Activity of eosinophils and immunoglobulin E concentration in the peritoneal fluid of women with endometriosis

AbstractAutoinflammatory phenomena, including autoantibody production and atopy, have been regarded as associated with endometriosis. The present study investigates the activity of eosinophils and the distribution of immunoglobulin E concentrations in the peritoneal fluid of women with early endometriosis. The study group consisted of 30 patients with laparoscopically diagnosed early endometriosis. The healthy control group consisted of 18 females with no evident changes in the abdominal cavity and no endometrial foci. Concentrations of immunoglobulin E in serum and peritoneal blood were determined by enzyme immunoassay. The activity of eosinophils was estimated according to the expression of the early activation molecule CD69 by the flow cytometry method. The concentrations of immunoglobulin E in the peripheral blood and peritoneal fluid were similar in both groups. However, the count of CD69+ eosinophils was higher in the peritoneal fluid of women with endometriosis. The results indicate that activated eosinophils accumulate in the peritoneal fluid in early endometriosis and can play a significant role in the pathogenesis of the disease.

Autoimmune diseases have been implicated in the development of intrinsic asthma, however little data are available on the role of autoimmunity in the pathogenesis of asthma. The purpose of this study was to investigate circulating autoantibodies against the high-affinity receptor for immunoglobulin E, FcεRI, in patients with asthma. Seventy-eight patients with asthma and 32 healthy control subjects were included. All individuals were tested using a triple-staining flow cytometry-based basophil activation test (BAT) for the potential presence of autoantibodies against FcεRI. Of the 78 asthma patients, 29 (37.2%) had a positive BAT result, indicating that their serum was able to activate basophils, compared with only four (12.5%) of the control group, a statistically significant between-group difference. These data suggest that some asthma patients have aberrant anti-FcεRI autoantibodies, which implies that autoimmunity may be one factor involved in the pathogenesis of intrinsic asthma.

Objective: This study aims to investigate the expression and clinical significance of Th17 cells and related factors in peripheral blood of patients with Autoimmune Hepatitis (AIH). Methods: A retrospective selection of 100 patients with AIH were included as a study group, and 100 healthy volunteers in the outpatient clinic were selected as the control group. The levels of IL- 17, IL-6, IL-21 and TNF-α in peripheral blood of all subjects were detected by enzyme-linked immunosorbent assay and the frequency of Th17 cells and Treg cells was detected by flow cytometry. Results: Results showed that the study group had higher levels of serum total bilirubin (TBil), alkaline phosphatase (ALP), γ -glutamyltranspeptidase (γ-GT), immunoglobulin G (IgG), immunoglobulin M (IgM), alanine aminotransferase (ALT) and aspartate aminotransferase (AST) than the control group, as well as higher levels of IL-17, IL-6, IL-21 and TNF-α in serum. The frequency of Th17 cells in peripheral blood was higher in the study group, while the frequency of Treg cells was lower. Also, serum IL-17, TNF-α levels and Th17 cells frequency were positively correlated with ALT and AST, whereas Treg cells frequency were negatively correlated with ALT and AST levels. Conclusion: Our finding demonstrates that Th17 cell frequency and their related factors IL-17 and TNF-α, are associated with liver damage, which might be used to monitor AIH disease severity.

AbstractInterferon-gamma (IFN-γ) is a cytokine involved in the pathogenesis of Takayasu’s arteritis (TAK). However, the source of IFN-γ in TAK patients is not fully clear. We aimed to investigate the source of IFN-γ in TAK. 60 TAK patients and 35 health controls were enrolled. The lymphocyte subsets of peripheral blood were detected by flow cytometry, cytokines were detected by Bio-plex. The correlation among lymphocyte subsets, cytokines and disease activity indexes was analyzed by person correlation. The level of serum IFN-γ in TAK patients was significantly increased (P < 0.05). The percentage of CD3+IFN-γ+ cells in peripheral blood CD3+ cells was significantly higher in TAK patients than that of healthy control group (P = 0.002). A higher proportion of CD3+CD8+IFN-γ+ cells/CD3+IFN-γ+ cells (40.23 ± 11.98% vs 35.12 ± 11.51%, P = 0.049), and a significantly lower CD3+CD4+IFN-γ+/ CD3+CD8+IFN-γ+ ratio (1.34 ± 0.62% vs 1.80 ± 1.33%, P = 0.027) were showed in the TAK group than that of control group. The CD3+CD8+IFN-γ+/CD3+IFN-γ+ ratio was positively correlated with CD3+IFN-γ+cells/ CD3+cells ratio (r = 0.430, P = 0.001), serum IFN-γ level (r = 0.318, P = 0.040) and IL-17 level (r = 0.326, P = 0.031). It was negatively correlated with CD3+CD4+IFN-γ+/CD3+IFN-γ+ ratio (r = − 0.845, P < 0.001). IFN-γ secreted by CD3+CD8 + T cells is an important source of serum IFN-γ in TAK patients.

Purpose – to improve the results of surgical treatment of patients with intra-abdominal infiltrates and abscesses through the introduction of the latest imaging methods and surgical technologies. Materials and methods. In the clinic of the Department of Surgical Diseases No 1, on the basis of the Surgery Center of the Kyiv City Clinical Hospital No. 1 from 2006 to 2019 218 patients with primary and secondary intra-abdominal infiltrates, abscesses and fluid formations were treated. The patients’ age ranged from 16 to 85 years. There were 107 (49.08%) male patients, 111 (50.92%) female patients. Depending on the time of hospitalization (by years), the patients were divided into two groups: the control group (CG) (2006–2012) 117 (53.67%) patients and the study group (SG) (2013–2019) 101 (46.33%) patients. The SG used the latest imaging technologies and improved methods of surgical treatment. Results. The patients were divided into two groups: primary in 191 (87.61%) and secondary postoperative infiltrates and abscesses in 27 (12.39%). The causes of primary infiltrates and abscesses were: complicated forms of appendicitis in 74 (33.94%), perforated stomach and duodenal ulcer in 48 (22.02%), complicated forms of cholecystitis in 69 (31.65%). Postoperative infiltrates and abscesses were observed in 27 (12.39%) patients who underwent urgent surgery: adgeolysis of adhesive ileus in 14 (6.42%) and complicated hernias of various localization in 13 (5.97%). Postoperative complications were diagnosed in 43 (19.72%) patients, of whom 34 (15.59%) from the surgical wound and 29 (15.18%) of the abdominal cavity, who required relaparotomy or laparoscopy, with destructive appendicitis in 10 (13.51%), perforated gastric ulcer and 12 duodenal ulcer in 6 (12.5%), destructive cholecystitis in 9 (13.04%), adhesive intestinal obstruction in 13 (19.12%) and with strangulated and complicated hernias in 14 (17.28%) of the examined patients. During relaparotomy, incompetence of the intestinal wall and intestinal sutures was established in 11 out of 32 patients, an ileostomy was imposed in 7, and cecostomy in 1 patient. Actually, in the control group, 8 (6.84%) patients died on the background of ongoing peritonitis, thrombosis of mesenteric vessels and multiple organ failure and concomitant ailments and in the study group 4 (3.96%) patients died. Conclusions. Surgical treatment is individualized depending on the disease, so with destructive appendicitis from 74 (38.74%) laparotomic in 42 (21.99%), laparoscopic in 32 (16.75%), and in 12 (6.28%) with conversion; perforated gastric ulcer and duodenal ulcer in 48 (25.13%) open laparotomy; with cholecystitis of 69 (36.13%) patients, 48 (25.13%) had laparotomy and 21 (11.00%) had laparoscopic examination. The use of the latest imaging and treatment technologies: Doppler ultrasonography, hydrojet scalpel and laparoscopy in 64 (33.51%), allowed to have better near and long-term results and to reduce postoperative mortality from 6.84% to 3.96%, with an average of 5.5%. The research was carried out in accordance with the principles of the Helsinki declaration. The study protocol was approved by the Local Ethics Committee of these Institutes. The informed consent of the patient was obtained for conducting the studies. The authors declare no conflicts of interests. The purpose of this study was to focus on the underlying relationship between the hyperactivity for the peripheral monocytes and heat stroke by investigating the inflammatory oxidative activity of and the expression of superficial molecules. Peripheral blood samples were collected from 10 healthy adult volunteers. Human blood monocytes were isolated by density gradient centrifugation and sequent adherent culture. The objectives were divided into four groups: 43°C heat stress combined with lipopolysaccharide (LPS) group, 43°C heat stress group, LPS group, and control group. There were 10 cases in each group. An enzyme-linked immunosorbent assay (ELISA) test was used to measure the concentrations of supernatant inflammatory mediators (tumor necrosis factor alpha (TNF-α), interleukin-1β (IL-1β) and interleukin-10 (IL-10)). After loaded by 2,7-Dichlorodi-hydrofluorescein-diacetate (DCFHDA) fluorescent probe, intracellular reactive oxygen species (ROS) levels were determined by a flow cytometry. After fluorescent microspheres incubation, the phagocytosis of monocytes was observed under a fluorescent microscope. Respectively, the flow cytometry and Western blot were used to evaluate the level of triggering receptor expressed on myeloid cells-1 (TREM-1) and Toll-like receptor-4 (TLR-4) on the monocytes. Furthermore, the mRNA expression of TREM-1 and TLR-4 was detected by real-time polymerase chain reaction (RT-PCR). The heat stress combined with LPS stimulation promoted the peripheral monocytes to produce inflammatory mediators (TNF-α, IL-1β, and IL-10) and release ROS. Otherwise, such complex strike significantly suppressed the phagocytic activity of monocytes in peripheral blood. Moreover, the expression of TREM-1, TLR-4 and CD86 was measured by the flow cytometry on peripheral monocytes which were respectively promoted by the union of heat stress and LPS. The results of Western blot and RT-PCR demonstrated the similar kinetics on these superficial molecules (TREM-1, TLR-4, and CD86) stimulated by the combination of heat stress and LPS. The underlying mechanism of the dysfunction for the peripheral monocytes may be related to the abnormal expression of superficial molecules TREM-1, TLR-4, and CD86 on the monocytes induced by heat stress and LPS.

Abstract Background: Endocannabinoids take part in physiology of neural and immune system. Last data showed that these compounds and their receptors play an important role in proliferation and apoptosis of different neoplastic cells. Cannabinoids were shown to increase the apoptosis in human neoplastic cells through a number of mechanisms including vanilloid receptors (Sanchez et al. 1998, Maccarone et al. 2000). The vanilloid receptor family of cation channels includes the capsaicin-sensitive, proton- and heat-activated vanilloid receptor type I (TRPV1). Furthermore Saunders et al. (2006) showed that TRPV1 are expressed on normal lymphocytes in human peripheral blood. Aims: The aim of our study was the assessment of receptor TRPV1 mRNA expression in the lymphocytes B derived from patients with newly diagnosed chronic lymphocytic leukaemia. Material and methods: The study group contains newly diagnosed, untreated adult patients with B-cell chronic lymphocytic leukaemia; 11 males and 10 females, aged from 46 to 74. The patients were in A-C stage according to Binet. We used 13 samples from healthy donors as a control group. The isolation of mononuclear cells from peripheral blood was carried out with the use of density centrifugation method. The resolution of mononuclear cell population to lymphocytes B subpopulation was done with negative isolation method utilizing magnetic microballs (Dynal). Total RNA was isolated from B-lymphatic cells of which 10ng was used in each reaction. Quantitative RT-PCR was carried out with the use of Light Cycler (Roche) and respective commercial kits. The nucleotides sequence of the studied receptor and parameters of the amplification process were based on method previously described by Qiao et al (2003). We used house keeping gene hexose-6-phosphate dehydrogenase (glucose 1-dehydrogenase) (G6PDH) as a reference gene. Amplification product was sequenced by ABI PRISM (Applied Biosystem). All results are presented as a mean concentration ratio of TRPV1 to G6PDH mRNA ± standard error. Statistical analysis was performed using Shapiro-Wilk, non parametric U Mann-Whitney Tests. Results: We found that concentration ratio of studied transcript was significantly lower in the study group in comparison to the control group (0,048± 0,012 vs. 106,836± 40,215 respectively. We found no differences between the subgroups irrespectively of gender, age, stage of the disease and some prognostic factors (LDH, lymphocyte doubling time, β2-microglobulin). Conclusions: The results confirm the existence of vanilloid receptors on the lymphocytes B of healthy individuals and for the first time show presence of this type of receptors in CLL group. The lower level of TRPV1 transcripts in the group of patient may suggests their potential role in the process of leukemic transformation. Thus the effect of endocannabinoids through this receptor may be altered in CLL. However, further studies are required to elucidate the nature of relationship between this type of receptor and neoplastic development of CLL.

Morphometry and flow cytometry for intraepithelial lymphocyte phenotyping were used to determine the changes in duodenal mucosae after administration of low doses of deoxynivalenol in chickens. Moreover, functions of phagocytes and immunocompetent cells in peripheral blood were evaluated by flow cytometry. In total, sixty chickens of Ross hybrid broilers 308 were used in this experiment. Two experimental groups of 20 birds were continually fed for 14 days a diet containing deoxynivalenol at a dose of 1 and 3 mg·kg-1; 20 birds of the control group were fed uncontaminated diet. Morphometry showed only tendency to decrease the height of villi and surface area of duodenal mucosae in chickens fed the diet supplemented with 3 mg·kg-1 deoxynivalenol. Phenotyping of intraepithelial lymphocytes showed a decrease of CD45+ (P < 0.034) in duodenum of birds fed diets supplemented with deoxynivalenol. Examination of white blood cells showed a decrease of monocytes (P < 0.020) in chickens fed 3 mg·kg-1 of deoxynivalenol. Both experimental groups revealed higher metabolic burst of peripheral blood heterophils (P < 0.001). Phenotyping of immunocompetent cells showed an increase (P < 0.003) of CD3+ and a decrease (P < 0.001) of MHC II+ cells in peripheral blood of chickens fed with 3 mg·kg-1 dose of deoxynivalenol. The experimental feeding of chickens with deoxynivalenol resulted in immunomodulation of immunocompetent cells in duodenum and blood with mild atrophy of intestinal villi, mainly after the feeding of the dose of 3 mg·kg-1. We proved that even low doses of deoxynivalenol can cause changes in haemathological, immunological and morphological profiles already during two weeks, and lead to the activation of compensatory-adaptive mechanisms with unfavourable impact on health and performance of birds.

Objective. To investigate the Th17/Treg immune balance in the ulcerative colitis (UC) patients with two Chinese syndrome: dampness-heat in large intestine (DHLI) and spleen and kidney Yang deficiency (SKYD).Methods. Ninety UC patients (45 were diagnosed with DHLI and 45 with SKYD syndrome) and 23 healthy people were recruited. The serumIL-17 and TGF-β1 levels of these participants were measured with ELISA; the expression of IL-17 and TGF-β1 in colonic mucosa tissue was determined with immunohistochemistry and the percentage of Th17 and Treg in peripheral blood with flow cytometry.Results. The levels of IL-17 and Th17 were significantly higher in both DHLI and SKYD groups than in healthy control group and higher in DHLI than in SKYD group (P<0.05). The levels of TGF-β1 and Treg were significantly lower in the two UC patients groups than in healthy control group; and lower in SKYD group than in DHLI group (P<0.05).Conclusions. UC with DHLI syndrome could be characterized by the elevation of Th17 and IL-17 levels, which indicated an accentuation of inflammatory reaction; UC with SKYD syndrome could be characterized by the reduction of serum Treg and TGF-β1 levels, which represented a depression of immune tolerance.

AbstractObjective:This study aimed to determine whether there was any relationship between tinnitus and mean platelet volume.Methods:This retrospective study was conducted between January 2013 and January 2014 in Ankara Atatürk Hospital and Ondokuz Mayıs University Hospital, Turkey, on a study group of 86 patients with tinnitus and a control group of 84 healthy subjects. Mean platelet volume was recorded and comparisons were made between the two groups.Results:Mean (± standard deviation) platelet volume was 7.67 ± 0.83 μm3 in the study group and 7.28 ± 0.56 μm3 in the control group. There was a statistically significant difference in mean platelet volume between the tinnitus patients and the healthy subjects (p < 0.05).Conclusion:The clinical findings indicated that tinnitus patients had a higher mean platelet volume than the healthy control subjects; however, the pathophysiological mechanism remains unclear.

Purpose: To study the combined effects of tenofovir and interferon α1b on viral load and peripheral blood regulatory T cell concentrations of chronic hepatitis B (CHB) subjects. Methods: Patients with chronic hepatitis B (86 cases) were randomly assigned to two groups: control group and study group. In control subjects, tenofovir was given orally (300 mg/kg bwt/day). In addition to tenofovir, the study group received interferon α1b injection intramuscularly at a dose of 50 μg/kg thrice a week. Liver function, serum hepatitis B viral (HBV) load, and serum levels of peripheral blood regulatory T-lymphocytes were determined. Clinical effectiveness and adverse reactions in both groups were also assessed. Results: After treatment, total effectiveness was higher in the study group (86.04 %) than in control patients (62.79 %) (p < 0.05). Serum aspartate transaminase (AST), alanine aminotransferase (ALT) and total bilirubin (TBIL) significantly decreased in the study group, relative to control, but HBV DNAnegative, HbeAg-negative and HbsAg-negative cells were markedly higher in patients in the study group (p < 0.05). Moreover, there were higher CD4+ T and CD8+ T counts, and CD4+ T/CD8+ T ratio in study subjects than in control subjects (p < 0.05). Conclusion: The combination of tenofovir with interferon α1b effectively improves liver functions in patients with CHB, reduces viral load, and exerts anti-HBV effect by regulating the levels of peripheral blood T-lymphocytes.
