Carotenoids are a widespread group of fat-soluble pigments, and their major nutritional importance comes from their pro-vitamin A activity and their antioxidant capacity. In this study, two different pumpkin cultivars (Cucurbita maxima, also named `Delica’ and Cucurbita moschata, also known as `Violina’) from the southern Po Delta area were investigated in terms of carotenoid content and the influence of food processing on compositional changes and carotenoid bioaccessibility. Quali- and quantitative determination of carotenoids in sample extracts were performed on a C30 column by means of an online coupled HPLC-UV/Vis-APCI-MS/MS technique. The identification of separated compounds was tentatively achieved by merging (i) chromatographic data, (ii) UV-Vis spectra, and (iii) MS/MS fragmentation spectra. The chromatographic profiles for the two cultivars showed qualitative differences. Two major carotenoids were considered for quantification purposes and further investigations: lutein and β -carotene. Quantification of target carotenoids was performed with external calibration through analytical standards. The concentration of lutein and β -carotene was higher in C. maxima than in the other variety, C. moschata. Carotenoids are susceptible to degradation (isomerization and oxidation) during food processing (i.e., cooking), and the concentration of lutein and β -carotene were monitored in oven-cooked and steam-cooked pumpkins. The steam-cooking process was superior in terms of limiting carotenoid loss. A complete functional profile of pumpkins as a source of carotenoids was gained with the evaluation of their in vitro bioaccessibility and their bioavailability after intake during human digestion. Bioaccessibility of lutein and β -carotene were estimated by an in vitro static digestion model that involved salivary, gastric, and duodenal phases. Bioaccessibility values progressively increased from the salivary to the duodenal phase for both pumpkin varieties and cooking methods. Bioaccessibility of lutein was always lower than β -carotene for both cultivars and for both cooking methods. Bioaccessibility values for lutein and β -carotene changed from 1.93% to 2.34% vs. 4.94% and 8.83% in the salivary phase, from 2.7% to 4.63% vs. 7.83% and 15.60% in the gastric phase, and from 10.04% to 13.42% vs. 25.81% and 35.32% in the duodenal phase. For both target compounds, bioaccessibility in the duodenal phase was more than twice the gastric values, and it underlined that the type of cooking did not influence release from the initial matrix.

Cucurbita species are delicious, nutritious, and delightful products. Cucurbita seeds remain in large quantities as a waste product that could be valorized since they are excellent sources of oil. The aim of this study was to compare the seed oil of two Cucurbita species (Cucurbita pepo and Cucurbita moschata) harvested in Bejaia (Algeria). The oil quality was evaluated by the determination of some physicochemical parameters, and the content of phenolic compounds. The antiradical capacity of the antioxidants present in the oils was also assessed using two methods. The oil yield was 42.85% and 40.47% from the seeds of Cucurbita pepo and Cucurbita moschata, respectively. The determined physicochemical parameters were close to those defined by the international standards. The phenolic contents of the methanolic extracts of both oils were 5.53 and 4.45 mg GAE/100 g for Cucurbita moschata and Cucurbita pepo, respectively. The best anti-DPPH power was attributed to the oil of Cucurbita moschata (44.7%), while the methanolic extract of the seed oil of Cucurbita pepo showed the highest percentage (41.02%) of the ABTS•+ radical inhibition. By this study we confirmed that the Cucurbita seeds oil are highly nutritious and offer some medicinal benefits.

Cucurbita maxima x Cucurbita moschata rootstock are used to prevent infection with Fusarium oxysporum f. sp. niveum in watermelon production; however, this rootstock is not effective against nematode attack. Because of their vigor, the grafted plants can be planted at lower plant densities than the non-grafted plants. The tolerance to Fusarium oxysporum f. sp. niveum and Meloidogyne incognita was assessed in watermelon plants grafted onto a hybrid of Citrullus lanatus cv Robusta or the Cucurbita maxima x Cucurbita moschata cv Super Shintoza rootstocks. The densities of plants were 2083 and 4166 plants ha-1. Non-grafted watermelons were the controls. The Crunchy Red and Sangría watermelon cultivars were used as the scions, it the latter as a pollinator. The experiments were performed for two production cycles in soils infested with Fusarium oxysporum f. sp. niveum and Meloidogyne incognita. The incidence of Fusarium oxysporum f. sp. niveum was significantly greater in the non-grafted than in the grafted plants. The grafted plants presented similar resistance to Fusarium regardless of the rootstock. The root-knot galling index for Meloidogyne incognita was significantly lower in plants grafted onto Citrullus lanatus cv Robusta than onto the other rootstock. The yields of plants grafted onto Citrullus lanatus cv Robusta grown at both plant densities were significantly higher than in the other treatments.

SummaryRates of transposition and excision of the Drosophila melanogaster retrotransposon elements mdg3, 297, Doc, roo and copia were estimated directly, by in situ hybridization analysis of their cytological insertion sites in 31 replicates of a highly inbred line that had accumulated spontaneous mutations for approximately 160generations. Estimated transposition rates of Doc, roo and copia were, respectively, 4·2 × 10−5, 3·1 × 10−3 and 1·3 − 10−3; no transpositions of 297 nor mdg3 were observed. Rates of transposition of copia varied significantly among sublines. Excisions were only observed for roo elements, at a rate of 9·0 × 10−6 per element per generation. Copy number averaged over these element families increased 5·9 %; therefore, in these lines the magnitude of the forces opposing transposable element multiplication were weaker than transposition rates.

Cucurbita species ( Cucurbita moschata and Cucurbita maxima) are rich sources of nutrients, especially carotenoids and carbohydrates. Metabolites in pulps of C. moschata and C. maxima lines were examined by high-performance liquid chromatography and gas chromatography–mass spectrometry. Our results revealed that glucose and sucrose were the dominant sugars in C. maxima and C. moschata, respectively. A correlation was found between Brix percentage and sucrose levels as compared with glucose and fructose. A greater amount of myo-inositol had accumulated in C. moschata lines compared with C. maxima. Conversely, total carotenoids and antioxidant activity were found to be higher in C. maxima lines than in C. moschata. A strong correlation of glucose, fructose, and sucrose with β-carotene and violaxanthin revealed that it will be difficult to develop pumpkin cultivars with both high inositol and carotenoid levels. In conclusion, the composition of carbohydrates and carotenoids was more diverse in C. moschata lines than in C. maxima lines. Our results will contribute to a better understanding of metabolite changes in the fruits of these as well as other pumpkin species.
