In recent years, there has been increasing demand for higher voltage SEMs, in the field of surface observation, especially that of magnetic domains, dislocations, and electron channeling patterns by backscattered electron microscopy. On the other hand, the resolution of the CTEM has now reached 1 ∼ 2Å, and several reports have recently been made on the observation of atom images, indicating that the ultimate goal of morphological observation has beem nearly achieved.

Abstract Background Cholestasis is the major cause of bile acid accumulation leading to liver damage. Chronic infection of worms can modulate the immune response towards T helper (Th)2-related cytokines. The present study aims to evaluate the protective impact of an ascarid nematode Toxocara vitulorum extract (TvE) against alpha-naphthylisothiocyanate (ANIT)-induced cholangitis male wistar rat model compared to ursodeoxycholic acid (UDCA) as a standard drug. Results Pretreatment with TvE and/or UDCA induced a marked reduction in the levels of liver function tests and malondialdehyde, while antioxidant markers were increased compared to cholestatic rats. Pretreatment with either TvE or combination before cholangitis induction attenuated the predominant Th1-related cytokines (IFN-γ and TNF-α) to Th2 (IL-13 and IL-10). TvE administration promoted higher expression levels of Bcl-2 protein and lower levels of caspase-3 compared to cholestatic rats. Conclusions Treatment with TvE has improved the liver functions and elevated the levels of oxidative stress markers. The upregulation of Th2-related cytokines and suppression of apoptosis through caspase-3 might be considered as a potential mechanism of TvE. Thereby, this natural extract revealed an opportunity for use in treatment of cholangitis disease.

ABSTRACTAmong many convergently evolved sequestrate fungal genera in Boletaceae (Boletales, Basidiomycota), the genus Octaviania is the most diverse. We recently collected many specimens of Octaviania subg. Octaviania, including several undescribed taxa, from Japan and the Americas. Here we describe two new species in subgenus Octaviania, O. tenuipes and O. tomentosa, from temperate to subtropical evergreen Fagaceae forests in Japan based on morphological observation and robust multilocus phylogenetic analyses (nrDNA ITS and partial large subunit [LSU], translation elongation factor 1-α gene [TEF1] and the largest subunit of RNA polymerase II gene [RPB1]). Based on specimens from the Americas as well as studies of the holotype, we also taxonomically re-evaluate O. asterosperma var. potteri. Our analysis suggests that O. asterosperma var. potteri is a distinct taxon within the subgenus Octaviania so we recognize this as O. potteri stat. nov. We unexpectedly collected O. potteri specimens from geographically widespread sites in the USA, Japan and Colombia. This is the first verified report of Octaviania from the South American continent. Our molecular analyses also revealed that the RPB1 sequence of one O. tenuipes specimen was identical to that of a closely related species, O. japonimontana, and that one O. potteri specimen from Minnesota had an RPB1 sequence of an unknown species of O. subg. Octaviania. Additionally, one O. japonimontana specimen had an unusually divergent TEF1 sequence. Gene-tree comparison and phylogenetic network analysis of the multilocus dataset suggest that these heterogenous sequences are most likely the result of previous inter- and intra-specific hybridization. We hypothesize that frequent hybridization events in Octaviania may have promoted the high genetic and species diversity found within the genus.

Aiming at effectively controlling nonylphenol (NP) pollution, three bacterial strains were isolated from activated sludge and landfill leachate, which could grow with nonylphenol as sole carbon and energy source. The three nonylphenol-degrading bacteria isolated were named as WN6, SLY9 and SLY10, respectively. The morphological observation and 16S rDNA identification revealed that the strains belonged to Serratia sp., Klebsiella sp. and Pseudomonas putida, respectively. WN6 and SLY9 contained ALK gene, while WN6 and SLY10 harbored C12O genes. The three strains were combined together to form complex microorganisms ZJF. The ratio of Serratia sp. to Klebsiella sp. to Pseudomonas putida was 2:1:2 (volume ratio of bacterial suspension). Under the conditions that temperature was 30 ℃, pH was 6, inoculation amount was 10% (volume ratio), initial concentration of NP solutions was 20 mg/L, NP degradation rate by ZJF reached 73.82%, compared with any single strain of the three bacteria, NP degradation rate by ZJF increased more than 15% during 6 days. Bioremediation of nonylphenol-polluted the Yangtze River and the Ancient Canal water by ZJF ware simulated. After a 6-day incubation period, the degrading rate of nonylphenol in Ancient Canal water was close to 80%, and the degrading rate of nonylphenol in Yangtze River water was 72.84%.

Two steroidal saponins, tigogenin hexasaccharide-1 (TGHS-1, (25R)-5α-spirostan-3β-yl 4-O-[2-O-[3-O- (α-L-rhamnopyranosyl)-β-D-glucopyranosyl]-3-O-[4-O-(α-L-rhamnopyranosyl)-β-D-glucopyranosyl]-β-D-glucopyranosyl]- β-D-galactopyranoside) and tigogenin hexasaccharide-2 (TGHS-2, (25R)-5α-spirostan-3β-yl 4-O-[2-O-[3-O- (β-D-glucopyranosyl)-β-D-glucopyranosyl]-3-O-[4-O-(α-L-rhamnopyranosyl)-β-D-glucopyranosyl]-β-D-glucopyranosyl]- β-D-galactopyranoside), were isolated from the fresh bulbs of Camassia cusickii. In murine leukemic L1210 cells, both compounds showed cytotoxicity with an EC50 value of 0.06 µM. The morphological observation revealed that TGHS-1 and TGHS-2 induced shrinkage in cell soma and chromatin condensation, suggesting apoptotic cell death. The cell death was confirmed to be apoptosis by Annexin V binding to phosphatidylserine in the cell membrane and excluding propidium iodide. A typical apoptotic DNA ladder and the cleavage of caspase-3 were observed after treatment with TGHS-1 and TGHS-2. In the presence of both the compounds, cells with sub-G1 DNA content were detected by flow cytometric analysis, indicating that TGHS-1 and TGHS-2 (each EC50 value of 0.1 µM) are the most powerful apoptotic saponins known. These results suggest that TGHS-1 and TGHS-2 induce apoptotic cell death through caspase-3 activation.Key words: steroidal saponin, tigogenin hexasaccharide, apoptosis, DNA fragmentation, murine leukemic L1210 cells.

AbstractIn this study, yeast was isolated from cherry wine lees by rose Bengal medium, and its species was identified through three-stage screening, morphological observation and molecular biological identification. Moreover, the tolerance of screened strains was studied. The results showed that 30 strains of yeast were isolated from cherry wine lees, and five strains of yeast were selected, which were named YJN10, YJN16, YJN18, YJN19 and YJN28. After preliminary appraisal, strain YJN10 was Saccharomyces kudriavzevii, strain YJN16 was Saccharomyces paradoxus, and strains YJN18, YJN19, YJN28 were Saccharomyces cerevisiae. In the tolerance study, the tolerable sugar concentrations of the five strains were 650, 650, 550, 600 and 600 g/L. The tolerable alcohol volume fractions were 20, 20, 16, 18 and 18%. The tolerable molar concentration of potassium chloride was 1.8, 1.8, 1.5, 1.5 and 1.5 mol/L. Finally, strains YJN10, YJN16, YJN19 and YJN28 showed good tolerance, which laid a foundation for subsequent application in cherry wine fermentation.

NiO nanoparticle was synthesized by a sol-gel method with three different pH values namely pH=1, 7 and 11, and then calcined at temperature of 450 ᵒC. The influence of different pH values on the physical properties of NiO nanoparticles were investigated by a particle size analyzer (PSA), field emission scanning electron microscope (FESEM) and X-ray diffractometer (XRD). Structural analysis confirmed that a cubic structure of NiO nanoparticle was obtained without any secondary phase for NiO powders prepared with pH=1, while the peak of secondary phase (Ni) appeared for NiO powders prepared with pH= 7 and 11. Morphological observation showed that the NiO nanoparticles prepared with pH=7 and 11 tend to form more agglomerates compared to one prepared with pH=1. The average diameter of NiO nanoparticles with pH 1, 7 and 11 were approximately in the range of 19-26 nm, 21-28 nm, and 24-30 nm, respectively. NiO powder that was synthesized with pH=1 was further used to prepare composite anode of NiO Nps-BaCe0.54Zr0.36Y0.1O2.95 (BCZY) powder. Unfortunately a composite of NiO Nps-BaCeO3-BaZrO3 was obtained instead of BCZY and governed by agglomerates with size in the range of 70-300 nm.

Flexible sensors and wearable electronics have become important in recent years. A good conductive and flexible textile is needed to develop a commercial wearable device. Conductive polymers have generally been used with limitation in reducing the surface resistance to a certain amount. In this research, a method for fabricating a stretchable highly conductive cotton/lycra knitted fabric is introduced by treating the fabric with polypyrrole (PPy), silver nanoparticles (SNPs) composites, and post-treating with poly (3,4-ethylenedioxythiophene) poly (styrenesulfonate) (PEDOT:PSS). Polypyrrole and SNPs were in situ fabricated on the cotton/lycra fabric by consecutive redox reaction of silver nitrate and pyrrole and finally covered by PEDOT:PSS solution through dip-coating. The coated textile was characterized by Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), field emission scanning electron microscopy (FESEM), X-ray mapping, and energy dispersive X-ray spectroscopy (EDX). Fourier transform infrared spectroscopy confirmed PPy-SNPs (P-S) composites on the fabric surface. Fourier transform infrared spectroscopy results, X-ray mapping, EDAX, and XRD analysis also confirmed the P-S composites and PEDOT:PSS polymeric layer on the fabric. Morphological observation showed a layer of PEDOT:PSS on the P-S caused the higher connection of coating on textiles which resulted in the higher electrical conductivity (43 s/m). Also morphological observations showed penetration of the silver particles inside fibers which represented improving in attachment and stability of the coating on the fibers. Further, the electrical conductivity of PPy-SNPs-PEDOT:PSS coated textile increased under the tension. Hence, the stretchable and highly conductive knitted cotton/lycra fabric has potentiality to be used for fabricating the flexible sensors or wearable electronics.
