Controlled Release of Natural Polyphenols in Oral Cavity Using Starch Pickering Emulsion

ABSTRACTThe goal of this study was to determine the potential use of starch Pickering emulsion as a vehicle to deliver a natural phenolic compound, curcumin in the oral cavity. To this end, an oil-in-water (o/w) emulsion was prepared using starch molecules as the stabilizer/emulsifier. The physical stability, oxygen permeability and release of curcumin from the starch Pickering emulsion in simulated saliva fluid (SSF) were determined. The results of this study showed that the starch stabilized o/w emulsions were stable for up to 2 weeks. The starch Pickering emulsion also provided better protection against oxidation than a surfactant-stabilized emulsion, and the digestion of the starch Pickering emulsion using amylase led to the complete disruption and phase separation of the emulsion.

One of the most complex problems in hair care formulations is the duality of the surfactants used. In this regard, such surfactants must be cationic so as to interact with the negatively charged cuticle surface of hair. However, these interdependencies typically lead to non-ideal values for the required hydrophilic–lipophilic balance (HLB) in the oil phase. This study was designed to evaluate the physicochemical properties of several oil-in-water emulsion prototypes for the potential use in hair conditioners. Here, a base formulation was utilized, incorporating binary mixtures of cationic surfactants in different proportions. The cationic surfactants employed were hydroxyethyl-behenamidopropyl-diammonium chloride, behentrimonium methosulphate, cetrimonium chloride, and (iv) Polyquaterniumpolyquaternium-70. The surfactants were evaluated for their capability to decrease the surface tension in an aqueous solution through contact angle measurements between the oily phase and the aqueous phase. The required HLB of the oil phase was also determined. The emulsification process was developed using standard preparation methods. For three months, the prototypes with high viscosity were packed in containers and stored in a stability chamber at accelerated conditions (40 ± 2 °C and 75 ± 5% RH). During this time, the size, size polydispersity, zeta potential, viscosity, rheological profile, and creaming index were all evaluated monthly. The results showed a slight change in the physical stability of the prototypes, where the droplet size increased moderately, however, did little to destabilize the formulations. This suggests that the mixtures of cationic surfactants used could be useful for technological developments in hair conditioning products.

Appropriate pretreatment of proteins and addition of xanthan gum (XG) has the potential to improve the stability of oil-in-water (O/W) emulsions. However, the factors that regulate the enhancement and the mechanism are still not clear, which restricts the realization of improving the emulsion stability by directional design of its structure. Therefore, the effects of whey protein micro-gel particles (WPMPs) and WPMPs-XG complexes on the stability of O/W emulsion were investigated in this article to provide theoretical support. WPMPs with different structures were prepared by pretreatment (controlled high-speed shear treatment of heat-set WPC gels) at pH 3.5–8.5. The impact of initial WPC structure and XG addition on Turbiscan Indexes, mean droplet size and the peroxide values of O/W emulsions was investigated. The results indicate that WPMPs and XG can respectively inhibit droplet coalescence and gravitational separation to improve the physical stability of WPC-stabilized O/W emulsions. The pretreatment significantly enhanced the oxidative stability of WPC-stabilized O/W emulsions. The addition of XG did not necessarily enhance the oxidative stability of O/W emulsions. Whether the oxidative stability of the O/W emulsion with XG is increased or decreased depends on the interface structure of the protein-XG complex. This study has significant implications for the development of novel structures containing lipid phases that are susceptible to oxidation.

ABSTRACTNB-003 and NB-003 gel formulations are oil-in-water nanoemulsions designed for use in bacterial infections.In vitrosusceptibility ofPropionibacterium acnesto NB-003 formulations and comparator drugs was evaluated. Both NB-003 formulations were bactericidal against allP. acnesisolates, including those that were erythromycin, clindamycin, and/or tetracycline resistant. In the absence of sebum, the MIC90s/minimum bactericidal concentrations (MBC90s) for NB-003, NB-003 gel, salicylic acid (SA), and benzoyl peroxide (BPO) were 0.5/2.0, 1.0/2.0, 1,000/2,000, and 50/200 μg/ml, respectively. In the presence of 50% sebum, the MIC90s/MBC90s of NB003 and BPOs increased to 128/1,024 and 400/1,600 μg/ml, respectively. The MIC90s/MBC90s of SA were not significantly impacted by the presence of sebum. A reduction in the MBC90s for NB-003 and BPO was observed when 2% SA or 0.5% BPO was integrated into the formulation, resulting in MIC90s/MBC90s of 128/256 μg/ml for NB003 and 214/428 μg/ml for BPO. The addition of EDTA enhanced thein vitroefficacy of 0.5% NB-003 in the presence or absence of 25% sebum. The addition of 5 mM EDTA to each well of the microtiter plate resulted in a >16- and >256-fold decrease in MIC90and MBC90, yielding a more potent MIC90/MBC90of ≤1/<1 μg/ml. The kinetics of bactericidal activity of NB-003 againstP. acneswere compared to those of a commercially available product of BPO. Electron micrographs ofP. acnestreated with NB-003 showed complete disruption of bacteria. Assessment of spontaneous resistance ofP. acnesrevealed no stably resistant mutant strains.

This study investigates the antioxidant behaviors of a hazelnut tetrapeptide, FSEY (Phe-Ser-Glu-Tyr), in an oil-in-water emulsion. The emulsion was prepared with stripped hazelnut oil at a ratio of 10%. O/W emulsions, both with and without antioxidants (FSEY and TBHQ), were incubated at 37 °C. The chemical stabilities, including those of free radicals and primary and secondary oxidation productions, along with the physical stabilities, which include particle size, zeta-potential, color, pH, and ΔBS, were analyzed. Consequently, FSEY displayed excellent antioxidant behaviors in the test system by scavenging free lipid radicals. Both primary and secondary oxidation products were significantly lower in the FSEY groups. Furthermore, FSEY assisted in stabilizing the physical structure of the emulsion. This antioxidant could inhibit the increase in particle size, prevent the formation of creaming, and stabilize the original color and pH of the emulsion. Consequently, FSEY may be an effective antioxidant additive to use in emulsion systems.

In this work, nanoemulsion-based delivery system was developed by encapsulation of fennel essential oil. A response surface methodology was used to study the influence of the processing conditions in order to obtain monomodal nanoemulsions of fennel essential oil using the microchannel homogenization technique. Results showed that it was possible to obtain nanoemulsions with very narrow monomodal distributions that were homogeneous over the whole observation period (three months) when the appropriate mechanical energy was supplied by microfluidization at 14 MPa and 12 passes. Once the optimal processing condition was established, nanoemulsions were formulated with advanced performance xanthan gum, which was used as both viscosity modifier and emulsion stabilizer. As a result, more desirable results with enhanced physical stability and rheological properties were obtained. From the study of mechanical spectra as a function of aging time, the stability of the nanoemulsions weak gels was confirmed. The mechanical spectra as a function of hydrocolloid concentration revealed that the rheological properties are marked by the biopolymer network and could be modulated depending on the amount of added gum. Therefore, this research supports the role of advanced performance xanthan gum as a stabilizer of microfluidized fennel oil-in-water nanoemulsions. In addition, the results of this research could be useful to design and formulate functional oil-in-water nanoemulsions with potential application in the food industry for the delivery of nutraceuticals and antimicrobials.

A pyromellitic dianhydride (PMDA) and 4,4′-oxydianiline (ODA)-based oligoimide (PMDA-ODA) was synthesized by a one-step procedure using water as a solvent. The PMDA-ODA particles showed excellent partial wetting properties and were stably dispersed in both water and oil phases. A stable dispersion was not obtained with comparison PMDA-ODA particles that were synthesized by a conventional two-step method using an organic solvent. Both oil-in-water and water-in-oil Pickering emulsions were prepared using the oligoimide particles synthesized in water, and the size of the emulsion droplet was controlled based on the oligoimide particle concentration. The oligoimide particles were tested to prepare Pickering emulsions using various kinds of oils. The oil-in-water Pickering emulsions were successfully applied to prepare microcapsules of the emulsion droplets. Our new Pickering emulsion stabilizer has the advantages of easy synthesis, no need for surface modification, and the capability of stabilizing both oil-in-water and water-in-oil emulsions.

Doxycycline was loaded with synthesized micelles composed of methyl Poly (ethylene glycol-block-poly (L-alanine–co–L-aspartate), or mPEG–Ala–Asp, and then characterized as a drug delivery carrier. The synthesis of the temperature-sensitive mPEG–Ala–Asp block copolymer was carried out by two-step ring-opening polymerization: firstly, the mPEG reacts with L-alanine N-carboxylic anhydride, and secondly the resulting mPEG–Ala reacts with benzyl aspartate N-carboxylic anhydride. The molecular structure of the copolymers obtained was determined by FT-IR and NMR spectroscopy methods and the micelles were characterized by SEM, TEM and DLS, respectively. The controlled release of Dox from hydrogel in the presence of PBS (8 to 9% by weight) lasts 6 to 7 days exhibiting stable release rates. The drug release mechanisms were studied: Higuchi and zero order models. The results and correlation coefficients applied to the Higuchi and zero-order models. The findings show the potential use of mPEG–Ala–Asp as an effective depot matrix to deliver anthracycline class drugs.

Chitosan has been widely used in industrial, food, pharmaceuticals and agriculture. Chitosan is a natural biocompatible polymers means that as nature does not have the side effect, non-toxic, can not be easily digested and broken down by microbes (biodegradable). This study aims to determine whether the chitosan may be formulated into dosage gel hand sanitizer that meets the requirements of the physical stability of the gel.Chitosan is formulated with 3 varying concentrations of Na CMC basis of 3%, 4.5% and 6%. Tests conducted gel formulation is the physical stability test which includes organoleptic test, homogeneity, pH test, test dispersive power, viscosity test and test consistency. Testing is done every week for 4 weeks of storage.Results of testing physical properties of chitosan gel hand sanitizer has the shape and color stable but the resulting aroma change during storage. pH gel meet the requirements, the consistency test of phase separation does not occur, the homogeneity test showed no homogeneous gel, gel dispersive power does not meet the requirements, the viscosity of the gel preparation third formula does not meet the requirements of viscosity gel
