In the chick embryo retina (days 3-4 of incubation), coinciding with an increase in cell death, specialized phagocytes characterized by intense acid phosphatase activity have been described. In these preparations, all free cells in the vitreal humor (vitreal cells) were strongly labeled. Conventional TEM and SEM techniques were used to characterize them and attempt to determine their relationship with retinal phagocytes.Two types of vitreal cells were distinguished. The first are located at some distance from the basement membrane of the neuroepithelium, and are rounded, with numerous vacuoles and thin cytoplasmic prolongations. Images of exo- and or endocytosis were frequent; the cells showed a well-developed Golgi apparatus (Fig. 1) In SEM images, the cells was covered with short cellular processes (Fig. 3). Cells lying parallel to or alongside the basement membrane are elongated. The plasma membrane is frequently in intimate contact with the basement membrane. These cells have generally a large cytoplasmic expansion (Fig. 5).

Suggestive evidence for the extragonadal origin of germ cells in birds was first presented by Swift (1914), who described primordial germ cells in the chick embryo at as early a stage as the primitive streak. According to Swift, primordial germ cells are originally located extra-embryonically in the anterior part of the blastoderm and occupy a crescent-shaped region (‘germinal crescent’) on the boundary between area opaca and area pellucida. Swift also found that primordial germ cells later enter into the blood vessels, circulate together with the blood throughout the whole blastoderm and finally penetrate into the genital ridges, where they become definitive germ cells. Swift's views have been confirmed in numerous descriptive and experimental investigations. Among the latter, the publications of Willier (1937), Simon (1960) and Dubois (1964a, b, 1965a, b, 1966) merit special attention. Dubois finally proved that the genital ridges exert a strong chemotactic influence on the primordial germ cells.

We studied ultrastructural localization of acid phosphatase in derepressed Saccharomyces cerevisiae cells transformed with a multicopy plasmid carrying either the wild-type PHO5 gene or a PHO5 gene deleted in the region overlapping the signal peptidase cleavage site. Wild-type enzyme was located in the cell wall, as was 50% of the modified protein, which carried high-mannose-sugar chains. The remaining 50% of the protein was active and core glycosylated, and it accumulated in the endoplasmic reticulum cisternae. The signal peptide remained uncleaved in both forms. Cells expressing the modified protein exhibited an exaggerated endoplasmic reticulum with dilated lumen.
