Abstract Background:Increasing evidences have unveiled the connection between microbiome and lung cancer. This study aims to identify the characteristics of microbial communities in the lung cancer tissues from patients in southwestern China, and to compare the distinct microbial genes at different clinical stages of lung cancer for uncovering potential immunotherapy targets.Methods:Forty samples of primary lung adenocarcinoma tissue were performed by 16S rRNA gene sequencing. The subjects were grouped according to TNM stages (T and N group), clinical stage and smoke status. To identify the taxa composition of each sample, Operational Taxonomic Units (OTUs) were classified on the Effective Tags with 97% identity. The linear discriminant analysis effect size (LEfSe) method was utilized to compare relative abundances of all bacterial taxa between non-metastasis group and metastasis group. The Shannon index of the 97% identity OTUs was calculated to evaluate alpha diversity. Beta diversity measurement was calculated using Principal Co-ordinates Analysis (PCoA).Results:A total of 951 OTUs were identified in the cancer tissues, including 224 overlapping genera. No significant difference has been found in the alpha diversity within all the groups. Beta diversity was significantly different in N group, T group and clinical stage group. By LEfSe analysis, nine differential species were identified in the N group, of which the relative abundance of genus Bifidobacterium was 10.78%±11.59% in the N0 group and 20.15%±13.44% in the N+ group (p<0.05). In the T1 and T2 group, the LEfSe result identified 4 phylum and 10 genera. The differential genera were Moraxella, Dolosigranulum, Corynebacteriaceae and Citrobacter in the T2 group and Bifidobacterium, Alistipes, Akkermansia, Blautia, Lactobacillus as well as Facelibacterium in the T1 group. Differential bacterial composition and abundance were also observed in the clinical stage group.Conclusions:In conclusion, by 16S RNA sequencing, we identified dominant species of lung cancer tissue in different groups of AD patients. Bifidobacterium plays important role both in lymph node metastasis and tumor progression, which could provide specific immunotherapy strategy for lung cancer.

Objective: To analysis of the effect of video-assisted thoracoscopic surgery on immune function and survival in patients with early stage lung cancer undergoing radical operation. Methods From August 2009 to May 2011, 96 cases of early stage lung cancer undergoing radical operation patients were studied in our hospital.According to the simple random method were divided into control group and observatio n group, each group of 48 patients. The control group underwent thoracotomy and resection of lung cancer and the observation group underwent endoscopic resection of lung cancer. Comparison of the two groups of patients with surgery, immune parameters, quality of life and prognosis. Results The operation situation of the two groups were compared, found no significant difference between the two groups of patients with operation time (P>0.05), but the amount of intraoperative bleeding, postoperative drainage, extubation time and postoperative ambulation time were significantly lower than the control group (P<0.05); two groups of patients before and after surgery, IgA IgM had no significant difference (P>0.05), and IgG patients aftersurgery is higher than the level before surgery, andCD3+,CD4+,CD8+T cell fraction less than before the surgery, compared with statistical significance (P<0.05); the patients in the observation group 1 days after IgG was significantly higher than control group, and CD3+,CD4+,CD8+T cell fraction in 1 days after surgery was lower than that of control group, compared with statistical significance (P<0.05); the control group of patients with 5 year survival rate was 43.75% and the observation group patients 5 year survival rate was 56.25%. Two groups of patients with 5 year survival rate, recurrence rate and distant metastasis showed no significant difference (p>0.05). But the recurrence time of the observation group was significantly higher than the control group (P<0.05); the two groups of patients after treatment, the physiological status, social / family status, emotional status, functional status, additional lung cancer status was better than that before treatment (P<0.05), and the observation group of patients with physiological status, social / family, emotional status, functional status, additional lung cancer status was significantly better than the control group (P<0.05). Conclusion Compared with thoracotomy, video assisted thoracoscopic surgery for early lung cancer resection, with small trauma, short recovery time of patients with, and has little effect on the immune function of patients, prolong the recurrence time of patients, improve the quality of life of patients.

AbstractObjective: To study the effects of single-nucleotide polymorphisms of the OPRM1 and ABCB1 genes on the analgesic effect and consumption of sufentanil after thoracoscopic-assisted radical resection of lung cancer.Methods: A total of 225 Chinese Han nationality patients undergoing thoracoscopic-assisted radical resection of lung cancer were enrolled in the present study. Among them, 132 were males (58.67%) and 93 (41.33%) were females having American Society of Anesthesiologists statuses classified as grades I or II. The rs1799971, rs563649 and rs1323040 genotypes of the OPRM1 gene and rs2032582, rs1045642 and rs1128503 genotypes of the ABCB1 gene were detected by Sanger sequencing. The state anxiety index and pressure pain threshold were assessed preoperatively. Sufentanil was administered intravenously to maintain anesthesia. The doses and side effects of sufentanil consumed 6 h (T1), 24 h (T2) and 48 h (T3) after surgery were recorded.Results: The sufentanil doses at T1, T2 and T3 were significantly higher in radical-operation lung cancer patients with mutant homozygous rs1799971 and rs1323040 loci in the OPRM1 gene and rs2032582 and rs1128503 loci in the ABCB1 gene. The doses of sufentanil consumed by mutant heterozygous lung cancer patients at T1, T2 and T3 were significantly higher than those consumed by patients without mutations, and the differences were statistically significant (P&lt;0.05). There was no significant difference in sufentanil doses consumed by lung cancer patients with mutant homozygous, mutant heterozygous and wild-type rs563649 locus of the OPRM1 gene and rs1045642 locus of the ABCB1 gene at T1, T2 and T3 (P&gt;0.05). There was no significant difference in the visual analog scale scores at T1, T2 and T3 for different genotypes of OPRM1 and ABCB1 genes in lung cancer patients (P&gt;0.05). No significant difference was found between the adverse reactions of OPRM1 and ABCB1 genotypes in patients undergoing radical resection of lung cancer (P&gt;0.05).Conclusion: The rs1799971 and rs1323040 polymorphisms of the OPRM1 gene and rs2032582 and rs1128503 polymorphisms of the ABCB1 gene are related to the analgesic effect and consumed dose of sufentanil in Chinese Han patients undergoing radical operation of lung cancer.
