The methanol extract of Actaea acuminata roots have beenpreviously reported to exhibit significant antianxiety, anticonvulsant andantidepressant activities. In present study fractionation of crude bioactivemethanol extract was carried out using different solvents employing standardprocedure. Various fractions were evaluated for antianxiety, anticonvulsantand antidepressant activities using elevated plus maze model (EPM), maximalelectroshock-induced convulsions model (MES) and forced / despair swim test(FST) respectively. Successive partitioning of bioactive methanol extract wasdone with ethyl acetate and 1-butanol. The ethyl acetate fraction (EAF; 100mg/kg), 1-butanol fraction (BF; 25 mg/kg) and remaining methanol extract(RME; 70 mg/kg) were subjected to neuropharmacological activities. TheEAF significantly enhanced entries and average time spent in open arms;significantly decreased duration of MES-induced tonic extension phase andsignificantly decreased duration of immobility time of rats in comparisonto control. BF and RME did not exhibit any neuropharmacological activity.Qualitative chemical tests confirmed presence of alkaloids and polyphenolsin EAF. It is finally concluded that alkaloids and/or polyphenols are bioactiveconstituents of A. acuminata which are responsible for neuropharmacologicalactivities.

Abstract:: HeMI (12.5–100 mg/kg, p.o.) was administered 1 h before subjecting the animal to the forced swim test (FST), tail suspension test (TST) and elevated plus maze tests (EPM).: HeMI (12.5–100 mg/kg, p.o.) treatment produced significant reduction in immobility time [F(6.56)=8.35, p<0.001], [F(6,56)=7.55, p<0.001] in the FST and TST, respectively. Moreover, co-administration of sub-therapeutic doses of imipramine or fluoxetine with HeMI (3.125 mg/kg) elicited significant reduction in time spent immobile in the FST. However, pretreatment of mice with parachlorophenylalanine, metergoline, yohimbine or sulpiride abolished the antidepressant-like effect elicited by HeMI. In the EPM, HeMI produced significant [F(5,42)=8.91, p<0.001] increase in open arms exploration by 75.55 % and this effect was blocked by pretreatment of mice with flumazenil or metergoline.: Findings from this study showed antidepressant-like effect of

Penelitian terhadap ekstrak metanol, fraksi etil asetat, dan fraksi butanol dari rumput laut Eucheuma spinosum menggunakan metode Disc Diffusion Test dilakukan untuk mengetahui peranannya sebagai antibakteri Escherichia coli ATCC 25922, dan Staphylococcus aureus ATCC 6538. Sebagai kontrol digunakan ekstrak (negatif) pelarut metanol, dan Amoxicilin. Konsentrasi sampel uji ditetapkan 10.000 ppm. Hasil pengujian menunjukkan adanya  aktivitas antibakteri yang ditandai dengan terbentuknya zona hambat terhadap bakteri Escherichia coli ATCC 25922.  Besarnya zona hambat secara berurutan untuk ekstrak metanol, fraksi  butanol, dan fraksi etil asetat, ialah : 6,98 mm, 7,85 mm, dan 7,88 mm.  Nilai zona hambat tersebut, relatif sama untuk setiap sampel uji. Sampel uji juga menunjukkan terdapat aktivitas antibakteri, ditandai dengan terbentuknya zona hambat terhadap bakteri Staphylococcus aureus ATCC 6538.  Besarnya zona hambat secara berurutan untuk ekstrak metanol, fraksi  butanol, fraksi etil asetat,  dan kontrol negatif pelarut metanol, ialah : 7,42 mm, 14.40 mm, 7,90 mm, dan 6,75 mm.  Zona hambat fraksi butanol lebih tinggi dibanding yang lain, ini menunjukkan bahwa aktivitas antibakteri Staphylococcus aureus pada fraksi butanol lebih tinggi dibanding fraksi lainnya.Kata kunci:  Antibakteri;  Eucheuma spinosum;  Escherichia coli; Staphylococcus aureus Antibacteria Activities of Extract Sea Algae  Eucheuma spinosum Fresh from Nain Island Waters North Sulawesi  ABSTRACTA research on antibacterial activity of methanol extract, ethyl acetate, and butanol fraction from Eucheuma spinosum seaweed using the disc diffusion method to know the role as  antibacterial activity was tested against Escherichia coli ATCC 25922 and Staphylococcus aureus ATCC 6538. Methanol was used as negative control while amoxicillin was used as positive control. The concentration of the test sample was set at 10,000 ppm. The antibacterial activity was indicated by the formation of an inhibition zone against the tested microorganisms. The inhibition zones against E.coli for methanol extract, butanol fraction, and ethyl acetate fraction were 6.98 mm, 7.85 mm, and 7.88 mm, respectively.  The value of the zone of inhibition is relatively the same for each sample.  In addition, the inhibition zones of methanol extract, butanol and ethyl acetate fraction against Staphylococcus aureus were 7.42 mm, 14.40 mm, 7.90 mm, and 6.75 mm, respectively. The inhibition zone of the butanol fraction is higher than the others, this indicated that the antibacterial activity for butanol fraction is higher than other fractions. Keywords: Antibacterial activity; Eucheuma spinosum; Escherichia coli; Staphylococcus aureus

Decrease in natural illumination in fall/winter months causes depressive-like seasonal affective disorders in vulnerable individuals. Obesity is another risk factor of depression. The lethal yellow (AY) mutation causes ectopic expression of agouti protein in the brain. Mice heterozygous for AY mutation (AY/a) are obese compared to their wild-type littermates (a/a). The main aims of the study were to investigate the effects of AY mutation, photoperiod and the interaction between these factors on daily activity dynamics, feeding, locomotor and exploratory activities, anxiety-related and depressive-like behaviors in mild stress condition. Six weeks old mouse males of AY/a and a/a lines were divided into four groups eight animals each and exposed to long- (14 h light and 10 h darkness) or short- (4 h light and 20 h darkness) day conditions for 28 days. Then the behavior of these mice was successively investigated in the home cage, open field, elevated plus-maze and forced swim tests. We did not observed any effect of AY mutation on the general activity, water and food consumption in the home cage; locomotion and exploration in the open field test; anxiety-related behavior in the open field and elevated plus-maze tests. At the same time, AY mutation increased depressive-like immobility time in the forced swim test (F1.28 = 20.03, p = 0.00012). Shortday conditions decreased nocturnal activity in the home cage, as well as locomotion (F1.28 = 16.33, p = 0.0004) and exploration (F1.28 = 16.24, p < 0.0004) in the open field test. Moreover, short-day exposition decreased time spent in the center of the open field (F1.28 = 6.57, p = 0.016) and in the open arms of the elevated plus-maze (F1.28 = 12.08, p = 0.0017) tests and increased immobility time in the forced swim test (F1.28 = 9.95, p = 0.0038). However, no effect of the interaction between AY mutation and photoperiod on immobility time in the forced swim test was observed. Therefore, short-day photoperiod and AY mutation increased depressive-like behavior in the forced swim test by means of different mechanisms.

The present investigation was planned to estimate total phenols and antioxidant activity in Clematis erecta aerial parts. Properly identified powdered plant was successively extracted in Soxhlet apparatus with solvents in order of increasing polarity viz., petroleum ether, chloroform, methanol and water. Ethyl acetate fraction and 1-butanol fraction of methanol extract were prepared using standardized procedure. Quantification of phenols was done on the basis of standard curve of gallic acid using standardized procedures. In vitro antioxidant activity was evaluated using DPPH assay in comparison to standard drug rutin. Maximum phenols content and antioxidant activity was found in ethyl acetate fraction followed by methanol extract and 1-butanol fraction, whereas negligible phenols content and antioxidant activity was observed in petroleum ether, chloroform and water extracts. The available literature reveals that polyphenols have been reported to possess antioxidant properties. Finally, it can be concluded that these polyphenols may be responsible for antioxidant properties.

Verbena officinalis L. (Vervain; family-Verbenaceae) has long been used in the treatment of mental disorders, but not validated systemically for traditional claims. The ethyl acetate fraction (rich in flavonoids) of V. officinalis aerial parts was subjected to bioactivity-guided fractionation to isolate main chemical constituents responsible for antianxiety activity. The column chromatography of ethyl acetate fraction yielded five fractions F1-F5, which were screened for antianxiety activity employing elevated plus maze model. F2 produced significant antianxiety activity at the dose of 16 mg/kg, p.o., upon acute administration in mice. F2 was also subjected to column chromatography, and yielded four sub fractions (F2.1 F2.4). Upon acute oral administration of sub fractions, antianxiety activity was evaluated. Only F2.3 exhibited significant antianxiety activity, which was statistically equivalent to the standard drug (Diazepam, 2 mg/kg, p.o.). Three compounds (VO-1, VO-2 and VO-3) were isolated during the process of fractionation of bioactive fraction and sub fraction. After antianxiety evaluation of compounds, it was observed that VO-2 and VO-3 exhibit antianxiety activity.

Lafoensia pacari A. St.-Hil. can be found from Amapá to Rio Grande do Sul states, and also in Paraguay and Bolivia. It is popularly known as pacari or mangava-brava and is used to promote weight loss, as an anti-thermal or tonic, to treat gastritis, ulcers, scarring, itching, discouragement, and cancer. In the open field tests, the hydroalcoholic extract from L. pacari stem bark (HEP) decreased the number of rearings, number of invaded squares, and increased immobility time compared to control animals. In the pentobarbital-induced sleep time test, HEP decreased latency time to sleep and increased sleeping time. In the rota-rod test, no changes in the studied parameters were observed. In the elevated plus maze, HEP increased the percentage time and percentage entries in the open arms, indicating that this extract exerts an anxiolytic-like activity.

Background: The antidepressant and anxiolytic effects of selenium (Se) have been proven in many studies. This work was aimed at confirming these activities of its inorganic form—sodium selenite—and examining the possible synergy of action with antidepressants and diazepam. Methods: The antidepressant- and anxiolytic-like activity of Se was assessed using forced swim tests (FSTs) and elevated plus-maze test (EPMs). Spontaneous locomotor activity was measured using photoresistor actimeters. The experiments were conducted on male Albino Swiss mice. Results: Sodium selenite (0.5 mg/kg) reduced the immobility time in the FSTs and extended time spent in the open arms of EPMs without affecting locomotor activity The combined administration of Se at an ineffective dose (0.25 mg/kg) together with imipramine (15 mg/kg), fluoxetine (5 mg/kg), tianeptine (10 mg/kg), but not with reboxetine (2.5 mg/kg), resulted in a reduction of immobility time in FSTs, and with a threshold dose of diazepam (0.25 mg/kg) led to the prolongation of time spent in the open arms of the EPM. Moreover, the antidepressant-like effect of Se (0.5 mg/kg) was significantly reduced by pretreatment with p-chlorophenylalanine (100 mg/kg). Conclusions: The results may indicate the participation of serotonergic transmission to antidepressant action of Se and GABA-ergic transmission to its anxiolytic effects.

ABSTRACT Allophylus edulis and Cupania vernalis (Sapindaceae) are Brazilian native trees used as medicinal plants for the treatment of respiratory, digestive, circulatory, and skin diseases. Ubiquitously distributed in the Brazilian territory, these species are indicated for mixed plantations aimed at the recovery of degraded ecosystems. In this study, the total phenolic content (TPC) and total flavonoid content (TFC), and the antioxidant activity of extracts and fractions obtained from A. edulis and C. vernalis leaves were assessed. The TPC and TFC was determined spectrophotometrically. Antioxidant activity was evaluated through radical scavenging activity of 2,2-diphenyl-1-picrylhydrazyl (DPPH). The extracts were obtained by two methods: maceration (method 1) and Soxhlet (method 2). Solvents of increasing polarity (hexane, dichloromethane, ethyl acetate, and n-butanol) were used to obtained the fractions. The results showed that the ethyl acetate fraction from A. edulis, obtained from the maceration method, had the highest TPC (442.0 ± 18.2 mg GAE g-1) and TFC (58.1 ± 0.4 mg RUE g-1), and antioxidant activity (EC50 = 43.6 ± 2.6 µg mL-1). By C. vernalis, superior results were obtained with the n-butanol fraction (TPC = 126.1 ± 5.8 mg GAE g-1, TFC = 37.7 ± 0.6 mg RUE g-1). The highest antioxidant potential was found in the crude hydroalcoholic extract (EC50 = 816.1 ± 50.9 µg mL-1) and butanol fraction (1,156.4 ± 3.8 µg mL-1). The results of this study show that the fractions obtained by maceration and liquid-liquid partition with more polar solvents (ethyl acetate and n-butanol) are the richest in TPC and TFC, and presented the greater antioxidant activity. Comparing the two plants, A. edulis showed the best results, with a high content of TPC, TFC, and antioxidant potential, and therefore may be used to treat diseases related to oxidative stress.
