The effect of a weight-reducing diet on the nitrogen metabolism of obese adolescents

Rates of whole body amino nitrogen flux were measured in 16 obese adolescents undergoing weight reduction with a high protein low energy diet. The subjects received approximately 2.5 g of animal protein per day per kilogram ideal body weight and maintained nitrogen balance throughout the 18 days on the diet. Flux rates were calculated separately from the cumulative excretion of 15N in urinary ammonia and urea following the administration of a single dose of [15N]glycine. The pattern of 15N label appearance in urinary ammonia and urea nitrogen was followed for 72 h after the administration of [15N]glycine. Significant amounts of label continued to be excreted in both urinary ammonia and nitrogen for 36–48 h after label administration. The weight-reducing diet accelerated 15N cumulative excretion in urinary urea, but not in ammonia nitrogen compared with the control diet. Whole body nitrogen flux rates increased rapidly and significantly on the diet. Using the urea end product, this increase was evident on the 4th diet day, but not by the 7th or subsequent days. On the other hand, using the ammonia end product, flux rate increased markedly (p < 0.0001) and remained elevated throughout the whole study. Our results demonstrate adaptive changes in whole body amino-nitrogen metabolism in response to the reducing diet. Different patterns of change are seen depending upon whether an ammonia or a urea end product is used. Our data thus add to the evidence for compartmentation of the body's amino-nitrogen pools.

Papain and trypsin partially hydrolyzed wheat gluten dispersed in 10% sodium salicylate, and pepsin partially hydrolyzed gluten dispersed in aluminium lactate. Trypsin was considerably less effective than either papain or pepsin. Papain produced non-protein nitrogen most rapidly during early stages of hydrolysis but pepsin produced the largest total after 48 h. The rate of release of terminal amino nitrogen was similar for papain and pepsin, and release was not complete after 48 h. The mean size of polypeptides was larger in papain than in pepsin hydrolysates during early stages of hydrolysis but definitely smaller after 48 h. Only pepsin released appreciable amounts of free amino acid nitrogen. Leucine and phenylalanine made up over 50% of the free amino acids after 48 h but several other amino acids were found in significant quantities.

For wine quality management, nitrogen metabolism should be considered as a key process in the system “grape – wine.” Nitrogen is one of the dominant elements a grapevine receives from the soil. It is important in many biological processes of the plant itself and of the microorganisms involved in fermentation. Nitrogen-containing compounds are nutrients necessary for yeast growth, in particular, for stable fermentation. This group of compounds directly and indirectly affects the aromatic and taste qualities of wine during its maturation and largely determines its stability. Nitrogen compounds are transferred to wine directly from grapes and yeast during fermentation. Since their role in the formation and maturation of wine is significant, it is highly important to regulate their metabolism in the fermenting must. On analysing literature references and summarising the information on the metabolism of nitric substances, a scheme has been developed reflecting how these substances influence the formation of the quality characteristics of grape wines. The paper presents the results of studying the metabolism of total nitrogen and amino nitrogen in grape must during its fermentation. The grape variety considered in the research was Sukholimansky White bred by the National Science Centre “Tairov Institute of Viticulture and Winemaking” and harvested in 2015–2017. It has been established that nitrogen metabolism during fermentation does not depend on the feedings added. However, the yeast race affects the physicochemical parameters, namely the content of volatile acids. It has been observed that during fermentation, the amine nitrogen concentration decreases by 90% and the total nitrogen concentration by 40–50%. Regarding the factors that effect on the quality characteristics of wines produced in the South of Ukraine, the physicochemical parameters of wine materials can be improved by using the active dry yeast Vitilevure Quartz and the nutritional supplements Actiferm 1 and Actiferm 2, in combination with aeration. This allows revealing fruity aromas, and achieving the right acidity and harmony of taste due to the presence of the descriptors (butter, apple, peach, apricot, and geranium) characteristic of the grape variety Sukholimansky White.

C-type natriuretic peptide (CNP) is a newly identified peptide that is structurally related to atrial (ANP) and brain natriuretic peptide (BNP). CNP exists as a 22-amino acid peptide and like ANP and BNP has a 17-amino acid ring formed by a disulfide bond. Unlike these two previously identified cardiac peptides, CNP lacks the COOH-terminal amino acid extension from the ring structure. ANP, BNP and CNP decrease cardiac preload, but unlike ANP and BNP, CNP is not natriuretic. While ANP and BNP have been localized to the heart, recent investigations have failed to detect CNP mRNA in the myocardium although small concentrations of CNP are detectable in the porcine myocardium. While originally localized to the brain, recent investigations have localized CNP to endothelial cells consistent with a paracrine role for CNP in the control of vascular tone. While CNP has been detected in cardiac tissue by radioimmunoassay, no studies have demonstrated CNP localization in normal human heart by immunoelectron microscopy.

ABSTRACT A protein isolated from ovine pituitary glands has been purified, and its homogeneity assessed by NH2- and COOH-terminal amino acid determination, ultracentrifugation studies, and polyacrylamide gel electrophoresis after carboxymethylation. Its chemical and immunochemical properties are closely similar to those of beef and pork neurophysins, less similar to those of human neurophysins. It contains no tryptophan (like other neurophysins) or histidine (like all except bovine neurophysin-I and human neurophysins). It has alanine at the NH2-terminus and valine at the COOH-terminus. Its amino acid composition is similar to, but not identical with those of porcine and bovine neurophysins.
