Background. The lncRNA taurine upregulated gene 1 (TUG1) is a recently identified potential biomarker in cancer. However, its prognostic role in various cancers is inconsistent among published data. We conducted this meta-analysis to comprehensively confirm the prognostic effect of TUG1 in malignant tumors. Methods. We systemically analyzed the prognostic-predictive capacity of TUG1 through amplifying sample sizes and cancer types. STATA 12.0 was applied for this meta-analysis. Results. A total of 57 eligible studies were included in our meta-analysis. The pooled results suggested that overexpression of TUG1 was significantly correlated with unfavorable overall survival (OS) ( HR = 1.70 , p < 0.001 ), shorter recurrence-free survival (RFS) ( HR = 2.40 , p ≤ 0.001 ), and shorter event-free survival (EFS) ( HR = 1.88 , p < 0.001 ) in patients with cancer. In the subgroup analysis by cancer type, elevated TUG1 expression was associated with poorer survival in patients with gastrointestinal cancer, urinary tumors, gynecological tumors, hematological tumors, and osteosarcoma. However, high expression of TUG1 in respiratory tumors indicated a better prognosis. There was no correlation between high TUG1 expression and OS in patients with head and neck neoplasms or melanoma. Additionally, overexpression of TUG1 was found to be correlated with low-grade tumor differentiation, advanced tumor stage, positive lymphatic metastasis, and positive distant metastasis. Conclusions. High TUG1 expression correlates with poor prognosis and advanced clinicopathological features, verifying the prognostic-predictive capacity of TUG1 in tumors, especially in gastrointestinal cancer, urinary tumors, gynecological tumors, hematological tumors, and osteosarcoma. Meanwhile, the prognostic role of TUG1 in respiratory tumor may be opposite to other tumors.

Several studies suggest that upregulated expression of the long non-coding RNA HOX transcript antisense RNA (HOTAIR) is a negative predictive biomarker for numerous cancers. Herein, we performed a meta-analysis to further investigate the prognostic value of HOTAIR expression in diverse human cancers. To this end, a systematic literature review was conducted in order to select scientific studies relevant to the association between HOTAIR expression and clinical outcomes, including overall survival (OS), recurrence-free survival (RFS)/disease-free survival (DFS), and progression-free survival (PFS)/metastasis-free survival (MFS) of cancer patients. Collectively, 53 eligible studies including a total of 4873 patients were enrolled in the current meta-analysis. Pooled hazard ratios (HRs) with their corresponding 95% confidence intervals (CIs) were calculated to assess the relationship between HOTAIR and cancer patients’ survival. Elevated HOTAIR expression was found to be significantly associated with OS, RFS/DFS and PFS/MFS in diverse types of cancers. These findings were also corroborated by the results of bioinformatics analysis on overall survival. Therefore, based on our findings, HOTAIR could serve as a potential biomarker for the prediction of cancer patient survival in many different types of human cancers.

BACKGROUND: Recent studies have shown that microRNAs (miRNA) have prognostic values in cancers. This meta-analysis seeks to summarize the global predicting role of miR-155 for survival in patients with a variety of carcinomas.METHODS: Eligible studies were identified through multiple search strategies. Data were extracted from studies investigating the relationship between miR-155 expression and survival in cancer patients. Combined hazard ratios (HRs) of miR-155 for outcome were analyzed.RESULTS: A total of 16 studies dealing with various carcinomas were included for this meta-analysis. For overall survival, higher miR-155 expression could significantly predict worse outcome with the pooled HR of 2.057 (95% CI: 1.392–3.039). For relapse or progress-free survival, elevated miR-155 was also a significant predictor, with a combined HR of 1.918 (95% CI: 1.311–2.806,). In addition, subgroup analysis showed that higher expression of miR-155 had the trends to predict worse outcome in lung cancer. However, the HRs did not reach the statistical significance.CONCLUSION: Our findings suggest that miR-155 detection has a prognostic value in cancer patients. Regularly measuring miR-155 expression may be useful in clinical practice.

Background: Recently, many studies have shown that microRNAs (miRNA) exhibit altered expression in various cancers and may serve as prognostic biomarkers. We performed a systematic review and meta-analysis to evaluate the prognostic role of miR-200c expression in different cancers. Methods: Studies were recruited by searching PubMed, Embase and the Cochrane Library (last search update was May 2014) and assessed by further quality evaluation. Results: A total of 25 studies dealing with various carcinomas were identified for systematic review. Among them, 18 studies were ultimately included in the meta-analysis. Our results indicated that the expression of tissue miR-200c was not associated with OS and PFS in various carcinomas; however, downregulation of tissue miR-200c did predict poor OS of patients with stage I disease (HR=0.41, 95% CI 0.25-0.68, P=0.001). Furthermore, overexpression of blood miR-200c was significantly related to poor OS and PFS (HR=3.07 95% CI 1.58-5.96 P=0.001, HR=2.26 95% CI 1.66-3.08 P<0.001, respectively), especially in patients with advanced disease. Conclusion: This systematic review and meta-analysis clarified that low expression of miR-200c in primary tissue was significantly associated with poor survival in cancer patients at early stage, whereas a high level of blood miR-200c predicted poor prognosis in patients with advanced tumors.

For the prognosis analysis of miR-155 in lung cancer, the pooled HRs and 95% CIs of miR-155 for overall survival/disease free survival/progression-free survival (OS/DFS/PFS) were calculated. In addition, Subgroup and meta-regression analyses were performed to distinguish the potential sources of heterogeneity between studies. Results: For the diagnostic analysis of miR-155 in lung cancer, the pooled SEN and SPE were 0.82 (95% CI: 0.72-0.88) and 0.78 (95% CI: 0.71-0.84), respectively. Besides, the pooled PLR was 3.75 (95% CI: 2.76-5.10), NLR was 0.23 (95% CI: 0.15-0.37), DOR was 15.99 (95% CI: 8.11-31.52) and AUC was 0.87 (95% CI: 0.84-0.90), indicating a significant value of miR-155 in the lung cancer detection. For the prognostic analysis of miR-155 in lung cancer, up-regulated miRNA-155 expression was not significantly associated with a poor OS (pooled HR = 1.26, 95% CI: 0.66-2.40) or DFS/PFS (pooled HR = 1.28, 95% CI: 0.82-1.97). PINX1(Pin2/TRF1 interacting protein X1, an intrinsic telomerase inhibitor and putative tumor suppressor gene) may represent a novel prognostic tumor biomarker. However, the results of previous studies are inconsistent and the prognostic value ofPINX1remains controversial. Therefore, we conducted a meta-analysis to determine whetherPINX1expression is associated with overall survival (OS), disease-specific survival (DSS), disease-free survival (DFS), recurrence-free survival (RFS), and clinicopathological characteristics in patients with malignant tumors. A systematic search was performed in the PubMed, Web of Science, and Embase databases in April 2018. Quality assessment was performed according to the modified Newcastle-Ottawa Scale. Pooled odds ratios (ORs) and hazard ratios (HRs) with 95.0% confidence intervals (CIs) were calculated to determine the relationship betweenPINX1expression and OS, DSS, DFS/RFS, and clinicopathological characteristics. Due to the heterogeneity across the included studies, subgroup and sensitivity analyses were performed. Fixed-effects models were used when the heterogeneity was not significant and random-effects models were used when the heterogeneity was significant. Fourteen studies of 16 cohorts including 2,624 patients were enrolled. LowPINX1expression was associated with poor OS (HR: 1.51, 95.0% CI: 1.03–2.20;P= 0.035) and DFS/RFS (HR: 1.78, 95.0% CI: 1.28–2.47;P= 0.001) but not DSS (HR: 0.80, 95.0% CI: 0.38–1.67;P= 0.548). LowPINX1expression was also associated with lymphatic invasion (OR: 2.23, 95.0% CI: 1.35–3.70;P= 0.002) and advanced tumor-node-metastasis stage (OR: 2.43, 95.0% CI: 1.29–4.57;P= 0.006). No significant associations were observed between lowPINX1expression and sex, depth of invasion, grade of differentiation, and distant metastasis. LowPINX1expression was associated with poor OS and DFS/RFS and lymphatic invasion and advanced tumor-node-metastasis stage, suggesting thatPINX1expression may be a useful predictor of prognosis in patients with malignant tumors.
