AbstractEastern North American migratory monarch butterflies (Danaus plexippus) have faced sharp declines over the last two decades. Although captive rearing has been used as an important tool for engaging the public and supplementing conservation efforts, a recent study that tested monarchs in a flight simulator suggested that captive-reared monarchs lose their capacity to orient southward during fall migration to their Mexican overwintering sites. We raised offspring of wild-caught monarchs on swamp milkweed (Asclepias incarnata) and, after eclosion, individuals were either tested in a flight simulator or radio-tracked in the wild using array of over 100 automated telemetry towers. While only 33% (7/39) of monarchs tested in the flight simulator showed strong southeast to southwest orientation, 97% (28/29) of the radio-tracked individuals were detected by automated towers south or southeast of the release site, up to 200 km away. Our results suggest that, though captive rearing of monarch butterflies may cause temporary disorientation, proper orientation is likely re-established after exposure to natural skylight cues.

The “Tulsi” plant or Indian basil occupies an important place in the Indian population. The plant grows in the wild in the tropics and other warm regions. The study followed an exhaustive literature survey and further concluded its uses in edibles as well as in therapeutic formulations. Leaves of Tulsi was found to be used in various pathological conditions like cough, respiratory disorders, eye disorders, depression and fever. Traditionally, tulsi is consumed as many practices like in dried powder, herbal tea and fresh leaf and/or mixed with ghee. Essential oil obtained from “Karpooratulasi” is commonly used for medicinal values as well as in herbal cosmetics.The active chemical constituents found in tulsi are eugenol, ursolic acid, oleanolic acid, rosmarinic acid, carvacrol, linalool and β-caryophyllene.

Abstract Captive-rearing conservation programs focus primarily on maximizing postrelease survival. Survival increases with size in a variety of taxa, often leading to the use of enhanced size as a means to minimize postrelease losses. Head-starting is a specific captive-rearing approach used to accelerate growth in captivity prior to release in the wild. We explored the effect of size at release, among other potential factors, on postrelease survival in head-started Mojave desert tortoises Gopherus agassizii. Juvenile tortoises were reared for different durations of captivity (2–7 y) and under varying husbandry protocols, resulting in a wide range of juvenile sizes (68–145 mm midline carapace length) at release. We released all animals (n = 78) in the Mojave National Preserve, California, United States, on 25 September 2018. Release size and surface activity were the only significant predictors of fate during the first year postrelease. Larger sized head-starts had higher predicted survival rates when compared with smaller individuals. This trend was also observed in animals of the same age but reared under different protocols, suggesting that accelerating the growth of head-started tortoises may increase efficiency of head-starting programs without decreasing postrelease success. Excluding five missing animals, released head-starts had 82.2% survival in their first year postrelease (September 2018–September 2019), with all mortalities resulting from predation. No animals with >90-mm midline carapace length were predated by ravens. Our findings suggest the utility of head-starting may be substantially improved by incorporating indoor rearing to accelerate growth. Target release size for head-started chelonians will vary among head-start programs based on release site conditions and project-specific constraints.

Wild stocks of Pacific salmonids have experienced sharp declines in abundance over the past century. Consequently, billions of fish are released each year for enhancing abundance and sustaining fisheries. However, the beneficial role of this widely used management practice is highly debated since fitness decrease of hatchery-origin fish in the wild has been documented. Artificial selection in hatcheries has often been invoked as the most likely explanation for reduced fitness, and most studies to date have focused on finding signatures of hatchery-induced selection at the DNA level. We tested an alternative hypothesis, that captive rearing induces epigenetic reprogramming, by comparing genome-wide patterns of methylation and variation at the DNA level in hatchery-reared coho salmon (Oncorhynchus kisutch) with those of their wild counterparts in two geographically distant rivers. We found a highly significant proportion of epigenetic variation explained by the rearing environment that was as high as the one explained by the river of origin. The differentially methylated regions show enrichment for biological functions that may affect the capacity of hatchery-born smolts to migrate successfully in the ocean. Shared epigenetic variation between hatchery-reared salmon provides evidence for parallel epigenetic modifications induced by hatchery rearing in the absence of genetic differentiation between hatchery and natural-origin fish for each river. This study highlights epigenetic modifications induced by captive rearing as a potential explanatory mechanism for reduced fitness in hatchery-reared salmon.

AbstractReintroduction is an important strategy to restore or re-establish wild populations of endangered species. Pre-release training is a necessary step to ensure post-reintroduction survival. However, studies reported contradicting outcomes after pre-release training of juveniles and adults. This study used farmed and feral American mink (Neovison vison) to analyze the influence of captive breeding on the morphology, structure and efficiency of the two major hindlimb levers, the femur and tibia pivoted by hip and knee joints that are essential for locomotion. Results showed that captive breeding did not alter the sexual dimorphism of the two levers that are related to survival in the wild. Captive-bred mink showed slightly altered morphology of the femur and fundamental structure of the hindlimb levers that improved efficiency, but this resulted in reduction of performance related to foraging in both terrestrial and aquatic environments, especially for females. These findings suggest that reintroduction of mustelid as exampled by the mink here should focus on juveniles because the skeletal alterations associated with captive rearing were recorded only among adults and are irreversible in adulthood. In contrast, captive-reared juveniles showed no skeletal alterations and would be expected to recovery from any atrophy of the muscular system caused by captive rearing for shorter durations. Our results support the application of pre-release training of juveniles in enriched environments as a method for alleviating structural alteration of appendages and enhancing locomotion to increase survival probability in complex habitats.

Our previous studies have demonstrated that abnormally high amounts of calcium (Ca) and zinc (Zn) can be accumulated in human retina-choroid under pathological conditions and that barium (Ba), which was not detected in the eyes of healthy individuals, is deposited in the retina pigment epithelium (RPE), and to a lesser extent in the sensory retina and iris. In an attempt to understand how these cations can be accumulated in the vertebrate eye, a morphological and microanalytical study of the uptake and loss of specific cations (K, Ca,Ba,Zn) was undertaken with incubated Rana catesbiana isolated retina and RPE preparations. Large frogs (650-800 gms) were dark adapted, guillotined and their eyes enucleated in deep ruby light. The eyes were hemisected behind the ora serrata and the anterior portion of the eye removed. The eyecup was bisected along the plane of the optic disc and the two segments of retina peeled away from the RPE and incubated.

Recently, the functional aspect as well as morphological aspect of the reserve cells in the cervix uteri drew much attention in view of the carcinogenesis in squamocolumunar junction. In this communication, the authors elucidate the ultrastructural features of the reserve cells in patients of various age groups visiting our university hospital and affiliated hospital.From conventional light microscopic point of view, the reserve cells tend to be pronounced in various pathological conditions, such as the persisting inflammation, proliferative disorders and irritation of hormones. The morphological patterns of the reserve cells from various stage and degree of irritation were observed.

The cervix is an important organ in reproduction. Its malfunction is frequently a factor for infertility. Ectocervix region does not appear to have received much attention although many studies have been reported on the endocervix. We report here our SEM observations on ectocervix in certain pathological conditions compared to normal ectocervix.Ectocervix specimens from human females with specific pathological disorders were processed for Scanning Electron Microscopy by conventional method and they were examined in a Philips SEM.The normal ectocervix is lined by flat layer of squamous epithelial cells with microridges (Fig. 1). These cells are known to be formed from columnar cells through metaplastic transformation. The cells of carcinoma-bearing ectocervix show a disorganised appearance (Fig. 2). In non-malignant tumour surface some cuboidal and few columnar cells were seen (Fig. 3). A cyst appears like an overgrowth on the surface of the squamous epithelium (Fig. 4). In ulcerated ectocervix a marked reduction of epithelial cells are observed (Fig. 5); the cells are devoid of microridges and, the large polygonal cells, as observed in normal tissues, have somehow acquired comparatively small hexagonal shape

Nectria haematococca mating population I (anamorph, Fusarium solani) macroconidia attach to its host (squash) and non-host surfaces prior to germ tube emergence. The macroconidia become adhesive after a brief period of protein synthesis. Recently, Hickman et al. (1989) isolated N. haematococca adhesion-reduced mutants. Using freeze substitution, we compared the development of the macroconidial wall in the wild type in comparison to one of the mutants, LEI.Macroconidia were harvested at 1C, washed by centrifugation, resuspended in a dilute zucchini fruit extract and incubated from 0 - 5 h. During the incubation period, wild type macroconidia attached to uncoated dialysis tubing. Mutant macroconidia did not attach and were collected on poly-L-lysine coated dialysis tubing just prior to freezing. Conidia on the tubing were frozen in liquid propane at 191 - 193C, substituted in acetone with 2% OsO4 and 0.05% uranyl acetate, washed with acetone, and flat-embedded in Epon-Araldite. Using phase contrast microscopy at 1000X, cells without freeze damage were selected, remounted, sectioned and post-stained sequentially with 1% Ba(MnO4)2 2% uranyl acetate and Reynold’s lead citrate. At least 30 cells/treatment were examined.
