Goods of plastic, due to their durability, universality and economical properties are broadly used in all spheres of life. On the whole, polymers are inert and nontoxic, but in the process of their production, various additives are used, which on contact or introduction into an organism has a negative effect on it. In our study, we determined the impact of some types of plastic (polyvinyl chloride, polysterene and polyethylene) on the organism of laboratory animals according to changes in their body weight, indices of mass of the internal organs, and blood parameters. For the experiment, we formed four groups of white male mice at the age of 3 weeks and average body weight of 50 g. For each group, we used different litter. For group I, the litter was sawdust; and for the other groups we added plastic products in different volumes to the sawdust; for group II finely cut polyvinyl chloride, for group III cut polyethylene, and for group IV granules of polystyrene. Every 3 days, we determined the body weight of the animals, and 32 days later we determined mass of the organs, clinical and biochemical parameters of the blood. Addition of polyvinyl chloride, polyethylene, and polystyrene into the substrate for mice did not have a significant effect on tempi of growth of body weight, and also relative mass of heart and lungs. Polyvinyl chloride and polystyrene have an immune-suppressive effect, and polyvinyl chloride affects both central and peripheral organs, and polystyrene mostly harms the peripheral organs. All used types of plastic cause leukocytopenia, following which neutrophilia of band neutrophils and monocytosis takes place as a result of damage to the biological barriers. We determined the systemic toxic effect of the studied types of plastic on the internal organs, which manifested in increase in their mass (liver, kidneys), steep increase in the activity of liver enzymes (AST, ALT), simultaneous decrease in activity of alkaline phosphatase and content of cholysterol and glucose in the blood serum of the mice. Also polyvinyl chlorine, polyethylene and polystyrene cause degeneration of the epithelium of the uriniferous tubule, which is manifested in reduction of globulins and creatinine in the blood of animals from the experimental groups following increase in relative mass of the kidneys. The results of our research allow us to state that different types of plastic can cause toxic effect on animals, as well as people who are in frequent contact with them.

Herbicides and food additives are included in many food products for humans. Non-used products or products beyond their expiry date are deposited in places of utilizatioin of solid municipal wastes, where they can take effects on the organisms of mouse-like rodents. Among the herbicides, glyphosphate takes first place in the world for volume of production, and is the most intensely used in agricultural farming. The discussion about negative impact on the organisms of mammals, especially against the background of using various substances and environmental factors, continues. In this study, we determined the combined effect of glyphosphate and food additives on the organism of laboratory animals, which manifested in changes in body weight, condition and indices of mass of the internal organs and blood parameters. Four groups of laboratory male rats were formed, which over 42 days received: unlimited access to clean water; 1% aqueous solution of glyphosate; 1% solution of glyphosphate and 1% solution sodium benzoate; 1% solution of glyphosphate with 1% solution of saccharin. Glyphosphate and glyphosphate with sodium benzoate and saccharin significantly reduced the daily increases in body weight of animals compared to the control group. The studied substances have notable suppressive effect on the immune system and haematopoiesis in general, which is manifested in reduce of relative mass of the thymus and spleen against the background of increase in the amount of lymphocytes in the peripheral blood. The inhibition of haematopoiesis is indicated by decrease in the amount of erythrocytes, neurophils and hemoglobin of blood of animals from the experimental groups. The impact on the digestive system of glyphosphate and food additives is indicated by occurance of the effect of “irritation” of mucous membranes, and, as a result, disorders in absorption followed by the disorder in metabolic processes. A dysbalance occurs in enzymic systems of the organism, which is manifested in distrophic processes, especially in the liver parenchyma, indicated by the activity of blood enzymes (ALT, AST, alkaline phosphatase), total number and ratio of proteins of blood plasma. We determined the impact of glyphosphate and its mixes with benzoate and saccharin on the pancreas, which manifests in severe pancreatitis with steep increase in the level of glucose of blood. The results of the study allow us to state that mixture of glyphosphate and food additives can cause toxic effect in animals and humans, which often contact with herbicides.

Diseases of the genitourinary system caused by pathogenic and potentially pathogenic microorganisms, which result into disbiosis of urinary organs, remain an urgent problem of dermatovenereology, gynecology and urology, despite the fact that there is a significant number of available and new medicines to treat them. The aim of the work was to determine the safety of the preparation Hexia within experiments on animals. The acute toxicity of Hexia has been determined on 20 adult female laboratory rats under the conditions of hourly intravaginal administration of the preparation with a dose of 70 mg/kg during 12 hours, as well as on 20 female laboratory rats and 20 female laboratory mice with a single intragastric administration in a dose of 145 mg/kg. The assessment of the impact of the preparation studied was performed on the basis of the following parameters: a) mortality (terms of death of animals in each group, on a daily basis); b) assessment of toxicity development (on a daily basis), including an assessment of the visual environment of the area of injection (the presence of irritation, redness, edema); c) dynamics of body weight changes (in the initial state, on the 4th, 7th and 14th day after application); d) macroscopy of internal organs, mass coefficients of internal organs within rats (on the 14th day). It has been found that intravaginal application and a single intragastric administration of the preparation Hexia in the form of pessaries, which contain chlorhexidine digluconate, does not result into death of rats and mice, brings no effect on body weight gain, integrative parameters of the functional state of laboratory animals as well as on the relative mass of internal organs, which implies the absence of significant toxic effect of the preparation. Thus, the results of the studies conducted indicate that the median lethal dose for Hexia in case of intravaginal application to rats or intragastric administration to rats and mice is beyond the rate of 500 mg/kg. According to the toxicological classification of substances Hexia belongs to the IV class of toxicity – low toxic substances.

Objective. To investigate the impact of the sorbtion-aspiration drainage system on background of hydroxyapatite and zinc oxide on experimental peritonitis. Materials and methods. The experiment was performed on 55 white laboratory rat males, ageing 2 - 3 mo, with the body mass 225 - 250 g, which were kept in the typical vivarium conditions. Suspension of E. coli in physiological solution, 1.0 - 1.5 ml in concentration 1010 , was used for the peritoneum contamination. The suspension was introduced into abdominal cavity in the dose, dependent on the animal body mass, using the syringe for insulin injection. Peritonitis in the rats was developed in 48 h and characterized by intoxication, which exaggerated rapidly, as well as by intestinal motor disorders and pronounced microcirculatory disturbances. The experimental animals were distributed into 5 Groups. In patients of Group I the interventions were not performed (control); in laboratory animals of Group II peritonitis was not simulated, but the abdominal cavity was drained, using the sorption-aspiration drainage system; in laboratory animals of Group III peritonitis was simulated; in the Group IV of laboratory animals on background of peritonitis the abdominal cavity was drained, using the sorption-aspiration drainage system; in the Group V animals the abdominal cavity was drained by a standard silicon drain on background of peritonitis. Results. Reduction of abdominal contamination by 43.12% and of histological features were produced in abdominal organs under the influence of the composite system, witnessing its bactericidal and sorption-antiinflammatory action. Conclusion. Experimentally there was confirmed the possibility of application of sorption-aspiration drainage system on background of hydroxyapatite and zinc oxide in treatment of an acute peritonitis.

AbstractCombinations of fructose- and fat-rich diets in experimental animals can model the human metabolic syndrome (MS). In rats, the increase in blood pressure (BP) after diet manipulation is sex related and highly dependent on testosterone secretion. However, the extent of the impact of diet on rodent hypophysial-testicular axis remains undefined. In the present study, rats drinking a 10% fructose solution or fed a high-fat (35%) diet for 10 weeks had higher plasma levels of luteinizing hormone (LH) and lower plasma levels of testosterone, without significant changes in circulating follicle-stimulating hormone or the weight of most reproductive organs. Diet manipulation brought about a significant increase in body weight, systolic BP, area under the curve (AUC) of glycemia after an intraperitoneal glucose tolerance test (IPGTT), and plasma low-density lipoprotein cholesterol, cholesterol, triglycerides, and uric acid levels. The concomitant administration of melatonin (25 μg/mL of drinking water) normalized the abnormally high LH levels but did not affect the inhibited testosterone secretion found in fructose- or high-fat-fed rats. Rather, melatonin per se inhibited testosterone secretion. Melatonin significantly blunted the body weight and systolic BP increase, the increase in the AUC of glycemia after an IPGTT, and the changes in circulating lipid profile and uric acid found in both MS models. The results are compatible with a primary inhibition of testicular function in diet-induced MS in rats and with the partial effectiveness of melatonin to counteract the metabolic but not the testicular sequelae of rodent MS.

Iron (IV) clathrochelate based on a macrobicyclic ligand of the hexahydrazide type is a unique compound that contains iron in a rare high valence IV. Preclinical and clinical studies of this complex, which were started for the first time in Ukraine, have an important theoretical and practical consequence as this complex can be recommended as an active substance in iron-containing drugs with antianemic action. In conducting preclinical studies of new drugs, pathomorphological studies are important because they are a necessary step in studying the biological response of animals to the action of test substances. It was found that some pathological changes develop in the body of white mice under conditions of experimental acute and chronic iron (IV) clathrochelate intoxication. They correlated with the dose of the test compound. During chronic intoxication, the microscopic changes in the liver and kidney of white mice treated with iron (IV) clathrochelate at a dose of 1/10 DL50 were similar to the microscopic changes in the liver and kidney of mice treated with the experimental drug at a dose of 1/5 DL50. However, the severity of these changes was lower, reflecting a lower degree of organ damage. In the myocardium of mice treated with iron (IV) clathrochelate at a dose of 1/5 DL50 on the 10th day, as in acute iron (IV) clathrochelate poisoning, only edema was recorded. The prospects for further research are the study of microscopic changes in the organs of laboratory animals of other species during experimental iron (IV) clathrochelate toxicosis.

This study aimed to determine the impact of Lactobacillus plantarum PC170 concurrent with antibiotic treatment and/or during the recovery phase after antibiotic treatment on the body weight, faecal bacterial composition, short-chain fatty acids (SCFAs) concentration, and splenic cytokine mRNA expression of mice. Orally administrated ceftriaxone quantitatively and significantly decreased body weight, faecal total bacteria, Akkermansia muciniphila, and Lactobacillus plantarum, and faecal SCFAs concentration. Ceftriaxone treatment also dramatically altered the faecal microbiota with an increased Chao1 index, decreased species diversities and Bacteroidetes, and more Firmicutes and Proteobacteria. After ceftriaxone intervention, these changes all gradually started to recover. However, faecal microbiota diversities were still totally different from control by significantly increased α- and β-diversities. Bacteroidetes all flourished and became dominant during the recovery process. However, mice treated with PC170 both in parallel with and after ceftriaxone treatment encouraged more Bacteroidetes, Verrucomicrobia, and Actinobacteria, and the diversity by which to make faecal microbiota was very much closer to control. Furthermore, the expression of splenic pro-inflammatory cytokine tumour necrosis factor-α mRNA in mice supplemented with PC170 during the recovery phase was significantly lower than natural recovery. These results indicated that antibiotics, such as ceftriaxone, even with short-term intervention, could dramatically damage the structure of gut microbiota and their abilities to produce SCFAs with loss of body weight. Although such damages could be partly recovered with the cessation of antibiotics, the implication of antibiotics to gut microbiota might remain even after antibiotic treatment. The selected strain PC170 might be a potential probiotic because of its contributions in helping the host animal to remodel or stabilise its gut microbiome and enhancing the anti-inflammatory response as protection from the side effects of antibiotic therapy when it was administered in parallel with and after antibiotic treatment.

This study examined the impact of early weaning on antioxidant function in piglets. A total of 40 Duroc × Landrace × Large White, 21-day-old piglets (half male and half female) were divided into suckling groups (SG) and weaning groups (WG). Piglets in WG were weaned at the 21st day, while the piglets in SG continued to get breastfed. Eight piglets from each group were randomly selected and slaughtered at 24th-day (SG3, WG3) and 28th-day old (SG7, WG7). The body weight, liver index, hepatocyte morphology, antioxidant enzymes activity, gene expression of antioxidant enzymes, and Nrf2 signaling in the liver of piglets were measured. The results showed that weaning caused decreased body weight (p < 0.01), lower liver weight (p < 0.01), and decreased the liver organ index (p < 0.05) of piglets. The area and size of hepatocytes in the WG group was smaller than that in the SG group (p < 0.05). We also observed that weaning reduced the activity of superoxide dismutase (SOD) and catalase (CAT) (p < 0.05) in the liver of piglets. Relative to the SG3 group, the gene expression of GSH-Px in liver of WG3 was significantly reduced (p < 0.05). The gene expression of Nrf2 in the SG3 group was higher than that in the WG3 group (p < 0.01). The gene expression of NQO1 in the SG7 group was higher than that in the WG7 group (p < 0.05). In conclusion, weaning resulted in lower weight, slowed liver development, and reduced antioxidant enzymes activity, thereby impairing liver antioxidant function and suppressing piglet growth.

Abstract Introduction There are a substantial number of lung cancer patients with negative mutations in Indonesia. This type of cancer is deemed to be the major contributor of lung cancer patient’s death. However, reseaerch related to therapy using vinorelbine combined with platinum-based compounds is still scarce in Indonesia. The aim of this study was to compare the efficacy and tolerability between vinorelbine and carboplatin with vinorelbin and cisplatin in stage III-IV epidermal growth factor receptor (EGFR) mutations-negative non-small cell lung cancer (NSCLC). Methods The participants were divided into two groups—group I(vinorelbine–carboplatin) and group II (vinorelbine–cisplatin). The participants were assessed based on several measurement criteria. Not only Eq-5D was performed, but the body weight and response evaluation criteria for solid tumors (RECIST) were also examined. The participants received chemotherapy for four cycles (1 cycle = 21 days). Results The quality of life was considered stable in 60% of group I and 60% of group II (p=0.255). In both groups, 46.67% of participants had an increased body weight, while the other 20.00% was stable (p = 1.000). In terms of RECIST evaluation after the second cycle, 80.00% of group I and 86.67% of group II were considered to have a stable disease, with 20% of group I and none of group II had partial response (p = 0.027). However, after the fourth cycle, there were no significant difference between the groups (p = 0.734). Conclusion In EGFR mutation-negative NSCLC patients, the combination of vinorelbine and carboplatin showed comparable outcomes to vinorelbine and cisplatin chemotherapy with no significant differences.

Suspension for injection “Amoxidev 15” is prescribed to fur-bearing animals (mink, fox), dogs and cats for the treatment of respiratory diseases (tonsillitis, tracheitis, pneumonia, bronchitis, rhinitis, sinusitis, bronchopneumonia), digestive (gastritis, enteritis, enteritis). genitourinary systems (nephritis, urethritis, urocystitis, mastitis, metritis, agalactia), musculoskeletal system (arthritis, osteoarthritis, joint injuries, tendonitis, hoof lesions), skin and soft tissues (eczema, dermatitis) caused by sensitive drug by microorganisms, including colibacillosis, streptococcus, bronchopneumonia, etc. Toxicological evaluation of the veterinary drug “Amoxidev 15” under the conditions of acute and subacute toxicological experiments on a model of white rats. According to the results of an acute toxicological experiment with intragastric administration of the drug “Amoxidev 15” white rats DL50 could not be calculated because the death of laboratory animals was not detected within 14 days after administration. The maximum administered dose (in absolute weight of the drug) was 20000.0 mg/kg body weight, which allows to refer the drug to class VI toxicity of relatively harmless substances (DL50 > 15000 mg/kg body weight), and the degree of safety to class IV – low-hazard substances (DL50 > 5000 mg/kg). According to the results of an acute toxicological experiment with subcutaneous administration of the drug “Amoxidev 15” white rats DL50 could not be calculated because the death of laboratory animals was not detected within 14 days after administration, the maximum dose was 5000.0 mg/kg body weight, therefore, the drug “Amoxidev 15” when administered subcutaneously by toxicity can be classified as class VI substances relatively harmless (DL50 Subcut > 4500.0 mg/kg). When administered subcutaneously to white rats, the drug “Amoxidev 15” under conditions of subacute toxicological experiment in doses of 0.1–1.0 ml/kg does not cause hemo-, hepato- and nephrotoxic effects on the body of laboratory animals, although 3-day administration of the drug in a dose 1.0 ml/kg body weight caused an increase in the activity of hepatospecific enzymes ALT and AST by 12.5 and 11.1 % (P < 0.05), respectively, relative to the control, which was restored to the control level 7 days after cessation.

Khat, Catha edulis, use is rampant in Eastern Africa and Middle East countries with associated reports of reproductive function impairment in the body of the user. Reports on recovery post long-term khat exposure are obscure. The present study investigated evidence of restoration of testicular and epididymal structure and function during withdrawal from cytotoxic damage caused by sub-chronic exposure of khat extract. Twenty-eight male rabbits were divided into 7 groups of 4 rabbits each. Group I (control) was administered normal saline while groups II, III and IV were administered 1.0 g/kg, 10 g/kg and 20 g/kg body weight of khat extract, respectively, via oral gavage on alternate days of the week for 12 weeks. Blood samples from animals were collected for hormonal assays followed by euthanasia using 26.4 mg/kg body weight of Sagatal sodium intramuscularly for testicular and epididymal histology. Group V, VI and VII were administered 1.0 g/kg, 10 g/kg and 20 g/kg body weight of khat extract, respectively, orally on alternate days of the week for 12 weeks followed by 1-month withdrawal period, blood samples collected for hormone assays and animals sacrificed for testicular and epididymal histology. High khat dose, 20 g/kg body weight, at sub-chronic exposure caused degeneration in spermatogenic cells with accompanying decrease in plasma FSH and testosterone. Histological output of Sertoli cells, Leydig cells and epididymal epithelium appeared unaffected in treatment groups. Post withdrawal data showed apparent regeneration of seminiferous epithelium and restoration of plasma FSH and testosterone comparable to control. It appears khat extract preferentially affected germ cell spermatogonia and subsequent daughter cells while stem cell spermatogonia were unaffected and contributed to regeneration of germinal epithelium and endocrine function.
