Criticisms addressed to the historiographical paradigm of the so-called ‘three quests’ by several scholars with different ideological backgrounds and who have worked independently have debunked it by proving its untenable character. The present paper makes a proposal for a new paradigm which allows us to understand the quest of the historical Jesus in a more comprehensive, lucid and explanatory way. This proposal has been articulated through a set of theses, accompanied in each case by an explanation (and, sometimes, by corollaries), accomplishing a threefold task: a broader characterization of historical Jesus research, a summary discussion of the main distortions contained in the ‘three-quests’ model, and, more importantly, an exposition of the basic principles of a new paradigm.

It has been 70 years since the name inhibin was used to describe a gonadal factor that negatively regulated pituitary hormone secretion. The majority of this period was required to achieve purification and definitive characterization of inhibin, an event closely followed by identification and characterization of activin and follistatin (FS). In contrast, the last 15–20 years saw a virtual explosion of information regarding the biochemistry, physiology, and biosynthesis of these proteins, as well as identification of activin receptors, and a unique mechanism for FS action—the nearly irreversible binding and neutralization of activin. Many of these discoveries have been previously summarized; therefore, this review will cover the period from the mid 1990s to present, with particular emphasis on emerging themes and recent advances. As the field has matured, recent efforts have focused more on human studies, so the endocrinology of inhibin, activin, and FS in the human is summarized first. Another area receiving significant recent attention is local actions of activin and its regulation by both FS and inhibin. Because activin and FS are produced in many tissues, we chose to focus on a few particular examples with the most extensive experimental support, the pituitary and the developing follicle, although nonreproductive actions of activin and FS are also discussed. At the cellular level, it now seems that activin acts largely as an autocrine and/or paracrine growth factor, similar to other members of the transforming growh factor β superfamily. As we discuss in the next section, its actions are regulated extracellularly by both inhibin and FS. In the final section, intracellular mediators and modulators of activin signaling are reviewed in detail. Many of these are shared with other transforming growh factor β superfamily members as well as unrelated molecules, and in a number of cases, their physiological relevance to activin signal propagation remains to be elucidated. Nevertheless, taken together, recent findings suggest that it may be more appropriate to consider a new paradigm for inhibin, activin, and FS in which activin signaling is regulated extracellularly by both inhibin and FS whereas a number of intracellular proteins act to modulate cellular responses to these activin signals. It is therefore the balance between activin and all of its modulators, rather than the actions of any one component, that determines the final biological outcome. As technology and model systems become more sophisticated in the next few years, it should become possible to test this concept directly to more clearly define the role of activin, inhibin, and FS in reproductive physiology.

Sphingolipid metabolism is tightly controlled by enzymes to regulate essential processes such as energy utilisation and cell proliferation. The central metabolite is ceramide, a pro-apoptotic lipid catabolized by ceramidase enzymes to ultimately produce pro-proliferative sphingosine-1-phosphate. Human ceramidases can be soluble proteins (acid and neutral ceramidase) or integral membrane proteins (alkaline ceramidases). Increasing ceramide levels to increase apoptosis has shown efficacy as a cancer treatment using small molecules inhibiting a soluble ceramidase. Due to the transmembrane nature of alkaline ceramidases, no specific small molecule inhibitors have been reported. Here, we report novel fluorescent substrates (FRETceramides) of ceramidases that can be used to monitor enzyme activity in real-time. We use FRETceramides to discover the first drug-like inhibitors of alkaline ceramidase 3 (ACER3) which are active in cell-based assays. Biophysical characterization of enzyme:inhibitor interactions reveal a new paradigm for inhibition of lipid metabolising enzymes with non-lipidic small molecules.

SummaryThe use of the concept of an exclusive Economie zone has increased since the adoption of the United Nations Convention on the Law of the Sea. However, the characterization of this zone varies greatly between States. This article presents an exhaustive survey of the concept of an exclusive Economie zone. The author discusses the types of jurisdiction exercised by States in their uses of an exclusive Economie zone. Disparity exists between the provisions of the Convention and State practice in some specific areas: for example, the provisions on the environment and on scientific research. Despite these exceptions, the author maintains that the basic tenets of the Convention are respected in State practice. State declarations as well as arbitral and judicial decisions show that the Convention and State practice are together evolving to reinforce the basic principles of the concept of an exclusive Economie zone.

The relationship of the preaching and teaching of Jesus to apocalyptic has been a vexed one ever since Albert Schweitzer's assault on the liberal “Lives of Jesus” and his advocacy of consistent eschatology along with his characterization of Jesus’ teachings as interim ethics. While many of the details of Schweitzer's hypothesis failed to be persuasive, his insistence that Jesus’ activity be seen in the context of apocalypticism has made a profound impact on subsequent historical Jesus scholarship and, in spite of his own noncommital stance with regard to the Two Document Hypothesis, on the theological characterization of Q.

DAPK (death-associated protein kinase) is a newly recognized member of the mammalian family of ROCO proteins, characterized by common ROC (Ras of complex proteins) and COR (C-terminal of ROC) domains. In the present paper, we review our recent work showing that DAPK is functionally a ROCO protein; its ROC domain binds and hydrolyses GTP. Furthermore, GTP binding regulates DAPK catalytic activity in a novel manner by enhancing autophosphorylation on inhibitory Ser308, thereby promoting the kinase ‘off’ state. This is a novel mechanism for in cis regulation of kinase activity by the distal ROC domain. The functional similarities between DAPK and the Parkinson's disease-associated protein LRRK2 (leucine-rich repeat protein kinase 2), another member of the ROCO family, are also discussed.

Abstract Ancestral sequence reconstruction (ASR) is the calculation of ancient protein sequences on the basis of extant ones. It is most powerful in combination with the experimental characterization of the corresponding proteins. Such analyses allow for the study of problems that are otherwise intractable. For example, ASR has been used to characterize ancestral enzymes dating back to the Paleoarchean era and to deduce properties of the corresponding habitats. In addition, the historical approach underlying ASR enables the identification of amino acid residues key to protein function, which is often not possible by only comparing extant proteins. Along these lines, residues responsible for the spectroscopic properties of protein pigments were identified as well as residues determining the binding specificity of steroid receptors. Further applications are studies related to the longevity of mutations, the contribution of gene duplications to enzyme functionalization, and the evolution of protein complexes. For these applications of ASR, we discuss recent examples; moreover, we introduce the basic principles of the underlying algorithms and present state-of-the-art protocols.

One of the less controversial points among Jesus scholars is the importance of Capernaum to the historical Jesus, variously described as his ‘hub,’ ‘headquarters,’ ‘centre,’ etc. This article instead suggests that the importance of Capernaum may be understood as a specific to Mark’s depiction of Jesus and that Mark’s redactional interest in Capernaum prematurely treated as a datum concerning the historical Jesus. Indeed, exegetical insights about Mark’s interest in Galilee have more recently developed into arguments that the Second Gospel was composed somewhere in that region. This article will survey Mark’s characterization of the region to not only argue that Capernaum is a distinctively Markan point of interest, but that there is ample reason to believe that the Gospel was composed in that village.

AbstractInternet-based scientific communities promise a means to apply distributed, diverse human intelligence towards previously intractable scientific problems. However, current implementations have not allowed communities to propose experiments to test all emerging hypotheses at scale or to modify hypotheses in response to experiments. We report high-throughput methods for molecular characterization of nucleic acids that enable the large-scale videogame-based crowdsourcing of functional RNA sensor design, followed by high-throughput functional characterization. Iterative design testing of thousands of crowdsourced RNA sensor designs produced near-thermodynamically optimal and reversible RNA switches that act as self-contained molecular sensors and couple five distinct small molecule inputs to three distinct protein binding and fluorogenic outputs—results that surpass computational and expert-based design. The paper reveals the current problems of production systems, their lack of readiness to function in the conditions of application and development of NBIC technologies. The methodological basis of the research consists of fundamental works and modern scientific and practical developments of national and foreign scientists on the problems of creation and functioning of production, technological and socio-economic systems in the conditions of development using NBIC technologies. The research was carried out using such general scientific and special methods as: historical-dialectical and abstract-logical; comparative-analytical and system analysis; synthesis of scientific approaches. The authors prove the need to create a new system paradigm. The new paradigm of production systems that realize the potential of NBIC technologies is based on a concept that finds its concrete expression, firstly, in the initial or original concepts, and secondly, in the system of basic principles, laws, or sufficiently plausible hypotheses. It is these elements that make up the conceptual core of the paradigm, and efforts should be directed to their search and justification when forming a new paradigm. The paper analyzes the main methods and tools for implementing a new approach to the design of production systems.
