ABSTRACTMonoclonal antibodies against meso-tetrakis(4-carboxyphenyl porphine)(TCPP) were prepared by immunizing Balb/c mice with TCPP bound keyhole limpet hemocyanin covalently and fusing the spleen cells with myeloma cells by using poly(ethylene glycol). The antibodies bind TCPP strongly with the dissociation constants of 10−6−2.5 × 10−7 M. One of the antibodies causes a large shift of the Soret band of TCPP to a longer wavelength and large induced Cotton effects(ICD) on TCPP. Quantitative analyses of the binding of TCPP to the antibody by ICD show that not only one-to-one binding but two-to one(binding site: TCPP) binding occurs in excess of the antibody over TCPP.

In a cytosolic extract from rat liver, the number and the concentration of ADP-binding sites as well as their dissociation constants were determined by using the rate-of-dialysis technique. Interfering cytosolic adenylate kinase was extracted from the cytosol by affinity chromatography on Ap5A-agarose, and remaining traces of enzyme activity were inhibited with (+)-catechin. Binding of ADP to cytosolic proteins was increased by poly(ethylene glycol) and decreased by EDTA. The effect of 0.1 mM-EDTA could be reversed by addition of equimolar concentrations of Mn2+ or Mg2+. In presence of 5% poly(ethylene glycol), added to increase local protein concentration, two binding sites for ADP were observed, with KD values of 1.9 microM (site I) and 10.8 microM (site II). The concentration of these binding sites, when extrapolated to cellular protein concentrations, were 30 microM (site I) and 114 microM (site II). It is concluded that a minimum of about 50% of total cytosolic ADP is bound to proteins, and that the ratio of free ATP/free ADP is at least twice that of total ATP/total ADP.

The B cell hybridomas producing monoclonal antibodies (E10, D7, F4, H6, and D4) were established by the fusion of P3U1 or NS-1 murine myeloma cell lines and spleen cells of B10.A(5R) mice hyperimmunized with mitomycin C-treated B10.A(3R) spleen and thymus cells. Two types of monoclonal antibodies specific for the products controlled by a gene in the I-Jb subregion of the H-2 complex were characterized: one specific for the private type of I-Jb determinant, the other recognizing the cross-reactive determinant between the I-Jb and I-Jd products. By using these monoclonal reagents, the I-J-encoded product on the antigen-specific suppressor T cells was found to be expressed on their soluble suppressor factors. Furthermore, the I-Jb products were successfully detected not only on the T cell hybridoma with suppressor activity specific for keyhole limpet hemocyanin (KLH), but also on KLH-primed suppressor T cells enriched by antigen-coated petri dishes and concanavalin A-induced thymocyte blasts of C57BL/6 mice by complement-dependent cytotoxic assays and membrane fluorescence techniques.

SBA-15 nanoporous silica was prepared by cooperation assembly of tetraethylorthosilicate  precursor in the presence of poly (ethylene glycol)–block- poly (propylene glycol)–block- poly (ethylene glycol) copolymer surfactant and functionalized with (3-aminopropyl) triethoxysilane (APTES) via sol-gel reaction to obtain APTES-SBA-15. Tetra-(p-chlorophenyl) porphyrin (TClPP) was synthesized using modified Alder- Longo method. Zinc was inserted into the TClPP using zinc acetate as a metal source The existance of zinc in porphyrin was confirmed by the appearance of Soret band and Q band in UV- vis spectra. ZnTClPP was also characterized using FTIR and NMR spectroscopy SBA-15 was modified with APTES to immobilize the metalloporphyrin on SBA-15 surface. The product (NH-SBA-15 ZnP) can be used as a heterogenous catalyst in various reactions. The formation of this material was confirmed by FTIR spectroscopy, XRD and BET adsorption.
