The spider, Porrhothele antipodiana, starved and provided with water, produced urine via the anal excretory system (Malpighian tubules, midgut diverticula and stercoral pocket) at a mean rate of about 2.5-5 μlg−1 day−1 and with a mean Na+/K+ ratio of about 1.0. Salts ingested from the prey were eliminated by two mechanisms. A K+-rich (Na+/K+ about 0.2) anal diuresis lasted about 3 days following a single meal and was maintained at more than 30μlg−1day−1 during feeding ad libitum. The second mechanism, interpreted as coxal secretion, functioned only during feeding itself and delivered Na+ into the prey at a constant rate of about 3%h−1 of total body Na+. This progressively raised the Na+/K+ ratio of the prey debris from 0.47 to 0.96 and, because of re-ingestion, recycled more Na+ than was originally present in the prey. Feeding was associated with large net increases in dry weight and ions, particularly K+, which were mainly stored in the diverticular tissue (midgut diverticula and Malpighian tubules embedded in adipose tissue). The stercoral fluid (final urine) was slightly hyposmotic to the haemolymph in starved and fed spiders. Only about half of its osmolarity was accounted for by Na+, K+ and Cl−. The volume of water gained from the meal was about equal to that lost in diuresis, and P. antipodiana drinks to maintain water balance because of relatively high transpirational and other losses. The primary function of the diuresis is probably elimination of ions from the meal, and not volume regulation.

1. The physiology of the excretory system and its role in salt and water balance have been studied in the desert locust, Schistocerca gregaria. 2. Collections and analyses were made of hindgut fluid (derived from the Malpighian tubules) and rectal fluid from locusts kept under various dietary régimes. 3. In starved locusts supplied with tap water very little fluid accumulates in the rectum, the output of the Malpighian tubules being almost completely reabsorbed. Relatively more salt than water is reabsorbed with the result that the rectal fluid attains a lower ionic concentration but higher osmotic pressure than the haemolymph. No fluid is voided. 4. In starved locusts supplied with hypertonic saline the rectum is distended with, fluid which has an osmotic pressure and ionic concentration considerably above that of the haemolymph. Very little fluid is voided. 5. Substantial elimination of fluid from the rectum occurs only in fed locusts, in association with the voiding of faeces. 6. These results are discussed in relation to the mechanism of osmotic and ionic regulation.

Visceral adipose tissue (VAT) accumulation, is a part of a polycystic ovary syndrome (PCOS) phenotype. Dual-energy x-ray absorptiometry (DXA) provides a gold standard measurement of VAT. This study aimed to compare ten different indirect methods of VAT estimation in PCOS women. The study included 154 PCOS and 68 age- and BMI-matched control women. Subjects were divided into age groups: 18–30 y.o. and 30–40 y.o. Analysis included: body mass index (BMI), waist circumference (WC), waist-to-hip ratio (WHR), waist-to-height ratio (WHtR), waist/height 0.5 (WHT.5R), visceral adipose index (VAI), lipid accumulation product (LAP), and fat mass index (FMI). VAT accumulation, android-to-gynoid ratio (A/G), and total body fat (TBF) was measured by DXA. ROC analysis revealed that WHtR, WHT.5R, WC, BMI, and LAP demonstrated the highest predictive value in identifying VAT in the PCOS group. Lower cut-off values of BMI (23.43 kg/m2) and WHtR (0.45) were determined in the younger PCOS group and higher thresholds of WHtR (0.52) in the older PCOS group than commonly used. Measuring either: WHtR, WHT.5R, WC, BMI, or LAP, could help identify a subgroup of PCOS patients at high cardiometabolic risk. The current observations reinforce the importance of using special cut-offs to identify VAT, dependent on age and PCOS presence.

A continuous blood volume monitoring (CBVM) device (Inline Diagnostics, Riverdale, USA) was used to study response to prescribed ultrafiltration during haemodialysis (HD) in 66 stabilised HD patients. Fifty percent of patients showed the expected linear decrease in BV right from the beginning of HD (group 1), 32% exhibited no decrease at all (group 2), while eighteen percent formed the transient group 3 which showed a plateau of varying length after which a decrease occurred. The correct setting of dry weight was verified through evaluation of the ratio of extracellular fluid volume to total body water (VEC/TBW) in 26 patients by means of whole body multifrequency impedometry MFI (Xitron Tech., San Diego, USA) and through measurement of the Vena Cava Inferior diameter (VCID) pre and post HD (in 6 and 5 patients from groups 1 and 3 and from group 2, respectively). The mean VEC/TBW in groups 1 and 3 was 0.56 pre and 0.51 post HD as compared to 0.583 and 0.551 in group 2. VCID decreased on average by 14.1% in groups 1 and 3 but remained stable in group 2. Both findings thus confirmed inadequately high estimation of dry weight. Since CBVM is extremely easy to perform it can be used as a method of choice in detecting inadequately high prescribed dry weight. The status of the cardiovascular system must always be considered before final judgement is made.

In male Sprague-Dawley rats, renal artery constriction in the presence of an inact contralateral kidney induced sodium retention (for 2-3 wk), moderate potassium loss,elevation of blood volume (BV), and an increase in water turnover. It is suggestedthat renal artery constriction activates the renin-angiotensin-aldosterone system, resulting in disordered regulation of salt and water balance and in blood pressure (BP) elevation. Subsequently, sodium balance was reestablished in one group of hypertensive rats. The previously retained sodium was kept in the body, and BV and reninactivity remained elevated. In a second group of animals, a malignant course of hypertension developed: BP surpassed a critical level of about 180 mmHg; sodium, potassium, and water were lost; BV declined; renin activity was further stimulated; and in the contralateral kidney malignant nephrosclerosis occurred. It is assumed that pressure diuresis and natriuresis induce a vicious circle: the increasing renin activity may maintain or further increase BP level, therby inducing further salt and water loss, etc.; high BP levels and high renin activities induce vascular damage and deterioration of renal function.

Four treatments, involving differences in method of feeding and interval between last feed and slaughter, were employed in a study of the effect of dietary chlortetracycline on protease, amylase, and cellulase activity in the intestinal and cecal contents of 16 pairs of weanling pigs. Most consistent results were obtained with six pairs which were limited pair-fed except for the final feeding during which feed was available ad libitum for a 4-hour period ending 18 hours before slaughter. The mean total activities of all three hydrolases in the contents of the small intestines and of the ceca of the antibiotic-fed animals of these six pairs were significantly greater than in those of the control animals. Expressed as activity per gram dry matter of intestinal contents, significantly higher values for protease and amylase, but not for cellulase, were observed in the pigs that had received chlortetracycline. When the combined results obtained from all 16 pairs of the experimental animals were analyzed, the results showed that on a basis of activity per gram dry matter of intestinal contents, ingested chlortetracycline was associated with significant increases in amylase and cellulase but not in protease activity. Protease, amylase, and cellulase activities per gram dry matter of cecal contents were higher for pigs fed the antibiotic than for their controls. The mean wet weight of the empty small intestine and the mean dry weight of the mucosa scraped from the anterior 3-meter section of the small intestine were lower for the chlortetracycline-fed animals, but the differences were not statistically significant.

The oxygen consumption of human adipose tissue (AT) was determined in 53 adults, lean and obese, and in nine lean boys. The oxygen consumption was positively related to fat cell weight and negatively to age and degree of obesity. Men and women did not differ with respect to oxygen consumption of AT. The positive relationship between oxygen consumption per cell and fat cell size was also demonstrated in size-separated cells from the same donors. Expressed per cell the oxygen consumption was higher in fat cells from obese than in cells from lean subjects, but expressed per gram of tissue the opposite result was found. The oxygen consumption of the total AT organ was higher in obese than in lean subjects. The energy expenditure of AT constituted approximately 4% of the estimated 24-h energy expenditure in both groups. It is concluded that obese subjects do not maintain their obesity because of a reduced energy expenditure of the total AT (or of the total body). After a partial weight reduction in five subjects, the energy metabolism tended to change in direction toward the conditions seen in lean subjects. However, it is still an open question whether the observed energy metabolic aberrations of obese human AT are only secondary to the obese state or partly primary and thus of etiological importance.

Antiadiposity effects of caloric restriction (CR) are associated with reduced insulin/IGF-1 signaling, but it is unclear whether the effects of CR would be additive to genetically reducing circulating insulin. To address this question, we examined female Ins1+/−:Ins2−/− mice and Ins1+/+:Ins2−/− littermate controls on either an ad libitum or 60% CR diet. Although Igf1 levels declined as expected, CR was unable to reduce plasma insulin levels in either genotype below their ad libitum-fed littermate controls. In fact, 53-week-old Ins1+/−:Ins2−/− mice exhibited a paradoxical increase in circulating insulin in the CR group compared with the ad libitum-fed Ins1+/−:Ins2−/− mice. Regardless of insulin gene dosage, CR mice had lower fasting glucose and improved glucose tolerance. Although body mass and lean mass predictably fell after CR initiation, we observed a significant and unexpected increase in fat mass in the CR Ins1+/−:Ins2−/− mice. Specifically, inguinal fat was significantly increased by CR at 66 weeks and 106 weeks. By 106 weeks, brown adipose tissue mass was also significantly increased by CR in both Ins1+/−:Ins2−/− and Ins1+/+:Ins2−/− mice. Interestingly, we observed a clear whitening of brown adipose tissue in the CR groups. Mice in the CR group had altered daily energy expenditure and respiratory exchange ratio circadian rhythms in both genotypes. Multiplexed analysis of circulating hormones revealed that CR was associated with increased fasting and fed levels of the obesogenic hormone, glucose-dependent insulinotropic polypeptide. Collectively these data demonstrate CR has paradoxical effects on adipose tissue growth in the context of genetically reduced insulin.
