In vitro experiments have shown that fermentation of carbohydrates prevents accumulation of nitrogen in the colon. Variable results have been obtained on modulation of dietary intakes in vivo. Lactose[15N,15N]-labeled ureide has been proposed as a tool to study colonic nitrogen metabolism. However, on oral administration of the marker, different urinary excretion patterns of the 15N label have been found. In this study, 50 mg lactose[15N,15N]ureide was directly instilled in the colon through an orocecal tube to investigate the colonic handling of this molecule in a direct way. In basal conditions, 42% (range, 37–48%) of labeled nitrogen administered as lactose[15N,15N]ureide was retrieved in urine after 72 h. A substantial variability in total urinary excretion of the label was found, but the urinary excretion pattern of the label was similar in all volunteers. When inulin, a fermentable carbohydrate, was administered together with the labeled marker, a significant decrease in urinary excretion of 15N after 72 h was found, to 29% (range, 23–34%). The effect of a smaller dose of inulin (250 mg) on colonic handling of lactose[15N,15N]ureide (50 mg), was investigated in another group of volunteers, and this time, fecal excretion of the marker was also evaluated. The results seem to indicate that fermentation of inulin causes an increased fecal excretion of the marker, thereby reducing urinary excretion but not retention in the human nitrogen pool. This instillation study shows that lactose[15N,15N]ureide is a tool with good properties to investigate the effect of different types of carbohydrates on nitrogen metabolism in the proximal colon in vivo.

Phalaenopsis merupakan salah satu anggrek yang terkenal karena keindahan bunganya dengan variasi warna yang tinggi dan periode pembungaan yang lama. Salah satu masalah dalam budidaya anggrek adalah Penyakit Busuk Lunak (PBL) yang disebabkan oleh bakteri Dickeya dadantii. Induksi resistensi dengan Asam Salisilat (AS) merupakan metode alternatif dalam mengendalikan PBL pada Phalaenopsis. Penelitian ini dilaksanakan pada bulan November 2018 sampai Maret 2019, di Laboratorium Kultur Jaringan Tanaman, Departemen Agronomi dan Hortikultura IPB. Penelitian terdiri dari dua percobaan. Percobaan 1 bertujuan untuk mengevaluasi keragaman respon planlet in vitro asal biji hasil penyerbukan sendiri P. amabilis terhadap D. dadanti dengan inokulasi pada daun terpisah dan menentukan tingkat ketahanannya. Sebanyak 7 dari 158 planlet yang diuji, memiliki respon agak tahan terhadap D. dadantii, sedangkan sisanya rentan atau sangat rentan. Percobaan 2 bertujuan untuk membandingkan respon bibit P. amabilis terhadap D. dadantii pada 1 hari setelah perlakuan asam salisilat (AS) pada konsentrasi 0, 15, 30, 45, 60, 75, dan 90 ppm menggunakan metode inokulasi daun pada bibit yang masih menempel di tanaman. Percobaan 2 menggunakan rancangan acak kelompok lengkap teracak (RKLT). Hasil penelitian menunjukkan perlakuan AS 75 ppm paling baik dalam menginduksi ketahanan bibit P. amabilis terhadap patogen penyebab busuk lunak D. dadantii. Tanaman pada perlakuan tersebut menghasilkan nilai CFU dan AUPDC lebih rendah dan aktivitas peroksidase lebih tinggi dibanding perlakuan lainnya. Nitrogen metabolism of Mycobacterium tuberculosis (Mtb) is crucial for the survival and virulence of this pathogen inside host macrophages but little is known about the nitrogen sources acquired from the host or their route of assimilation. Here we performed a systems-based analysis of nitrogen metabolism in intracellullar Mtb and developed 15N-Flux Spectral Ratio Analysis (FSRA) to probe the metabolic cross-talk between the host cell and Mtb. We demonstrate that intracellular Mtb acquires nitrogen from multiple amino acids in the macrophage including glutamate, glutamine, aspartate, alanine, glycine and valine, with glutamine being the predominant nitrogen donor. Each nitrogen source is uniquely assimilated into specific intracellular pools indicating compartmentalised metabolism. This was not observed for in vitro-grown Mtb indicating that there is a switch in nitrogen metabolism when the pathogen enters the intracellular environment. These results provide clues about the potential metabolic targets for development of innovative anti-tuberculosis therapies.
