An ascospore germination method was developed and validated to assess the sensitivity of bulk samples of Mycosphaerella fijiensis to trifloxystrobin. Using this method, the sensitivity of 142 ascospore samples from banana plantations not treated with strobilurins was analyzed to establish a baseline of pathogen sensitivity. A bulk method was utilized for monitoring purposes because it avoids potential complications due to the isolation and propagation of single-spore isolates and enables the testing of larger samples. Following intensive use of strobilurins (6 to 11 applications per year) over 4 years, under conditions of high disease pressure and the absence of sanitary measures at a development site in Costa Rica, bulk samples with 50% effective concentration (EC50) resistance factors (RFs) in excess of 500 compared with the mean baseline sensitivity were detected. Single-ascospore isolates derived from spores germinating at the discriminatory dose of 3 μg/ml were also resistant, suggesting that the frequency of resistant individuals in bulk samples could be estimated from the relative numbers of ascospores growing at this dose. The resistance of selected isolates was confirmed in planta. In vitro tests with four resistant and two sensitive single-ascospore isolates collected from different locations and times indicated possible cross-resistance of trifloxystrobin to azoxystrobin, famoxadone, and fenamidone, but not to propiconazole.

Penelitian dilaksanakan untuk menambah wawasan, pengalaman teknis, keterampilan kerja, manajerial, dan secara khusus mempelajari serta menganalisis pengelolaan perawatan tanaman dan buah pisang cavendish. Penelitian dilaksanakan di Lampung pada bulan Februari hingga Juni 2017. Metode yang digunakan untuk mendapat data primer dengan cara mengikuti dan mengamati kegiatan teknis di lapang dan wawancara. Data sekunder diperoleh dari laporan dan arsip kebun. Hasil penelitian menunjukkan kualitas buah pisang dipengaruhi pemangkasan daun, penyuntikan jantung, pembrongsongan, pembuangan bunga, pembuangan buah, pembuangan penghalang buah, dan pemasangan sekat buah. Penyakit pisang yang menjadi kendala di lokasi penelitian adalah Black Leaf Streak (BLS) yang disebabkan oleh Mycosphaerella fijiensis dan banana freckle yang disebabkan oleh Guignardia musae. Kehilangan buah ekspor terbesar karena kondisi No Functional Leaf (NFL) karena Banana Freckle. Blok dengan kondisi NFL terbesar adalah blok 27A dengan jumlah 7420 tanaman. Indeks tenaga kerja sebesar 2.08.

La sigatoka negra (Mycosphaerella fijiensis Morelet) es la enfermedad más limitante del cultivo del banano a escala mundial, su severidad se magnifica a niveles epidémicos y aumenta con el monocultivo de cultivares y clones genéticamente uniformes. En Colombia ocasiona pérdidas en regiones exportadoras de Urabá y Magdalena donde su manejo depende del uso de agroquímicos, sin lograr su erradicación. En la búsqueda de métodos para reducir la presión del patógeno en la zona bananera del departamento del Magdalena–Colombia, se determinó la relación entre el clima, propiedades edáficas y la incidencia de la enfermedad. Se utilizaron los sistemas de información geográfica, se digitalizaron los datos de las variables edáficas reportadas en el estudio de suelos IGAC (2009), los registros de las estaciones meteorológicas existentes en la región y la información de incidencia del patógeno. Como resultado se establecieron 3 áreas (alta, media y baja incidencia) y se evidenció correlación positiva entre incidencia de la enfermedad con precipitación (r=0,56), magnesio intercambiable Mg+2 (r=0,45), microporosidad (r= 0,40), el contenido de arcilla (r= 0,54) y la evaporación (r= 0,51), se observó que existen condiciones edáficas que inciden en la presencia de la enfermedad, variables a tener en cuenta en el manejo del cultivo.
