Regeneration from mesenteric arteries in short-term culture

The investigations which are described here were originally undertaken in order to investigate the pathogenesis of early hypertension at a time when no morphological changes were evident in adrenergic arteries on light microscopy. Evidence of functional change was therefore sought by comparing the growth patterns of cells from hypertensive and normal mesenteric arteries in culture. The study of the cells which migrated from the explants form the basis for this paper. Month-old Wistar laboratory-strain albino rats were used for the experiment. Young rats were deliberately selected, as fat could be more cleanly dissected away from vessel walls. Also: ‘Tissues from old animals are often more difficult to grow than tissues from young animals and exhibit a longer lag period before growth commences’ (Paul, 1965a). Each week, over a period of 14 weeks, hypertension was induced in a group of rats according to the method of Loomis (1946).

Objective: Acute anorexia nervosa (AN) leads to reduced gray (GM) and white matter (WM) volume in the brain, which however improves again upon restoration of weight. Yet little is known about the extent and clinical correlates of these brain changes, nor do we know much about the time-course and completeness of their recovery. Methods: We conducted a meta-analysis and a qualitative review of all magnetic resonance imaging studies involving volume analyses of the brain in both acute and recovered AN. Results: We identified structural neuroimaging studies with a total of 214 acute AN patients and 177 weight-recovered AN patients. In acute AN, GM was reduced by 5.6% and WM by 3.8% compared to healthy controls (HC). Short-term weight recovery 2–5 months after admission resulted in restitution of about half of the GM aberrations and almost full WM recovery. After 2–8 years of remission GM and WM were nearly normalized, and differences to HC (GM: –1.0%, WM: –0.7%) were no longer significant, although small residual changes could not be ruled out. In the qualitative review some studies found GM volume loss to be associated with cognitive deficits and clinical prognosis. Conclusions: GM and WM were strongly reduced in acute AN. The completeness of brain volume rehabilitation remained equivocal.

To assess the functional change in adenylyl cyclases (AC) associated with the diabetic state, we investigated AC-mediated relaxations and cAMP production in mesenteric arteries from rats with streptozotocin (STZ)-induced diabetes. The relaxations induced by the water-soluble forskolin (FSK) analog NKH477, which is a putative AC5 activator, but not by the β-adrenoceptor agonist isoproterenol (Iso) and the AC activator FSK, were reduced in intact diabetic mesenteric artery. In diabetic rats, however, Iso-, FSK-, and NKH477-induced relaxations were attenuated in the presence of inhibitors of nitric oxide synthase and cyclooxygenase. To exclude the influence of phosphodiesterase (PDE), we also examined the relaxations induced by several AC activators in the presence of 3-isobutyl-1-methylxanthine (IBMX; a PDE inhibitor). Under these conditions, the relaxation induced by Iso was greatly impaired in STZ-diabetic rats. This Iso-induced relaxation was significantly attenuated by pretreatment with SQ-22536, an AC inhibitor, in mesenteric rings from age-matched controls but not in those from STZ-diabetic rats. Under the same conditions, the relaxations induced by FSK or NKH477 were impaired in STZ-diabetic rats. Neither FSK- nor A-23187 (a Ca2+ ionophore)-induced cAMP production was significantly different between diabetics and controls. However, cAMP production induced by Iso or NKH477 was significantly impaired in diabetic mesenteric arteries. Expression of mRNAs and proteins for AC5/6 was lower in diabetic mesenteric arteries than in controls. These results suggest that AC-mediated relaxation is impaired in the STZ-diabetic rat mesenteric artery, perhaps reflecting a reduction in AC5/6 activity.

Seasonal growth patterns of mature loblolly pine (Pinustaeda L.) trees over the interval 1988–1993 have been analyzed to evaluate the effects of ambient ozone on growth of large forest trees. Patterns of stem expansion and contraction of 34 trees were examined using serial measurements with sensitive dendrometer band systems. Study sites, located in eastern Tennessee, varied significantly in soil moisture, soil fertility, and stand density. Levels of ozone, rainfall, and temperature varied widely over the 6-year study interval. Regression analysis identified statistically significant influences of ozone on stem growth patterns, with responses differing widely among trees and across years. Ozone interacted with both soil moisture stress and high temperatures, explaining 63% of the high frequency, climatic variance in stem expansion identified by stepwise regression of the 5-year data set. Observed responses to ozone were rapid, typically occurring within 1–3 days of exposure to ozone at ≥40 ppb and were significantly amplified by low soil moisture and high air temperatures. Both short-term responses, apparently tied to ozone-induced increases in whole-tree water stress, and longer term cumulative responses were identified. These data indicate that relatively low levels of ambient ozone can significantly reduce growth of mature forest trees and that interactions between ambient ozone and climate are likely to be important modifiers of future forest growth and function. Additional studies of mechanisms of short-term response and interspecies comparisons are clearly needed.

Anabolic androgenic steroids (AAS) are used by some athletes to enhance performance despite the health risk they may pose in some persons. This work was carried out to evaluate the possible structural and functional alterations in the heart using two-dimensional, M-mode, tissue Doppler imaging (TDI) and strain rate imaging (SRI) in athletes using supraphysiological doses of AAS. Additionally, the histological and ultrastructural changes in cardiac muscles of adult albino rats after injection of sustanon, as an example of AAS, were studied. Fifteen male bodybuilders using anabolic steroids constituted group 1, five male bodybuilders who are not using anabolic steroids constituted group 2, and five nonathletic males constituted negative control group (group 3). They were investigated by two-dimensional, M-mode, TDI and SRI. This study was performed on 30 adult albino rats. They were divided into two groups. Group I (Control group) (10) was subdivided into negative control, subgroup 1a (5), and subgroup 1b (5), which received 0.8 ml olive oil intramuscular once a week for 8 weeks. Group II (Experimental group) (20) received sustanon 10 mg/kg intramuscularly once a week for 8 weeks. The heart specimens were prepared for light microscopy and transmission electron microscopy. Echocardiographic results showed that bodybuilders who use steroids have smaller left ventricular dimension with thicker walls, impaired diastolic function, as well as higher peak systolic strain rate in steroid-using bodybuilders as compared to the other two groups. Light microscopy examination of cardiac muscle fibers showed focal areas of degeneration with loss of striations and vacuolation in the experimental group. Ultrastructural examination showed disturbance of the banding pattern of the cardiac muscle fiber with disintegration, loss of striations, dehiscent intercalated disc, and interrupted Z-bands. Administration of supraphysiological doses of AAS caused severe deleterious effects in the myocardium both in athletes and in experimental animals. The SRI shows promise in the early detection of systolic dysfunction in those athletes who use steroids.
