ABSTRACTParasites can account for a substantial proportion of the biomass in marine communities. As such, parasites play a significant ecological role in ecosystem functioning via host interactions. Unlike macropredators, such as large piscivores, micropredators, such as parasites, rarely cause direct mortality. Rather, micropredators impose an energetic tax, thus significantly affecting host physiology and behaviour via sublethal effects. Recent research suggests that infection by gnathiid isopods (Crustacea) causes significant physiological stress and increased mortality rates. However, it is unclear whether infection causes changes in the behaviours that underpin escape responses or changes in routine activity levels. Moreover, it is poorly understood whether the cost of gnathiid infection manifests as an increase in cortisol. To investigate this, we examined the effect of experimental gnathiid infection on the swimming and escape performance of a newly settled coral reef fish and whether infection led to increased cortisol levels. We found that micropredation by a single gnathiid caused fast-start escape performance and swimming behaviour to significantly decrease and cortisol levels to double. Fast-start escape performance is an important predictor of recruit survival in the wild. As such, altered fitness-related traits and short-term stress, perhaps especially during early life stages, may result in large scale changes in the number of fish that successfully recruit to adult populations.

Studying the sublethal effects of catch-and-release (C&R) is challenging, as there are several potential sources of bias. For example, if behavioural alterations immediately after the release event are to be studied, separation of tagging effects from actual C&R effects is required, which is a challenge in the wild, particularly in marine environments. To investigate the effects of C&R on Atlantic cod (Gadus morhua) in their natural environment, 80 cod were caught in fyke nets, fitted with acoustic transmitters, and released. After recovery from tagging and handling for at least 14 days, nine individuals were recaptured and released at least once during experimental angling, following best release practice. All cod survived the C&R event and did not show any large-scale behavioural changes (i.e., changes in diel vertical migrations). However, analysis of small-scale vertical movements showed that three individuals underwent short-term alterations (e.g., reduced or increased swimming activity). This study showed that pretagging fish with acoustic transmitters before experimental angling is an option when investigating fish behaviour immediately after the release event in marine environments. Moreover, release guidelines for cod should be developed, as cod can recover quickly if caught in shallow waters (<20 m) and properly handled and released.

AbstractStandard registration algorithms need to be independently applied to each surface to register, following careful pre-processing and hand-tuning. Recently, learning-based approaches have emerged that reduce the registration of new scans to running inference with a previously-trained model. The potential benefits are multifold: inference is typically orders of magnitude faster than solving a new instance of a difficult optimization problem, deep learning models can be made robust to noise and corruption, and the trained model may be re-used for other tasks, e.g. through transfer learning. In this paper, we cast the registration task as a surface-to-surface translation problem, and design a model to reliably capture the latent geometric information directly from raw 3D face scans. We introduce Shape-My-Face (SMF), a powerful encoder-decoder architecture based on an improved point cloud encoder, a novel visual attention mechanism, graph convolutional decoders with skip connections, and a specialized mouth model that we smoothly integrate with the mesh convolutions. Compared to the previous state-of-the-art learning algorithms for non-rigid registration of face scans, SMF only requires the raw data to be rigidly aligned (with scaling) with a pre-defined face template. Additionally, our model provides topologically-sound meshes with minimal supervision, offers faster training time, has orders of magnitude fewer trainable parameters, is more robust to noise, and can generalize to previously unseen datasets. We extensively evaluate the quality of our registrations on diverse data. We demonstrate the robustness and generalizability of our model with in-the-wild face scans across different modalities, sensor types, and resolutions. Finally, we show that, by learning to register scans, SMF produces a hybrid linear and non-linear morphable model. Manipulation of the latent space of SMF allows for shape generation, and morphing applications such as expression transfer in-the-wild. We train SMF on a dataset of human faces comprising 9 large-scale databases on commodity hardware.

Abstract Background Captive European bison (Bison bonasus) play an active role in conservation measures for this species; this includes education, which may conflict with these animals’ welfare. The effect of the presence of visitors on the welfare of captive animals can be negative, positive or neutral. However, the response of a given species to visitors is difficult to predict, since even closely related species display varying levels of tolerance to captivity. The aim of the study was to compare immunoreactive fecal cortisol levels (regarded as an indicator of the level of physiological stress) in groups of captive European bison that differed in terms of their social structure and the level of visitor pressure. The second aim was to determine if there was a correlation between intestinal parasitic burden and immunoreactive fecal cortisol levels. Results Immunoreactive fecal cortisol levels were not influenced by sex or age. However, study site and the interaction between study site and visitor pressure were statistically significant. European bison in one enclosure presented higher levels of immunoreactive fecal cortisol on weekdays than at weekends. In the other two study sites, the levels did not differ between weekdays and weekends. No correlation was found between parasitological infestation and immunoreactive fecal cortisol levels. Conclusions Measurement of fecal cortisol metabolites could be a valuable method for further research into the welfare of European bison in captivity. More subtle factors such as individual animal characteristics, feeding systems, and the arrangement of enclosures can be of great importance in terms of the effect of visitors on animals. The results of this study can be used in guidelines for the management of European bison populations.

The euryhaline milkfish (Chanos chanos) is an excellent subject for studies of the physiological and behavioral processes involved in salinity adaptation. In this study, energy partitioning for metabolism, activity and growth, maximal activity performance and blood osmotic concentrations were assessed at two activity levels in juvenile milkfish fed equal rations and maintained at a relatively constant temperature (262 C) and at salinities(15, 35 and 55 ?) that represented a wide range of osmoregulatory challenges. Changes in the measured parameters were not consistently related to the magnitude of the trans-integumentary osmotic gradients. Routine oxygen consumption rates were high in 35 ? salinity (mean 1 s.e.m. 1678 mg O2 kg-1 h-1) and comparably low in 15 and 55 ? salinity (1336 and 1273 mg O2 kg-1 h-1, respectively). Routine activity levels (relative swimming velocity) were highest in 35 ? salinity (0. 960.04 L s-1), where L is standard length, intermediate in 15 ? salinity (0.770.03 L s-1) and lowest in 55 ? salinity (0.670.03 L s-1). Growth was significantly higher in 55 ? salinity (3.40.2 % increase in wet body mass per day) than in 35 ?salinity (2.40.2 % increase per day) and intermediate in 15 ? salinity(2.90.5 % increase per day). Maximum swimming velocities decreased with increases in salinity, from 9.90.7 L s-1 in 15 ? salinity to 6.60. 5 L s-1 in 55 ? salinity. Sustained swimming activity above routine levels for 2 h resulted in an increase in blood osmotic concentrations in milkfish in 55 ?salinity, but osmoregulation was re-established during the second 2 h of activity. Thus, patterns of variation in metabolic rate and growth were largely parallel to variations in routine activity although, comparing 15 and 55 ? salinity, elevated maintenance costs for osmoregulation at the high salinity were detectable. Reduced osmoregulatory abilities and reductions in maximal swimming performance suggest that high salinity may constrain activity. The results demonstrate that investigations of salinity adaptation in euryhaline fishes should take into account the interactive effects of salinity on physiology and behavior.

Background: Large-scale health surveys often consider sociodemographic characteristics and several health indicators influencing physical activity that often vary across units (regions or states). Data in a survey for some small units are often not representative of the larger population. This study developed a relatively simple multilevel regression and poststratification (MRP) model to estimate the proportion of leisure-time physical activity across Brazilian state capitals, based on the Brazilian cross-sectional national survey VIGITEL (2018). Methods: We used various approaches to evaluate whether the MRP approach outperforms single-level aggregated estimates, with various subsample proportions tested. Results: The mean absolute errors were consistently smaller for the MRP estimates than single-level regression estimates, particularly with smaller sample sizes. MRP consistently had predictions closer to the estimation target than single-level aggregated estimations. MRP presented substantially smaller uncertainty estimates compared to aggregated estimates. Conclusions: Our results confirm that MRP is a promising strategy to derive disaggregated data for health-related outcomes and, in particular, physical activity indicators from aggregated-level surveys. Overall, the MRP is superior to single-level aggregated estimates and disaggregation, yielding smaller errors and more accurate estimates. MRP significantly expands the scope of issues for which researchers can better address participation bias and interpret interactions to estimate descriptive population quantities. The observations present in this study highlight the need for further research, potentially incorporating more information in the models to better interpret interactions and types of activities across target populations.

Emerging infectious diseases (EIDs) are typically characterized by novelty (recent detection) and by increasing incidence, distribution, and/or pathogenicity. Ophidiomycosis, also called snake fungal disease, is caused by the fungus Ophidiomyces ophidiicola (formerly “ophiodiicola”). Ophidiomycosis has been characterized as an EID and as a potential threat to populations of Nearctic snakes, sparking over a decade of targeted research. However, the severity of this threat is unclear. We reviewed the available literature to quantify incidence and effects of ophidiomycosis in Nearctic snakes, and to evaluate whether the evidence supports the ongoing characterization of ophidiomycosis as an EID. Data from Canada remain scarce, so we supplemented the literature review with surveys for O. ophidiicola in the Canadian Great Lakes region. Peer-reviewed reports of clinical signs consistent with ophidiomycosis in free-ranging, Nearctic snakes date back to at least 1998, and retrospective molecular testing of samples extend the earliest confirmed record to 1986. Diagnostic criteria varied among publications (n = 33), confounding quantitative comparisons. Ophidiomycosis was diagnosed or suspected in 36/121 captive snakes and was fatal in over half of cases (66.7%). This result may implicate captivity-related stress as a risk factor for mortality from ophidiomycosis, but could also reflect reporting bias (i.e., infections are more likely to be detected in captive snakes, and severe cases are more likely to be reported). In contrast, ophidiomycosis was diagnosed or suspected in 441/2,384 free-ranging snakes, with mortality observed in 43 (9.8 %). Ophidiomycosis was only speculatively linked to population declines, and we found no evidence that the prevalence of the pathogen or disease increased over the past decade of targeted research. Supplemental surveys and molecular (qPCR) testing in Ontario, Canada detected O. ophidiicola on 76 of 657 free-ranging snakes sampled across ~136,000 km2. The pathogen was detected at most sites despite limited and haphazard sampling. No large-scale mortality was observed. Current evidence supports previous suggestions that the pathogen is a widespread, previously unrecognized endemic, rather than a novel pathogen. Ophidiomycosis may not pose an imminent threat to Nearctic snakes, but further research should investigate potential sublethal effects of ophidiomycosis such as altered reproductive success that could impact population growth, and explore whether shifting environmental conditions may alter host susceptibility.

Mutations giving rise to anatomical defects in the inner ear have been isolated in a large scale screen for mutations causing visible abnormalities in the zebrafish embryo (Haffter, P., Granato, M., Brand, M. et al. (1996) Development 123, 1–36). 58 mutants have been classified as having a primary ear phenotype; these fall into several phenotypic classes, affecting presence or size of the otoliths, size and shape of the otic vesicle and formation of the semicircular canals, and define at least 20 complementation groups. Mutations in seven genes cause loss of one or both otoliths, but do not appear to affect development of other structures within the ear. Mutations in seven genes affect morphology and patterning of the inner ear epithelium, including formation of the semicircular canals and, in some, development of sensory patches (maculae and cristae). Within this class, dog-eared mutants show abnormal development of semicircular canals and lack cristae within the ear, while in van gogh, semicircular canals fail to form altogether, resulting in a tiny otic vesicle containing a single sensory patch. Both these mutants show defects in the expression of homeobox genes within the otic vesicle. In a further class of mutants, ear size is affected while patterning appears to be relatively normal; mutations in three genes cause expansion of the otic vesicle, while in little ears and microtic, the ear is abnormally small, but still contains all five sensory patches, as in the wild type. Many of the ear and otolith mutants show an expected behavioural phenotype: embryos fail to balance correctly, and may swim on their sides, upside down, or in circles. Several mutants with similar balance defects have also been isolated that have no obvious structural ear defect, but that may include mutants with vestibular dysfunction of the inner ear (Granato, M., van Eeden, F. J. M., Schach, U. et al. (1996) Development, 123, 399–413,). Mutations in 19 genes causing primary defects in other structures also show an ear defect. In particular, ear phenotypes are often found in conjunction with defects of neural crest derivatives (pigment cells and/or cartilaginous elements of the jaw). At least one mutant, dog-eared, shows defects in both the ear and another placodally derived sensory system, the lateral line, while hypersensitive mutants have additional trunk lateral line organs.

Viruses pose substantial challenges for society, economy, healthcare systems, and research. Their distinctive pathologies are based on specific interactions with cellular factors. In order to develop new antiviral treatments, it is of central importance to understand how viruses interact with their host and how infected cells react to the virus on a molecular level. Invading viruses are commonly sensed by components of the innate immune system, which is composed of a highly effective yet complex network of proteins that, in most cases, mediate efficient virus inhibition. Central to this process is the activity of interferons and other cytokines that coordinate the antiviral response. So far, numerous methods have been used to identify how viruses interact with cellular processes and revealed that the innate immune response is highly complex and involves interferon-stimulated genes and their binding partners as functional factors. Novel approaches and careful experimental design, combined with large-scale, high-throughput methods and cutting-edge analysis pipelines, have to be utilized to delineate the antiviral innate immune landscape at a global level. In this review, we describe different currently used screening approaches, how they contributed to our knowledge on virus–host interactions, and essential considerations that have to be taken into account when planning such experiments.

Micropropagation is an alternative technique to propagate at large scale plants to meet global plant demand. Various researchers have worked on the micropropagation technique to regenerate bamboo species by using nodal segments from years. Contamination, browning, necrosis, and acclimatization with physiological stress are the extreme problems of the micropropagation technique. But, many numbers of papers have been published on micropropagation of the bamboo species through nodal segments as explants. The proliferation of the bamboo shoots is dependent on the season of collection, size of explants, the position of explants, diversity of plants, concentration and combination of plant growth regulators, most adequate culture medium, environmental condition of the equipment, handling, and individual species. Bamboo is a monocarpic fast-growing, tall perennial grass and having the high potential to generate economic and social benefits. It helps to maintain land patterns and control soil erosion.  The long life cycle of the bamboo produces a huge amount of seeds but unfortunately, mostly, they are non-viable. So, bamboos are propagated from vegetative by cutting and air layering. However, these methods are only for a small scale and they also tend to destroy large mother plant stocks and difficult to be transported. So, the in vitro propagation technique is useful to obtain large progenies from desired genotypes. Mostly, BAP and TDZ growth hormones are widely used for shoot multiplication and IBA, NAA and IAA are used for root initiation as per developed protocols in tissue culture for large scale production. This review intends to explore an overview of the recent literature reports to summarize the importance of micropropagation by using nodal segments of bamboo species and factors influencing it.
