The numerical density of GABA immunopositive and GABA immunonegative neurons was quantitatively determined in 0, 12, 30 and 90 day-old neocortical transplants, derived from E17 rat embryos and transplanted into adult hosts. It was found that the original, very high neuronal density in the fetal transplant declined steadily after transplantation to the somatosensory cortex of adult rat. The decline in numerical density of GABA-positive neurons, however, was disproportionately larger than that of GABA-negative nerve cells: At 90 days the proportion of GABA-positive cells was 2.3% (in contrast to the 11.8% in the adult host cortex). The density of GABA-negative neurons, on the other hand, remained slightly higher than comparable values in the control cortex. The decline in density Of GABA-positive neurons was continuous until the 90th post-transplantation day, while final, close to normal density values of GABA-negative nerve cells were already reached in 30 day-old grafts, with no significant change afterwards.

Abstract To cryopreserve cells, it is essential to avoid intracellular ice formation during cooling and warming. One way to achieve this is to convert the water inside the cells into a non-crystalline glass. It is currently believed that to accomplish this vitrification, the cells must be suspended in a very high concentration (20–40%) of a glass-inducing solute, and subsequently cooled very rapidly. Herein, we report that this belief is erroneous with respect to the vitrification of one-cell rat embryos. In the present study, one-cell rat embryos were vitrified with 5 μL of EFS10 (a mixture of 10% ethylene glycol, 27% Ficoll, and 0.45 M sucrose) in cryotubes at a moderate cooling rate, and warmed at various rates. Survival was assessed according to the ability of the cells to develop into blastocysts and to develop to term. When embryos were vitrified at a 2,613 °C/min cooling rate and thawed by adding 1 mL of sucrose solution (0.3 M, 50 °C) at a warming rate of 18,467 °C/min, 58.1 ± 3.5% of the EFS10-vitrified embryos developed into blastocysts, and 50.0 ± 4.7% developed to term. These rates were similar to those of non-treated intact embryos. Using a conventional cryotube, we achieved developmental capabilities in one-cell rat embryos by rapid warming that were comparable to those of intact embryos, even using low concentrations (10%) of cell-permeating cryoprotectant and at low cooling rates.

AbstractFloods in Ghana have become a perennial challenge in the major cities and communities located in low-lying areas. Therefore, cities and communities located in these areas have been classified as potential or natural flood-prone zones. In this study, the Digital Elevation Model (DEM) of the Accra Metropolis was used to assess the drainage density and elevation patterns of the area. The annual population estimation data and flood damages were assessed to understand the damages and population trend. This research focused primarily on the elevation patterns, slope patterns, and drainage density of the Accra Metropolis. Very high drainage density values, which range between 149 and 1117 m/m2, showed very high runoff converging areas. High drainage density was also found to be in the range of 1117–1702 m/m2, which defined the area as a high runoff converging point. The medium and low converging points of runoff were also found to be ranging between 1702–2563 m/m2 and 2563–4070 m/m2, respectively. About 32% of the study area is covered by natural flood-prone zones, whereas flood-prone zones also covered 33% and frequent flood zones represent 25%. Areas in the Accra Metropolis that fall in the Accraian and Togo series rock types experience high floods. However, the lineament networks (geological structures) that dominate the Dahomeyan series imply that the geological structures in the Dahomeyan series also channel the runoffs into the low-lying areas, thereby contributing to the perennial flooding in the Accra Metropolis.

The rate of conversion of [1,3-14C]glycerol into 14CO2 was measured in the presence and absence of unlabelled alternative substrates in whole homogenates from the brains of young (4-6 and 18-20 days old) and adult rats. Unlabelled glucose decreased 14CO2 production from [1,3-14C]glycerol by about 40% at all ages studied. Unlabelled 3-hydroxybutyrate significantly decreased the 14CO2 production from both low (0.2 mM) and high (2.0 mM) concentrations of glycerol in 4-6- and 18-20-day-old rat pups. However, the addition of 3-hydroxybutyrate had no effect on the rate of 14CO2 production from 2.0 mM-glycerol in adult rats, suggesting that the interaction of 3-hydroxybutyrate with glycerol in adult rat brain is complex and may be related to the biphasic kinetics previously reported for glycerol oxidation. Unlabelled glutamine decreased the production of 14CO2 by brain homogenates from 18-20-day-old and adult rats, but not in 4-6-day-old rat pups. In the converse situation, the addition of unlabelled glycerol to whole brain homogenates had little effect on the rate of 14CO2 production from [6-14C]glucose, 3-hydroxy[3-14C]butyrate and [U-14C]glutamine, although some significant differences were noted. Collectively these results suggest that glycerol and these other substrates may be metabolized in separate subcellular compartments in brain such that the products of glucose, 3-hydroxybutyrate and glutamine metabolism can dilute the oxidation of glycerol, but the converse cannot occur. The data also demonstrate that there are complex age-related changes in the interaction of glycerol with 3-hydroxybutyrate and glutamine. The fact that glycerol oxidation was only partially suppressed by the addition of 1-5 mM-glucose, -3-hydroxybutyrate or -glutamine could also suggest that glycerol may be selectively utilized as an energy substrate in some discrete brain region.

To unravel the mechanisms behind the higher resistance to light damage of juvenile (JR) versus adult (AR) rats, Sprague Dawley rats were exposed to a bright luminous environment of 10, 000 lux. The light-induced retinopathy (LIR) was assessed with histology, electroretinography and immunohistochemistry (IHC). In JR, 2 days of exposure induced the typical LIR, while >3 days added little LIR. IHC revealed a subtle migration of microglia (Iba1 marker) from the inner to the outer retina after 3 days of exposure in JR contrasting with the stronger reaction seen after 1 day in AR. Similarly, in JR, the Müller cells expressed less intense GFAP, CNTF and FGF2 staining compared to AR. Our results suggest that in JR the degree of retinal damage is not proportional to the duration of light exposure (i.e., dose-independent retinopathy), contrasting with the dose-dependent LIR reported in AR. The immature immune system in JR may explain the delayed and/or weaker inflammatory response compared to AR, a finding that would also point to the devastating contribution of the immune system in generating the LIR phenotype, a claim also advanced to explain the pathophysiology of other retinal degenerative disorders such as Age-related Macular Degeneration, Diabetic Retinopathy and Retinitis Pigmentosa.

The susceptibility of postnatal day 15 and adult rat diaphragms (DIAs) to acute injury after repetitive isovelocity lengthening activations was examined in vitro. Forces were measured during two phases of each stimulation protocol: 1) isometric phase: during the first 300 ms of each 500-ms train, DIA length was not changed; and 2) isovelocity lengthening phase: during the remaining 200 ms of each stimulus train, DIA was lengthened at a constant velocity from 90 to 110% of optimal length. At maximal activation (75 Hz and a lengthening velocity of 1.0 optimal length per second), the maximum force measured during the isometric phase and that measured during the isovelocity lengthening phase were both greater in adult DIAs than in day 15 DIAs but both declined to a greater extent in adults with repetitive activation. Ultrastructural analysis showed that after lengthening activations muscle fiber injury was very evident in adult but much less prevalent in day 15 DIAs. This difference in susceptibility between the adult and day 15 DIAs did not depend on differences in peak force or absolute velocity of lengthening. We conclude that lengthening activations result in DIA injury and that the adult is more susceptible than its younger counterpart.

An orchard trial was established by planting an orchard with between-row intervals of 4 m. The French Axe was trained for trees with intervals in the row of 1 and 1.5 m. The hedgerow was used for treatments of 2–2.5 and 3 m between trees in the row. Semi-dwarf rootstock of Bud54-118 and dwarf one Bud62-396 were used. The growth of of these rootstocks was analogous to MM106 and M26, respectively. The trunk cross-sectional area of 7-year trees on 54-118 rootstock was 2.3 times more than on 62-396 for cv. Antey and 1.5 times more for cv. Tellisaare. The height of tree with French Axe crown at 7 years after planting on 54-118 rootstock reached 3.5–4 m. The height of tree was 0.5 m smaller on 62-396. The crown habit of tree on 62-396 rootstock was more comfortable for high -density orchard than trees on 54-118. The sum length of twigs that were cut out during 1993–96 to attain of normal density of crown was 2-4 times more than on 62-396 rootstock. Commercial fruiting of cv. Antey started at the 3rd leaf, but it was on 4th leaf for the more-dwarf rootstock 62-396. Average yield of fruit at 3–6 years after planting of cv. Antey for treatment of distance between trees in the row of 2 or 1.5 m was 6.8 kg/tree per year-1 for 54-118 rootstock, 3.4 and 3.5, respectively, for 62-396 rootstock. Yield at the 7th year after planting reached 24 and 32 kg on 54-118 rootstock, 16 and 15 kg on 62-396, respectively. Analogous date obtained for cv. Tellisa are. cv. Spartan on both rootstocks started to fruiting at 5-6 years after planting. The fruit quality was very high in all treatments of the trial.

In the length of time, a wide variety of drug combinations emerged in the management of post-transplantation therapy in order to improve the survival of the recipient and graft. However, the efficacy and safety of the applied combinations regarding the rejection and other complications are continuing to be the subject of research. In our study, our aim is to compare the effects of various combinations, namely cyclosporine with mycophenolate mofetil/mofetil mycophenolic acid with prednisolone and tacrolimus with mycophenolate mofetil/mofetil mycophenolic acid with prednisolone in a length of time. Methods. A total of 204 patients included into the study who received post-renal transplantation treatment in Scientific-Research Institute of Heart Surgery and Organs Transplantation and followed-up over a 10-year period. The estimated survival probabilities in the study were determined by the Kaplan-Meier method; whereas intragroup comparisons were evaluated by Log-rang, Breslow, and Tarone-Ware tests. Complications occurred in patients with combinations were analyzed by Chi-square and its alternatives. Hazard risk factors were tested by Cox regression analysis. Results. Of these 204 patients, 36 received Cyclosporin combination (CCG) and 168 Tacrolimus combination (TCG). The estimated life expectancy of the patients of TCG was significantly longer than the CCG ones. Furthermore, gender and age did not have a significant effect on survival depending on time, however, gender and age-related hazard factor showed a significant difference in the groups. It was determined that chronic rejection was significantly different in patients who used tacrolimus combinations, the difference was close to the significant value in acute rejection analysis. Other adverse events, namely, infection, tumour and organ damage were statistically less common in the patient group treated with tacrolimus combinations. Conclusion. In general, TCG showed better results in contrast to CCG.

NG2 immunoreactive cells (NG2 cells) are found in the brain and peripheral tissues including the skin, intestinal tracts, and bladder. In a previous study, we observed the presence of NG2 cells in the stomach using bioluminescence imaging techniques in NG2-firefly luciferase (fLuc) transgenic (Tg) rats. Here, we aimed to identify and characterize NG2 cells in the adult rat stomach. Immunohistochemical studies showed that NG2 cells were mainly present in the lamina propria and most of the cells were gastric telocytes, co-expressing CD34, and platelet-derived growth factor receptor alpha (PDGFRα), with a small oval-shaped cell body and extremely long and thin cellular prolongations. In the rat stomach, NG2-expressing telocytes comprised two subpopulations: NG2+/CD34+/PDGFRα+ and NG2+/CD34+/PDGFRα-. Furthermore, we showed that the expression of NG2 gene in the aged rat stomach decreased relative to that of the young rat stomach and the decline of NG2 expression in aged rats was mainly observed in NG2+/CD34+/PDGFRα+ telocytes. These findings suggested age-related alterations in NG2+/CD34+/PDGFRα+ telocytes of rat stomach.
