Neem (Azadirachta indica A. Juss) is one of the tropical plants found in Indonesia that has been used to prevent and treat various diseases. This study aimed to investigate the effect of the ethanol extract of neem leaves on the concentration of aspartate aminotransferase (AST), alanine aminotransferase (ALT), urea, and creatinine in male rats. Twenty-four male Wistar rats were randomly divided into four groups (T0, T1, T2, and T3) with 6 rats in each group. T0 is the control group, and T1, T2, and T3 are the treatment groups that were administered 100, 200, and 300 mg/kg body weight of neem leaf ethanolic extracts for 48 days, respectively. On day 49, blood samples were collected to measure the concentration of AST, ALT, creatinine, and urea followed by an evaluation of liver and kidney histology. The results showed that the ethanolic extract of neem leaves did not affect the concentration of AST, ALT, and creatinine, The ethanol leaves reduced extract on the urea concentration, no abnormal changes were observed in the liver and kidney organs. In the future, it is required to carry out a comprehensive safety evaluation of the neem leaf ethanol extract for herbal medicines.

The aim of this study was to determine the effect of neem leaf extract (Azadirachta indica) on parasitemia of rats infected with Trypanosoma evansi (T. evansi) Aceh local isolate. A total of 24 male rats aged three months were used in this study and randomly divided into six treatment groups equally. The negative control group (K0) without T. evansi infection and neem leaf extract, the positive control group (K1) was infected with T. evansi but no neem leaf extract given, group K2, K3, K4, and K5 were infected with 5x104 T. evansi and were given neem leaf extract after patent infection with dose of 50, 100, 400, and 800 mg/kg BW respectively. The extract was given orally for three consecutive days. On the fourth day, rat blood was drawn for parasitemia examination. The results showed that no T. evansi detected in rats in negative control group (K0), while parasitemia in group K1; K2; K3; K4; and K5 was 12,295 x106/mL; 10,495 x106/mL; 9,360 x106/mL; 5,080x106/mL; and 2,398x106/mL of blood, respectively. Percentage of inhibition of parasitemia in K2, K3, K4, and K5 reached 14.64, 23.78, 58.68, and 80.50%, respectively. Based on the result of the study, neem leaf extract of 800 mg/kg BW gave the highest reduction of parasitemia in rats infected with T. evansi.

Tobacco (Nicotiana tabacum L.) was a popular plant, usually used as cigarette material, traditional medicine, chewy tobacco and natural insecticide. This study aims to investigate toxicity effects of tobacco to mice (Mus musculus). The study had been done from March to July 2015 in Plant Physiology Laboratory of Mathematic and Natural Sciences Unimed and Phatology Laboratory USU. The study used male mice as the animal test. Twenty-five mices were divided in to 5 groups. One control group and four groups were given the ethanolic extracts of tobacco in four doses 10%; 15%; 20%; dan 25% orally. Evaluation of toxic effects was 24 hours until 3 days after administration. Evaluation included physical and physiological adverse, behaviour changes, and histopathologycal adverse of liver and kidney. According to the results of study, ethanolic extract of tobacco made a systemic toxicity. Most of doses (except control) caused acute toxicity symptoms and immediately death to the tested animals. It affected mices in physic, behaviour and anatomy. The toxic symptoms were uncontrolable movement, aggressivity, tail abnormality, coordination damages, pupil extention, and refflects damages. Another effects were gastric, intestinal and testis inflammations. The liver and kidney were highly susceptible to toxicant. Histopathologycal examination showed that toxic affected congestion and necrosis in liver and kidney. Keywords: ethanol extract, toxicity, tobacco, histopathology, mice

Chronic kidney disease in the world is currently experiencing an increase and become a serious health problem. Doxorubicin clinical efficacy is hampered by dose-related organotoxic (heart, liver, and kidney) potential. The purpose of this study was to determine the nephroprotective activity of mengkudu fruit ethanol extract against the rats induced by doxorubicin. Mengkudu fruit ethanol extract was obtained by maceration. Nephroprotective activity test is done by measuring urea and creatinine. Animals were induced with doxorubicin (DOX) 5 mg/kgbw on day 1, 7, 14 and 20th. Administration of mengkudu extract 100 mg/kgbw, 300 mg/kgbw, and 500 mg/kgbw given from day 1 to day 20 and on the 21st day blood serum levels of urea and creatinine. Mengkudu dose of 100 mg/kg BW, 300 mg/kgbw and 500 mg/kgbw have nephroprotective activity against male rats induced by doxorubicin. The effective dose of mengkudu as nephroprotective is at a dose of 500 mg/kgbw with a serum creatinine level of 0.570 ± 0.030 mg/dl and a serum urea level of 28.333 ± 6.210 mg/dl which shows a significant difference (p <0.05) of negative controls and not significantly different (p> 0.05) from positive control (Vitamin E). In the positive control group and the administration of mengkudu 500 mg/kgbw, the kidney tissue appeared normal. In the treatment group, mengkudu 500 mg/kgbw did not occur kidney tissue damage because mengkudu was able to repair kidney damage due to doxorubicin induction.

This research was conducted to determine the immunomodulatory effect of ethanol extract of neem leaves (Azadirachta indica) to the number of peritoneal macrophages in mice wich induced by BCG vaccine. Bacillus Calmette-Guerin (BCG) vaccine contained an attenuated Mycobacterium bovis. Mycobacterium bovis belongs to the Mycobacterium Tuberculosis Complex (MTC) group that has a similar phenotype characteristic with Mycobacterium tuberculosis and similar clinical manifestations of tuberculosis.The type of the research that used in this study is laboratory experimental research with Post Test Design Design Only Control Group Design. The research was conducted at the Faculty of Veterinary Medicine of Airlangga University in July 2017 using 25 male mice divided into five groups. The dosage of ethanol extract of the neem leaves given was 200 mg / Kg BW with variation for two days, four days, and six days are given.In the result of statistical data analysis using Kruskal-walis test, it is known that the significance value p = 0,03 (p <0,05), that means immunomodulatory of ethanol extract of neem leaves (Azadirachta indica) give an effect to peritoneal macrophage cell number in mice wich induced by BCG vaccine, so that neem leaves ethanol extract can be applied to tuberculosis patients.

Aim: The aim of the present study was to investigate the antibacterial effect of ethanolic extract of neem (Azadirachta indica) leaf against Gram-negative, gastric pathogen, Helicobacter pylori (H. pylori). Methodology: Extracts of neem leaf were prepared in different solventslike hexane, dichloromethane, chloroform, ethyl acetate, acetone and ethanol. Antibacterial activity was estimated in terms of zone of inhibition by performing Agar cup diffusion assay. Depending on the diameter of zone of inhibition, ethyl acetate, acetone and ethanol extract of neem leaves were selected for Thin Layer Chromatography. The presence of photochemicals were detected using iodine fumigation. Elution Assay was done to detect the bioactive components of the ethanol extract. Results: Out of sixsolvents used, ethanol extract of neem leavesshowed the maximum zone of inhibition against H. pylori. TLC separation of ethyl acetate, acetone and ethanol extract of plant products showed dark brown bands of phytochemicals on silica-gel G 60 plates. The contact bioautography assay showed a zone of 15 mm. Elution assay and agar cup bioassay was performed against H. pylori and the loading spot showed a zone of 11 mm. Interpretation: The findings of the present study revealed the anti-bacterial potency of ethanolic extracts of neem (Azadirachta indica) leaf against Gram-negative gastric pathogen H. pylori. The ethanolic extract of neem leaf can be used as an effective natural remedy in combating H. pylori infection.

Objective: This study aimed to observe the effect of the 70% ethanol extract of Orthosiphonis stamineus Benth leaves on the pharmacokineticparameters of furosemide in white male rats.Methods: 18 Sprague–Dawley male rats were divided into three groups: The normal control group was given only 1% carboxymethyl cellulose,the furosemide group was given 7.2 mg/200 g body weight (BW) suspension of furosemide, and the combination group was given 700 mg/kg BWsuspension of the 70% ethanolic extract of O. stamineus Benth leaves for 4 days followed by a 7.2 mg/200 g BW suspension of furosemide. On the4th day of treatment, we performed orbital sinus blood sampling on the eyes of the rats and analyzed the levels of furosemide in plasma using highperformanceliquid chromatography.Results: Therefore, the results showed that the administration of the 70% ethanol extract of O. stamineus Benth leaves improves the pharmacokineticparameters of furosemide on Cpmax and the area under the curve (p<0.05).Conclusion: This study concludes that the 70% ethanol extract of O. stamineus Benth leaves improves the pharmacokinetic parameters of furosemidein white male rats.

Pimpinella pruatjan Molkis a local indegineuos plant speciescommonly used  aphrodisiac.This studywas performed to analyze the effect of acute administration of purwoceng (Pimpinella pruatjan Molk.) roots ethanol extract to liver and kidney histological damagein rats. The study used an experimental approachusing post test only with control group design. Rats were randomly divided into five groups; 3 rats in each group. Group A as a control group received aquadest, group B, C, D, and E were given purwoceng roots ethanol extract dose of 5 mg/kgBW, 50 mg/kgBW, 300 mg/kgBW, and 2000 mg/kgBW respectively.Liver histological damage was assessed by a modification of the Roenigk score, whereaskidney damage was by the semiquantitative scoring of proximal tubular necrosis. UV test was used to quantify the AST and ALT levels, the measurement of blood urea levels was using the Urease-GLDH method, and Jaffe methodwas used to access the creatinine levels.Kruskal-Wallis test showed that liver and kidney histologicalparameterswere not significantly affected, as well as theblood urea and creatinine levels (p>0.05).Meanwhile,ALT level wasonly parameters which showed the significant test (p <0.05)among groups. Study concluded that the liver and kidney histological appearance, AST, blood urea, and creatinine levels in the male albino rat were not significantly affected by acute administration of Purwoceng roots in various dosesbut  the ALT level was significantly affected

Abstract Objectives Several studies have established the ethnobotanical benefits of Pupalia lappacea (PL) in laboratory animals without extensive toxicological evaluation of its safety profiles. Thus, an extensive toxicological investigation of sub-chronic oral administration of the hydroethanol leaf extract of P. lappacea in rodents was carried out in this study. Methods Different groups of rats were treated orally with the extract (10, 50 and 250 mg/kg) daily for 90 consecutive days. The control group received distilled water (10 mL/kg). After 90 days, some rats were left for additional 30 days without treatment for reversibility study. Blood and organs samples were collected for different evaluations at the end of study periods. Results The extract decreased the bodyweights, feeding and water intakes in female rats. PL increased the weights of the liver and kidney in male rats. PL increased the red blood cell (RBC), packed cell volume (PCV), hemoglobin (Hb), triglycerides (TRIG), cholesterol and high density lipoprotein (HDL) contents in rats. PL (250 mg/kg) significantly reduced the sperm motility and serum testosterone level. Cyto-architectural distortions of the testes, liver and spleen were visible. Conclusions The findings showed that P. lappacea is relatively safe at lower doses but cautions should be taken at higher dose.

The toxicity of lead acetate (L. A.) concerned to public health disruptor due to its persistence in the environment and it has the adverse influence on the human and animal health as well. It causes physiological,biochemical, and neurological dysfunctions in humans. Histologically it has a negative effect on the liver which is considered one of the major target organs where acts as detoxification machine by elimination the toxic substance from the blood in rich with it.  As well as it affects kidneys that are the two of the most filtering organs. Therefore the present study was aimed to investigate the histopathological effect of L.A. on liver and kidney tissues in male rats. Twenty male rats involved in the study were equally and randomly divided into two groups each of them involved 10 animals. Group I (castrated rats) and Group II (control) each group received 80mg/L of lead acetate dissolved in one liter distilled water by drinking for 15 days. Histological sections showed some alterations including abnormal architecture, cell degeneration, nuclear degeneration, hyperchromatic hepatocytes, immune cells, degeneration in tubules, dilation in sinusoids, dilation in central vein of liver increased bowman's space glomerular atrophy degeneration of tubular cells in liver and kidney tissues of rats in castrated rats from control group. But the size of degenerated tissue was more severe in castrated male rats. It was concluded that the castration process could produce a hypogonadism and decreased testosterone which owns many receptors in kidney and liver may produce adverse influence with L.A. administration.

Syzygium campanulatum and Syzygium aromaticum contains antioxidant components suchas flavonoids, phenolic, and terpenoids. May have hepatoprotective properties in reducing SGPT and SGOT activity. This research wants to determine the potency of hepatoprotective of ethanolic extract of Syzygium campanulatum (Korth) and Syzygium aromaticum leaf compared with curcuma tablets. This research uses 24 male Wistar rats divided into 6 groups: I, II, III (as a normal, induction, and compared control), group IV, V, VI were treated 105, 210, and 420 mg/kg BW respectively. The study was conducted for 9 days. After 7 days of treatment, treated groups were exposed by hepatotoxic dose of paracetamol (2000 mg/kg BW). The SGPT and SGOT activity of all groups was measured by enzimatic assay. The result can be concluded that Syzygium campanulatum extract was found to be active as hepatoprotective agent with 210 mg/kg BW dosage (SGPT 21.76 ± 3.98 U/L and SGOT 7.32±6.74U/L) as eff ective as with the curcuma tablets (SGPT 23.91 ± 4.41 U/L and SGOT 14.12±5.37 U/L) and the hepatoprotective activity of Syzygium campanulatum extract at a dosage 420 mg/kg BW better than curcuma tablets (SGPT 12.43 ± 6.51 U/L and SGOT 6.64 ± 5.88 U/L). While the hepatoprotec Syzygium campanulatum and Syzygium aromaticum contains antioxidant components such as flavonoids, phenolic, and terpenoids.May have hepatoprotective properties in reducing SGPT and SGOT activity.
