reaching translational applications, including in fertility preservation for women whose ovarian follicles may be damaged by disease or its treatment or in wildlife conservation. Two- and three-dimensional follicle culture systems have been developed and are rapidly evolving. It is clear from a review of the literature on isolated follicle culture methods published over the past two decades (1980–2018) that protocols vary with respect to species examined, follicle isolation methods, culture techniques, culture media and nutrient and hormone supplementation, and experimental endpoints. Here we review the heterogeneity among these major variables of follicle culture protocols.

Air is one the major source for all living organisms to sustain on the earth. Nowadays due to heavy traffic and industrial population most of the cities are affected by air pollution. In this research the Marudhamalai Hill Shrine was selected as the study area to monitor the air quality in its vicinity. Marudhamalai is located on the Western Ghats in Coimbatore district. It is famous for the Murugan temple located on it as people from Coimbatore and other parts of India visit every year. From the results it was clear that the forest surrounding the hill shrine not have medicinal plants but these plants help in to maintain the air quality parameters resulting in the quality to be within the prescribed limits.

Keberhasilan kegiatan budidaya perikanan harus ditunjang dengan ketersediaan benih yang berkesinambungan. Oleh karena itu, diperlukan juga ketersediaan pakan alami larva berupa rotifera (Brachionus rotundiformis). Desain percobaan berupa rancangan faktorial dengan dua faktor dan lima ulangan diaplikasikan dalam penelitian ini. Sebagai perlakuan berupa suhu ruang penyimpanan (suhu kamar, suhu ruang AC, dan suhu refrigerator/lemari es) dan pakan mikroalga konsentrat (monospesies dan multispesies). Bakteri probiotik juga digunakan sebagai pengontrol kualitas air. Spesies mikroalga yang digunakan adalah Nannochloropsis sp., Dunaliella sp., Isochrysis sp., dan Pavlova sp. Parameter yang diukur adalah kelimpahan rotifera dan parameter kualitas air media kultur (pH, salinitas, DO, dan NH3). Analisis data terdiri atas analisis regresi, analisis ragam, dan uji keparalelan. Hasil pengukuran parameter kualitas air selama penyimpanan menunjukkan kondisi media yang relatif stabil dan merupakan kisaran optimum bagi pertumbuhan B. rotundiformis. Kelimpahan maksimum tertinggi dari B. rotundiformis baik pada perlakuan pakan monospesies maupun multispesies alga adalah pada suhu kamar. Dari interaksi kedua perlakuan, diperoleh kelimpahan akhir tertinggi pada suhu ruang AC–pakan multispesies. Hal ini menunjukkan bahwa rotifera dapat disimpan lebih lama pada suhu ruang AC dengan pemberian pakan multispesies alga.The success of any aquaculture practices should be supported by sustainable supply of fish fry. Therefore, the availability of rotifers (Brachionus rotundiformis) as natural feed for fish larvae is required. The research was arranged in factorial design with two treatments and five replications. Treatments consisted of different room storage temperatures (refrigerator, room temperature, and room with air conditioner/AC) and microalgae concentrate added as rotifer feed (monospecies and multispecies algae). Probiotic bacteria was used to control water quality. Mikroalgae species consisted of Nannochloropsis sp., Dunaliella sp., Isochrysis sp., and Pavlova sp. Parameters measured were rotifer density and water quality of rotifer media (pH, salinity, DO, and NH3). Data analysis included regression analysis, analysis of varians and parallel testing. The results of water quality parameters during rotifer storage showed that media conditions were relatively stable and optimal for B. rotundiformis growth. The result of treatments interaction showed that the highest maximum density of rotifer at the end of the research was achieved by rotifer stored in air conditioned room fed with multispecies algae. This showed that rotifers can be stored longer in room storage with AC and fed by multispecies algae.
