AbstractExtracellular vesicles (EVs) reflect the cell of origin in terms of nucleic acids and protein content. They are found in biofluids and represent an ideal liquid biopsy biomarker source for many diseases. Unfortunately, clinical implementation is limited by available technologies for EV analysis. We have developed a simple, robust and sensitive microscopy-based high-throughput assay (EVQuant) to overcome these limitations and allow widespread use in the EV community. The EVQuant assay can detect individual immobilized EVs as small as 35 nm and determine their concentration in biofluids without extensive EV isolation or purification procedures. It can also identify specific EV subpopulations based on combinations of biomarkers and is used here to identify prostate-derived urinary EVs as CD9-/CD63+. Moreover, characterization of individual EVs allows analysis of their size distribution. The ability to identify, quantify and characterize EV (sub-)populations in high-throughput substantially extents the applicability of the EVQuant assay over most current EV quantification assays.

An experiment has been conducted to assess the real time utility and abundance of organic minerals in cultivation of common wheat, Triticum aestivum L. in Bundelkhand region of Uttar Pradesh with the aim of finding the effect of available soil organic minerals on various biochemicals or chemical characters including yield attributes. For the experiment, the cultivar namely RAJ-4037 which is best for dry land area and crop matures in 120 days. This variety is suitable for bakery and beverage industry, has been taken. The pre availability of manures were measured and the application of various fertilizers have been done as per the recommended dose for the cultivation. The data of various biochemical characters like Dry matter accumulation, Protein content, NPK in grain, NPK in straw, yield and biological yields have been recorded as per standard methods. The results revealed that organic minerals had a significant impact to influence the various biochemical traits such as dry matter accumulation maximum in FYM, Protein content (11.18), NPK (0.60; 0.35 and 0.36) were recorded maximum in vermin compost treatment.

Ninety-nine F4 lines from the cross Lennox × Stadler of common wheat (Triticum aestivum L.) ranged widely in both kernel hardness and kernel protein content. A nonsignificant correlation of r = −0.17 showed the absence of any tendency for hardness and high protein to be associated, contrary to what might be expected from the association found in commercial cultivars.

SummarySome morphological, biochemical and functional parameters of platelet population in children with cyanotic congenital heart disease (CCHD) were studied by making comparisons of the normal platelet population in both CCHD patients and controls. The mean volume of the platelets from cyanotic patients was greater than from normals. The platelet size distribution curves demonstrated a shift towards larger than normal size in the case of CCHD. The mean protein content, as well as the mean PF3 content of platelets was increased in CCHD. Following addition of kaolin, PF3 release was more rapid and of shorter duration with platelets from CCHD patients as compared to normal platelets. They also released more PF3 than did normal platelets. After addition of ADP, collagen, or adrenalin, platelets of CCHD patients were more responsive than similarly treated platelets from normals. Platelets from CCHD showed an increased initial rate of aggregation and greater maximum aggregation. These data suggested that the platelet population of CCHD patients consists of larger, younger and functionally more active platelets than does the platelet population of normals.

Aim. The aim of this study was to analyze quality indices in mutants at gliadin loci developed on the basis of the common wheat cultivar Bezostaya 1. Methods. Acid polyacrylamide gel electrophoresis and SDS-electrophoresis of storage proteins were performed to check mutations. Grain quality indices (SDS sedimentation volume, grain hardness, seed plumpness, and grain protein content) were analyzed: Results. Both mutants at Gli-R1 with the intensified secalin component and with the changed mobility of the secalin component showed lower grain hardness in comparison with the respective non-mutant lines. The mutation of the allele Gli-B1b causing the absence of synthesis of the g-gliadin and the minor w-gliadin resulted in the decrease in grain hardness and in the increase in the SDS-sedimentation volume. Seeds of the Gli-B1b mutant with the absence of synthesis of the major w-gliadin were less plump and showed higher protein content than in the non-mutant line. Conclusions. The effects of mutations of the allele Gli-B1b associated with the absence of certain gliadin components and Gli-R1 mutations on grain quality indices were revealed.
