The cellular contributions of blastema and stump to 180° supernumerary limbs in the axolotl

By inverting triploid blastemas onto diploid stumps (and vice versa) the cellular contributions to supernumerary limbs so generated have been assessed. The four classes of 180° supernumerary limbs each had a different mixture of stump and blastemal cells. The mesoderm of normal supernumeraries was composed entirely or almost entirely of stump cells and were always of stump handedness. The mesoderm of symmetrical supernumeraries was of variable composition, it could be mostly stump, mostly blastema or half and half. In part normal/part symmetrical supernumeraries the normal part was usually of stump origin and the abberant symmetrical part of blastemal origin. In part normal/part inverted supernumeraries the normal part came from the stump and the inverted part from the inverted blastema. The handedness of each part of these supernumeraries corresponded with its cellular origin. The epidermis of the supernumeraries was not of the same relative composition as the mesoderm, it tended to have a larger stump component. The black/white marker was also used and this too tended not to conform to the mesodermal contribution patterns. These results are discussed in terms of rules for generating supernumeraries and it is concluded that with the exception of symmetrical supernumeraries the cellular contributions of stump and blastema determine their structure.

The hypothesis that a specialized polarizing zone controls the pattern of the anterior-posterior axis during limb development in Xenopus has been tested by analysing the cellular contribution to supernumerary limbs. Supernumerary limbs were generated by grafting hindlimb buds contralaterally between X. borealis and X. laevis to appose anterior and posterior limb tissues. Cells derived from these two species of Xenopus are readily identified by staining with quinacrine. The analysis of cellular contribution showed that supernumerary limbs consist of approximately half anterior-derived (57%) and half posterior-derived (43%) cells. These data are not consistent with the polarizing zone theory but are consistent with the hypothesis that both supernumerary limbs and normally developing limbs arise from intercalary interactions between limb bud cells with different positional values.

The origin of lipid containing cells in atheromatous lesion has been disputed. Geer in his study on atheromatous lesions of rabbit aorta, suggested that the early lesion is composed mainly of lipid-laden macrophages and the later lesion has a mixed population of macrophages and smooth muscle cells. Parker on the other hand, was able to show evidence that the rabbit lesion is primarily composed of lipid-laden cells of smooth muscle origin. The above studies and many others were done on an intact lesion without any attempt of cellular isolation previous to their ultrastructural studies. Cell isolation procedures have been established for atherosclerotic lesions through collagenase and elastase digestion Therefore this procedure can be utilized to identify the cells involved in rabbit atheroma.

В статье рассмотрена актуальная проблема перевоспитания военнослужащих в дисциплинарных частях в Российской империи как организованного и структурированного процесса. Изучено использование их потенциала для поддержания дисциплины и правопорядка среди личного состава. Определено, что к военнослужащим предусмотрено применение специальных видов уголовных наказаний, кроме того, допускается изъятие в применении отдельных наказаний к данной категории лиц. Авторами раскрывается порядок реализации наказания в виде лишения свободы в частях с воинской организацией в отношении военнослужащих, совершивших преступления. Указывается на то, что перевоспитание осужденных военнослужащих было направлено на повышение уровня их военной и строевой подготовки, принуждение их к выполнению требований военной присяги и воинских уставов. В качестве негативного момента перевоспитания осужденных военнослужащих указывается отсутствие специальной подготовки у штатных офицеров и нижних чинов дисциплинарных частей. В статье рассматривается порядок реализации наказания в виде лишения свободы в арестантских частях, а также дисциплинарных частях. Раскрыто понятие арестантских рот, их руководящий и личный состав, а также порядок содержания осужденных. Показаны особенности комплектования дисциплинарных частей как постоянным составом, так и переменным (осужденными), а также прохождения ими службы. Авторами отмечается, что в перевоспитании осужденных военнослужащих превалировала принудительная функция в ущерб нравственному воздействию. Авторы приходят к выводу о том, что дисциплинарные и штрафные части со строевой организацией в целом справляются с задачей перевоспитания осужденных военнослужащих, а опыт функционирования дисциплинарных частей царской армии был использован при создании военно-карательного аппарата Советской армии.The article deals with the actual problem of re-education of military personnel in disciplinary units in the Russian Empire as an organized and structured process. The use of their potential for maintaining discipline and law enforcement among personnel was studied. It is determined that the use of special types of criminal penalties is provided for military personnel, in addition, exceptions are allowed in the application of certain penalties to this category of persons. The author reveals the order of realization of punishment in the form of imprisonment in re-lations with the military organization in relation to the military personnel who have committed crimes. It is pointed out that the re-education of convicted servicemen was aimed at increasing the level of their military and drill training, forcing them to fulfill the requirements of the military oath and military regulations. As a negative aspect of the re-education of convicted servicemen, the lack of special training of regular officers and lower ranks of disciplinary units is indicated. The article deals with the procedure for the implementation of punishment in the form of im-prisonment in prison units, as well as disciplinary units. The concept of convict companies, their leadership and personnel, as well as the order of detention of convicts is revealed. The peculiarities of completing disciplinary units with both permanent and variable composition (convicts), as well as their service are shown. The author notes that in the re-education of convicted servicemen, forced functioning prevailed to the detriment of moral influence. The author comes to the conclusion that disciplinary and penal units with drill organization in General cope with the task of re-education of convicted servicemen, and the experience of functioning of disciplinary units of the tsarist army was used in the creation of the military punitive apparatus of the Soviet army.

Abstract Rhabdoid tumors (RT) are rare but highly aggressive pediatric neoplasms. These tumors carry homozygous loss-of-function alterations of SMARCB1 in almost all cases with an otherwise low mutational load. RT arise at different intracranial (ATRT) as well as extracranial (MRT) anatomical sites. Three main molecular subgroups (ATRT-SHH, ATRT-TYR, ATRT-MYC) have been characterized for ATRT which are epigenetically and clinically diverse, while MRT show remarkable similarities with ATRT-MYC distinct from ATRT-SHH and ATRT-TYR. Even though there are hypotheses about various cells of origin among RT subgroups, precursor cells of RT have not yet been identified. Previous studies on the temporal control of SMARCB1 knockout in genetically engineered mouse models have unveiled a tight vulnerable time frame during embryogenesis with regard to the susceptibility of precursor cells to result in RT. In this study, we employed single-cell RNA sequencing to describe the intra- and intertumoral heterogeneity of murine ATRT-SHH and -MYC as well as extracranial MYC tumor cells. We defined subgroup-specific tumor markers for all RT classes but also observed a notable overlap of gene expression patterns in all MYC subgroups. By comparing these single-cell transcriptomes with available single-cell maps of early embryogenesis, we gained first insights into the cellular origin of RT. Finally, unsupervised clustering of published human RT methylation data and healthy control tissues confirmed the existence of different cells of origin for intracranial SHH tumors and MYC tumors independent of their anatomical localizations.
