A soluble NADH dehydrogenase (NADH:ferricyanide oxidoreductase) has been obtained by simple disruption of cells of Thermus aquaticus strain T351, and purified. The enzyme is of low molecular mass, 50 000 Da, and displays many of the properties of the membrane-bound enzyme, including inhibition by both NADH and ferricyanide, and the same Km for ferricyanide. The enzyme contains 0.05 mol of FMN, 0.16 mol of labile sulphur and 2.2 mol of iron per mol of protein. The enzyme is inhibited by NAD and cupferron competitively with ferricyanide, and by ATP (but not ADP) competitively with NADH. The enzyme is particularly thermostable, having a half-life at 95 degrees C of 35 min. The effect of temperature on the molar absorption coefficient and the stability of NADH was determined.

The rate of exchange of deuterium for protium in the 4-methyl groups of 2-imino-1,4-dimethyl-1,2-dihydropyrimidine (1), 1,4-dimethyl-2-pyrimidone (2), and 4-methyl-2-amino-pyrimidine (3) has been determined in aqueous solution over an acidity range of some 21 pH(H0) units. The various exchange routes involve attack by base (water, hydroxide ion, buffer anion) on substrate (neutral, singly protonated, doubly protonated) and the identities of the principal components across the acidity spectrum have been established for all three compounds. The Brønsted slope, kinetic isotope effect, and activation parameters for 1 have also been determined. Protonating 1 activates it toward exchange by a factor of 103; addition of a second proton has a further effect of >105. The activating effects of the imino group, the carbonyl group, and the protonated imino group in these compounds are in the ratio 10−1:1:102.

A sensitive, specific and precise method for the determination of ammonia in sea water is presented. The method depends upon the oxidation of ammonia by chloramine-T and subsequent coupling with phenol to produce indophenol blue in an n-hexanol extract. Molar absorption coefficient ca 84,000. Values of ammonia measured in samples from the English Channel are given and the effect of freezing on samples for ammonia analysis is discussed.

Abstract In this paper, a series of click reaction-based Bodipy derivatives have been synthesized as part of a preliminary study. Following the characterization of the four challenger molecules, the photophysical properties (absorption, emission, excitation, molar absorption coefficient, quantum yield, etc.) were examined in various organic solvents. The results showed that the energy transfer from the Bodipy cores to other moieties was calculated between 71% and 93% depending on different π-electron configurations and chromophore groups.
