Association of Sweet Taste Receptor Gene Polymorphisms with Dental Caries Experience in School Children

Sweet taste is a powerful factor influencing food acceptance. The peripheral taste response to sugar is mediated by the TAS1R2/TAS1R3 taste receptors. The aim of the study was to determine the relationship between TAS1R2 (rs35874116 or rs9701796) and/or TAS1R3 (rs307355) single nucleotide polymorphisms with dental caries experience in schoolchildren. A total of 184 schoolchildren aged between 7 and 12 years (101 girls, 83 boys) were included in the study. Genomic DNA was extracted from saliva samples and the genotypes were identified by qPCR. The genotype frequencies were as follows: 6.6% for homozygous wild type, 41.8% for heterozygous and 51.6% for homozygous polymorphic genotype carriers of TAS1R2 gene rs35874116; 27.8% for heterozygous and 72.2% for homozygous polymorphic genotype carriers of TAS1R2 gene rs9701796, and 83.1% for homozygous wild type and 16.9% for heterozygous genotype carriers of TAS1R3 gene rs307355 polymorphism. A significant association was observed between total caries experience (dft + DMFT - decayed filled primary teeth + decayed, missing and filled permanent teeth) and TAS1R2 rs35874116 (p = 0.008) and TAS1R3 rs307355 (p = 0.04) gene polymorphisms but not for TAS1R2 gene rs9701796 polymorphism. TAS1R3 gene rs307355 polymorphism has been found to be an independent risk factor for dental caries experience by logistic regression analysis and to have increased the risk of caries. Moderate caries experience (4-7 caries) was found to be associated with TAS1R3 rs307355 heterozygous genotype, whereas high-risk caries experience (>8 caries) was found to be associated with TAS1R2 rs35874116 homozygous polymorphic genotype.

The aim of this study was to analyze the association between BMP2 (rs1884302) and DLX3 (rs2278163) gene polymorphisms and dental caries in primary and permanent dentitions. A total of 914 subjects were genotyped by the TaqMan methods: 176 caries-free children (with Decayed/Missing/Filled Teeth, DMFT = 0), 542 patients with dental caries in permanent dentition (DMFT ≥1), 83 caries-free children with primary teeth (with decayed/missing/filled teeth, dmft = 0), and 113 children with early childhood caries (ECC, dmft ≥1). There were no significant differences in allele/genotype frequencies between patients with caries in permanent dentition/ECC and caries-free children or between patients with very low (DMFT = 0-2), low (DMFT = 3-5), moderate (DMFT = 6-8), or high (DMFT ≥9) caries experience. Variability in BMP2 and DLX3 was not associated with caries in the Czech population.

Objective: The aim of the present study was to investigate the relationships between cariogenic bacterial infection and single nucleotide polymorphisms (SNPs) in candidate genes associated with dental caries, and to explore the factors related to caries in children.Study design: Children aged 3 to 11 years were selected. Detection of cariogenic bacteria (Streptococcus mutans, Streptococcus oralis, Streptococcus sobrinus and Lactobacillus) from the plaque of each patient, and SNP analyses of five candidate genes (MBL2, TAS2R38, GLUT2, MMP13 and CA6) were performed using DNA isolated from buccal mucosal cells. The dental caries experience in primary and permanent teeth was determined using the decayed, missing and filled teeth (DMFT) index, and the effects of the observed factors on the DMFT value were analyzed by multiple regression analysis. Results: The results of the multiple regression analysis showed that the DMFT value significantly increased in the presence of S. mutans or S. sobrinus (p < 0.001), while the dmft/DMFT value decreased in the presence of nucleobase C in MBL2 (p < 0.05). Conclusion: These results suggest that the MBL2 gene is related to the pathogenesis of dental caries.

The DMF index provides one source of information on past and present dental caries experience; however, important limitations hinder its ability to characterize fully the impact of dental caries. The purpose of this paper is to describe a measure of selected restoration and tooth conditions that supplements information from the DMF index and to report on the application of this measure as part of the Third National Health and Nutrition Examination Survey, Phase 1, conducted between 1988 and 1991. Data from this survey were used to estimate the prevalence and severity of selected disaggregated physical and biological oral conditions among dentate adults aged 18 to 74 years. Trained, standardized, and calibrated dentist examiners assessed 28 permanent teeth or tooth spaces for each of 6,767 subjects. Teeth or tooth spaces were classified based on criteria for: defective intracoronal restorations, crowns, or bridges; gross loss of tooth structure associated with a restoration; pulpal involvement; or retained roots. Approximately 40.5%, or 61.6 million, dentate adults had at least one tooth or tooth space that met the criteria. Among all persons, an average of 0.9 teeth or tooth spaces met the criteria for at least one category. Adults with at least one scored tooth or tooth space had an average of 2.2 such teeth or tooth spaces. Data from this assessment supplement information available from the DMF index to provide a broader profile of the impact of dental caries on permanent teeth of US adults.

Background: Celiac disease is an autoimmune chronic disease that affects the human’s intestine and subsequently reflects its effect on the entire body health by retardation the absorption and immune mediated complications cause the involvement of oral health. The present study intended to evaluate the impact of the histopathological disease activity upon dental enamel defects and dental caries. Subjects and methods: Forty celiac-diseased patients aged 7-11 years were collected from 3 different teaching hospitals in Baghdad classified by means of the histopathological activity of the intestinal disease according to modified Marsh-Rostami classification. Dental enamel defects were measured by Aine’s classification, while dental caries experience and severity were measured using d1-4mfs/t and D1-4MFS/T. Results: The majority of the sample came with partial villous atrophy of the small intestine (Marsh III-a) and almost half of the sample were with no celiac disease specific dental enamel defects (Aine’s 0), while Aine’s I was the most predominant than Aine’s II. Most missed surfaces due to dental caries in permanent teeth came with Marsh II. Conclusion: The more the severity of celiac disease histopathological activity the more the severity of celiac specific dental enamel defects and the less experienced dental caries. SummaryBackground/Aim: Individuals make food choices based on a number of physiological, nutritional, environmental and socioeconomic factors but sensory qualities of food namely the taste has priority as the determinant in food selection. The purpose of the study is to evaluate sweet, salty, sour and bitter taste perceptions of school children and compare them in term of caries experience.Material and Methods: Two hundred children aged from 6-13 were included in the study. The dental examinations of children were performed using DMF(T), DMF(S), df(t) and df(s) indices. Questionnaires were presented to parents to record the socioeconomic and educational levels, oral health knowledge, child’s general health, oral health habits and fluoride exposure. Children rinsed sucrose (12g/L; 24g/L), sodium chloride (2g/L; 4g/L), citric acid (0,6g/L; 1,20g/L) and caffeine (0,27g/L; 0,54g/L) solutions randomly and the taste perception was recorded. Relationship between the taste perception and caries experience were evaluated.Results: A significant correlation was found between 2 g/L of NaCl and age (p= 0,007, r= 0,178). When occlusal or approximal caries of the children aged from 6 to 9 were discriminated, the Spearman’s test found a weak positive correlation between occlusal caries and higher sweet taste (24 g/L sucrose) (r= 0,232; p= 0,021) and a weak negative correlation between approximal caries and higher salty taste (4 g/L NaCl) (r= –0,225; p= 0,025).Conclusions: Age should be considered in the assessment of taste perception of children. Additionally, there is a weak relationship between taste perceptions and dental caries. These data suggest that further studies need to focus on the effect of taste preferences on dental caries.

Objectives: The aim of this study was to consider the potential association between the formation of salivary streptococcus biofilms and the incidence of caries, as well as with the occurrence of antibiotic resistance among adult mothers in Sana'a, Yemen.  Study design: A total of 261; 25-35 year old females were chosen. Clinical examination of females were performed to estimate dental caries experience with the Silness-Loe index, as well as stimulated saliva were collected to assess biofilm production by the phenotypic method i.e. Tissue culture palate methods (TCPM). Finally, antibiogram susceptibility pattern of isolated S. mutans was done by Kirby-Bauer disc diffusion method for 6  β-Lactam antibiotics (ampicillin, penicillin, amoxicillin, cefotaxime, methicillin and cefazolin) and 4 non β-Lactam antibiotics (clindamycin, erythromycin, lincomycin and vancomycin). Results: When isolated S. mutans were exposed to biofilm detection by TCP method, 31 (12.2%) showed high biofilm formation capacity, 46 (18%) showed moderate biofilm formation capacity, while 184 (72.2%) showed non / weak formation capacity of biofilm. The overall rate of biofilm formation was 30.2%. There was an escalation in the rate of formation of S. mutans biofilms with an increased degree of caries index. The S. mutans biofilms positve showed a higher rate of resistance than non/weak biofilm formation e.g ampicillin (28.6% versus 12.9%,  p = 0.002), amoxicillin (77.9% versus 18%, p <0.0001), and penicillin (79.2 % versus 23%, <0.0001) etc.  Conclusion: The present study proved that S. mutans is still the major bacteria isolate from the oral cavity, but few persons might not have significant number of S. mutans in oral cavity. The S. mutans biofilm - producers were more able to cause dental caries compared to the S. mutans biofilm-non-producers. Background: The autism spectrum disorder (ASD) has been introduced as one of the complicated developmental disabilities impairing communication and behavioral, intellectual as well as social functioning describes diverse symptoms, such as difficulties in communication skills and social interactions. The present research has been performed to assess seriousness of the dental caries in conjunction with the oral cleanliness amongst children suffering from autism in comparison with a control group according to age groups and gender. Materials and Methods: This research involved 30 children aged 3-14 years (male, female) who suffered from autism and attended autism centers in Welfare Children Teaching Hospital in Baghdad province, Iraq, were selected for the study compared to 30 healthy children with the same age group. Plaque (PlI), Decayed, missing, and filled surfaces (dmfs, DMFS), calculus (CI) as well as Gingival (GI) indices have been applied for measuring the status of oral health for these two groups. The data of our research has been analyzed by SPSS 26. Results: The entire autism group was caries active. For primary dentition, a lower dmfs values were recorded for study in comparison with the controls, differences have been not significant in dmfs, while for permanent dentition, a higher DMFS values were recorded for study in comparison with the control group with statistically significant concerning DMFS (P < 0.05). Moreover, caries experience (DS and DMFS) among both genders were higher in the study group in comparison with the controls with a significant difference in female only. Total mean value of PlI in the study group has been greater than the controls with no statistically significant difference. In addition, total mean value of GI in the study group has been lower than the controls with no statistically significant difference. Furthermore, correlation coefficient between the caries experience of primary and permanent teeth with PlI and GI among study and control group showed no significant correlations seen in the study and control groups, all of them were positively correlated except (ds) with (GI) in control group which was negatively correlated with no significant correlation. Conclusion: children with ASD had higher dental caries severity regarding permanent teeth compared to normal subjects, plaque higher in autistic group than control group. Dental care planners must design preventing strategies for avoiding caries; procedures for oral care as well as educational programs for oral care and promotion for addressing diverse challenges facing the oral care in ASD.

Most mammals prefer the sweet taste of sugars, which is mediated by the heterodimeric T1R2+T1R3 taste receptor. Sugar appetite is also enhanced by the post-oral reinforcing actions of the nutrient in the gut. Here, we examined the contribution of gut T1R3 (either alone or as part of the T1R3+T1R3 receptor) to post-oral sugar reinforcement using a flavor-conditioning paradigm. We trained mice to associate consumption of a flavored solution (CS+) with intragastric (IG) infusions of a sweetener, and a different flavored solution (CS-) with IG infusions of water (23 h/day); then, we measured preference in a CS+ vs. CS- choice test. In experiment 1, we predicted that if activation of gut T1R3 mediates sugar reinforcement, then IG infusions of a nutritive (sucrose) or nonnutritive (sucralose) ligand for this receptor should condition a preference for the CS+ in B6 wild-type (WT) mice. While the mice that received IG sucrose infusions developed a strong preference for the CS+, those that received IG sucralose infusions developed a weak avoidance of the CS+. In experiment 2, we used T1R3 knockout (KO) mice to examine the necessity of gut T1R2+T1R3 receptors for conditioned flavor preferences. If intact gut T1R3 (or T1R2+T1R3) receptors are necessary for flavor-sugar conditioning, then T1R3 KO mice should not develop a sugar-conditioned flavor preference. We found that T1R3 KO mice, like WT mice, acquired a strong preference for the CS+ paired with IG sucrose infusions. The KO mice were also like WT mice in avoiding a CS+ flavor paired with IG sucralose infusions These findings provide clear evidence that gut T1R3 receptors are not necessary for sugar-conditioned flavor preferences or sucralose-induced flavor avoidance in mice.

Elevated alcohol consumption is associated with enhanced preference for sweet substances across species and may be mediated by oral alcohol-induced activation of neurobiological substrates for sweet taste. Here, we directly examined the contribution of the T1r3 receptor protein, important for sweet taste detection in mammals, to ethanol intake and preference and the neural processing of ethanol taste by measuring behavioral and central neurophysiological responses to oral alcohol in T1r3 receptor-deficient mice and their C57BL/6J background strain. T1r3 knockout and wild-type mice were tested in behavioral preference assays for long-term voluntary intake of a broad concentration range of ethanol, sucrose, and quinine. For neurophysiological experiments, separate groups of mice of each genotype were anesthetized, and taste responses to ethanol and stimuli of different taste qualities were electrophysiologically recorded from gustatory neurons in the nucleus of the solitary tract. Mice lacking the T1r3 receptor were behaviorally indifferent to alcohol (i.e., ∼50% preference values) at concentrations typically preferred by wild-type mice (5–15%). Central neural taste responses to ethanol in T1r3-deficient mice were significantly lower compared with C57BL/6J controls, a strain for which oral ethanol stimulation produced a concentration-dependent activation of sweet-responsive NTS gustatory neurons. An attenuated difference in ethanol preference between knockouts and controls at concentrations >15% indicated that other sensory and/or postingestive effects of ethanol compete with sweet taste input at high concentrations. As expected, T1r3 knockouts exhibited strongly suppressed behavioral and neural taste responses to sweeteners but did not differ from wild-type mice in responses to prototypic salt, acid, or bitter stimuli. These data implicate the T1r3 receptor in the sensory detection and transduction of ethanol taste.
