Bacterial Macrofibers and Bionites: Materials of Natural and Synthetic Design

ABSTRACTTwo fiber forms can be obtained from cells of the rod-shaped bacterium,Bacillus subtilis, one called macrofibers, the other bacterial thread. Macrofibers are highly organized, multicellular structures, millimeters in length that selfassemble in a unique way. Each fiber is essentially a single chain of cells linked end-to-end that has repeatedly folded upon itself and twisted into helical form. The growth of individual cells yields both the material of the macrofiber and the forces required for its assembly. The forces involved stem from twisting motions caused by cell growth geometry. The folding process is akin to negative supercoiling. New approaches have been used to estimate the magnitude of forces. Torque generated by single filaments has been estimated from snapopening motions resulting from aborted attempts at folding to be in the range of 10−10to10−8dyne-cm. In contrast, multifilament fibers carrying small wires in their loops must have generated a torque of at least 10−5dyne-cm and a supercoiling force of at least 10−5dyne in order to have moved the wires in viscous solutions at the rates observed. The second bacterial fiber form, bacterial thread, and its mineralized derivatives, called bionites, are man-made materials. They are produced by the drawing and drying of bacterial cell filaments from cultures grown in the form of a textile-like web. The material properties of bacterial thread reflect primarily those of the strength-bearing cell wall polymer, peptidoglycan. A variety of new fiber-like materials have been produced by mineralizing the cell walls in situ in web cultures and drawing the products. Iron, copper, calcium, and potassium phosphate-containing bionites have been obtained in this manner. We are currently searching for order in the bionite crystal forms that may reflect the electrostatic nature of the wall polymer structural templates.

ABSTRACT Previous work, using solubilization of yeast cell walls by carboxymethylation, before or after digestion with β(1-3)- or β(1-6)glucanase, followed by size chromatography, showed that the transglycosylases Crh1p and Crh2p/Utr2p were redundantly required for the attachment of chitin to β(1-6)glucan. With this technique, crh1Δ crh2Δ mutants still appeared to contain a substantial percentage of chitin linked to β(1-3)glucan. Two novel procedures have now been developed for the analysis of polysaccharide cross-links in the cell wall. One is based on the affinity of curdlan, a β(1-3)glucan, for β(1-3)glucan chains in carboxymethylated cell walls. The other consists of in situ deacetylation of cell wall chitin, generating chitosan, which can be extracted with acetic acid, either directly (free chitosan) or after digestion with different glucanases (bound chitosan). Both methodologies indicated that all of the chitin in crh1Δ crh2Δ strains is free. Reexamination of the previously used procedure revealed that the β(1-3)glucanase preparation used (zymolyase) is contaminated with a small amount of endochitinase, which caused erroneous results with the double mutant. After removing the chitinase from the zymolyase, all three procedures gave coincident results. Therefore, Crh1p and Crh2p catalyze the transfer of chitin to both β(1-3)- and β(1-6)glucan, and the biosynthetic mechanism for all chitin cross-links in the cell wall has been established.

In a recent study of the superplastic forming (SPF) behavior of certain Al-Li-X alloys, the relative misorientation between adjacent (sub)grains proved to be an important parameter. It is well established that the most accurate way to determine misorientation across boundaries is by Kikuchi line analysis. However, the SPF study required the characterization of a large number of (sub)grains in each sample to be statistically meaningful, a very time-consuming task even for comparatively rapid Kikuchi analytical techniques.In order to circumvent this problem, an alternate, even more rapid in-situ Kikuchi technique was devised, eliminating the need for the developing of negatives and any subsequent measurements on photographic plates. All that is required is a double tilt low backlash goniometer capable of tilting ± 45° in one axis and ± 30° in the other axis. The procedure is as follows. While viewing the microscope screen, one merely tilts the specimen until a standard recognizable reference Kikuchi pattern is centered, making sure, at the same time, that the focused electron beam remains on the (sub)grain in question.

SummaryUrokinase (UK) could be purified to apparent homogeneity starting from crude urine by sequential adsorption and elution of the enzyme to gelatine-Sepharose and agmatine-Sepharose followed by gel filtration on Sephadex G-150. The purified product exhibited characteristics of the high molecular weight urokinase (HMW-UK) but did contain two distinct entities, one of which exhibited a two chain structure as reported for the HMW-UK while the other one exhibited an apparent single chain structure. The purification described is rapid and simple and results in an enzyme with probably no major alterations. Yields are high enough to obtain purified enzymes for characterization of UK from individual donors.

Stress is the crucial factor of ferromagnetic material failure origin. However, the nondestructive test methods to analyze the ferromagnetic material properties’ inhomogeneity on the microscopic scale with stress have not been obtained so far. In this study, magnetic Barkhausen noise (MBN) signals on different silicon steel sheet locations under in situ tensile tests were detected by a high-spatial-resolution magnetic probe. The domain-wall (DW) motion, grain, and grain boundary were detected using a magneto-optical Kerr (MOKE) image. The time characteristic of DW motion and MBN signals on different locations was varied during elastic deformation. Therefore, a time-response histogram is proposed in this work to show different DW motions inside the grain and around the grain boundary under low tensile stress. In order to separate the variation of magnetic properties affected by the grain and grain boundary under low tensile stress corresponding to MBN excitation, time-division was carried out to extract the root-mean-square (RMS), mean, and peak in the optimized time interval. The time-response histogram of MBN evaluated the silicon steel sheet’s inhomogeneous material properties, and provided a theoretical and experimental reference for ferromagnetic material properties under stress.

We report two infants with the acquired immunodeficiency syndrome (AIDS) and rectal bleeding due to cytomegalovirus (CMV) ileitis and colitis with minimal focal mucosal ulceration but with extensive leiomyolysis of the muscularis propria. Immunostaining and in situ hybridization for CMV showed numerous viral inclusions in the myocytes of the muscularis propria and vascular endothelium/smooth muscle with only occasional inclusions present in the muscularis mucosae. Colectomy was curative in one patient; in the other the bowel was only examined at postmortem.

When one hears the term “ground cover,” one immediately thinks of “grasses.” This perception is so deep-seated that paleobotanists even have been overheard to proclaim that “there was no ground cover before grasses.” Today grasses are so predominant in many environments that this perception is perpetuated easily. On the other hand, it is difficult to imagine the absence or lack of ground cover prior to the mid-Tertiary. We tested the hypothesis that different forms of ground cover existed in the past against examples from the Recent and the fossil record (Table 1). The Recent data were obtained from a large number of sources including those in the ecological, horticultural, and microbiological literature. Other data were derived from our knowledge of Precambrian life, sedimentology and paleosols, and the plant fossil record, especially in situ floras and fossil “monocultures.” Some of the data are original observations, but many others are from the literature. A detailed account of these results will be presented elsewhere (Pfefferkorn and LePage, in preparation).

SUMMARYFifty-six Merino d'Aries and 56 Romanov × Merino crossbred lactating ewes were divided into two treatment groups, one (T1) with a ewe-to-ram ratio of 48: 1 (24 Merino and 24 crossbred) and the other (T2) of 64: 1 (32 Merino and 32 crossbred).Intravaginal sponges with 40 mg FGA (Searle) were inserted in two subgroups at 8-day intervals and withdrawn after 12 and 16 days in situ, in four equal groups at 4-day intervals. A dose of 400 IU pregnant mare's serum gonadotrophin (PMSG) was applied at sponge withdrawal.Fertility after two oestrous cycles was equally satisfactory in both treatments, though it was lower in treatment group T2. This could be due to the significantly low (P<0·01) number of Merino ewes raddled by the T2 ram (0·33 Merino against 0·92 crossbred) suggesting preferential behaviour by the ram in this treatment.Comparisons between breeds show a slight superiority in the fertility of crossbred ewes and a highly significant superiority in prolificacy These results suggest that using the described method, 2% of rams seems more than adequate to mate with progestagen-synchronized ewes with a satisfactory fertility level. It is, however, recommended to use homogeneous groups of ewes.

In situ high pressure energy dispersive X-ray diffraction measurements on layered perovskite-like manganate Ca 3 Mn 2 O 7 under pressures up to 35 GPa have been performed by using diamond anvil cell with synchrotron radiation. The results show that the structure of layered perovskite-like manganate Ca 3 Mn 2 O 7 is unstable under pressure due to the easy compression of NaCl-type blocks. The structure of Ca 3 Mn 2 O 7 underwent two phase transitions under pressures in the range of 0~35 GPa. One was at about 1.3 GPa with the crystal structure changing from tetragonal to orthorhombic. The other was at about 9.5 GPa with the crystal structure changing from orthorhombic back to another tetragonal.
