Oridonin (ORI) is a natural active ingredient with strong anticancer activity. But its clinical use is restricted due to its poor water solubility, short half-life, and low bioavailability. The aim of this study is to utilize the metal organic framework material MOF-5 to load ORI in order to improve its release characteristics and bioavailability. Herein, MOF-5 was synthesized by the solvothermal method and direct addition method, and characterized by Scanning Electron Microscopy (SEM), X-Ray Diffraction (XRD), Fourier Transform Infrared Spectrometer (FTIR), Thermogravimetric Analysis (TG), Brunauer–Emmett–Teller (BET), and Dynamic Light Scattering (DLS), respectively. MOF-5 prepared by the optimal synthesis method was selected for drug-loading and in vitro release experiments. HepG2 cells were model cells. MTT assay, 4′,6-diamidino-2-phenylindole (DAPI) staining and Annexin V/PI assay were used to detect the biological safety of blank carriers and the anticancer activity of drug-loaded materials. The results showed that nano-MOF-5 prepared by the direct addition method had complete structure, uniform size and good biocompatibility, and was suitable as an ORI carrier. The drug loading of ORI@MOF-5 was 52.86% ± 0.59%. The sustained release effect was reliable, and the cumulative release rate was about 87% in 60 h. ORI@MOF-5 had significant cytotoxicity (IC50:22.99 μg/mL) and apoptosis effect on HepG2 cells. ORI@MOF-5 is hopeful to become a new anticancer sustained release preparation. MOF-5 has significant potential as a drug carrier material.

Abstract Two-dimensional (2D) nanomaterials generally exhibit enhanced physiochemical and biological functions in biomedical applications due to their high surface-to-volume ratio and surface charge. Conventional cancer chemotherapy based on nanomaterials has been hindered by their low drug loading and poor penetration in tumor tissue. To overcome these difficulties, the novel materials systems are urgently needed. Hereby, the lanthanide-based porphyrin metal organic framework (MOF) nanosheets with promising cancer imaging/chemotherapy capacities are fabricated, which displays superior performance in the drug loading and tumor tissue penetration. The biodegradable PPF-Gd nanosheets deliver an ultrahigh drug loading (>1500%) and demonstrate the stable and highly sensitive stimuli-responsive degradation/release for multimodal tumor imaging and cancer chemotherapy. Meanwhile, PPF-Gd NSs also exhibits excellent fluorescence and magnetic resonance imaging capability in vitro and in vivo. Compared to the traditional DOX chemotherapy, the in vivo results confirm the evident suppression of the tumor growth by the PPF-Gd/DOX drug delivery system with negligible side effects. This work further supports the potential of lanthanide-based MOF nanomaterials as biodegradable systems to promote the cancer theranostics technology development in the future.

AbstractChemo-photothermal therapy based on nanoparticles has emerged as a promising strategy for cancer treatment. However, its therapeutic efficacy and application potential are largely subjected to the uncontrollability and biotoxicity of functional nanoplatforms. Herein, a novel biocompatible and biodegradable metal organic framework (MOF), which was constructed by growing crystalline zeolitic imidazolate framework-8 on gold nanoroad (Au@ZIF-8), was designed and fabricated for efficient drug loading and controlled release. Owing to the large surface area and guest-matching pore size of ZIF-8, doxorubicin (DOX) was successfully loaded into the Au@ZIF-8 with a high drug loading efficiency of ~ 37%. Under NIR light or weakly acidic environment, the ZIF-8 layer was quickly degraded, which resulted in an on-demand drug release in tumour site. More importantly, under the irradiation of near infrared (NIR) laser, highly efficient cancer treatment was achieved in both in vitro cell experiment and in vivo tumour-bearing nude mice experiment due to the synergistic effect of photothermal (PTT) therapy and chemotherapy. In addition, the in vivo study revealed the good biocompatibility of Au@ZIF-8. This work robustly suggested that Au@ZIF-8 could be further explored as a drug delivery system for chemo-photothermal synergistic therapy.

Tujuan dari penelitian ini adalah untuk mengetahui nilai loading ibuprofen pada material UiO-66 (Zr-Metal Organic Framework). Pada penelitian ini, dilakukan variasi lama perendaman 12, 24, 36, 48, 60 dan 72 jam, sehingga didapatkan nilai drug loading yang optimal. Nilai drug loading ibuprofen diukur menggunakan spektrofotometer UV-VIS. Adapun penyimpanan ibuprofen dalam UiO-66 dikonfirmasi dengan XRD, FTIR, dan SEM-EDX. Hasil analisa gugus fungsi sampel dengan Fourier-Transform Infra Red (FTIR) dari Zr-MOF menunjukkan bahwa puncak khas dari UiO-66 juga muncul pada 663, 748 dan 547 cm-1 yang menunjukkan adanya interaksi Zirkonium (Zr) dengan ligan H2BDC, setelah UiO-66 digunakan untuk loading Ibuprofen tidak mengubah posisi dari puncak-puncak UiO-66. Namun, terjadi penurunan intensitas dan hilangnya puncak disekitar 1300 cm-1 yang dikarenakan interaksi antara UiO-66 dengan ibuprofen. Pola difraktogram UiO-66  sebelum dan sesudah loading ibuprofen yang  menunjukkan bahwa UiO-66 yang terbentuk memiliki puncak karakteristik UiO-66 pada 7,3o. Mikrograf UiO-66 sebelum dan setelah loading  ibuprofen memiliki morfologi kubus. Namun, setelah  loading  ibuprofen terdapat bagian seperti karang putih, ukuran yang terlihat lebih besar, serta pengurangan ruang kosong permukaan material. Uji Drug Loading Ibuprofen pada UiO-66 menunjukkan loading optimum ibuprofen dalam Zr-MOF mencapai 82,79% pada waktu pengadukan 72 jam.
