Endophytic fungi Fusarium sp. LCJ273 capable of producing L-asparaginase was isolated from the medicinal plant Adhatoda vasica. The aim of the present study was to maximize L-asparaginase production by submerged fermentation through statistical optimization. L-Asparaginase production by Fusarium sp. LCJ273 was studied in five different media. Various nutritional parameters specifically carbon, nitrogen and inducers were optimized for enhancing the production of L-asparaginase. In addition, different statistical based experimental designs were also applied to increase the production of L-asparaginase by Fusarium sp. LCJ273. Dextrose, ammonium sulphate and wheat bran were found to be effective for growth and higher yield of L-asparaginase in Modified Czapek’s Dox Broth. Dextrose at a concentration of 3.0 g/L increased L-asparaginase production up to 9.18±0.9 U/mL, ammonium sulphate at the concentration of 20 g/L showed maximum L-asparaginase production up to 13.69±0.4 U/mL and wheat bran at 2.5 g/L yielded up to 14.24±0.5 U/mL. The maximum L-asparaginase production was observed by Fusarium sp. LCJ273 on 5th day. The study revealed that through optimization, a 2 fold increase in L-asparaginase could be achieved.

Phylloplane biodiversity and endophytic fungi is considered one of the rich origins of novel biological activity compounds and high-level structural variation on the leaf surface. Plant leaves surface is a diverse terrestrial ecosystem, including filamentous fungi. This study aims to study the isolation and enumeration of Sapindus mukorossi phylloplane and endophytic fungal diversity. The Sapindus mukorossi isolated 14 fungal species from 9 genera of phylloplane and endophytic fungi. Cladosporium herbarum, Penicillium expansum, Fusarium oxysporum, Fusarium sp., Alternaria alternate, Collectotrichum orbiculare, Torulla herbarium, Epicoccum nigrum and Candida sp. as a phylloplane fungi. Aspergillus niger, A. flavus, Epicoccum nigrum. Penicillum digitatum, Penicillum sp. were identified as endophytic fungi.

Three enzymes FH3, S13 and LR1 from three different sources showed fibrinolytic activities. Two were from endophytic fungal cultures and one from the sclerotium of Lignosus rhinocerus mushroom (LR1). FH3, S13 cultures and LR1, the crude extract of the sclerotium were concentrated and purified by ammonium sulphate precipitation, ion-exchange chromatography and gel-filtration. The molecular weights of the FH3, S13 and LR1 purified enzymes were estimated to be approximately 34kDa, 34kDa and 10kDa, respectively. Maximum fibrinolytic activities were observed for FH3 at pH 7 and 30°C, S13 at pH 8 and 40°C and LR1 at pH 6 and 40°C.  In our earlier paper we identified FH3 as Fusarium sp. and S13 as Penicilium citrinum.

Protease has been in increasing demand in industries due to its hydrolytic nature. In industries, high yield of enzyme is required to meet the industrial need at a relatively cheaper cost. In the present study, the protease from Bacillus sp. MSK-01 was produced in large quantity by submerged fermentation. Statistical techniques including Plackett-Burman and Response surface methodology are useful tools for optimizing many parameters at a time and are used for increasing the protease production from Bacillus sp. MSK-01. 19 different parameters were chosen, out of which 15 factors had positive effect on protease yield. Four maximum influencing factors were peptone, magnesium sulphate, skim milk powder and casein were chosen to further increase the protease yield. 397.3 IU ml-1 of enzyme yield was obtained under optimized conditions which lead to 198 fold increase in the yield of protease from unoptimized condition.
