In the present study, pure line seeds of mungbean (Vigna radiata L. Wilczek) were treated with four doses (0.2%, 0.4%, 0.6% and 0.8% as w/v) of ethyl methane sulphonate (EMS) to analyse the genetic variability and heritability for some prime agro-metrical traits in second mutant (M2) generation. The analysis of variance revealed highly significant differences among the studied characters. The phenotypic coefficient of variation (PCV) was higher than its genotypic counterpart (GCV) for all the studied traits. The highest GCV (0.537) for number of seeds per pods and highest PCV (0.635) for plant height were recorded in 0.4% EMS treatment. The lowest GCV (0.179) and PCV (0.214) were recorded in the control set for number of pods per plant. The highest broad sense heritability estimate (92.33%) was recorded on 0.4% EMS for plant height. The expected genetic advance was high (42.39%) in 0.4% dose for days to flowering. The lowest heritability (38.43) and genetic advance (5.37) were noticed in control for the total seed yield per plant. EMS at 0.4% and 0.6% concentration gave the maximum values of all the genetic parameters. The increased genetic variability in treated population for these traits has a high scope for selection and can be exploited for the further improvement of mungbean.

Induced variability was studied in the quantitative characters of mung bean (Phaseolus aureus Roxb.) after treatment with gamma rays, ethyl methane sulphonate (EMS), and hydrazine hydrate (HZ) in M1, M2, and M3 generations. The characters evaluated were seeds per pod, 100-seed weight, and total plant yield. The variability increased in almost all of the characters in the M1 generation. The mean number of seeds per pod and 100-seed weight decreased, whereas the plant yield did not show any particular trend. There was an increase in the mean values after gamma irradiation and EMS treatments in the M2 generation. However, HZ fails to show the same response. However, the mean values increased in the M3 generation. Estimates of heritability were higher for 100-seed weight, followed by plant yield, and then seeds per pod in the M3 generation. Coefficient of genotypic variation and genetic advance were high for the plant yield in M2 and M3 generations. In general, the genotypic coefficient of variation values, heritability, and genetic advance increased more in M3 as compared with M2, indicating that the significant gain could possibly be achieved through selection in M3 generation.Key words: quantitative, heritability, mutagenesis.

Soybean (Glycine max (L.) Merr.) var. Co-1 seeds were treated with physical mutagen namely such as gamma rays and chemical mutagen namely such as Ethyl Methane Sulphonate (EMS) and Diethyl Sulphate (DES). Various concentrations of gamma rays 10, 20, 30, 40, 50 and 60 KR, EMS (0.1, 0.2, 0.3, 0.4, 0.5 and 0.6 %) and DES (0.1, 0.2, 0.3, 0.4, 0.5 and 0.6 %). The morphological parameters such as days to first flower, plant height, number of cluster per plant, number of pod per plan, seed yield per plant, number of branches per plant, number of leaves per plant, protein content and oil content increasing with increasing level of some doses gamma rays, Ethyl methane sulphonate and Diethyl sulphate treatment in M2 and M3 generation were carried out. A number of mutants were recorded in M2 and M3 generations for one or more traits viz., plant height, maturity, branching, pigmentation, fruit size and yield were isolated in M2 and M3 generation. Mean for various characters increased at mutagenic treatment than control. The yield parameters like plant height, number of cluster per plant, number of seeds per plant and seed yield per plant were recorded the moderated and high mean value in the 50 KR of gamma rays 0.5% of EMS and 0.4% of DES treated population with compared to control plants.

Abstrak Penelitian ini bertujuan mengamati pengaruh lama perendaman biji dengan 1% EMS terhadap persentase munculnya bibit dan karakter pertumbuhan tanaman cabai rawit pada umur 4 minggu setelah tanam (MST). Benih cabai rawit direndam dalam air selama 6 jam, selanjutnya direndam 1% EMS dalam buffer fosfat pH 7, selama 6 jam, 9 jam dan 12 jam. Tiap biji disemai dalam bumbungan kertas. Munculnya bibit diamati setiap hari. Setelah berumur 3 minggu, bibit dipindahkan ke bedengan. Hasil menunjukkan perlakuan 1% EMS memperlambat munculnya bibit. Pada 10 hari setelah semai (HSS) munculnya bibit pada perlakuan dapat mencapai 100%. Perlakuan 1% EMS berpengaruh nyata terhadap tinggi tanaman dan jumlah daun, sedangkan panjang, dan lebar daun dengan perendaman 6 jam tidak berbeda dengan kontrol, namun berbeda dengan perendaman 9 jam dan 12 jam. Kata kunci : EMS, bibit, pertumbuhan, Capsicum frustescens L.   Abstract This research aimed to evaluate the influence of different exposure durations of 1% EMS on seedling emergence and growth characters of C, fustescent at 4 week after planting (WAT). Seeds were soaked in water for 6 hours, then soaked in 1% EMS in phosphate buffer pH 7, for 6 hours, 9 hours and 12 hours. Each seed was then sowed in a single paper tube. Seedling emergence was observed every day. At 3 weeks after sowing, seedlings were transferred to field. Results showed that soaking seeds in 1% EMS inhibited seedling emergence. At 10 days after sowing, the percentages of seedling emergence at control and treated seed were 100%. Treatments of 1% EMS have a significant effect on plant height, and the number of leaf, while length, and width of leaf with 6 hour exposure were not different with control, but differ from exposure of 9 hours and 12 hours. <p>Estimation of Genetic Variability and Correlation Among Early Maturity Mungbean Yield Components. Ratri T. Hapsari. Early maturity mungbean [Vigna radiata (L.) Wilczek] is very important to avoid drought stress, pest and disease attack as well as increase the index planting. The aims of this research was to estimate genetic variability and correlation. The genetic study included heritability, coefficient of genetic variability, genetic advance and correlation among yield components so that it can be used as selection criteria for early maturity mungbean. A total of 145 accessions of mungbean were tested at Muneng farm station in March-June 2010 using a randomized block design, with two replicates. Each accession was planted at 0.8 m x 4 m with spacing 40 cm x 10 cm, with two plants/hole. Fertilization was done by adding 50 kg urea, 75 kg SP36, and 75 kg KCl/ha, at the time of planting. The results showed that mungbean accesions had significant differences in all characters tested. The genetic variance value of all characters was broad with high broadsense heritability estimates, except for number of pods/ cluster and seed number/pod. Genetic advance of all characters were high, except for seed number/pod. The phenotypic correlation between 1000 seeds weight and pod length with seed yield were positive significant while plant height, flowering days, days to maturity, and number of pods per plant had negative significant correlation with its yield. Therefore, plant height, days to maturity, pod lenght, 1000 seeds weight and seed yield could be used as selection criteria based on estimating value of genetic variability, correlation with yield and economic value. There were five genotype which have index value above 20, i.e MLGV 0353, MLGV 0362, MLGV 0354, MLGV 0358, and MLGV 0351.</p><p> </p><p><strong>Abstrak</strong></p><p>Kacang hijau [Vigna radiata (L.) Wilczek] berumur genjah berperan penting untuk menghindari cekaman kekeringan, serangan hama penyakit, dan meningkatkan indeks pertanaman. Penelitian ini bertujuan untuk mengetahui nilai duga parameter genetik dan korelasi antar komponen hasil sehingga dapat digunakan sebagai kriteria seleksi kacang hijau berumur genjah. Sebanyak 145 genotipe kacang hijau diuji di KP Muneng pada bulan Maret sampai dengan Juni 2010 menggunakan Rancangan Acak Kelompok dengan dua ulangan. Setiap aksesi ditanam pada plot 0,8 m x 4 m dengan jarak tanam 40 cm x 10 cm, dua tanaman/lubang. Pemupukan dilakukan dengan 50 kg urea, 75 kg SP36, dan 75 kg KCl per hektar pada saat tanam. Parameter yang diamati adalah tinggi tanaman, umur 50% berbunga, umur 80% masak, jumlah polong/tangkai, jumlah polong/tanaman, panjang polong, jumlah biji/polong, bobot 1.000 biji, dan bobot biji/plot. Hasil penelitian menunjukkan bahwa genotipe yang diuji memiliki keragaman semua sifat yang diamati. Keragaman genetik dan fenotipik tergolong luas. Heritabilitas arti luas tergolong tinggi, kecuali jumlah polong/tangkai dan jumlah biji/polong tergolong sedang. Kemajuan genetik seluruh karakter tinggi, kecuali jumlah biji/polong. Korelasi antara bobot 1.000 biji dan panjang polong bernilai positif nyata dengan bobot biji/plot, sedangkan tinggi tanaman, umur berbunga, umur masak, dan jumlah polong per tanaman berkorelasi negatif nyata. Berdasarkan nilai duga parameter genetik, korelasi antarhasil, dan nilai ekonomisnya, maka tinggi tanaman, umur masak, panjang polong, bobot 1.000 biji dan bobot biji per plot dapat dijadikan kriteria seleksi indeks. Terdapat lima genotipe memiliki nilai indeks lebih dari 20, yaitu MLGV 0353, MLGV 0362, MLGV 0354, MLGV 0358, dan MLGV 0351.</p>

Background: Induction of mutation plays an important role in the breeding programme among modern methods of plant breeding. Genetic variability is prerequisite for crop improvement and through induced mutation it was found to be very effective for creating variability in the quantitative and qualitative characters. Hence, the present study was aimed to induce genetic variability within short time. In the present investigation positive as well as negative impact on quantitative traits were recorded. Methods: The experiment material comprised of seeds of horsegram [Macrotyloma uniflorum (Lam.) Verdc] variety Paiyur 2 which was subjected to ethyl methane sulphonate (02, 0.3, 0.4 and 0.5%), gamma radiation (100, 200, 300 and 400 Gy) and combination treatments. The mutations affecting gross morphological changes in growth and yield characters such as plant habit, flowering, pod morphology, maturity and seed yield were scored as quantitative characters. The micro mutations at population level can be easily detected in the form of increased variations for quantitative traits in the segregation of mutagen treated populations. Micro mutations can alter morpho-physiological characters hence they are of a particular interest to the plant breeders. ANOVA test was performed to determine the significant differences and duncan’s multiple range test (p=0.05) to compare the differences among treatment means. Result: Among the twenty-five treatments studied both the mutagens, gamma radiations and Ethyl Methane Sulphonate proved to be very effective to induce variability in quantitative traits in M2 and M3 generations. The results of Duncan’s multiple range test analysis revealed that the treatment of mutagens in combinations induced more positive effects as compared to that of using alone. The combined treatments were found positive for improvement of primary branches per plant, no. of pods per plant, pod length, no. of seeds/pod and seed yield per plant in M2 and M3 generations whereas the treatment of gamma irradiation imparted its significant role for improvement of plant height, days required for 50% flowering, days to maturity and 1000 seed weight in both M2 and M3 generations whereas the treatment with Ethyl Methane Sulphonate is useful for improving no. of pods per plant and no. of seeds/pod in M2 generation alone.

Seeds of two pigeonpea varieties namely BDN 708 and BSMR 853 were treated with Gamma rays, ethyl methane sulphonate (EMS) and Sodium azide (SA). The treated as well as control plant populations were screened to study the induced variability for quantitative characters. Positive shift in mean values for number of primary branches per plant was observed except 0.10% EMS in variety BSMR 853 and BDN 708 in M2 generation. The effect of all the mutagenic treatments on pods per plant revealed statistically significant negative as well as positive shifts in mean values in BSMR 853 in M2 and M3 generations.
