AbstractTwelve mature and six 2-year-old Arabian horses were used to determine the effect of dietary long-chain polyunsaturated omega-3 fatty acid supplementation on plasma fatty acids and lameness. Lameness scores and stride lengths were measured on day 0. Horses were striated and pair-matched according to age, gender, stride length and, for mature horses, lameness score, and each horse was fed either a treatment diet containing 5.95 g of stabilized omega-3 fatty acids plus a fat carrier (FA), for a total of 19.4 g fat, or a control diet containing 49 g of corn oil (CO) for 75 days. Horses were exercised 5 d week− 1, and blood samples were drawn and body weights recorded on days 0, 25, 50 and 75. Lameness scores and stride lengths were recorded again on day 75. Total plasma omega-3 fatty acid concentrations were higher on all days in FA horses than in CO horses. Total plasma omega-6 fatty acids increased from days 0 to 25, remained elevated through day 50 and returned to baseline on day 75 in all horses. The ratio of plasma omega-6:omega-3 fatty acids was lower in FA horses. Horses on FA had increased plasma docosahexaenoic acid (DHA) on days 25, 50 and 75. No difference in walk stride length was noted; however, FA horses tended to have a longer trot stride after supplementation when compared with CO horses. No differences were seen in prostaglandin E2 (PGE2) metabolite or tumour necrosis factor-α as measured in blood serum. In summary, supplementing omega-3 fatty acids increases plasma DHA, although there was no overall increase in omega-3 in FA horses. While a trend to increase trot stride length was seen, no differences in lameness scores between treatments were noted.

Diets high in saturated fatty acids (SFAs) are associated with the development of circadian dysregulation, obesity, and Type 2 diabetes mellitus. Conversely, polyunsaturated fatty acids (PUFAs) have recently been identified to improve insulin sensitivity, reduce weight gain, and relieve obesity-induced inflammation. While saturated fatty acids, such as the prevalent dietary fatty acid palmitate, have been implicated in circadian disruption, there is a paucity of studies regarding the effects of PUFAs on circadian parameters. Therefore, the immortalized murine neuronal model, mHypoE-37, was utilized to examine the effects of the SFA palmitate and omega-3 PUFA docosahexaenoic acid (DHA) on circadian rhythms. The mHypoE-37 neurons express the core clock genes, Bmal1, Per2, and Rev-erbα, in a circadian manner. 25 μM of palmitate significantly increased the transcriptional expression of Bmal1, without altering the expression of inflammatory markers TLR4, IκBα, and IL-6, nor the orexigenic neuropeptide AgRP, suggesting that the observed disruption of the molecular clock is the result of a mechanism distinct from that of hypothalamic cellular inflammation. Furthermore, treatment with the PUFA DHA resulted in alterations in the circadian expression profile of Bmal1, although differentially from the effects of palmitate. In the presence of DHA, the disruptive effects of palmitate on Bmal1 were less pronounced, suggesting a protective effect of DHA. These studies are the first to identify the potential for omega-3 PUFAs to protect against palmitate-mediated dysregulation of circadian parameters and will ultimately improve the understanding of circadian control mechanisms.

Feeding docosahexaenoic acid to pigs reduces triglycerides in blood and induces gene expression for fat oxidation in liver and adipose but not in muscle. Can. J. Anim. Sci. 91: 601–612. The essential fatty acids required in diets of humans are linoleic acid (18:2n-6:LA) and α-linolenic acid (18:3n-3: ALA), and these can be elongated and desaturated to form long-chain omega-6 or omega-3, respectively. Even though not considered essential, consumption of long-chain omega-3 fatty acid is recommended for health benefits, including protection against cardiovascular disease. The omega-3 fatty acid, docosahexanoic acid (DHA), was supplemented in pig diets as a dried biomass of the microalgae Schizochytrium to see if there are unique physiological changes associated with DHA feeding. Pigs were fed a diet with 330 mg (low), 3600 mg (medium) or 9400 mg (high) DHA per day for the last 25 d before slaughter at market weight (∼110 kg). Blood triglycerides (TG) were assayed colormetrically and tissue samples were analyzed for gene expression patterns of RNA by quantitative real-time polymerase chain reaction. Fatty acids were analyzed by gas chromatography. Animal performance appeared to increase with DHA, as shown by a 14% improved feed:gain ratio of 2.74±0.27 (P<0.05). Blood triglycerides were reduced significantly from 0.40±0.23 mM to 0.20±0.09 mM. Pigs accumulated 14 times more DHA in their subcutaneous fat (SQ) (10.67 mg g−1) on the high diet compared with the control diet (0.75 mg g−1). Gene analysis showed that the expression of the fat oxidation biomarkers acyl CoA oxidase 1 (ACOX1), peroxisome proliferator-activated receptors alpha (PPARα) and gamma (PPARγ) were stimulated in the SQ and liver. The delta-6 desaturase (D6D) and elongase (Elov5), which are genes involved in the endogenous synthesis of DHA, were unchanged. Fatty acid synthase (FASN) was stimulated in the liver and muscle of pigs on the high DHA diet. Analysis of gene transcription activity suggested fat metabolism was stimulated in the liver and SQ fat, but the genes involved in the endogenous production of DHA remained unchanged.

Bryophytes (mosses, liverworts and hornworts) often produce high amounts of very long-chain polyunsaturated fatty acids (vl-PUFAs) including arachidonic acid (AA, 20:4 △5,8,11,14) and eicosapentaenoic acid (EPA, 20:5 △5,8,11,14,17). The presence of vl-PUFAs is common for marine organisms such as algae, but rarely found in higher plants. This could indicate that bryophytes did not lose their marine origin completely when they landed into the non-aqueous environment. Vl-PUFA, especially the omega-3 fatty acid EPA, is essential in human diet for its benefits on healthy brain development and inflammation modulation. Recent studies are committed to finding new sources of vl-PUFAs instead of fish and algae oil. In this review, we summarize the fatty acid compositions and contents in the previous studies, as well as the approaches for qualification and quantification. We also conclude different approaches to enhance AA and EPA productions including biotic and abiotic stresses.

Background: High oxidative stress, which is caused by smoking, can alter omega-3 fatty acid concentrations. Since omega-3 fatty acids play a role in dopaminergic neurotransmission related to dependence, it is important to understand their effects on nicotine dependence. Methods: This research comprised 2 studies. The first one consisted of a cross-sectional evaluation, in which the levels of the most important omega-3 fatty acids, eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), were compared between smokers and non-smokers in a sample of 171 individuals; of them, 120 were smokers and 51 were non-smokers. The other study was a clinical, double-blind, randomized, placebo controlled, in which 63 smokers received daily treatment with capsules of fish oil (a source of omega-3/3 g/day) or mineral oil (used as placebo, also 3 g/day), taken 3 times a day for 90 days. Each fish oil capsules contained approximately 210.99 mg EPA and 129.84 mg of DHA. The outcome was evaluated by means of psychometric and biological measures as well as self-reports of tobacco use. The evaluations were carried out at the beginning of treatment and once a month thereafter (total of 4 times). Outcomes: The omega-3 fatty acid lipid profile showed that smokers present lower concentrations of DHA. After treatment, the omega-3 group showed a significant reduction in their levels of dependence. Interpretation: Smokers showed lower peripheral levels of omega-3, and treatment with the most important omega-3 fatty acids brought about a reduction in nicotine dependence.
