The purpose of this study is to investigate the significance of alpha-enolase (ENO1) expression in squamous cell carcinoma of the lung (LUSC), its prognostic value, and prospective molecular mechanism. Using multiplatforms data, including in-house immunohistochemistry, in-house real-time fluorescence quantitative polymerase chain reaction (RT-qPCR), in-house microarray, and public high-throughput data, the expression significance and prognostic role of ENO1 in LUSC tissues were analyzed comprehensively. With the combination of all eligible cases, compared with 941 non-LUSC lung tissues, ENO1 was significantly overexpressed in 1163 cases of LUSC (standardized mean difference (SMD) = 1.23, 95% confidence interval (CI) = 0.76–1.70, P  < 0.001). ENO1 also displayed a great ability to differentiate LUSC tissues from non-LUSC lung tissues (AUC = 0.8705) with the comprehensive sensitivity being 0.88 [0.83–0.92], and comprehensive specificity being 0.89 [0.84–0.94]). Moreover, in 1860 cases of LUSC with survival information, patients with higher expression of ENO1 had poorer prognosis (hazard ratio (HR) = 1.20, 95% CI = 1.01–1.43, P  = 0.043). ENO1 and its related genes mainly participated in the pathways of cell division and proliferation. In conclusion, the upregulation of ENO1 could affect the carcinogenesis and unfavorable outcome of LUSC.

Introduction. The associative role of the bacterial factor in the development of both primary malignant tumors of the oral mucosa and relapses is being actively discussed today. In the article published earlier, we have already described the possible mechanisms of action of periodontopathogenic microflora and its connection with the development of squamous cell carcinoma of the oral mucosa through cell proliferation, intracellular accumulation of pathogen, DNA replication and affect the signaling pathways of MARK (mitogen-activated protein kinase).Objective – to analyze the impact of periodontal pathogens on the risk of oral squamous cell carcinoma and its recurrence, as well as to evaluate the role of polymerase chain reaction and bacterial culture in the diagnosis of squamous cell carcinoma.Materials and methods. This study included 35 patients with stage Т3–4 squamous cell carcinoma, whose tumor tissue samples were tested for periodontal pathogens using culture and PCR. We analyzed 5 paraffin-embedded and 30 frozen tissue blocks from newly diagnosed (n = 15) and re-treatment (n = 15) patients.Results. We found that PCR was more sensitive than culture for the detection of possible etiological agents and predictors of squamous cell carcinoma (including Porphyromonas gingivalis, Tannerella forsythia, and Treponema denticola) (р <0.001). For example, Tannerella forsythia and Treponema denticola were detected only using PCR, whereas all cultures were negative. However, conventional culture proved to be more effective than PCR for the detection of Prevotella intermedia. Between 3 % and 100 % of newly diagnosed patients tested positive for Treponema denticola and some other periodontal pathogens (PCR) developed relapses, whereas among re-treatment patients, this proportion was 66.6 %.Conclusion. The combination of the culture method and the polymerase chain reaction method in the study of periodontopathogenic microflora has shown high efficiency in identifying possible predictors of squamous cell carcinoma of the oral mucosa and in preventing the development of chronic infectious periodontal diseases.

Objective. The purpose of this study was to evaluate the prognostic role of the cytotoxic T-lymphocyte-associated antigen-4 (CTLA-4) expression level and the platelet lymphocyte ratio (PLR) level in esophageal squamous cell carcinoma (ESCC) patients. Methods. 84 ESCC patients who received surgical treatment in our hospital were enrolled in the study. The correlation of each biomarker’s level with ESCC patients’ clinicopathological characteristics and overall survival (OS) was assessed. Results. The elevated expression rate of T-CTLA-4 (tumor cell CTLA-4) and I-CTLA-4 (interstitial lymphocyte CTLA-4) was 48.8% and 44.0%, respectively. The number of enrolled patients with a higher PLR level (≥119) was 48. The prognostic value of T-CTLA-4, I-CTLA-4, and PLR in ESCC patients was not detected. However, patients with both a low T-CTLA-4 expression level and a low PLR level that had longer OS (p=0.023) were found. The prognostic role of T-CTLA-4(-) +PLR (-) status in ESCC patients was also confirmed in multivariate analyses (p=0.027). Conclusion. These results demonstrated the potential prognostic value of combined analysis of CTLA-4 and PLR in ESCC patients.
