Ionizing radiation has been applied in several areas of knowledge, among them the study of the radiomodifier activity of natural substances. These substances can modify the cellular response to the damage induced by the radiation. Therefore, this work aimed to evaluate the radiomodifier action of Pityrocarpa moniliformis extract on Biomphalaria glabrata embryos exposed to 60Co gamma radiation. Initially, toxicity tests were performed on the extract against the B. glabrata embryos for the choice of concentration that did not cause death and embryonic malformation. Then, the antioxidant activity of the P. moniliformis extract with flavonoids and phenolic compounds was evaluated by means of the ABTS method. To evaluate the radiomodifier activity of the extract, embryos were selected in the blastula stage and irradiated with 7.5 Gy in a 60Co source (gammacell-Co60). Then, the embryos were exposed for 24 h to the extract of P. moniliformis at a concentration of 250 μg/mL. The results showed that the extract of P. moniliformis presents flavonoids and enzymatic inhibition by ABTS, which demonstrates the presence of antioxidant compounds. However, the tests of the radiomodifier activity did not present radioprotective effect for embryos exposed to ionizing radiation.

Cyanide (CN) through leaching process in the gold processing could produce tailings that have negative impacts on the aquatic environment. To determine the acute and chronic effects of cyanide on aquatic organisms, toxicity tests have been conducted in the laboratory using Daphnia sp.This zooplankton organism is recommended by APHA as standard test animals. In this study the test concentrations refer to the Ministerial Decree No. 202 of 2004 and Government Regulation No. 82 of 2001. Cyanide was analyzed as free CN and WADS CN. The result of acute toxicity test using cyanide solution showed that Daphnia could survive a maximum concentration of 0.2 ppm. LC50 values for 24 and 72 hours were 0,59 mg/L and 0,10 mg/l, respectively. The LT50 was found at 42 hours. Daphnia sp. produced different numbers of offspring at different CN concentrations, and changed their reproduction pattern from parthenogenesis to sexual reproduction after exposure to a cyanide concentration of 0.1 ppm for 24 hours©  Sianida (CN) melalui proses leaching dalam proses pengolahan emas menghasilkan limbah yang dapat memberi dampak negatif bagi lingkungan perairan. Zooplankton ini direkomendasikan oleh APHA sebagai salah satu hewan uji standar. Dalam penelitian ini konsentrasi uji yang dipakai mengacu pada peraturan yang berlaku, yaitu Keputusan Menteri No. 202, Tahun 2004, dan Peraturan Pemerintah No. 82, Tahun 2001. Analisis sianida yang dihitung adalah nilai free CN dan WADS CN. Hasil penelitian uji toksisitas akut dengan menggunakan larutan sianida didapati Daphnia mampu bertahan hidup sampai pada konsentrasi 0,2 ppm dan nilai LC50 berada pada konsentarsi 0,1 ppm serta LT50 pada jam ke 42. Hasil uji efek kronis, Daphnia sp. menghasilkan jumlah anakan yang berbeda pada konsentrasi CN yang berlainan, dan mengalami perubahan pola reproduksi dari partenogenesis menjadi seksual setelah dipaparkan pada kosentrasi sianida 0,1 ppm selama 24 jam©

Metallic elements copper (Cu), zinc (Zn), nickel (Ni) and manganese (Mn) are some of the most commonly found in water and sediment samples collected from the Danube - Danube Delta. These elements are important as essential micronutrients, being normally present at low concentrations in biological organisms, but in high concentrations they become toxic with immediate and delayed effects. The role of this metals is still controversial, that�s why bioconcentration potential is so important. In this non-clinical study, we tested in vitro effect of heavy metals on carp, Cyprinus carpio, reproducing in vivo presence of Cu, Zn, Ni and Mn in the Romanian�s surface water. The toxicity tests were performed according to OECD 203 by detecting the average (50%) lethal concentration - LC50 on aquatic organisms (freshwater fish) at 96h. The results pointed out that, copper value for LC 50 at 96h was estimated as 3.4 mg/L (concentrations tested in the range of 0.1 - 4.75 mg/L). Zinc value for LC 50 at 96h was estimated as 20.8 mg/L (concentrations tested in the range of 0.028 � 29.6 mg/L). Nickel value for LC 50 at 96h was estimated as 40.1 mg/L (concentrations tested in the range of 0.008 - 84.5 mg/L). For manganese the mortality effects has recorded at LC 50 at 96h at estimated value higher than 53 mg/L (concentrations tested in the range of 0.04 - 53.9 mg/L). The accuracy of the testing metals concentration was insured by the screening of the dilution water, as well as food and control fish, acclimated in laboratory conditions.

Abstract The primary goal of this study was to investigate the possibility of using early life stages of white sturgeon (Acipenser transmontanus) (eggs, larvae and fry) as a species relevant to the Fraser River, B.C., for the acute and sublethal toxico-logical testing of forest industry effluents. Here we report the first successful acute toxicity tests for 8-day-old larvae and 42-day-old fry exposed to several chemicals known to be released into the Fraser River (i.e., 6 monochlorovanillin [6 MVAN], 4,5 dichloroguaiacol [4,5 DCG], 4,5 dichlorocatechol [4,5 DCAT], pentachlorophenol [PCP], and didecyldimethylammonium chloride [DDAC]). In most cases, white sturgeon fry were at the lower end of the range for acute toxicity values for chlorinated phenolic compounds, when compared with other juvenile fish species, and they were extremely sensitive to DDAC. The larval stage was usually more sensitive than the fry stage. Acute toxicity tests with fertilized eggs were unsuccessful. A postexposure growth study was inconclusive because neither control nor toxicant-exposed larvae and fry withstood the additional handling used for measuring body mass. At 62-days-old, fry were more tolerant of handling. This allowed measurement of their swimming performance. Although we have concerns about the reliability of using larvae for acute toxicity testing at this time, 60-day-old white sturgeon fry would appear to be both a sensitive and relevant species for assessing environmental impacts relevant to the Fraser River.

Abstract At many mining sites process (milling) and drainage waters escape treatment and cause receiving waters to become contaminated above avoidance or even toxic levels for fish. The present know-how on chemical agents which can be used to complex with copper and zinc to form non-toxic compounds is limited to chelating agents such as NTA or EDTA. Preferential reaction with trivalent ions such as iron means that such ions must be tied up before complexation of copper and zinc can occur. As the amount of iron in contaminated water is usually two to eight times higher than that of copper and zinc, high dosages of chelating agents are usually required. In this project, the use of salts of anthranilic acid, especially calcium anthranilate, was investigated. The consumption of anthranilateions was found to be about the same as that of NTA or EDTA, i.e. four milligrams per milligram of copper or zinc. The total dosage to be applied to contaminated waters would be, however, several times lower as iron is not involved in the reactions and copper and zinc are complexed in that order. Toxicity tests to compare the efficiency and dasages of calcium anthranilate and NTA or EDTA are presently being carried out.

A biomonitoring program has been developed in support of the National Policy for the Development of Water Quality-Based Permit Limitations for Toxic Pollutants. The program focuses on the use of laboratory toxicity tests on aquatic plants and animals to predict ecosystem impact caused by toxic pollutants. Both acute and chronic toxicity tests were developed to test effluents and ambient waters. Laboratory and biological field studies were conducted at nine sites. Single species laboratory toxicity tests were found to be good predictors of impacts on the ecosystem when two or more species were used. Biomonitoring can be undertaken either on effluents and/or on the receiving waters. In that toxicity related to seeps, leachates and storm sewers has often been found upstream from dischargers, it is beneficial to conduct both effluent and ambient biomonitoring.

With the increasing focus on environmental issues, the objective of this study is to evaluate the potential impact of contaminated sediments of the Blesbok Spruit near Witbank - which receives acid mine drainage (AMD) inter alia - on biota. Direct transfer of chemicals from sediments to organisms is considered to be a major route of exposure for many species, and therefore focusing attention on sediment contamination and highlighting the fact that sediments are an important resource. Acute toxicity tests were performed on Daphnia pulex using both extracted sediment interstitial water and surface water. Chemical analyses were also performed on the sediment, interstitial water and surface water samples. The toxicity results suggest that metal toxicity adds significantly to the toxicity of the stream water which is enhanced by the effect of pH. The pH of the stream and interstitial water was consistently below 4.5.

Ethnopharmacological Relevance:Mucuna pruriens (Mp) belongs to Leguminosae family, it is native of tropical regions and used to treat several maladies such as urinary, neurological, and menstruation disorders, constipation, edema, fever, tuberculosis, ulcers, diabetes, arthritis, dysentery, and cardiovascular diseases. Mp seeds are rich in bioactive compounds, for instance, lectins, a heterogeneous group of proteins and glycoproteins with a potential role as therapeutic tools for several conditions, including gastric disorders. This study investigated the acute toxicity, gastroprotective, and antioxidant activities of a lectin from Mucuna pruriens seeds (MpLec) on ethanol-induced gastropathy model in mice.Materials & Methods:Mice received MpLec (5 or 10 mg/kg; i.v.) and were observed for acute toxicity signs; in another experimental series, mice were pre-treated with MpLec (0.001; 0.01 or 0.1 mg/kg, i.v.), ranitidine (80 mg/kg, p.o.), or saline (0.3 mL/30g, i.v.) before ethanol 99.9% (0.2 mL/animal, p.o.), and euthanized 30 min after ethanol challenge. Macroscopic and microscopic gastric aspects, biochemical parameters (tissue hemoglobin levels, iron-induced lipid peroxidation, GSH content, SOD activity, and gastric mucosal PGE2) were measured. Additionally, pharmacological tools (yohimbine, indomethacin, naloxone, L-NAME) were opportunely used to clarify MpLec gastroprotective mechanisms of action.Results:No toxicity signs nor death were observed at acute toxicity tests. MpLec reduced ethanol-induced gastric damage, edema, and hemorrhagic patches formation, as well as decreased lipid peroxidation, SOD activity, and increased GSH content. Yohimbine and indomethacin prevented MpLec effects, suggesting the involvement of alpha-2 adrenoceptors and prostaglandins in the MpLec-mediated effects.Conclusion:MpLec does not present toxicity signs and shows gastroprotective and antioxidant activities via alpha-2 adrenoceptors and prostaglandins in the ethanol-induced gastropathy model.

The benthic foraminifers Bulimina denudata and Eggerelloides advenus are commonly abundant in offshore regions in the Pacific Ocean, especially in waste-discharge sites. The relationship between their abundance and standard macrofaunal sediment toxicity tests (amphipod survival and sea urchin fertilization) as well as sediment chemistry analyte measurements were determined for sediments collected in 1997 in Santa Monica Bay, California, USA, an area impacted by historical sewage input from the Hyperion Outfall primarily since the late 1950s. Very few surface samples proved to be contaminated based on either toxicity or chemistry tests and the abundance of B. denudata did not correlate with any of these. The abundance of E. advenus also did not correlate with toxicity, but positively correlated with total solids and negatively correlated with arsenic, beryllium, chromium, lead, mercury, nickel, zinc, iron, and TOC. In contrast, several downcore samples proved to be contaminated as indicated by both toxicity and chemistry data. The abundance of B.denudata positively correlated with amphipod survival and negatively correlated with arsenic, cadmium, unionized ammonia, and TOC; E. advenus negatively correlated with sea urchin fertilization success as well as beryllium, cadmium, and total PCBs. As B. denudata and E. advenus are tolerant of polluted sediments and their relative abundances appear to track those of macrofaunal toxicity tests, their use as cost- and time-effective marine sediment toxicity tests may have validity and should be further investigated.
