To understand the differences in mineral nutrient storage within tissues and organs of rice (Oryza sativa L.) grains, the distribution of globoids in rice embryo and endosperm tissues was examined using light and transmission electron microscopy and energy dispersive X-ray microanalysis was used to study globoid composition. Globoids were found in most embryo tissues, including provascular cells, and their location and size in sections of protein bodies is described. While P, Mg, and K were commonly detected in all globoids, other elements such as Ca, Mn, Fe, and Zn were sometimes detected in globoids of specific tissues and (or) regions. High peak-to-background ratios for P were obtained in globoids of scutellar and aleurone cells, and moderately high values were detected in ground meristem regions of the mesocotyl and coleoptile. Relatively high K levels were found in globoids in parenchyma cells of the scutellum and coleorhiza; in provascular cells of the radicle; and in ground meristem cells from the mesocotyl, coleoptile, and plumule. Calcium was mainly detected in globoids of the aleurone layer. Iron was mostly found in radicle tissue globoids. Zinc was commonly found in globoids of the scutellar epithelium and in provascular tissues of the mesocotyl, coleoptile, and radicle. Manganese was distributed throughout most of the tissues examined, but the highest levels of Mn were detected in globoids from the coleoptile tip regions and the plumule. A novel finding was that, in the provascular tissues of the coleoptile tip, distinctive differences were found in Mn, Fe, and Zn storage between globoids in the future xylem and the future phloem. In one of our earlier papers (Wu et al. 1978), we suggested that glutelin is the major composition of the round storage protein bodies although they also contain relatively more prolamine than the angular one does. Immunochemical studies of Krishnan et al. (1986) later showed the presence of glutelin in the irregularly-shaped (angular) protein bodies while the prolamines were found in the round ones. Our recent experiment using protein A-gold technique found that prolamine is mainly deposited into the angular protein bodies.Small blocks (1 mm3) of 7 DAF (days after flowering) caryopsis of Orvza perennis were fixed with 3% paraformaldehyde and 3% glutaraldehyde in 0.1M sodium phosphate, pH7.4, dehydrated in a graded ethanol series and infiltrated with Spurr’s resin. Thin sections, after gold labeling, were stained with uranyl acetate and lead citrate. Rabbit antibodies were raised against purified prolamine. Protein A-gold sol complex was prepared based on the technique of Horisberger et al. (1977).

Keragaan morfologi dan kandungan antosianin padi beras merah (Oryza sativa L.) pada Kecamatan Munte dan Kecamatan Payung di Kabupaten Karo. Penelitian ini bertujuan untuk mendapatkan informasi morfologi dan kandungan antosianin padi beras merah (Oryza sativa L.). Penelitian ini dimulai dari Juni 2017dan selesai pada Januari 2018 di kecamatan Munte dan Kecamatan Payung Kabupaten Karo.Metode survei deskriptif menggunakan panduan International Rice Research Institute (IRRI). Teknik penentuan lokasi secara sengaja dan pengamblan sampel secara kebetulan.Uji kandungan antosianin menggunakan metode analitik dengan menghomogenkan sampel. Hasil eksplorasi didapatkan 72 genotipe yang dibagi menjadi tiga lokasi lahan. Berdasaran uji kandungan antosianin didapatkan lahan A memiliki kandungan tertinggi yaitu 0,5 mg/100 g dan terendah yaitu lahan B 0,08 mg/100 g.

Experiments were carried out to assess some insecticide modules against major insect pests of rice. Each module consists of a basal application of carbofuran 3G @ 1 kg a.i ha-1 at 20 DAT and Rynaxypyr 20 SC @ 30 g a.i ha-1 at 45 DAT except untreated control. All modules differ with each other only in third treatment which was applied in 65 DAT. The third treatment includes: Imidacloprid 17.8 SL @ 27 g a.i ha-1, Pymetrozine 50 WG @ 150 g a.i ha-1, Triflumezopyrim 106 SC @ 27 g a.i ha-1, Buprofezin 25 SC @ 250 g a.i ha-1; Glamore (Imidacloprid 40+Ethiprole 40% w/w) 80 WG @ 100 g a.i. ha-1, Thiacloprid 24 SC @ 60 g a.i ha-1, Azadirachtin 0.03 EC @ 8 g a.i ha-1, Dinotefuran 20 SG@ 40 g a.i ha-1 and untreated control. All the treated plots recorded significantly lower percent of dead heart, white ear- head caused by stem borer and silver shoot caused by gall midge. Module with Pymetrozine 50 WG @ 150 g a.i ha-1 treated plot recorded significantly higher per cent reduction of plant hoppers (>80% over untreated control) and produced higher grain yield (50.75 qha-1) than the other modules. Among the different treated modules the maximum number of spiders was found in Azadirachtin 0.03 EC @ 8 g a.i ha-1 treated module plot followed by other treatments.
