Background: The oleo-gum-resin of Commiphora myrrha (myrrh) has a long history of therapeutic use in traditional medicine. The aim of this study was to seek for the scientific evidence to determine whether the ethanolic extract of myrrh (EEM) has any beneficial effects on Streptozotocin (STZ) induced testicular impairments, and explore the possible mechanisms underlying such actions. Methods: Forty-eight severe and complicated diabetic rats (fasting blood glucose above 350 mg/dL), randomly were divided into six equal groups (n=8). Besides, eight healthy rats allocated as a normal control group and only treated with vehicle solution. The diabetic animals orally received the extract (100, 200, 300, and 500 mg/kg), metformin (500 mg/kg) or vehicle solution for 28 days. As a final point, plasma glucose and insulin, circulatory sex hormones, sperm parameters including sperm concentration, motility and viability and also testicular malondialdehyde (MDA) levels were assessed. Furthermore, quantitative histological evaluation of seminiferous tubules area and determination of germinal cells apoptosis were performed. Results: None of the studied doses of EEM showed anti-diabetic effects. However, the extract mainly at the maximum dose (500 mg/kg) exhibited beneficial effects on reproductive impairments. The EEM treated rats mainly at 500 mg/kg had significantly higher sperm concentration, sperm motility, sperm viability, sex hormones and lower testicular MDA and germ cell apoptosis index than untreated diabetic rats. Conclusion: These results indicated that EEM may have beneficial effects against reproductive dysfunction induced by diabetes. The mechanisms behind the effects might be associated with the EEM sex hormone booster potential, antioxidant and anti-apoptotic properties.

The effects of a polyherbal mixture containingAllium sativum,Cinnamomum zeylanicum,Citrullus colocynthis,Juglans regia,Nigella sativa,Olea europaea,Punica granatum,Salvia officinalis,Teucrium polium,Trigonella foenum,Urtica dioica, andVaccinium arctostaphyloswere tested on biochemical parameters in diabetic rats. The animals were randomized into three groups: (1) normal control, (2) diabetic control, and (3) diabetic rats which received diet containing 15% (w/w) of this mixture for 4 weeks. Diabetes was induced by intraperitoneal injection of streptozotocin (55 mg/kg). At the end of experiment, the mixture had no significant effect on serum hepatic enzymes, aspartate aminotransferase, and alanine aminotransferase activities. However, the level of fasting blood glucose, water intake, and urine output in treated group was lower than that in diabetic control rats (P<0.01). Also, the levels of triglyceride and total cholesterol in polyherbal mixture treated rats were significantly lower than those in diabetic control group (P<0.05). Our results demonstrated that this polyherbal mixture has beneficial effects on blood glucose and lipid profile and it has the potential to be used as a dietary supplement for the management of diabetes.

AbstractObjectivesThe present study was conducted to examine antidiabetic effects of Artemisia absinthium ethanolic extract [A. absinthium] and to investigate its effects on oxidative stress markers and the expression of TLR4, S100A4, Bax and Bcl-2 genes in the kidney of STZ-induced diabetic rats.MethodsThirty six rats (weight 200–250 g) were randomly divided into diabetes and control groups. Induction of diabetes was performed using STZ (55 mg/kg.bw). Biochemical parameters and oxidative stress markers (SOD and MDA) were measured using spectrophotometry after 60 days of treatment. The expression of TLR4, S100A4, Bax and Bcl-2 were analyzed by real-time PCR. One-way analysis of variance (ANOVA) and Bonferroni post hoc test were used to compare the data.ResultsDiabetes significantly impairs the serum fasting blood glucose (FBG), lipid profile, urea, creatinine and albumin. At the end of treatment with A. absinthium extract, these parameters were close to the normal range. The results showed that the A. absinthium extract significantly decreased the kidney expression of TLR4, S100A4, Bax and increased the expression of Bcl-2 and improved oxidative stress markers (SOD and MDA) in the kidney tissues of treated rats. Also, all of these beneficial effects of the A. absinthium were dose-dependent.ConclusionsThe extract of A. absinthium possesses antidiabetic effects. A. absinthium decreased the expression of TLR4, S100A4, Bax and increased the expression of Bcl-2 and improved oxidative stress. Therefore, this herbal extract can be used as an adjuvant treatment for diabetic complications.

The purpose of this project is to study the anti-diabetic effect of on a diabetic rat model. A total of Twenty male Sprague rats were used and it randomly distributed into four groups which are Group I: , Group II: negative control, Group III: and Group IV: and . In diabetic model were induced with via injection at the dosage of 65mg/kg. and FBG (Fasting Blood Glucose) level of diabetic rats were assessed every three days. Blood was collected via cardiac puncture at day 21 after the induction of treatment. Insulin level of the rats was assessed with the Mercodia Rat Insulin ELISA kit. FBG level of group I (12.16 ±3.96, p&lt;0.05) and group IV (11.34 ±3.67, p&lt;0.05) were significantly decreased. Meanwhile, the for all rats did not show any significant increase. However, the insulin level was escalated in group IV (0.74+0.25, p&lt;0.05) significantly. The present study shows that the and the combination of and lowered blood glucose level and enhanced insulin secretion.

Background: Ficus glomerata (F. glomerata) Linn. Family Moraceace is a large tree found all over India including outer Himalayan ranges, Punjab, Chota Nagpur, Bihar, Orissa, West Bengal, Rajasthan, Deccan and also as a common plant in South India. It is planted around the home and temples. It is cultivated throughout the year, distributed in evergreen forests and moist localities. Objective: The Ethanolic Extract of roots of F. Glomerata (EEFG) belonging to the family Moraceace, was investigated for its antidiabetic activity using alloxan induced diabetic rats. Methods: Thirty rats were divided into 5 groups having 6 rats in each group. The alloxan was administered to the rats of all groups except normal control group through intraperitoneal route at a concentration of 140mg/kg body weight. A dose of 100mg/kg and 200 mg/kg body weight of EEFG was administered to alloxan induced diabetic rats. The administration of the extract was lasted for 11 days. Effectiveness of the extract on glucose, cholesterol, triglycerides, and high density lipoprotein and protein concentrations was analyzed. Results: Significant (p<0.05) reduction in the levels of glucose, cholesterol, triglyceride of the diabetic rats was observed after treatment with ethanolic extract. After subjecting to oral glucose tolerance test EEFG also showed significant improvement in glucose tolerance. Conclusion: F. glomerata root ethanolic extract showed that it possesses antidiabetic effect and can be found useful for the management of diabetes mellitus.

Introduction: Cardiac apoptosis is one of the most important cardiovascular complications of diabetes. We aimed to investigate the changes of caspase-8, Bcl-2, and N-terminal pro B-type natriuretic peptide (NT-proBNP) in cardiac tissue after 4 weeks of aerobic exercise in male rats with diabetes. Methods: Forty adult male rats were randomly allocated to healthy control, diabetes, control + exercise and exercise + diabetes groups. Diabetes was induced by intraperitoneal injection of streptozotocin (STZ) solution (55 mg/kg). Two weeks after injection, fasting blood glucose levels were measured. After the induction of diabetes, the exercise program was performed for 4 weeks (5 sessions per week) at a speed of 15 to 18 m/min for 25 to 44 minutes. Forty-eight hours after the last training session, the subjects were anesthetized and the heart muscle was removed. Caspase-8, Bcl-2 and NT-proBNP levels were measured by ELISA method. Results: The induction of diabetes in the control group resulted in a significant increase in caspase-8, and NT-proBNP levels while an insignificant increase was observed for Bcl-2 levels (P<0.05). In non-diabetic groups, exercise caused no changes in caspase-8, NT-proBNP and Bcl-2 (P<0.05). Exercise in diabetic groups significantly decreased NT-proBNP while no changes were observed in caspase-8 and Bcl-2 (P<0.05). Conclusion: Our findings showed that diabetes increases the pro-apoptotic and anti-apoptotic agent. In addition, 4 weeks of regular aerobic exercises can be used as a non-pharmacological strategy to reduce the complications of apoptosis in diabetic cardiomyocytes.

 Objective: Lawsonia inermis (Linn.) leaves are one of the alternative medicines to treat diabetes mellitus in Indonesia. We investigated the blood glucose level (BGL) of the L. inermis (Linn.) leaves ethanolic extract (LLEE) leaves and evaluated the histopathological alterations in diabetic rats.Methods: This study was an experimental study with posttest - only control group design. Alloxan (120 mg/kg, intraperitoneally)-induced diabetic rats. 35 of Wistar rats (Rattus norvegicus) were divided randomly into five groups, i.e. K: Normal control, P1: Diabetic control, and P2, P3, and P4 (200 mg/kg body weight [BW], 400 mg/kg BW, and 600 mg/kg BW of LLEE, orally) for 28 days. At the end of the treatment, the rats were sacrificed to obtain the kidney for histopathological evaluation using hematoxylin and eosin technique. BGLs were conducted using a glucose meter (GlucoDR).Results: One-way ANOVA test showed that dose 400 mg/kg BW of the LLEE was related to BGL of alloxan-induced diabetic rats (p=0.000). The histopathological of kidney showed glomerular inflammation (GI), epithelial membrane lining degeneration, vascular congestion, and interstitial tubule hemorrhage at diabetic control (P1). Meanwhile, treated with 600 mg/kg BW of LLEE (P4) showed increase cellular regeneration as normal architecture of the kidney.Conclusion: The LLEE at dose 400 mg/kg BW effective decreased BGL and was able to restore the kidney destruction of alloxan-induced diabetic rats at dose 600 mg/kg BW.

Background: The exhibit, the treatment for diabetes, for the most part, involves insulin and different oral hypoglycemic drugs. In chronic treatment medications are losing viability, therefore searching of more effective and safer natural anti-diabetic agents has continued to be an important area of investigation Objective: The present study aimed to investigate the effect of ethanolic and aqueous extracts of Schleichera oleosa (S.oleosa) in diabetic rats and isolate and characterize the active compound from an effective extract. Method: The ethanolic and aqueous extracts were administered orally (200 and 400 mg/kg, for 28 days) to streptozotocin-nicotinamide-induced diabetic rats. Hypoglycemic effects, OGTT, alteration in body weight, lipid profile, biochemical parameters, hepatic enzymes and histopathological examination were assessed. Then isolate the active constituents from effective extract using column chromatography and preliminary structure features were investigated by FT-IR, 1H NMR, 13C NMR and LC/MS. Results: The extracts treated rats showed a significant reduction in the fasting blood glucose level (FBGL), total cholesterol, triglycerides, LDL, VLDL and stress markers (MDA, SGOT, SGPT) and significant increase in the concentrations of HDL and bodyweight when compared to the diabetic rat. And also the extract restored the altered level of antioxidant enzymes and glucose metabolizing hepatic key enzymes to close normal level. Histopathological evaluation of the pancreas revealed the regeneration of the β-cells in diabetic rats which was earlier necrosed by streptozotocin. In this study, an active constituent SMK/SO/01 isolated from effective ethanolic extract and spectral studies confirmed that it’s belonging to the polyphenolic category. Conclusion: These findings provide scientific evidence that SMK/SO/01 has to be a potential drug candidate for diabetes mellitus and its complication. Also, these results prove that S.oleosa leaves of ethanolic extract is an effective anti-diabetic activity which provides the scientific proof for the folklore medicine

Abstract Background Sex hormone, adiponectin and its receptors interacted in the testes. Diabetes can also interfere with this interaction. Regular exercise has some effects on the diabetes side effects. This study purpose was to investigate the effects resistance and endurance training on adiponectin gene expression and its receptors and sex hormones in those rats with diabetes.MethodsIn this experimental study, 48 male Wistar rats were divided into 6 groups by random. After performing the trainings, adiponectin gene expression and its receptors in the testis were evaluated using real time PCR, and blood serum was then used in order to assess FSH, LH and testosterone.Results The induction of diabetes mellitus STZ-NA significantly increased the serum level of fasting blood glucose, the gene expression of adiponectin and also the AdipoR 1 in the testicles of diabetic rats in comparison with healthy controls. In addition, diabetes resulted in a significant reduction in serum testosterone levels and LH in diabetic group, but it has no effect on FSH.Conclusions Resistance and endurance training decreased the blood glucose with a significant amount reduction in adiponectin levels and AdipoR 1 gene rats, and also increased the serum testosterone levels and LH in diabetic rats. Resistance and endurance training can improve expression of adiponectin and AdipoR 1 gene by increasing the serum testosterone and LH levels in type 2 diabetic rats.

The study was performed to compare the blood glucose lowering effect of Swietenia mahagoni seeds with an oral antidiabetic drug, rosiglitazone in experimentally induced diabetic rats. Twentyfour healthy Long Evans Norwegian strain of rats were included in the study and divided into four groups (A, B, C and D) comprising 6 rats each. Group A (control group) received standard rat food for 14 days. Diabetes was induced by a single intraperitoneal administration of alloxan 120mg/kg body weight in Group B, C and D. Group B was given standard food for 10 days and considered as diabetic control. Group C and D were treated with ethanolic extract of Swietenia mahagoni seeds 1000mg/kg and rosiglitazone 10mg/kg orally respectively. Administration of ethanolic extract of Swietenia mahagoni seeds in group C and rosiglitazone in group D produced a significant reduction in blood glucose level as compared to diabetic control (group B). Histological examination of pancreas showed destruction of beta cells in Islets of pancreas in group B whereas retaining of islets and few degranulations of beta cells of pancreas found in group C and group D. The observations and results of the present study provide information that ethanolic extract of Swietenia mahagoni seeds has hypoglycaemic effect in experimentally induced diabetic rats which requires further investigation. Introduction: Euphorbia antiquorum is a fleshy shrub belonging to the family Euphorbiaceae. It is widely distributed in the hotter parts of India. It is used as a substitute for the Ayurvedic drug ‘Snuhee’ whose accepted botanical source is Euphorbia neriifolia Linn. The leaf, stem, latex and root of Snuhee are used in treatment of diseases and conditions like, diabetes, coryza, and oedema. Objective: The present study is aimed to investigate the renal protective effect of  E. antiquorum alcoholic extracts of the stem   on alloxan (150mg/kg) induced diabetic Wistar rats. Methods: All animals were treated for 21 days and sacrificed on the last day. Unwanted placebo effect in case of the control group was blocked by executing appropriate vehicle treatment and same animal handling pattern. Extract and standard drug doses given orally were suspended in 0.9% normal saline. Blood samples were drawn from retro orbital sinus using Diethyl-ether anesthesia at weekly intervals for 3 weeks. Blood samples were labeled in such a way that analysis team was kept blind about groups and nature of the study design. Body weight measurement and blood sampling were done on 1st, 7th, 14th and 21st day of the study. Conclusion: 21 days daily treatment with a test drug of ethanolic extract of stems of Euphorbia antiquorum significantly reduced the elevated Blood glucose level, Urinary Albumin, urine creatinine, serum creatinine in alloxan induced diabetic rats.
