Title (Bahasa Indonesia): Analisis pertumbuhan dan kualitas karagenan rumput laut Kappaphycus alvarezii pada lokasi berbeda di Wilayah Perairan Banggai ProvinsiSulawesi Tengah Seaweed is a potential of coastal resources. Carrageenan is a polysaccharide extracted from seaweed or some species of red algae (Rhodophyceae). Seaweed growth is strongly influenced by two factors: internal and external factors. But twthat determine the success of the seaweed growth is the management carried out by people working on it. Banggai Regency is one of the largest seaweed production centers in Central Sulawesi. The main objective of this studyis toexamine the potential of seaweed cultivation (Kappaphycus avarezii) by looking at the growth and the carrageenan, inBanggai waters, Central SulawesiProvince. The temperature range obtained during this study r25to 31ºC. The results of carrageenananaliysis wasvery different due to differences inlocation, showed by content.  The highest and lowest ashcontentwere obtained from two locations, 1,8% (Jayabakti) and 2.8% (Liang), respectively.Rumput laut merupakan sumberdaya pesisir yang sangat potensial. Karagenan merupakan polisakarida yang diekstraksi dari beberapa spesies rumput laut atau alga merah (rhodophyceae). Pertumbuhan rumput laut sangat dipengaruhi oleh dua faktor yaitu faktor internal dan faktor eksternal. Namun yang sangat menentukan keberhasilan pertumbuhan rumput laut yaitu pengelolaan yang dilakukan oleh manusia. Kabupaten Banggai merupakan salah satu sentral produksi rumput laut terbesar di Sulawesi Tengah. Tujuan utama penelitian ini mengkaji tentang potensi budidaya rumput laut (Kappaphycus alvarezii) yang dikembangkan dengan melihat pertumbuhan dan analisis karaginannya di perairan Kabupaten Banggai Provinsi Sulawesi Tengah. Kisaran suhu yang didapat selama penelitian ini adalah berkisar 25–31ºC. Hasil analisa rendemen karagenan ini sangat berbeda yang disebabkan oleh perbedaan lokasi memberikan pengaruh nyata terhadap kandungannya. Nilai kadar abu tertinggi dan terendah berturut-turut yang di peroleh dari kedua lokasi ini adalah 1,8% (Jaya Bakti) dan 2,8% (Liang).

Abstract Fish meat is more nutritious source of protein and relatively safe for human consumption. Fish meat more easily damaged than red meat product quality, so it is necessary that the process of pickling fish maintained in fresh condition and suitable for consumption. The ethanol extract of red algae Kappaphycus alvarezii been known have antioxidant and antimicrobial effects, so need some research on the use of red algae extract as a preservative. Red algae extract with a concentration of 400 ppm, 500 ppm, 600 ppm and 1 % formalin, each sample is used as a meat preservative for tilapia fillets as 40 grams. Samples of tilapia meat is soaked in an preservative solution as long as 60 minutes. During immersion, the samples of tilapia meat stored at room temperature. Observation of tilapia meat quality is done about six hours after meat removed from the preservative solution. Fish meat quality were analyzed to describe the organoleptic quality, methods of the TPC, and proximate analysis. The results showed that the ethanol extract of red algae Kappaphycus alvarezii 600 ppm has potential as a natural preservative based on test results of TPC, proximate and organoleptic test.

Bacteria are microscopic organism found living in marine algae.  So far, species of bacteria in marine algae are not well known.  In this study, epiphytic bacteria in algal species of Kappaphycus alvarezii (red algae) were isolated to amplify their 16S-rRNA gene. Sample K.alvarezii was collected from the island of Nain.  The isolated epiphytic bacteria from the red algae K.alvarezii were grown in Nutrient Broth (NB) media.  DNA extraction was carried out using InnuPREP DNA Mini Kit.  16SrRNA genes was performed using primer pair of 8F and 1492R. Two different character of epiphytic bacteria were successfully isolated from K. alvarezii.  16S-rRNA genes from the two isolates was successfully amplified, indicated by the presence of DNA band in each sample gel electrophoreses at around 1500 bp.Keywords : Amplification, 16S-rRNA Genes, Kappapchycus alvarezii, Bacterial, Epiphytic  Abstrak Bakteri adalah organisme mikroskopis yang ditemukan hidup di alga laut. Sampel K.alvarezii dikumpulkan dari pulau Nain. Bakteri epifit terisolasi dari alga merah K.alvarezii ditanam dalam media Nutrient Broth (NB). Ekstraksi DNA dilakukan dengan menggunakan InnuPREP DNA Mini Kit. Gen 16S-rRNA dilakukan dengan menggunakan pasangan primer 8F dan 1492R. Dua karakter bakteri epifit yang berbeda berhasil diisolasi dari K. alvarezii. Abstract Mackerel (Rastrelliger sp.) is a kind of fish that have high economic value and good nutrition but susceptible to decay. People often to use formaldehyde as a preservative so that the fish does not quickly to decay. The use of formaldehyde could be replaced by natural ingredients that contain antibacterial compounds, one of which is Kappaphycus alvarezii. This study aims to determine the effect of red algae extract (K.alvarezii) against the total number of bacteria and organoleptic value of mackerel (Rastrelliger sp.). The research design that done in this study is Completely Randomized Design (CRD). The treatments that given are immersion of mackerel in a solution of extract of K. alvarezii with a concentration of 0 ppm, 600 ppm, 700 ppm, 800 ppm and 1% solution of formaldehyde with four repetitions in each treatment. Data analyzes using Analysis of Variants (ANOVA) and followed by Duncan's Multiple Range Test to determine differences between treatments. The results showed that the extract of red algae (K.alvarezii) at a concentration of 0 ppm, 600 ppm, 700 ppm and 800 ppm significantly (p <0.05) against the total number of bacteria of mackerel (Rastrelliger sp.). The ability of extract of K. alvarezii at a concentration of 800 ppm equivalent to the ability of formaldehyde as an antibacterial ingredient in mackerel. K.alvarezii extract could inhibit the growth of bacteria but have not been able to maintain the quality of mackerel based on organoleptic test.

Rhodophyta (red algae) comprises over 6000 species, however, there have only been a few comparative transcriptomic studies due to their under-representation in genomic databases. Kappaphycus alvarezii, a Gigartinales algae, is a valuable source of carrageenan and is extensively cultivated in many countries. The majority of seaweed farming in Southeast Asia is done in intertidal zones under varying light (i.e., spectra and irradiance) and carbon dioxide (CO2) conditions, which affects the rate of photosynthesis. This study conducted transcriptome profiling to investigate the photosynthetic mechanisms in K. alvarezii exposed to different wavelengths of light (i.e., blue, green, and red light, in comparison to white light) and CO2 availability. We analyzed the responses of photosynthetic protein complexes to light and observed that light of different wavelengths regulates a similar set of photosynthetic apparatuses. Under CO2 enrichment, genes encoding C3 and C4 enzymes were found to be actively transcribed, suggesting the likely shift in the carbon metabolism pathway or the involvement of these genes in adaptive physiological processes. This study contributes to the understanding of the regulatory mechanisms of photosynthetic carbon metabolism in red algae and has implications for the culture and commercial production of these economically valuable macroalgae.

Marine natural products from seaweeds have been the focus of research for novel products of pharmacological interest. Bioactivities of the phytochemicals derived from marine algae are important ingredients in many products, such as cosmetics and drugs for treating cancer and other lifestyle disorders. In this context, the aim of the present study is to compare the anti-inflammatory, analgesic, antipyretic properties of the purified terpenoids from the selected red algae Hypnea musciformis, Gracilaria dura and Kappaphycus alvarezii. Initially, the acute toxicity of the purified terpenoid extract was analyzed and was greater than 4000 mg/kg bw. In vitro BSA denaturation assay revealed significant % inhibition as compared to control. In the carrageenan induced paw edema, the purified terpenoids extract of H. musciformis exhibited remarkable soothing activity with 68.8% percentage of inhibition, which was comparable with that of indomethacin. Meanwhile G. dura and K. alvarezii showed 53 and 50% of inhibition respectively. Analgesic activity was determined by hot plate and acetic acid induced writhing test. In the hot plate test, terpenoids extract of H. musciformis and G. dura significantly increased the hot-plate latency as compared to normal saline, which reflects their analgesic efficacy. In the writhing test, terpenoid extracts from H. musciformis, G. dura and K. alvarezii inhibited the writhing response induced by acetic acid in a concentration dependent manner that suggests its varied mode of inhibition of stretching episodes. Lastly, the antipyretic activity was analyzed. Terpenoid extracts from H. musciformis, G. dura and K. alvarezii revealed a significant (P < 0.01) antipyretic activity up to 180 min.  In summary, the study demonstrates the anti-inflammatory, analgesic and antipyretic effects of terpenoids extracts from H. musciformis, G. dura and K. alvarezii on experimental models, suggesting its therapeutic potential in the treatment of peripheral painful and inflammatory pathologies.
