Alcap Delivery Devices and the Sustained Release of Difluoronethylornithine (DFNO) in Vitro and in Vivo Using Male Rats

AbstractSprague-Dawley albino male rats (25) were divided into five groups consisting of five rats each. Polymer (polylactic acid) impregnated ALCAP capsules filled with 40 mg DFMO were implanted subcutaneously (SC) or intraperitoneally (IP) in Group I and II rats respectively. Rats in Group III were implanted with empty polymer impregnated ALCAP capsules (ALCAP control). Group IV rats were administered orally 3% DFMO in drinking water. Rats in Group V served as controls. Blood samples were collected every week for nine weeks via the tail artery. The concentration of DFNO in the plasma was determined. Data obtained showed that the levels of DFMO in the serum of rats in groups I, I, and IV were 64.71 ±4.08. 219.18 ± 14.48, 16.71 ± 5.21 ug ml−1, respectively at the end of nine weeks. Body weights of the controls and DFMO treated rats were not significantly different (p<0.05). The diarrhea often noted in rats treated orally with DFHO was not observed in rats implanted with ALCAP or ALCAP capsules filled with DFMO. The results of this study suggest that: (1) polymer impregnated ALCAP ceramic implants can be used to deliver DFMO in vivo in a sustained manner for long durations of time, and (2) a ceramic system can be designed to deliver DFNO and drugs such as DFMO in a sustained manner over long durations of time in humans.

Pyrethroid cyhalothrin (PC) is an insecticide that is used worldwide for pest control in agriculture and household use. Samoa extract (SE) is a potent antioxidant protecting cells from oxidative stress. The present study investigates the protective and therapeutic effect of SE on PC-induced changes in sperm quality in male rats. Fifty adult male albino rats were divided into five groups: group I: served as control; group II: received PC i.p. only (6.2 mg/kg b.wt.); group III: received SE only (100 mg/kg b.wt., p.o.) for eight weeks; group IV: received SE as a protective agent daily for eight weeks, then followed by the administration of PC (i.p.) three times a week for two weeks; group V: exposed to PC (i.p.) three times a week for two weeks, then treated with the SE daily for 8 weeks. Results showed that PC caused markedly impaired sperm quality (a count, viability, motility, and abnormality). Compared to PC-treated animals, SE in the protective group markedly restored the alteration of sperm indices. However, SE in the curative group was found to be less effective in restoring PC-induced alterations. In conclusion, the data of this study revealed that the SE as a protective agent is more effective than as a therapeutic agent. Keywords: Samoa; Pyrethroid; Sperm quality; Rat

ABSTRACT Aim The purpose of this study was to evaluate the fracture strength of roots instrumented with the self-adjusting file (SAF; ReDent-Nova, Ra'anana, Israel) and the Reciproc reciprocating file and that were and were not obturated using the warm vertical lateral compaction technique. Materials and methods In total, 75 mandibular premolar teeth were sectioned at or below the cementoenamel junction to obtain roots 13 mm in length. The roots were balanced with respect to buccolingual and mesiodistal diameters and weight. They were distributed into four experimental groups and one control group (n = 15): No instrumentation (group I), instrumentation with SAF files but no obturation (group II), instrumentation with SAF files and obturated with warm vertical lateral compaction (group III), instrumentation with Reciproc File but no obturation (group IV), and instrumentation with Reciproc File and obturated with warm vertical lateral compaction (group V). AH Plus sealer (Dentsply DeTrey, Konstanz, Germany) was used along with gutta-percha points. One week later, a vertical load was applied to the specimen's canal until fracture occurred. Data were statistically analyzed using one-way analysis of variance (p = 0.05). Results The mean fracture load was 312.83 N for group I, 297.35 N for group II, 359.15 N for group III, 231.51 N for group IV, and 275.81 N for group V. Conclusion The fracture resistances exhibited a statistically significant difference between all the groups. Teeth instrumented by SAF exhibited a better fracture resistance. This study evaluated in vitro the capacity of debris removal from the apical third of flattened root canals, using different final irrigation protocols. Thirty human mandibular central incisors with a mesiodistal flattened root were prepared using rotary instrumentation by Endo-Flare 25.12 and Hero 642 30.06, 35.02, 40.02 files, irrigated with 2 mL of 1% NaOCl after each file. The specimens were randomly distributed into 5 groups according to the final irrigation of root canals: Group I: 10 mL of distilled water (control), Group II: 10 mL of 1% NaOCl for 8 min, Group III: 2 mL of 1% NaOCl for 2 min (repeated 4 times), Group IV: 10 mL of 2.5% NaOCl for 8 min, and Group V: 10 mL of 2.5% NaOCl for 2 min (repeated 4 times). The apical thirds of the specimens were subjected to histological processing and 6-μm cross-sections were obtained and stained with hematoxylin-eosin. The specimens were examined under optical microscopy at ×40 magnification and the images were subjected to morphometric analysis using the Scion image-analysis software. The total area of root canal and the area with debris were measured in square millimeters. Analysis of variance showed no statistically significant difference (p>0.05) among the groups GI (2.39 ± 3.59), GII (2.91 ± 2.21), GIII (0.73 ± 1.36), GIV (0.95 ± 0.84) and GV (0.51 ± 0.22). In conclusion, the final irrigation protocols evaluated in this study using the Luer syringe presented similar performance in the removal of debris from the apical third of flattened root canals.

Abstract Cholesterol is needed in certain concentration as to avoid health effects. Efforts to handle hypercholesterolemia can be done by utilizing the content of resistant starch in Uter bananas flour. The aim of this study was to prove the hypocholesterolemic effect of Uter Banana flour in hypercholesterolemic Spraque-Dawlwy rats. Therefore, experimental research using animals to determine the hypocholesterolemic effect of Uter banana flour was carried out for 28 days, in Spraque Dawlwy male rats aged 2 months, weighing ± 200 grams. Rats were divided into 5 treatment groups. Group I was normal rats group, group II was hypercholesterol rats without treatment, group III was hypercholesterol rats with pulp fruit flour diet, group IV was hypercholesterol rats with whole fruit flour diet, group V was hypercholesterol rats with peel fruit flour diet. Each diet contained 100 mg / kgBB. Statistical analysis showed that a diet of pulp fruit flour and whole fruit flour can reduce levels of total cholesterol, triglycerides, and LDL significantly (p<0.05). Digesta characteristics differed among the treatment group and the hypercholesterolemia group without treatment (p <0.05). The whole fruit flour has ability to lower cholesterol better than pulp fruit flour, whereas, peel fruit flour has no hypocholesterolemic effects. Upaya penanganan hiperkolesterolemia dapat dilakukan dengan memanfaatkan kandungan pati resisten dalam pisang Uter. Tujuan dari penelitian ini adalah membuktikan efek hipokolesterolemik tepung Pisang Uter pada tikus Sprague Dawley hiperkolesterol. Untuk itu penelitian eksperimen dengan hewan coba untuk mengetahui efek hipokolesterolemik tepung pisang Uter dilakukan selama 28 hari, pada tikus Spraque Dawlwy jantan umur 2 bulan, dengan berat ± 200 gram. Tikus dibagi menjadi 5 kelompok perlakuan. Kelompok I adalah kelompok tikus normal, kelompok II adalah tikus hiperkolesterol tanpa perlakuan, kelompok III tikus hiperkolesterol dengan diet tepung daging buah, kelompok IV adalah tikus hiperkolesterol dengan diet tepung buah utuh, kelompok V adalah tikus hiperkolesterol dengan diet tepung kulit buah, masing-masing diberikan diet 100 mg/kgBB. Hasil uji statistik menunjukkan bahwa diet tepung daging buah dan tepung buah utuh dapat menurunkan kadar kolesterol total, trigliserida, dan LDL secara nyata (p<0,05). Karakteristik digesta berbeda antara perlakuan dan kelompok tikus hiperkolesterol tanpa perlakuan (p<0,05).Tepung buah utuh mempunyai kemampuan menurunkan kolesterol lebih baik dibandingkan tepung daging buah. Tepung kulit buah tidak mempunyai efek hipokolesterolemik. Kata kunci: Hiperkolesterol, pati resisten, tepung Pisang Uter

Cancer is a disease in which a group of abnormal cells grow uncontrollably by disregarding the normal rules of cell division. Across several cancers, Hepatocellular carcinoma (HCC) is one of the most aggressive cancers in worldwide. It is held responsible for up to 1 million deaths globally per annum. HCC is an inflammation-related cancer, as a chronic inflammatory state is necessary for cancer appearance. In this study, the drug astaxanthin and encapsulated astaxanthin was tested against HCC. Mice were divided into 7 groups; Group I: control, Group II: DEN induced, Group III: DEN + 50 mg/kg astaxanthin, Group IV: DEN + 100 mg/kg astaxanthin, Group V: DEN + 50 mg/kg encapsulated astaxanthin, Group VI: DEN + 100 mg/kg encapsulated astaxanthin, Group VII: DEN + 10 mg/kg sorafenib. Regular diet was given. Body weight, Food intake, water intake was noted. Other biochemical parameters such as ALP, AST, Albumin, proteins and TNF-α was determined. Finally, the liver was removed from each mice of different group by sacrificing them and histopathology was done. In vivo evaluation in mice models showed significant antitumor activities by both encapsulated and non-encapsulated astaxanthin at 100 mg/kg as compared with the control, DEN induced group and positive drug sorafenib. This research suggested that encapsulated astaxanthin can also be used as chemotherapeutic agent for the treatment of Hepatocellular carcinoma (HCC).

The protective effect of magnesium as magnesium sulfate (MS) on sodium-metavanadate- (SMV-) induced lipid peroxidation (LPO) underin vivoandin vitroconditions was studied. The 18-week SMV intoxication (Group II, 0.125 /mL) enhanced spontaneous malondialdehyde (MDA) generation in rat liver, compared with the control (Group I) and MS-supplemented animals (Group III, 0.06 /mL). Coadministration of SMV with MS (Group IV, SMV-MS) caused a return of the MDA level to the control value range. The effect seems to result from the -independent action and its antagonistic interaction with . Thein vitrotreatment of liver supernatants (LS) obtained from all the tested animals groups with selected exogenous concentrations of or exhibited enhanced MDA production, compared with spontaneously formed MDA. It also showed -stimulating effect on LPO (LS I, Group I) and revealed that the changes in the MDA generation in LS IV (Group IV) might have resulted from the synergistic interactions of with and and from the antagonistic interactions of with and . The findings allow a suggestion that adequate Mg intake for a specific period in the conditions of SMV exposure may prevent V-induced LPO in the liver.

AbstractObjectives:To determine the effect of royal jelly (RJ) on the oxidant-antioxidant balance in rats with N-methyl-N-nitrosourea (MNU) induced breast cancer and to compare this with the chemotherapeutic agent paclitaxel.Material and methods:Fifty-six female Sprague Dawley rats were divided into five groups. Except control group (n=8, Group I) others received MNU (50 mg/kg, a single dose, i.p.) to develop breast cancer: Group II (n=8) untreated, Group III (n=7) treated with paclitaxel (15 mg/kg/week, 3 times, i.p.), Group IV (n=7) with RJ (by oral gavage, 100 mg/kg/day, for 30 days), and Group V (n=7), with paclitaxel+RJ. At the end of 30 days, histopathological and biochemical parameters were evaluated in breast tissues.Results:Levels of protein carbonyl (PC) and 8-hydroxy-deoxyguanosine (8-OHdG) were higher in Group V than in Group II while malondialdehyde (MDA) levels were lower in groups IV and V compared to Group II (p<0.05). Levels of catalase (CAT) in Group V and glutathione (GSH) in Group III were higher than Group II (p<0.05). Glutathione peroxidase (GPx) and superoxide dismutase (SOD) levels did not significantly different. Decreasing effect of RJ on CA15-3 levels was relevant to histopathological results.Conclusion:Although RJ (with or without paclitaxel) had increasing effect of antioxidant status it was insufficient to reduce oxidative stress in breast cancer.

Edible bird's nest (Aedrodramus fuchipagus) contains EGF (Epidermal Growth Factor) and sialic acid which is useful in wound healing. The aim of this study was to investigate the effect of edible bird's nest on the injured Alloxan-induced rats to form optimal ointment formulation of edible bird's nest (Aedrodramus fuchipagus) which can heal wounds. The method used in this research is experimental using 25 Sprague Dawley rats divided into 5 groups in which each group consists of 5 rats; Group I was the control group (Vaseline + Betadine®), Group II, III, and IV were the group of EBN + Betadine® with concentrations of 10%, 20%, and 30%; and Group V was of the Sanoskin Meladerm® (SM) + Betadine® group. The rats were modeled for diabetes by inducing Alloxan (150mg/kgBW/i.p) and exposed with DM when the blood sugar level was > 200 mg/dL. The back skins were sliced as long as 2 cm and 2 mm depth and were given treatment according to the group, 1 x daily for 10 days. Parameters that were noticed were length wound, length of the dry wound, and the number of days of scab formation. The results obtained in the control group were 10%, 20%, and 30% on the EBN group, and the SM group on the 9th day were 1.3±0.2449, 0.34±0.427, 0.56±0.4586, 0±0, 0±0. The results of the average length of the dry wound in the control group were 10%, 20%, and 30% on the EBN group, and the SM group were 1±0.5, 2±1.5, 4±0, 8, 1±0.8, and 2±0.63. While the result of the average day of scab formation on the control group was 10%, 20%, and 30% on the EBN groups, SM Groups were 1.3±0.47, 2.2±1.6, 3.8±0.8, 1±0, and 2±0.63. Based on Kruskal Wallis test, there were significant differences in the wound length among treatment groups (sig 0.013) and significant differences in dry wounds among treatment groups (sig 0.046), but there was no difference in the time of scab formation among treatment groups (sig 0.066). In conclusion, edible bird’s nest (Aedrodramus fuchipagus) ointment concentration of 30% is the most optimal ointment to wound healing in Alloxan-induced male rats when compared to Sanoskin Melladerm.

It is anticipated that Japanese quail /Coturnix coturnix japonica/ will provide animal proteins in long term space flights. Consequently this species of birds is of research interest of international space program INTERCOSMOS. In the year 1987 we reported on an experiment /2/ in which the effect of chronic acceleration of 2 G hypergravitation, the hypodynamy and the simultaneous effect of chronic acceleration and the location in the centre of the turntable of the centrifuge on the protein fractions in skeletal muscles was studied. The ultrastructure of the heart muscle was now in this experiments examined as well.Japanese quail cockerels, aged 48 days were exposed to 2 G hypergravitation /group IV/ in a 6,4 m diameter centrifuge, to hypodynamy /group III/ and their combination /group V/, respectively for 6 days / Fig.1/. The hypodynamy in group III was achieved by suspending the birds in jackets without contact the floor. The group II was located in the centre ofthe turntable of the centrifuge. The control group I. was kept under normal conditions. The quantitative ultrastructure of myocard was evaluated by the methods of Weibel/3/ - this enables to determine the number, relative size and volume of mitochondria volume of single mitochondria, defficiency of mitochondrial cristae and volume of myofibrils.

Stamina-enhancing effects of human adipose derived stem cells (hADSCs) were investigated in young Sprague-Dawley rats. Ten-day-old male rats were transplanted intravenously (IV) or intracerebroventricularly (ICV) with hADSCs (1 × 106 cells/rat), and physical activity was measured by locomotor activity and rota-rod performance at post-natal day (PND) 14, 20, 30, and 40, as well as a forced swimming test at PND 41. hADSCs injection increased the moving time in locomotor activity, the latency in rota-rod performance, and the maximum swimming time. For the improvement of physical activity, ICV transplantation was superior to IV injection. In biochemical analyses, ICV transplantation of hADSCs markedly reduced serum creatine phosphokinase, lactate dehydrogenase, alanine transaminase, and muscular lipid peroxidation, the markers for muscular and hepatic injuries, despite the reduction in muscular glycogen and serum triglycerides as energy sources. Notably, hADSCs secreted brain-derived neurotrophic factor (BDNF) and nerve growth factor in vitro, and increased the level of BDNF in the brain and muscles in vivo. The results indicate that hADSCs enhance physical activity including stamina not only by attenuating tissue injury, but also by strengthening the muscles via production of BDNF.
