Human embryonic diploid lung fibroblasts were capable of growing in a medium consisting of Eagle's MEM vitamins plus 0.1 µg/ml biotin, Earle's salt solution, 4 mM glutamine, 0.4 mM cystine, 0.5 mM tyrosine, 10 % undialysed calf serum and 1 mM pyruvate. The growth rate and percent cell attachment in this medium was slightly less than in Eagle's MEM but the longevity was about the same. Addition of 0.5-1.0 mM histidine stimulated growth but addition of O.2 mM phenylalanine, methionine or valine decreased longevity markedly. Maintaining a constant pH of about 7.5-7.6 with 20 mM of the organic buffers HEPES, TES or TRICINE yielded a growth rate equal to or better than that of cells growing in unbuffered medium. Some buffers gave good growth initially but eventually the longevity decreased (20mM BIS-TRIS, 20 mM BICINE, 10 mM TRIS). In almost all cases the initial exposure to the buffers yielded the best growth results.

Only 2 amino acids and one vitamin were found to be essential for the growth of human embryonic diploid lung fibroblasts when 10% undialysed calf serum was used as a medium supplement. These amino acids were either glutamine + cysteine or serine + homocysteine. Replacing cysteine or homocysteine with cystine or homocystine, respectively, reduced growth. The growth rate in the glutamine + cysteine medium was slightly less than that in Eagle's medium, but the in vitro life time was similar. Folic acid was the only vitamin needed to support cell growth in vitro. The addition of other vitamins had no stimulatory effect with the possible exception of nicotinamide. When other amino acids were added to glutamine + cysteine none showed stimulatory effects but tryptophan was either toxic or inhibitory for the 3 human diploid strains examined. Serine was inhibitory for WI-38 but not for MRC5 cells. Subtle nutritional differences appear to exist between fibroblasts of the same type obtained from different embryos.

The bioactivity and biocompatibility of a zinc phosphate luting cement (HARVARD, Richter & Hoffmann, Dental-GmbH, Berlin) which was modified by Bioglass® (PerioGlas® Synthetic Bone Graft Particulate, US Biomaterials), was evaluated in vitro with human lung fibroblasts (MRC-5), baby hamster kidney fibroblasts (BHK) and rat pulp cells (RPC) by XTT and BrdU assays. A thin Ca-P layer was grown on the surface of Bioglass®-modified zinc phosphate cement specimens after immersion in SBF for 7 days and remained constant after 16 days immersion time. The incorporation of Bioglass® powder in zinc phosphate specimens resulted in equal or increased cell attachment and activity for almost all cell lines examined without any apparent impact on mechanical or physicochemical properties of the cement, although this needs further documentation. The combination of these two methods in determining the biocompatibility of Bioglass®-modified zinc phosphate cements showed that cells not only attached well on modified specimens but were actively synthesizing DNA after 72h of incubation.

Abstract Background: The post-mastectomy changes to the locomotor system are related to the scar and adhesion or to the lymphatic edema after amputation which, in turn, lead to local and global distraction of the work of the muscles. These changes lead to body statics disturbance that changes the projection of the center of gravity and worsens motor response due to changing of the muscle sensitivity. Objective: The aim of the study was to evaluate the static balance of women after undergoing mastectomy. Methods: The study included 150 women, including 75 who underwent mastectomy (mean age: 60±7.6) years, mean body mass index (BMI): 26 (±3.6) kg/m2) and 75 who were placed in the control group with matched age and BMI. The study was conducted using a tensometric platform. Results: Statistically significant differences were found for almost all parameters between the post-mastectomy group and group of healthy women, regarding center of foot pressure (COP) path length in the Y and X axes and the mean amplitude of COP. Conclusions: First, the findings revealed that balance in post-mastectomy women is significantly better than in the control group. Second, physiotherapeutic treatment of post-mastectomy women may have improved their posture stability compared with their peers.

The shell growth of small coastal Mytilus edulis L. was measured at three different constant low salinities over short periods of time. Growth was significantly depressed in 6·4 and 16‰ S but not in 22·4‰ S. Fluctuating salinities between 0 and 32‰ S depressed growth whether the fluctuations were of sinusoidal or abrupt form. After 1 week of preconditioning to constant 32‰ S the growth of coastal (Bangor) mussels was better than estuarine (Conwy) mussels. However, after two weeks’ preconditioning to 32‰ S the estuarine mussels displayed the best growth. In the fluctuating regime, both coastal and estuarine mussels exhibited poor growth rates. The long-term response of the shell growth of coastal M. edulis was followed over a period of 44 days. Salinities in the range 1·8–9·6‰ S were lethal to the mussels within 10 days. In 12·8 and 16‰ S growth was initially delayed but recovered eventually. There was a gradual decline in the growth rate of the mussels exposed to the higher salinities (19·2–32‰) and an improvement in the growth of the mussels living in lower salinities (12·8 and 16‰) to levels nearly matching that of the high salinity animals by day 37. This suggests that acclimation of the shell growth of M. edulis to salinities in the 12·8–28·8‰ S range was achieved by the mussels during the experimental period.

Five Phytophthora species comprising a total of 243 isolates (77 P. cinnamomi, 23 P. citrophthora, 18 P. multivora, 18 P. pini, and 107 P. plurivora) were screened for sensitivity to mefenoxam, fosetyl-Al, dimethomorph, dimethomorph + ametoctradin and fluoxastrobin using amended agar assays. Mefenoxam-insensitive isolates were detected within P. cinnamomi (4%), P. multivora (11%), and P. plurivora (12%) even at approximately 2.5x the recommended label rate. These isolates were also insensitive to higher (off-label) concentrations of fluoxastrobin. Concentrations of dimethomorph (400 g/mL) and dimethomorph + ametoctradin (100 g/mL) were mostly effective in mycelial growth inhibition, but two P. plurivora isolates were insensitive, suggesting that resistance management is required. All mefenoxam-insensitive isolates were sensitive to fosetyl-Al at the label rate. Surprisingly, the populations of P. cinnamomi from mid-Atlantic oak forests included insensitive isolates. With almost all species, isolates recovered from asymptomatic hosts (e.g., soil/potting media collected of randomly selected asymptomatic hosts) had a significantly greater relative growth rate when compared to isolates recovered from symptomatic hosts (e.g., isolates recovered from lesions or wilted plants). These findings suggest that mefenoxam should no longer be used to manage oomycetes in Maryland ornamental nurseries and that the use of fluoxastrobin should be limited.

Aim: To assess the knowledge and attitude of students of dental schools in Southern India towards Basic Life Support (BLS), as well as to study the retention of BLS knowledge over time. Subjects and methods: A prevalidated questionnaire was employed, which contains 12 knowledge Questions and five questions designed to assess the students perspective towards BLS. It was hosted on Google Forms, and the URL was distributed to the Students. Results: A total of 267 responses were received, of which 66 were males and 201 were female students. Out of a maximum possible score of 12, the mean knowledge score of the group was 4.54. It was found that those who attended a BLS workshop within the last 2 years scored significantly better than those who either took it more than 2 years ago or had never undergone BLS training. Almost all participants (96%) agreed that there is a need for BLS to be learned by all dental students and hence BLS training should be part of the dental curriculum. 76.8% of respondents expressed reluctance to perform CPR on strangers. Conclusion: The findings show that the dental students who participated in the study had inadequate knowledge in BLS, but had a positive attitude towards the adoption of BLS into the dental curriculum.

During lung injury, fibroblasts migrate into the alveolar spaces where they can be exposed to pulmonary surfactant. We examined the effects of Survanta and surfactant protein A (SP-A) on fibroblast growth and apoptosis and on type I collagen, collagenase-1, and tissue inhibitor of metalloproteinase (TIMP)-1 expression. Lung fibroblasts were treated with 100, 500, and 1,000 μg/ml of Survanta; 10, 50, and 100 μg/ml of SP-A; and 500 μg/ml of Survanta plus 50 μg/ml of SP-A. Growth rate was evaluated by a formazan-based chromogenic assay, apoptosis was evaluated by DNA end labeling and ELISA, and collagen, collagenase-1, and TIMP-1 were evaluated by Northern blotting. Survanta provoked fibroblast apoptosis, induced collagenase-1 expression, and decreased type I collagen affecting mRNA stability ∼10-fold as assessed with the use of actinomycin D. Collagen synthesis and collagenase activity paralleled the gene expression results. SP-A increased collagen expression ∼2-fold and had no effect on collagenase-1, TIMP-1, or growth rate. When fibroblasts were exposed to a combination of Survanta plus SP-A, the effects of Survanta were partially reversed. These findings suggest that surfactant lipids may protect against intraluminal fibrogenesis by inducing fibroblast apoptosis and decreasing collagen accumulation.

SUMMARYBirthcoat samples from Lincoln and Welsh Mountain lambs born in an ova transfer experiment (Weiner & Slee, 1965) were examined to determine the fibre type arrays and any effect of ova transfer on manifestation of the prenatal check or the base (Dry, 1965).In the Lincoln breed arrays were Truncated Ravine or Truncated Valley thus showing central checking. Pre-curly tip fibres were mainly confined to central primary follicles. Post-natal medullation was plentiful, especially in peak curly-tips, but kemp was absent. In the Welsh lambs all identifiable arrays were either Plateau or Saddle, with the lateral primary fibres more checked than the centrals. Pre-curly tip fibres occupied almost all the primary follicles, with only a few curly-tips in primary laterals. Post-natal medullation was strongest in pre-curly tips and their successors.The effect of ova transfer was to decrease the manifestation of check in Lincoln-in-Welsh and increase it in Welsh-in-Lincoln, without affecting the base. Although this, in each case, changed the check of the lamb in the direction of that of the foster breed, it is suggested that this may not be due to any influence of maternal check on foetal check. The effect on pre-natal check may be due to foetal size, and in particular skin expansion, during a short period immediately following completion of primary follicle initiation. As compared to their respective controls, the Welsh transfer lambs had a higher S/P ratio at birth, whilst that of the transferred Lincolns was reduced.If increased prenatal fibre check is associated with increased foetal growth rate at the relevant period, it may also be correlated with increased lamb size at birth and therefore have been favoured in selection for meat production, at least in some breeds.

The trend towards shorter rotations in planted conifer stands has resulted in a reduction in the proportion of mature wood relative to juvenile core wood, raising concerns that the mechanical performance of sawn battens will be affected. The potential to improve the wood quality of the juvenile core of Sitka spruce (Picea sitchensis (Bong.) Carrière) without compromising growth rate was investigated. Rapid and inexpensive indirect methods of assessing wood properties on standing trees using a Pilodyn gun and acoustic velocity were compared with direct measurements made on wood samples cut from the juvenile core. Strong genetic correlations were observed between Pilodyn gun values and direct measures of density (–0.76) and between the square of acoustic velocity and modulus of elasticity (0.73). The genetic correlation between the square of acoustic velocity and microfibril angle was also strong (–0.84). These results suggest that indirect assessments of wood properties within juvenile core wood are sufficiently reliable for these techniques to be used in the Sitka spruce breeding programme. Although a strong negative genetic correlation between diameter at breast height and density was noted (–0.79), sufficient variation exists within the breeding population to select families with both good growth rate and high modulus of elasticity wood.

The data on the dynamics of body weight changes, absolute and average daily gains, frequency rate of increase in body weight, relative growth rate and intensity of body weight growth of Limousine and Volyn Meat breeds heifers. Both studied breeds characterized by different body weight at different age periods. Newborn Limousine breed heifers are weighed 2,7 kg more (P < 0.05) than Volyn Meat breed heifers; at 3 months age the difference was 8.5 kg (P < 0.05) at 6 months – 14.6 kg, at 9 month – 20.8 kg (P < 0.05), at 12 months – 25,6 kg (P < 0.05), at 15–months – 31.9 (P<0,05), and at 18 months – 23.5 kg. Total and average daily gains in animals of both breeds were the highest for a period of 3 to 6 months of age. In the period from birth to 15 months of age preference for average daily gains were in Limousine, however, the difference was statistically significant only for the period of 0 – 3 months and amounted to 63,9 g (P < 0,05). From 15 to 18 months of age Limousine slightly conceded to Volyn Meat breeds on this parameter. In animals of both breeds magnification of body weight increased with age, but over the entire period (from birth to 18 months) this parameter in Volyn Meat heifers was 0.6 times better than Limousine heifers. The coefficients of relative intensity and tension increase of body weight in animals of both breeds were highest in the period from birth to 3 months of age. With age, these indicators declined. Mainly, the advantage was in Volyn meat breed heifers, but the difference was not statistically significant.
