Prolonged Food Deprivation Abolishes the Circadian Adrenocortical Rhythm, but Not the Endogenous Rhythm in Rats

Berghia stephanieae is a stenophagous sea slug that preys upon glass anemones, such as Exaiptasia diaphana. Glass anemones host photosynthetic dinoflagellate endosymbionts that sea slugs ingest when consuming E. diaphana. However, the prevalence of these photosynthetic dinoflagellate endosymbionts in sea slugs appears to be short-lived, particularly if B.stephanieae is deprived of prey that host these microalgae (e.g., during bleaching events impacting glass anemones). In the present study, we investigated this scenario, along with food deprivation, and validated the use of a non-invasive and non-destructive approach employing chlorophyll fluorescence as a proxy to monitor the persistence of the association between sea slugs and endosymbiotic photosynthetic dinoflagellates acquired through the consumption of glass anemones. Berghia stephanieae deprived of a trophic source hosting photosynthetic dinoflagellate endosymbionts (e.g., through food deprivation or by feeding on bleached E. diaphana) showed a rapid decrease in minimum fluorescence (Fo) and photosynthetic efficiency (Fv/Fm) when compared to sea slugs fed with symbiotic anemones. A complete loss of endosymbionts was observed within 8 days, confirming that no true symbiotic association was established. The present work opens a new window of opportunity to rapidly monitor in vivo and over time the prevalence of associations between sea slugs and photosynthetic dinoflagellate endosymbionts, particularly during bleaching events that prevent sea slugs from incorporating new microalgae through trophic interactions.

POMC neurons integrate metabolic signals from the periphery. Here, we show in mice that food deprivation induces a linear current-voltage relationship of AMPAR-mediated excitatory postsynaptic currents (EPSCs) in POMC neurons. Inhibition of EPSCs by IEM-1460, an antagonist of calcium-permeable (Cp) AMPARs, diminished EPSC amplitude in the fed but not in the fasted state, suggesting entry of GluR2 subunits into the AMPA receptor complex during food deprivation. Accordingly, removal of extracellular calcium from ACSF decreased the amplitude of mEPSCs in the fed but not the fasted state. Ten days of high-fat diet exposure, which was accompanied by elevated leptin levels and increased POMC neuronal activity, resulted in increased expression of Cp-AMPARs on POMC neurons. Altogether, our results show that entry of calcium via Cp-AMPARs is inherent to activation of POMC neurons, which may underlie a vulnerability of these neurons to calcium overload while activated in a sustained manner during over-nutrition.

Temperature and hunger mediate sablefish (Anoplopoma fimbria) feeding motivation: implications for stock assessment

Laboratory experiments were conducted to test the hypothesis that temperature (2–8 °C) and recent feeding history (1–6 days of food deprivation) influence feeding motivation and attacks on baits by adult sablefish (Anoplo poma fimbria). Activity level after introduction of a bait-related olfactory cue increased with increasing water temperature and food deprivation. Fish at the lowest temperature (2 °C) attacked and consumed fewer squid baits (33–71%) than those at higher temperatures (100% at 8 °C). Baits were frequently inspected or handled multiple times before consumption in low temperatures, and time to attack (seconds to >40 min), time to consume, and time to handle baits (2–20 s) all decreased significantly with increasing temperature, with some temperature × feeding history interactions. The total ration consumed increased (<1–8% of body weight) with increasing water temperature. The observed variability in activity and feeding motivation in sablefish may have a large impact on the effective area of baited gear but is rarely considered in stock assessment models. In particular, population sizes based upon baited gear surveys may be underestimated in conditions where temperature is low, natural prey density is high, or where other environmental variables reduce feeding motivation.
