<p>For the correction of reproductive function of cows with ovarian hypofunction practices use a number of hormones. Recently, to stimulate reproductive function using herbal medicines that have gonadotropic effect or stimulate secretion of steroid hormones who try to use to increase fertility. Therefore, we carried out an attempt to develop a method of regulation of reproductive function of the ovaries of cows using combination therapies that can provide effective treatment by studying the biochemical parameters of animals.</p> <p>The cows were divided depending on the treatment to control and two experimental groups of 5 animals in each group. Groups were formed by the following treatment regimens indicated pathology. Cows control group treated by next scheme: day 1 — intramuscular injection drug in vitro at a dose of 10 ml; day 2 —PMSG intramuscular administration of the drug at a dose of 500 IU; day 3 —intramuscular injection drug Surfahon at a dose of 50 mg. Cows from experimental group 1 was injected intramuscularly liposomal drug based on herbal (Rhodiola rosea, Salvia); Animals from second experimental group were injected intramuscularly liposomal drug based on phyto-substances (Rhodiola rosea, Salvia) with gonadotropin-releasing hormone (Surfahon).</p> <p>Analysis of biochemical parameters of blood serum of cows with ovarian hypofunction found low concentrations of estradiol-17-β and progesterone. Between the control and experimental groups concentration of progesterone and estradiol-17-β differ within 10%, which indicates the same level of disease in all animals selected. Level carotene, ascorbic acid and cholesterol in all groups was within the physiological norm and differed slightly.</p> <p>It was established that the treatment of cows with hypofunction ovaries in the experimental group 1 progesterone level 7 days after treatment was 11.5, and 2 - on 41,4% (p &lt;0,01) higher than in the control group animals, indicating that the revitalization of the ovary as a result of treatment and efficacy of liposomal drug on the basis of phyto-substances with gonadotropin-releasing hormone.</p> <p>The level of cholesterol on 7th day after treatment cows with ovarian hypofunction in the first experimental group was higher by 25,5% (p &lt;0,05) and slightly exceeded the physiological value of this indicator in cows. In the second experimental group, it also exceed the physiological norm and exceeded the level in blood of cows in the control group at 18,6% (p &lt;0,05), indicating the possibility of using cholesterol for the synthesis of progesterone.</p> <p>All the cows in the period from 30 to 60 days after the treatment have sign of oestrus and were artificially inseminated.</p>

Iodine deficiency in soil and water in many countries leads to its low content in plants and animal feed produced from them. The limited intake of this element in animals can lead to endemic diseases. The lack of iodine in animal feeding is most often compensated for by the use of additives with an inorganic form of iodine, which can be poorly absorbed by the body. Feed products with an organic form of iodine have a great biological effect. A study on the effect of iodine-containing additives on the physiological and biochemical parameters of the animal organism was carried out on two groups of 15 cows. The experimental group was fed an organic form of iodine as part of the additive, the control group received an inorganic form of iodine. For all animals during the experiment the biochemical parameters of blood, milk productivity and reproductive ability were studied. During the experiment, an increase in metabolic and redox processes in the body was noted in the experimental group. An increase in mineral metabolism, including iodine content, was found. Due to the sufficient intake of organic iodine in the animals of the experimental group, there was an increase in the activity of the main hepatic enzymes AST and ALT. An increase in the process of gluconeogenesis due to an increase in the glucose content in the blood of experimental cows was revealed. An increase in milk production and reproductive function of animals of the experimental group was established. Within 30 days after the termination of feeding the supplement with the organic form of iodine in the experimental group, a prolonging effect was observed, consisting in higher values of blood biochemical parameters, improved reproductive function and milk productivity relative to equivalent animals in the control. As a result, it was concluded that the use of an additive with an organic form of iodine in the amount of 1.5 g in feeding cows for 60 days increases the iodine content in the blood serum of animals and increases the performance of all types of metabolism, which may indicate the best stimulating effect of the organic form of iodine on thyroid activity glands. This in turn helps to improve the productive and reproductive qualities of animals. To recommend the studied drug as a means to replenish iodine deficiency in animals and to more accurately analyze its effect on the hormone-forming function of the thyroid gland, it is necessary to study the level of thyroid hormones in the blood.

The present study aimed to investigate the prophylactic effect of Cymbopogon proximus and Alhagi maurorum on Sulfadimidine induced urolithiasis in rabbits . Thirty New Zealand male rabbits were allocated into six equal groups (each of five): Group (1) was used as a negative control. Group(2) were administered sulfadimidine (200mg/kg) by intramuscular injection.Groups(3) and (4) were administered sulfadimidine(200mg/kg) by intramuscular injection and 330mg/kg of Cymbopogon proximus alcoholic and aqueous extracts respectively orally.Groups(5) and (6) were administered sulfadimidine(200mg/kg) by intramuscular injection and 400mg/kg of Alhagi maurorum alcoholic and aqueous extracts respectively orally. The period of experiment was 10 days. Blood and urine samples were collected from rabbits on the 10th day. The results recorded a significant decrease in serum creatinine, urea, uric acid and crystalluria in Cymbopogon proximus and Alhagi maurorum groups compared to sulfadimidine treated group.We conclude that Cymbopogon proximus and Alhagi maurorum have a nephroprotective and antiurolithiatic effects against sulfadimidine induced crystalluria.

ABSTRACT The gonadotropins luteinizing hormone (LH) and follicle-stimulating hormone (FSH) are produced in the embryonic pituitary in response to delivery of the hypothalamic gonadotropin releasing hormone (GnRH). GnRH has a pivotal role in reestablishing gonadotropin levels at puberty in primates, and for many species with extended reproductive cycles, these are reinitiated in response to central nervous system-induced GnRH release. Thus, a clear role is evident for GnRH in overcoming repression of these genes. Although the mechanisms through which GnRH actively stimulates LH and FSH β-subunit (FSHβ) gene transcription have been described in some detail, there is currently no information on how GnRH overcomes repression in order to terminate reproductively inactive stages. We show here that GnRH overcomes histone deacetylase (HDAC)-mediated repression of the gonadotropin β-subunit genes in immature gonadotropes. The repressive factors associated with each of these genes comprise distinct sets of HDACs and corepressors which allow for differentially regulated derepression of these two genes, produced in the same cell by the same regulatory hormone. We find that GnRH activation of calcium/calmodulin-dependent protein kinase I (CaMKI) plays a crucial role in the derepression of the FSHβ gene involving phosphorylation of several class IIa HDACs associated with both the FSHβ and Nur77 genes, and we propose a model for the mechanisms involved. In contrast, derepression of the LH β-subunit gene is not CaMK dependent. This demonstration of HDAC-mediated repression of these genes could explain the temporal shut-down of reproductive function at certain periods of the life cycle, which can easily be reversed by the actions of the hypothalamic regulatory hormone.

AbstractThis trial represents the final stage of the CARESSES project which aimed to develop and evaluate a culturally competent artificial intelligent system embedded into social robots to support older adult wellbeing. A parallel group, single-blind randomised controlled trial was conducted across older adult care homes in England and Japan. Participants randomly allocated to the Experimental Group or Control Group 1 received a Pepper robot for up 18 h across 2 weeks. Two versions of the CARESSES artificial intelligence were tested: a fully culturally competent system (Experimental Group) and a more limited version (Control Group 1). Control Group 2 (Care As Usual) participants did not receive a robot. Quantitative outcomes of interest reported in the current paper were health-related quality of life (SF-36), loneliness (ULS-8), and perceptions of robotic cultural competence (CCATool-Robotics). Thirty-three residents completed all procedures. The difference in SF-36 Emotional Wellbeing scores between Experimental Group and Care As Usual participants over time was significant (F[1] = 6.614, sig = .019, ηp2 = .258), as was the comparison between Any Robot used and Care As Usual (F[1] = 5.128, sig = .031, ηp2 = .146). There were no significant changes in SF-36 physical health subscales. ULS-8 loneliness scores slightly improved among Experimental and Control Group 1 participants compared to Care As Usual participants, but this was not significant. This study brings new evidence which cautiously supports the value of culturally competent socially assistive robots in improving the psychological wellbeing of older adults residing in care settings.

The reproductive traits of sows are of paramount importance in the production of high-quality pork. The world experience of pig farming shows the need to solve first of all the feed problem. Diets unbalanced in the content of vitamins restrain the manifestation of genetically inherent high prolificacy of sows. The purpose of the research was to study the influence of feeding the vitamin feed additive “LipoCar” on the reproductive traits of sows and the biochemical parameters of blood serum. The results of research on the introduction of vitamin feed additive “LipoCar” into the diet of pregnant sows have been presented in the article. The experiment has been carried out in the production conditions in JSC “Linevsky pedigree farm” on sows of Large White breed. Pregnant sows have received a basic diet, balanced for all the normalized elements of nutrition. Sows of the experimental group in the composition of the basic diet fed the feed additive “LipoCar”. The dosage of the drug “Lipocar” was 2,1 g/head/day. “Lipocar” has been fed for 20 days. The total duration of the experiment was 3 months. When introducing the vitamin feed additive “LipoCar” into the main diet of sows in the second half of pregnancy the increase in the number of piglets in the litter by 1,6-6,8 % and the weight of the litter – by 5,6-56,2 % (P ≤ 0,001) have been revealed in comparison with the control group of animals. There was a higher content of carotene in the blood serum of sows of the experimental group by 50,0 % (P ≤ 0,01), vitamin A - by 48,7 % (P ≤ 0,05), and total protein – by 4,9 % (P ≤ 0,05).

In order to study the effect of dietary supplement of linseed oil on polyunsaturated fatty acids (PUFAs) in egg yolk of Gallus domestiaus, total 160 healthy Gallus domestiaus of 26-week old were randomly selected and divided into 4 groups, each of which included 40 chickens fed outdoors with an area of 63 m2 and free food and drinking water. The control group (CK) was fed the basic diet, and the experimental group was added 1, 3 and 5% of linseed oil in the fundamentals of diet (experimental group 1, 2 and 3, abbreviated as P1, P2, P3), respectively. The content of PUFA and the value of n-6 PUFA and n-3 PUFA in egg yolk were determined by gas chromatography with 24 eggs randomly selected in each group after 10th, 25th and 40th days of the commence of the experiment. We found that the levels of α-linolenic acid (C18:3n3, ALA) and docosahexaenoic acid (C22:6n3, DHA) in egg yolk were clearly higher than the CK when 3% linseed oil or 5% linseed oil were added in the diet during the three experiment periods, in which the contents of linoleic Acid (18:2n6c, LA) and arachidonic acid (C20:4n6, ARA) in egg yolk differ little between P1, P2 and P3 and the CK while the value of n-6 PUFA and n-3 PUFA in egg yolk of the P2 and P3 groups decreased significantly. The comprehensive comparisons suggested that the content of C18:3n3 and C22:6n3 in egg yolk could be clearly increased in the P2 while the value of n-6 PUFA and n-3 PUFA decreased. This study provides theoretical basis for the production of functional eggs enriched with C22:6n3 and C18:3n3.

Изучено влияние на репродуктивную функцию коров белково-витаминно-минеральных концентратов (БВМК) с кормовой добавкой «AMG-COMPLEX», состоящей из эфирных масел и растительных экстрактов. Для проведения опыта были сформированы контрольная и опытная группы по 70 голов с учетом количества дней до отела из коров второй и третьей стельности. Животным контрольной группы скармливали основной рацион. Для коров опытной группы были составлены рационы с БВМК, которыми заменяли пропорциональное количество концентрированных кормов и часть витаминно-минерального премикса. Они получали БВМК для сухостойных коров в количестве 300 г на 1 голову в сутки с 60-го по 20-й день до отела и по 200 г на 1 голову в сутки с 20-го дня до отела. После отела 60 дней им скармливали БВМК для раздоя в количестве 1200 г на 1 голову в сутки. Использование БВМК в рационах коров опытной группы способствовало улучшению у них репродуктивной функции и снижению частоты гинекологических заболеваний после отела в 4,5 раза. Индекс осеменения в опытной группе составил 1,73 (P≤0,05), что на 0,76 ниже, чем у животных контрольной группы. Средняя продолжительность сервис-периода в опытной группе составила 87,71 дня (P≤0,01) и была короче, чем в контроле, на 14,72 дня. The effect on the reproductive function of cows of the protein-vitamin-mineral concentrate with the feed additive ʺAMG-COMPLEXʺ, consisting of essential oils and plant extracts, was studied. To experiment, taking into account the number of days before calving, control and experimental group of 70 heads each were formed from cows of the second and third pregnancy. In the scientific and economic experiment, the cows of the control group consumed the feed of the main rations. For the cows of the experimental group rations were made with PVMC, which replaced the proportional amount of concentrated feed and a part of the vitamin-mineral premix. They received PVMC deadwood in the amount of 300 g per 1 head per day from 60th to 20th day before calving and 200 g per 1 head per day 20 days before calving. 60 days after calving they received PVMC lactation in the amount of 1200 g per head per day. Feeding PVMC to cows of the experimental group contributed to the improvement of reproductive function and a decrease in the frequency of gynaecological diseases after calving by 4.5 times. The insemination index in the experimental group was 1.73 (P≤0.05), which is 0.76 lower than in the animals in the control group. The average duration of the service period in the experimental group was 87.71 days (P≤0.01) and was shorter than in the control by 14.72 days.

The aim: The aim of the study was to improve the results of treatment of patients with endometriosis by using a combination method of therapy. Materials and methods: For two years, 136 women of reproductive age who underwent laparoscopic surgeries for ovarian endometriosis were monitored: Group I (n = 24) did not receive any hormonal treatment in the perioperative period; Group II (n = 32) – received gonadotropin-releasing hormone agonists within 3 months after surgery; Group III (n = 80) prior to laparoscopic removal of the ovarian cyst used gonadotropin-releasing hormone agonists – Triptorelin 3.75 mg intramuscularly for 2 months, as well as three months after surgery. The control group consisted of 30 healthy women of reproductive age with regular menstrual periods. All patients underwent transvaginal ultrasound, counting the number of antral follicles before and after treatment. Serum hormone levels (FSH, prolactin, thyrotropic hormone, anti-Mullerian hormone, inhibin B) were determined by enzyme-linked immunosorbent assay on Cobas e-411 analyzer (Roche Diagnostics, Switzerland) on day 2-3 of the menstrual cycle and on day 2–3 of the first menstrual period after the end of treatment. Laparoscopic removal of the cyst was performed with exfoliation of the cyst, hemostasis on the wound surface of the bed of the cyst was performed with a bipolar electrocoagulator. Bipolar coagulation and resection of the ovarian tissue with no potential was used during surgical treatment of the ovaries, which made it possible to preserve the intact portion of the ovary as much as possible. Results: Analysis of ovarian reserve indices, namely number of antral foliculs, number of antral follicles, AMG, and inhibin B levels in all examined patients with ovarian endometriomas were significantly lower than those of the control group before the start of treatment: in the ovarian endometrial group group 1.26 times (p <0.01), inhibin B – 1.5 times (p <0.01), the number of antral follicles – 1.2 times (p <0.01), due to the development dystrophic changes of the follicular apparatus due to prolonged compression, hypoxia, fibrosis in the ovaries. Patients who planned pregnancy were advised to have an active sexual life before menstruation was restored. In 23 (46.9%) of 49 patients who had reproductive plans, pregnancy occurred without first menstruation after a course of gonadotropin-releasing hormone agonists, 12 (24.5%) women became pregnant during the first three menstrual cycles. Extracorporeal fertilization was recommended for women who did not have pregnancy within 6 months of surgery. For two years in women who did not plan pregnancy, recurrence of endometriosis was not observed. Conclusions: The combination of laparoscopic treatment with gonadotropin-releasing hormone agonists in patients with endometriosis with infertility allowed to restore reproductive function in 71.4% of women, which indicates the effectiveness of the treatment method used. In addition, it helps to achieve lasting remission and addresses the socio-social problems of women’s health and maternity.

We previously reported that follicular wave synchronization and follicular growth treatment (FGT) before ovum pick-up (OPU) were effective in improving oocyte competence, which was associated with an increase in related embryos obtained by somatic cell nuclear transfer (Sugimura et al. 2012 Cell. Reprogram. 14, 29–37). However, oxygen consumption in oocytes remained unknown. The present study was designed to examine the differences in oxygen consumption between bovine oocytes obtained by OPU with or without FGT after in vitro maturation. Holstein dry cows (n = 8) were reared under the same feeding and environmental conditions. Two OPU sessions were conducted in each cow to collect immature oocytes, as described by Sugimura et al. (2012). The first OPU session (OPU group) was performed in cows on arbitrary days of the oestrous cycle, using a 7.5-MHz linear transducer with the needle connected to an ultrasound scanner. Follicles larger than 8 mm in diameter were then aspirated and a controlled internal drug release device (CIDR) was inserted on Day 5 (the day of the first OPU session = Day 0). Then 30 Armour units (AU) of FSH (Antrin, Kyoritsu Seiyaku, Tokyo, Japan) was administrated to cows twice a day from Day 7 to 10 in decreasing doses (6, 6, 4, 4, 3, 3, 2, 2 AU day–1). Cloprostenol (prostaglandin F2α; 0.75 mg) was administered in the morning of Day 9. The second OPU session (FGT-OPU group) was performed 48 h after prostaglandin F2α administration (Day 11), and only follicles larger than 5 mm in diameter were aspirated. The CIDR was removed from the cows just before OPU. Collected cumulus–oocyte complexes in the OPU and FGT-OPU groups were matured in vitro as described by Imai et al. [2006 J. Reprod. Dev. 52(Suppl.), S19–S29]. To collect in vivo-matured oocytes (control group), the CIDR was inserted into the cows on arbitrary days of the oestrous cycle (= Day 0), and oestradiol benzoate (0.8 mg) was administered on Day 1. The cows received the FGT treatment (as described above) from Day 6 to 10; however, the CIDR was removed in the evening of Day 8. Buserelin (gonadotropin-releasing hormone; 200 µg) was then administrated in the morning of Day 10, and OPU was performed at 24 h after gonadotropin-releasing hormone administration (Day 11). Oxygen consumption of matured oocytes was measured noninvasively with a scanning electron microscopy system (HV-405SP; Hokuto Denko Co., Tokyo, Japan). Data were analysed by ANOVA followed by a Tukey-Kramer test. There was no difference in the mean oxygen consumption between the FGT-OPU group (0.34 ± 0.02 × 10–14 mol–1, mean ± SEM) and control group (0.40 ± 0.01 × 10–14 mol–1). However, oxygen consumption in the FGT-OPU and control groups was significantly lower (P < 0.01) than that in the OPU group (0.50 ± 0.02 × 10–14 mol–1). These results revealed significantly lower oxygen consumption in OPU-derived in vitro-matured bovine oocytes after FGT treatment compared with those obtained without FGT treatment. Oxygen consumption of oocytes obtained from FGT-OPU was similar to that of in vivo-matured oocytes, which may reflect their cytoplasmic maturation status with high developmental competence.

Treatment with gonadotropin-releasing hormone (GnRH) has been used to induce ovulation in prepubertal heifers. The objective of this study was to evaluate whether peripubertal heifers will continue to ovulate at regular intervals (i.e. attain puberty) after GnRH treatment. Prepubertal crossbred Hereford heifers, 11.0 ± 0.5 months of age, 344 ± 26 kg of body weight, and at random stages of ovarian follicular wave status, were assigned to 2 groups (age- and weight-matched) and given GnRH (n = 24) or no treatment (Control, n = 22) on Day 0. Ovarian ultrasonography was performed every second day from Day 0 to 44 to record the size of 2 largest follicles and the corpus luteum (CL). Thereafter, examinations were done every 4 days until the third ovulation or Day 100, whichever came first. The first and second interovulatory intervals (IOI) were categorized as short (<12 days), normal (16-24 days), or long (>24 days), and the onset of the puberty was defined by the occurrence of 2 consecutive short or normal IOI (i.e. 3 uninterrupted ovulations). Proportional data were compared among groups by chi-squared test. Single-point measurements were compared by analysis of variance and multiple comparisons were made using Tukey’s test. Two heifers (one in each group) failed to ovulate during the experiment. The diameter of the largest follicle on Day 0 did not differ (P = 0.31) between the GnRH and Control groups (12.6 ± 0.37 mm and 13.1 ± 0.29 mm, respectively). The proportion of heifers that ovulated by Day 4 after treatment was higher in the GnRH than in the Control group (9/24 v. 1/22; P < 0.01). However, the proportion of heifers in the GnRH v. Control groups that ovulated twice (19/24 v. 18/22; P = 0.60) or 3 times (9/24 v. 9/22; P = 0.81) did not differ. The age at first ovulation tended to be younger in the GnRH group than in the Control (12 ± 0.97 v. 13 ± 0.90 months; P = 0.07), but the effect was attributed only to those heifers that ovulated in response to GnRH treatment (n = 9). The age at first ovulation was 11.2 ± 0.50, 12.7 ± 0.73, and 12.6 ± 0.90 months in GnRH-responders, non-responders, and the control group, respectively (P < 0.0001). One heifer that ovulated in response to GnRH treatment failed to ovulate again during the study period (Day 100). In 87.5% (7/8) of the heifers that responded to GnRH treatment and ovulated at least twice, the first IOI was long (55.5 ± 8.3 days). When data were combined between the GnRH non-responders and the control group (n = 35), the first IOI was short (8.9 ± 0.4 days) in 77% of the heifers, and the second IOI was of normal length (18.8 ± 0.9 days) in 93% of heifers. The age at the second ovulation in GnRH responders (12.7 ± 0.3 months) was similar (P = 0.82) to the age at first ovulation in GnRH non-responders (12.9 ± 0.1 months) and the control group (12.7 ± 0.2 months), indicating that GnRH treatment did not hasten the onset of continuous cyclicity. In conclusion, although GnRH treatment induced first ovulation in some peripubertal heifers, treatment did not hasten the onset of puberty.
