Transcriptomic information provides fundamental insights into biological processes and can be used to determine gene expression in cell, tissue, or organism under specific physiological conditions, or in response to any environmental perturbation. Extraction of high quality RNA is a challenging step mainly in non-traditional organisms, and protocols for preservation and isolation need to be adjusted in many cases. In the present work, we aimed to develop a protocol for preservation and isolation of high-quality and quantity of RNA from blood and liver tissues of Caiman latirostris. Three preservation methods were tested: 1) flash freezing (LN2) and storage at –80°C; 2) RNAlater® conservation with progressive cooling up to –80°C); 3) preservation in TRIzol® reagent, flash freezing in LN2 and storage at –80°C. Methods 1 and 2 were tested for liver, while 2 and 3 for blood. Our results showed that both preservation methods resulted in excellent outcomes for liver samples. For blood samples however, TRIzol® preservation was an efficient procedure for adequate RNA quality, quantity, and integrity, while conservation in RNAlater® solution was inadequate in both quality and quantity for an optimal RNA extraction. Appropriate protocols were established for each tissue and are being used now for transcriptomic studies in this sentinel organism.

При производстве качественной пищевой продукции особое внимание необходимо уделять ее упаковыванию и хранению. Витаминно-минеральные концентраты, используемые для приготовления пищевых продуктов и получения качественной безопасной продукции животноводства, содержат комплекс лабильных биологически активных веществ, для сохранности которых необходимо минимизировать окислительные процессы. Это обусловливает повышенные требования к упаковке таких продуктов. С целью выбора упаковочного материала и способа упаковки витаминно-минерального концентрата проведен тематический обзор публикаций отечественных и зарубежных ученых. Рассмотрены тенденции в области применения упаковочных материалов, указаны преимущества и недостатки биополимеров. На основе проведенного анализа установлено, что для упаковывания витаминно-минерального концентрата целесообразно в качестве материала упаковки использовать полимерные пленки, обладающие высокой свето- и газонепроницаемостью, позволяющей снизить скорость протекания окислительных процессов и, следовательно, сократить потери содержащихся в продукте лабильных биологически активных веществ. В качестве способа упаковки можно рекомендовать применять вакуумирование, обеспечивающее отсутствие кислорода воздуха в упаковке. When producing high-quality food products, special attention should be paid to their packaging and storage. Vitamin and mineral concentrates used for food preparation and production of high-quality safe livestock products contain a complex of labile biologically active substances, for the safety of which it is necessary to minimize oxidative processes. This leads to increased requirements for the packaging of such products. In order to select the material and method of packaging of vitamin and mineral concentrates, a thematic review of publications of domestic and foreign scientists was conducted. Trends in the use of packaging materials are considered, advantages and disadvantages of biopolymers are indicated. It was found that for packaging vitamin and mineral concentrate, polymer pellicle with high light and gas tightness, which allows reducing the rate of oxidative processes and, consequently, reducing the loss of labile biologically active substances contained in the product, should be used as a packaging material. Vacuuming, which ensures the absence of oxygen in the air in the package, can be recommended as a method of packaging.

Ostrich meat is a high-quality dietetic product, however, it is very sensitive to deterioration during storage. The aim of this study was to assess the effect of packaging systems on the fatty acid (FA) profiles in ostrich meat during refrigerated storage. The systems were: Vacuum packaging (VP) and modified atmosphere packaging (MAP) in two combinations of gases: MAP1 (40% O2/40% CO2/20% N2) and MAP2 (60% O2/30% CO2/10% N2). Samples were taken from the M. ilifibularis (IF) muscles of eight ostriches in each treatment group. The packs were stored in a refrigerator at 2 °C and analyzed at 0, 4, 8, 12 and 16 days. The packaging conditions and storage time had an impact on the concentration of bioactive compounds such as polyunsaturated fatty acids (PUFA), including n-3 such as C18:3, C20:5 (EPA) and C22:6 (DHA). The least changes in composition of n-3 and the sum of PUFA were recorded in ostrich meat packaged in vacuum, followed by that packaged using MAP1 and MAP2. The sum of n-6 PUFAs decreased significantly by 2.1% for MAP2, and only by 0.7% for vacuum packaging as the experiment progressed. A significant deterioration of these compounds was observed in all package systems, especially from day 12 until day 16 of storage.

Innovation in food packaging is mainly represented by the development of active and intelligent packing technologies, which offer to deliver safer and high-quality food products. Active packaging refers to the incorporation of active component into the package with the aim of maintaining or extending the product quality and shelf-life. The intelligent systems are able to monitor the condition of packaged food in order to provide information about the quality of the product during transportation and storage. These packaging technologies can also work synergistically to yield a multipurpose food packaging system. This review is a critical and up-dated analysis of the results reported in the literature about this fascinating and growing field of research. Several aspects are considered and organized going from the definitions and the regulations, to the specific functions and the technological aspects regarding the manufacturing technologies, in order to have a complete overlook on the overall topic.

Straw is a waste produced from the threshing and harvesting of rice. So far, rice straw has only been removed. In fact, it can be used as a potential energy source. This paper aims to examine the potential of rice straw as a renewable energy source in Indonesia. Opportunities and challenges were evaluated based on extensive and in-depth literature review. The paper discussed rice straw potential, its characteristics and benefits of using straw energy. Technology to convert rice straw into energy was also discussed. Results showed that renewable energy potential of rice straw is 28.8 TJ/year. Conversion technologies that can be applied include densification, combustion, gasification, and biogas. The biggest challenge for using rice straw energy is collection and transportation. Densification may overcome density problem by producing high quality pellets or briquettes that reduce transportation and storage costs. Straw characteristics need to be improved to increase its combustion properties.

Abstract PurposeLong-term electrocardiogram (ECG) monitoring is an essential approach for the early diagnosis of cardiovascular diseases. Flexible dry electrodes that contains electrolyte without water could be a potential substitution of wet electrodes for long-term ECG monitoring. Therefore, this paper developes a long-term, portable ECG patch based on flexible dry electrodes, namely SEUECG-100.MethodA device consists of analog-front-end acquisition, data acquisition, and storage modules is developed and tested. An impedance test was conducted to compare the skin-electrode impedance of the flexible dry electrode and the Ag/AgCl wet electrode. The ECG signals were simutanously collected from the same subject using the SEUECG-100 and Shimmer device , which were then compared and analyzed from the perspective of ECG morphology, RR interval, and signal quality indices (SQI).ResultsThe experimental results reveal that the flexible dry electrode has the characteristics of low skin-electrode impedance. SEUECG-100 could collect high-quality ECG signals. The ECG signals collected by the two devices have a high RR interval correlation (r=0.999). SQI results show that SEUECG-100 is better than the Shimmer device in overcoming baseline drift. Long-term ECG acquisition and storage experiments show that SEUECG-100 could collect ECG signals with good stability and high reliability.ConclusionThe implementation of the proposed system design with dry electrodes could can effectively record long-term ECG monitoring with high quality in comparison to systems with wet electrodes from both impedance characteristics and signal morphology aspects.

A semi-product for cooking a smoothie drink was chosen as a subject of the study. It included strawberries, dried apples and oat flocks. A sample was frozen in a low-temperature calorimeter to –20 °С. It was established, that freezing at–20±2 °С favors the full conservation of a sample and storage during 270 days. The results of the organoleptic evaluation of the semi-product before freezing and after refrigeratory storage during 270 demonstrated that the product is characterized by a high quality. Microbiological parameters of the fresh and frozen semi-product were studied for a quantity of mesophilic aerobic and facultative-anaerobic microorganisms (QMAFMAnM). And also for bacteria of the colibacillus group (BGCB), pathogenic microorganisms of bacteria of Salmonella genus, molds and yeast. The gram-negative microflora – bacteria of the colibacillus group, pathogenic microorganisms including bacteria of Salmonella genus, were not observed in samples of frozen semi-products. The studies demonstrated freezing advantages from both aspects of minimization of losses of valuable biocomponents of the raw material and of achieving high organoleptic parameters of products after freezing and defrosting. The observance of optimal conditions of freezing excludes the necessity of the additional use of artificial preservatives, provides high-quality products and safety that corresponds to all principles of healthy nutrition.
