The article gives the results of the research on the influence of a developed complex liposomal preparation on the dynamics of morphological and biochemical indicators of the blood of rats subjected to modeled oxidative stress caused by intermuscular injection of 50% carbon tetrachloride at a dose of 0.25 ml per 100 g of body mass. We found that in conditions of intoxication by carbon tetrachloride, the physiological level of hematological indicators of the researched animals’ body was disrupted. This is indicated by the reduction in the amount of erythrocytes, hemoglobin content, concentration of hemoglobin per erythrocyte, increase in the amount of leucocytes, mass of hemoglobin per erythrocyte and increase of colour indicator. Also, we detected suppression of protein synthesis function of the liver. The levels of total protein and albumin fraction were below the normal physiological level. High indicators were observed in the levels of creatinine, urea and total bilirubin. In order to normalize the functional state of the liver under oxidative stress it is reasonable to use a liposomal preparation which contains butafosfan, interferon, thistle and vitamins. In order to normalize morphological and biochemical indicators of the blood of rats subject to intoxication of carbon tetrachloride it is reasonable to use a liposomal preparation that contains butafosfan, selenium, thistle, methionine and vitamins. Using the liposomal preparation Butaselmevit for rats under oxidative stress the morphological and biochemical indicators in the blood normalized. On the 14th day the indictors of the number of erythrocytes, the hemoglobin content, the white blood cell count and red blood indexes compared to control came within the physiological indicators, indicating a recovery of hematopoietic function of the bone marrow and normalized indicators of the functional state of the liver.

The article presents a study of the hepatoprotective activity of a tricyclic heterocycle, which refers to 5, 6, 7, 8-tetrahydroquinolines. The effect of 8, 8-dimethyl-5-p-tolyl-8, 9-dihydro-2H-pyrido [4, 3, 2-de] cinnolin-3 (7H) was studied on rats under the influence of the model of toxic hepatosis induced by carbon tetrachloride to find out the indicators of peroxidation and biochemical indicators. Biochemical studies have shown that modelling toxic fat hepatosis caused by the inception of carbon tetrachloride to rats increased the activity of alanine aminotransferase by 2.5 times more compared with the intact group, indicating the development of oxidative stress induced by the treatment of pyrido [4, 3, 2] Cinnol I that reduced the toxic effect of CTC by 79.9 %. Mexidol had a less pronounced hepatoprotective effect: the activity of Alanine aminotransferase on animals of the second group was lower by 29.2 % than on rats from the control group. Thus, a new compound with hepatoprotective activity has been developed and studied.

This article presents the results of research into the influence of the complex liposomal preparation Butaintervit on protein synthesis function in the livers of rats under the influence of carbon tetrachloride poisoning. Intramuscular injection of carbon tetrachloride into rats at a dose of 0.25 ml per100 gof body weight causes antigenic load on the body and leads to disruption of protein synthesis function in the liver. This is shown by reduction in blood levels of total protein and its fractions. Thus, the level of albumin in the serum of rats under the conditions of oxidative stress was 70% lower than in clinically healthy animals. However, the level of total protein in the serum was only 10% lower. This is because, along with the decrease of albumin content in the serum, the levels of globulin protein fraction increased by 8.8%. This has led to albumin/globulin disparities in the serum of sick animals. As a result, the value of A/G coefficient was 0.28 ± 0.03, compared to 0.52 ±0.02 inclinically healthy rats. For the normalization of functional state of the liver under oxidative stress it is advisable to apply the liposomal preparation Butaintervite, which in its structure contains butafosfan, interferon, thistle and vitamins A, D and E. Under conditions of oxidative stress and under the action of the liposomal preparation in the rats from the second experimental group we have found significant increase in the levels of total protein and albumins and a decrease in serum globulin in the animals on the fifth and tenth days of the experiment. On the fourteenth day of the experiment under the conditions of oxidative stress and under the action of the liposomal preparation in the rats from the second experimental group the normalization of protein synthesis function in the liver was observed. The level of indicators of total protein, albumin, globulin and the coefficient of albumin/globulin compared with the control group of animals were within normal values.

Introduction. Digital technologies are actively used in the work of specialists of medical centers of various profiles, which causes the impact on employees of a number of professional factors that determine the characteristics of their work. The aim of the study is to evaluate the influence of professional factors on the functional state of the employees' body and the glutathione system as an objective indicator of stress based on the analysis of the subjective perception of the medical center employees of the specifics of working with personal computers and psychophysiological testing. Materials and methods. The study involved 109 specialists of medical centers (25 ophthalmologists, 31 representatives of the secondary medical staff - nurses and paramedics, 53 office employees). The anonymous survey concerned the assessment of working conditions and their impact on the functional state of the body of the respondents. Performance and stress tolerance indicators were evaluated using three standard questionnaires. The study of 66 subjects' levels of glutathione in the blood: total, reduced, oxidized and the ratio of reduced and oxidized - was used to analyze the relationship of the neuro-emotional nature of labor with the reaction of oxidative stress. Statistical processing of the obtained data was carried out using traditional methods of variation statistics and calculating the value of the odds ratio (OR) with a 95% confidence interval (CI). Results. The survey revealed a significant proportion of people in all groups of respondents who constantly use computer technology in their work. Describing the quality of the processed information, doctors more than representatives of other groups note its complexity, importance, negative emotional color, a high degree of responsibility and tension. Doctors more often than average medical staff and office workers associate the manifestation of fatigue with an uncomfortable state of the visual organ, nervous system, and musculoskeletal system. The subjects showed reduced performance and stress tolerance with the most pronounced negative trends in the group of doctors. Changes in the glutathione system were detected, indicating the presence of oxidative stress in 40% of ophthalmologists and office workers, and in half of nurses. Conclusions. The work of specialists of medical centers of various profiles in accordance with their assessment is characterized by high nervous and emotional stress, due to the need to process a significant amount of complex and important professionally significant information. Subjectively noted by respondents the influence of working conditions on the functional state of the body is confirmed by indicators of the glutathione system, which can be used as indicators of nervous and emotional stress.

Background and Objective : A Tibetan traditional herb named Swertia mussotii Franch., also called “Zangyinchen” by the local people of Qinghai-Tibet area, has been used to protect the liver from injury for many years. However, the curative effect and molecular mechanism of the herb have not been demonstrated clearly. Materials and Methods: In our study, serum alanine aminotransferase, aspartate aminotransferase, total bilirubin levels were examined after S. mussotii Franch. treatment in the acute liver injury of the carbon tetrachloride-induced rat model. Then, Proteome Analysis was applied to explore the potential mechanism of SMT for hepatoprotective effects after iTRAQLC-MS/MS analysis (isobaric tag for relative and absolute quantification-liquid chromatograph-mass spectrometer with tandem mass spectrometry). Results: Serum results showed, alanine aminotransferase, aspartate aminotransferase, total bilirubin levels of rats with acute liver injury were all improved with SMT treatment. Moreover, Proteome Analysis suggested that, with S. Mussotii Franch. treatment, the levels of lipid catabolic process and lipid homeostasis were all enhanced. And the results of protein-protein interaction (PPI) analysis illustrated that these proteins assembled in PPI networks were found almost significantly enriched in response to lipid, negative regulation of lipase activity, response to lipopolysaccharide etc. Furthermore, the downregulated MRP14 and MRP8 proteins were found involved in the lipid metabolism, which may indicate the mechanism of SMT protection liver from ALI induced by carbon tetrachloride. Conclusion: SMT herb could play a role in hepatoprotection and alleviate the effect of acute liver injury by impacting the lipid metabolism associated biological process.

Background: Schizophrenia is associated with a deficiency of dietary antioxidants like vitamin B6, B9, and B12 resulting in defective methylation leading to hyperhomocysteinemia. Hyperhomocysteinemia causes mitochondrial DNA damage, oxidative stress, vascular damage, and lipid peroxidation. Oxidative stress and increase in reactive oxygen species result in 8-oxodG production which induces apoptosis of both astrocytes and thyrocytes thus predisposing them to thyroid dysfunction and neurodegeneration. Furthermore, the presence of excessive free radicals increases thyroid thermogenesis causing hyperthyroidism or its excess may cause hypothyroidism by inhibiting iodide uptake. In the present study, we evaluated the various biomarkers associated with thyroid dysfunction in schizophrenics.Materials and Methods: 288 patients suffering from schizophrenia and 100 control subjects were screened for liver function tests (LFTs) such as alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), and total bilirubin (TB). Also, the stress markers, namely malondialdehyde (MDA), homocysteine, cysteine, methionine, the thyroid profile including triiodothyronine (T3), thyroxine (T4), thyroid-stimulating hormone (TSH), thyroxine peroxide antibody (TPO-Ab); TSH receptor-Ab (TSHr-Ab), dietary antioxidants, lipids, cytokines, aminoacids and hormones, vitamins and trace elements, and other biochemical parameters.Results: The LFTs showed elevated levels of ALT (45.57 ± 4.87 Vs. 26.41 ± 3.76 U/L), AST (40.55 ± 1.34 Vs. 21.92 ± 3.65 U/L), ALP (121.54 ± 4.87 Vs. 83.76 ± 5.87 U/L), and total bilirubin (2.63 ± 0.987 Vs. 1.10 ± 0.056 mg/dl), in schizophrenics than controls. Increased levels of MDA (3.71 ± 0.967 Vs. 1.68 ± 0.099) and homocysteine (17.56 ± 2.612 Vs. 6.96 ± 1.987 μmol/L were observed in schizophrenics compared to the controls, indicating increased stress. Levels of cysteine and methionine were decreased in schizophrenics than the controls (1.08 ± 0.089 Vs. 4.87 ± .924 μmol/L and 17.87 ± 1.23 Vs. 99.20 ± 5.36 μmol/L). The levels of TPO-Ab (IU/ml), Tg-Ab (pmol/L), and TSHr-Ab (IU/L) were observed to be higher in the patients’ group as compared to control subjects (9.84 ± 2.56 Vs. 5.81 ± 1.98, 55.50 ± 2.98 Vs. 32.95 ± 2.87 and 2.95 ± 0.0045 Vs. 1.44 ± 0.0023 respectively). Levels of Vitamin B6, B9, and B12 were also significantly decreased in the patients compared to the healthy controls.Conclusion: The schizophrenics, demonstrated altered liver function, increased stress markers, and decreased dietary antioxidants. Reduced primary and secondary antioxidant levels, may result in hyperhomocysteinemia and cause further DNA and mitochondrial damage. Therefore, homocysteine and/or prolactin levels may serve as candidate prognostic markers for schizophrenia. Also, both neurological symptoms and the susceptibility to thyroid disorders may be prevented in the initial stages of this debilitating disorder by appropriate dietary supplementation of antioxidants which can rectify a reduction in primary and secondary antioxidants, and disturbed prolactin-serotonin-dopamine interactions in schizophrenics.

Abstract Background Titanium-based compounds have been incorporated as promising antineoplastic metals. In our previous studies, dithiophenolato titanium (IV) Complex "DBT" and its chitosan nanocomposite "DBT-CSNPs" were prepared and we showed that these compounds have antibacterial activities, cytotoxic, and have abilities to bind with DNA helixes. Therefore, in this study, we evaluated the LD50 values of dithiophenolato titanium (IV)-complex (DBT) and its high thermal stable chitosan nanoparticles (DBT-CSNPs). Then their therapeutic effects against liver injuries induced by carbon tetrachloride (CCl4) were assessed and compared with cisplatin treatment. Additionally, the anti-proliferative activity of DBT and DBT-CSNPs against human liver cancer (HepG2) cell lines through the analysis of the cell cycle was evaluated. Methods Nine groups of rats were prepared: normal, DBT, DBT-CSNPs, CSNPs, CCl4, CCl4-DBT, CCl4-DBT-CSNPs, CCl4-CSNPs and CCl4-cisplatin. Liver histopathology and the biochemical markers involving oxidative stress, apoptosis, liver and kidney functions, and lipid profile were determined. Results The results revealed that the treatment with DBT-CSNPs and DBT after CCl4 administration abolished liver damage since it reduced the apoptosis induced by CCl4 via the reduction of DNA fragmentation, Bax and caspase- 8 with an elevation of Bcl2 and Bcl2/Bax ratio. Also, these treatments caused nonsignificant changes in the markers of oxidative stress. Therefore, liver histopathology and functions, lipid profile, and kidney functions were improved. Cisplatin treatment reduced liver injury with a degree less than DBT-CSNPs and DBT, but it induced nephrotoxicity. Administration of DBT-CSNPs and DBT to healthy rats for 14 days has no adverse effect. Also, the results showed that DBT-CSNPs and DBT inhibited the proliferation of HepG2 cells by arresting cells in the G2/M phase and inducing cell death. Conclusion DBT-CSNPs and DBT have a therapeutic effect against CCl4-induced liver injuries via the reduction of apoptosis induced by CCl4. Moreover, both compounds have antineoplastic activities against the HepG2 cell line. In all cases, DBT-CSNPs have a greater effect due to their nanostructure. Therefore, both compounds can be used in the pharmacological fields, particularly DBT-CSNPs.
