Ayurvedic literature claims that Inula racemosa Hook. f. roots are beneficial for the liver. The study’s aim was to test the hepatoprotective effect of Inula racemosa Hook. f. roots ethanolic extract (IRE) against paracetamol (PCM) induced hepatotoxicity in rats. Silymarin (100 mg/kg/day) was used for 7 days, then PCM was orally administered (3 g/kg b.wt.) on the eighth day. 24 hours after the last PCM dosing, blood was withdrawn from the retro-orbital plexus and later on the rats were sacrificed. Aspartate Aminotransferase (AST), Alanine Aminotransferase (ALT), Alkaline Phosphatase (ALP), Albumin (ALB), Total Protein (TP), liver weight and relative liver weight were determined. Histopathology of the liver was performed. In the study, IRE was found to have comparable protective effects against PCM-induced hepatotoxicity as Silymarin. Gallic acid was isolated for the first time from Inula racemosa Hook. f. roots. IRE exhibited a hepatoprotective effect because gallic acid was present in it.

This study is an attempt to evaluate the hepatoprotective activity ofTabernaemontana divaricataagainst DEN and Fe NTA induced liver necrosis in rats. Ethanolic extract of the whole plant ofTabernaemontana divaricataat doses of 200 and 400 mg/kg body weight and 5-fluorouracil (standard drug) was orally administered to male Wistar Albino rats once daily for 24 weeks, simultaneously treated with the carcinogen DEN and Fe NTA. In simultaneously treated animals, the plant extract significantly decreased the levels of uric acid, bilirubin, AST, ALT, and ALP in serum and increased the levels of liver marker enzymes in liver. Treatment with the extracts resulted in a significant increase in the levels of antioxidants accompanied by a marked reduction in the levels of malondialdehyde when compared to DEN and Fe NTA treated group. When compared with 200 mg/kg bw rats, 400 mg/kg bw rats and 5-fluorouracil treated rats showed better results in all the parameters. The histopathological studies confirmed the protective effects of extract against DEN and Fe NTA induced liver necrosis. Thus, it could be concluded that the use ofTabernaemontana divaricataextract in the treatment of carcinogen induced hepatic necrosis.

Tannase plays a crucial role in many fields, such as the pharmaceutical industry, beverage processing, and brewing. Although many tannases derived from bacteria and fungi have been thoroughly studied, those with good pH stabilities are still less reported. In this work, a mangrove-derived yeast strain Rhodosporidium diobovatum Q95, capable of efficiently degrading tannin, was screened to induce tannase, which exhibited an activity of up to 26.4 U/mL after 48 h cultivation in the presence of 15 g/L tannic acid. The tannase coding gene TANRD was cloned and expressed in Yarrowia lipolytica. The activity of recombinant tannase (named TanRd) was as high as 27.3 U/mL. TanRd was purified by chromatography and analysed by SDS-PAGE, showing a molecular weight of 75.1 kDa. The specific activity of TanRd towards tannic acid was 676.4 U/mg. Its highest activity was obtained at 40 °C, with more than 70% of the activity observed at 25–60 °C. Furthermore, it possessed at least 60% of the activity in a broad pH range of 2.5–6.5. Notably, TanRd was excellently stable at a pH range from 3.0 to 8.0; over 65% of its maximum activity remained after incubation. Besides, the broad substrate specificity of TanRd to esters of gallic acid has attracted wide attention. In view of the above, tannase resources were developed from mangrove-derived yeasts for the first time in this study. This tannase can become a promising material in tannin biodegradation and gallic acid production.

A preliminary chemical characterization of main components of ethanolic extract with dried rhizomes of Smilax domingensis Wid. that grow in Cuba was done using a GCMS-QP2010 Ultra Shimadzu and the mass spectra of the compounds found in the extract was matched with the National Institute of Standards and Technology (NIST) library. After sample derivatization 125 chemical compounds were registered by the equipment and from them, 35 different chemical components were characterized and reported for the first time from this part of the plant in our country. The results demonstrate the developed method could be employed as a rapid and versatile analytical technique for identification of chemical constituents and quality control of Smilax domingensis.

The species Inga laurina is native to the Brazilian Cerrado. There are no studies about the chemical composition and biological activities of extracts of this endangered species. The ethanolic extract and its successive fractions are rich in phenolic compounds and presented good antifungal activities. HPLC/MS-MS/MS and H1/C13 analysis led to the identification of seventeen compounds, most of which are gallic acid derivatives, myricetin and quercetin glycosides. The ethyl acetate fraction (EAF) contained high levels of total phenolics, expressed in milligrams of gallic acid equivalents per gram of extract (475.3 ± 1.9 mg GAE gextract-1) and flavonoids expressed in milligrams of quercetin equivalents per gram of extract (359.3 ± 10.6 mg QE gextract-1). This fraction was active against fungi of the Candida genus. The EAF showed MIC value 11.7 μg mL−1 against C. glabrata and a selectivity index of 1.6 against Vero cells. The flavonol glycoside myricetin-3-O-rhamnoside was isolated for the first time from the Inga laurina. These results make I. laurina a promising plant as a source of pharmaceutical and biological active antifungal compounds.

The present study aimed to analyze the phytoconstituents of Neptunia triquetra (Vahl) Benth. Anti-inflammatory and hepatoprotective activities of ethanol (EE), chloroform (CE) and dichloromethane (DCME) of stem extracts were evaluated using in vivo experimental models. The extracts were analyzed for phytoconstituents using GC-HRMS. Anti-inflammatory activity of CE, EE and DCME was accessed using carrageenan-induced paw oedema, cotton pellet-induced granuloma and the carrageenan-induced air-pouch model in Wistar albino rats. The hepatotoxicity-induced animal models were investigated for the biochemical markers in serum (AST, ALT, ALP, GGT, total lipids and total protein) and liver (total protein, total lipids, GSH and wet liver weight). In the in vivo study, animals were divided into different groups (six in each group) for accessing the anti-inflammatory and hepatoprotective activity, respectively. GC-HRMS analysis revealed the presence of 102 compounds, among which 24 were active secondary metabolites. In vivo anti-inflammatory activity of stem extracts was found in the order: indomethacin > chloroform extract (CE) > dichloromethane extract (DCME) > ethanolic extract (EE), and hepatoprotective activity of stem extracts in the order: CE > silymarin > EE > DCME. The results indicate that N. triquetra stem has a higher hepatoprotective effect than silymarin, however the anti-inflammatory response was in accordance with or lower than indomethacin.

Objective: To evaluate the hepatoprotective effect of ethanolic extract of the root (REE) of Punica granatum.Methods: This study was conducted on adult albino Wistar rats of either sex weighing 150-200 g. Animals were divided into five groups (n=5). Liver injury was produced by carbon tetrachloride (CCl4) 1 ml/kg dissolved in olive oil (1:1) given intraperitoneally on day 1 and day 4 of the study duration of 14 days. Silymarin (50 mg/kg/d) orally was used as standard drug. Test groups received an REE of P. granatum (REE) at doses of 200 and 400 mg/kg/day orally along with CCl4. On the 15th day, all animals were sacrificed, and blood was collected. Liver was sent for histopathological examination. The hepatoprotective effect of REE was evaluated by assessment of physical parameters, histopathological examination and biochemical parameters such as aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), and total serum bilirubin.Results: The administration of REE of P. granatum at doses of 200 and 400 mg/kg/day orally, exhibited a highly significant decrease in the rise of mean serum AST, ALT, ALP, and total bilirubin as compared to CCl4 treated group (p<0.001). Histopathological examination of the liver also suggested hepatoprotective effect of REE of P. granatum by restoration of hepatic architecture toward normal. Decrease in the extent of centrilobular necrosis was observed in REE (200 and 400 mg/kg/day) treated rats when compared to CCl4 treated group.Conclusion: This study demonstrated hepatoprotective activity of REE of P. granatum against CCl4 induced liver injury in rats.

Terminalia chebula Gertn. (Combetraceae) is an important herbal drug in Ayurvedic pharmacopea. In the present study, a 95% ethanolic extract of T. chebula (fruit) (TC extract), which was chemically characterized on the basis of chebuloside II as a marker, was investigated for hepatoprotective activity against anti-tuberculosis (anti-TB) drug-induced toxicity. TC extract was found to prevent the hepatotoxicity caused by the administration of rifampicin (RIF), isoniazid (INH) and pyrazinamide (PZA) (in combination) in a sub-chronic mode (12 weeks). The hepatoprotective effect of TC extract could be attributed to its prominent anti-oxidative and membrane stabilizing activities. The changes in biochemical observations were supported by histological profile.

Helicteres isora L. is a medicinal plant which is used in several diseases, such as snake-bite, dog-bite, diarrhoea and constipation in a new born baby, gastrointestinal disorders, diabetes, cancer, and infections. This plant has also been used in the management of liver damage through traditional medicine. However, the hepatoprotective activity of H. isora L. ethanolic extract has not been reported so much. The present work was carried to investigate the hepatoprotective effect of H. isora L. against paracetamol-induced liver injury in Swiss mice. Paracetamol (PCM) is widely used as an analgesic and antipyretic drug which at high dose can lead to undesirable side effects, such as hepatotoxicity. Paracetamol induce hepatotoxicity was evaluated by an increase (P<0.05) in AST and ALT serum activity. Paracetamol hepatotoxicity was also manifested by an increase in (P<0.05) lipid peroxidation and depletion of reduced glutathione (GSH) in liver tissue. Ethanol extract of H. isora L. (250, 500 and 1000 mg/kg bw/day) significantly restored the PCM-induced alterations in the biochemical activities of blood and liver tissues. The hepatoprotective effect of H. isora L. was also confirmed by the histopathological examination of liver tissue. Histopathological examination of liver sections in mice administered with 1000 mg/kg bw/day doses of the extract were perfectly protected almost similar to those of untreated mice. The results indicated the hepatoprotective nature of studied plants extract against paracetamol induced toxicity. Our study scientifically validates the folkloric claim as well as traditional uses of H. isora L. as hepatoactive medicine. The results of this study suggests a new direction in the treatment of liver disease in future.
