Background. Body mass index, measured at colorectal cancer (CRC) diagnosis has been associated with recurrence and survival outcomes. Computed tomography- (CT-) defined body compositions accurately reflect body mass, but there was no consistent perspective on the influence of visceral adipose tissue (VAT) and skeletal muscle mass (SM) on the prognosis of nonmetastasis CRC, especially in the patients underwent surgery and regularly standard chemotherapy. Methods. We investigated the associations of CT-quantified body composition (VAT and SM) with CRC patients successively underwent surgery and regular 8-12 of periods standard chemotherapy. All of the CT images were obtained at the level of the L3/4 spinal level. The prognostic value of the body compositions was analyzed using the Cox regression model, and precise clinical nomograms were established. Results. In XELOX-treated patients, progression-free survival (PFS) ( P = 0.025 ) and overall survival (OS) ( P = 0.032 ) were lower in the high-SM than in the low-SM group. The univariate analysis demonstrated that compared with low-SM patients, patients with high-SM showed a strikingly poor prognosis in both OS ( P = 0.0512 ) and PFS in the T4 subgroup ( P = 0.0417 ), while contrary to the T2-3 subgroup. Conclusions. CT-quantified body compositions have a significant influence on CRC patients successively underwent curative resection and regularly standard chemotherapy with the endpoints of 1-year, 3-year, and 5-year both OS and PFS. Patients with high-SM showed a strikingly poor prognosis in OS and PFS in the T4 subgroup; however, the prognosis role of body composition was opposite in T2-3 patients.

Introduction: Body mass index (BMI) provides little information on body composition. For example, two people with the same BMI might have different body compositions. In this sense, the development of a new BMI able to provide body composition information is of clinical and scientific interest. The aim of the study was to suggest a new modified BMI formula.Material and methods: A total of 108 subject, females 56 and males 52, 0-73 years old, in various physiopathological conditions were evaluated. Data were collected and processed by a program that through anthropometric measurements calculates classic BMI, volume, surface, V/S (that we can defined like a body-thickness “pseudospessore”) and the new BMI-BFMNU.Results: The basic formula (BMI =Body Mass [kg]/Height [m2]) uses the height squared as the value of the body surface, although this is only an approximation of the real surface, whereas using the real surface instead, the new BMI reflects better the ratio between the body volume and its surface. The ratio called "pseudospessore" is already used in literature from the BFMNU (Italian acronym refereed to Biologia e Fisiologia Modellistica della Nutrizione Umana) method and has been shown to be related to the amount of fat.Conclusions: Using the BMI-BFMNU, it is possible to obtain an indication of the body structure related to the amount of fat. The consequence is that the obtained numerical values do not coincide with the traditional BMI’s values and will refer to different normal ranges. For instance, a person may be in the range of normal weight for both BMI measurements, but only the BMI-BFMNU detects whether a person has a higher or lower fat content considering the individual’s category. This study opens up to new possible future developments on the application of the new BMI that will allow a more accurate assessment and classification of patients.

The main goal of our study was to determine how the age of children, puberty and anthropometric parameters affect the formation of body composition and faulty body posture development in children. The secondary goal was to determine in which body segments abnormalities most often occur and how gender differentiates the occurrence of adverse changes in children’s body posture and body composition during puberty. The study group consisted of 464 schoolchildren aged from 6–16. Body posture was assessed with the Zebris system. The composition of the body mass was tested with Tanita MC 780 MA body mass analyzer and the body height was measured using a portable stadiometer PORTSTAND 210. The participants were further divided due to the age of puberty. Tanner division was adopted. The cut-off age for girls is ≥10 years and for boys it is ≥12 years. The analyses applied descriptive statistics, the Pearson correlation, stepwise regression analysis and the t-test. The accepted level of significance was p < 0.05. The pelvic obliquity was lower in older children (beta = −0.15). We also see that age played a significant role in the difference in the height of the right pelvis (beta = −0.28), and the difference in the height of the right shoulder (beta = 0.23). Regression analysis showed that the content of adipose tissue (FAT%) increased with body mass index (BMI) and decreased with increasing weight, age, and height. Moreover, the FAT% was lower in boys than in girls (beta negative equal to −0.39). It turned out that older children (puberty), had greater asymmetry in the right shoulder blade (p < 0.001) and right shoulder (p = 0.003). On the other hand, younger children (who were still before puberty) had greater anomalies in the left trunk inclination (p = 0.048) as well as in the pelvic obliquity (p = 0.008). Girls in puberty were characterized by greater asymmetry on the right side, including the shoulders (p = 0.001), the scapula (p = 0.001) and the pelvis (p < 0.001). In boys, the problem related only to the asymmetry of the shoulder blades (p < 0.001). Girls were characterized by a greater increase in adipose tissue and boys by muscle tissue. Significant differences also appeared in the body posture of the examined children. Greater asymmetry within scapulas and shoulders were seen in children during puberty. Therefore, a growing child should be closely monitored to protect them from the adverse consequences of poor posture or excessive accumulation of adipose tissue in the body.

Abstract This study is to establish NMF (nonnegative matrix factorization) typing related to the tumor microenvironment (TME) of colorectal cancer (CRC) and to construct a gene model related to prognosis to be able to more accurately estimate the prognosis of CRC patients. NMF algorithm was used to classify samples merged clinical data of differentially expressed genes (DEGs) of TCGA that are related to the TME shared in The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO) datasets, and survival differences between subtype groups were compared. By using createData Partition command, TCGA database samples were randomly divided into train group and test group. Then the univariate Cox analysis, Lasso regression and multivariate Cox regression models were used to obtain risk model formula, which is used to score the samples in the train group, test group and GEO database, and to divide the samples of each group into high-risk and low-risk groups, according to the median score of the train group. After that, the model was validated. Patients with CRC were divided into 2, 3, 5 subtypes respectively. The comparison of patients with overall survival (OS) and progression-free survival (PFS) showed that the method of typing with the rank set to 5 was the most statistically significant (p=0.007, p<0.001, respectively). Moreover, the model constructed containing 14 immune-related genes (PPARGC1A, CXCL11, PCOLCE2, GABRD, TRAF5, FOXD1, NXPH4, ALPK3, KCNJ11, NPR1, F2RL2, CD36, CCNF, DUSP14) can be used as an independent prognostic factor, which is superior to some previous models in terms of patient prognosis. The 5-type typing of CRC patients and the 14 immune-related genes model constructed by us can accurately estimate the prognosis of patients with CRC.

The purpose of this study was to assess the body composition of male and female basketball athletes (n = 323) across season, year, and sport-position using air displacement plethysmography. An independent sample t-test assessed sport-position differences. An analysis of variance was used to assess within-subjects across season (pre-season, in-season, and off-season), and academic year (freshman, sophomore, and junior). For both men and women basketball (MBB, WBB) athletes, guards had the lowest body fat, fat mass, fat free mass, and body mass. No seasonal differences were observed in MBB, but following in-season play for WBB, a reduction of (p = 0.03) in fat free mass (FFM) was observed. Across years, MBB showed an increase in FFM from freshman to sophomore year, yet remained unchanged through junior year. For WBB across years, no differences occurred for body mass (BM), body fat (BF%), and fat mass (FM), yet FFM increased from sophomore to junior year (p = 0.009). Sport-position differences exist in MBB and WBB: Guards were found to be smaller and leaner than forwards. Due to the importance of body composition (BC) on athletic performance, along with seasonal and longitudinal shifts in BC, strength and conditioning practitioners should periodically assess athletes BC to ensure preservation of FFM. Training and nutrition programming can then be adjusted in response to changes in BC.

Abstract Introduction: The aim of the present study was to evaluate the influence of body composition on the postural sway during quiet standing. Our hypothesis is that men and women do not have the same relation between body composition and postural sway during quiet standing. Materials and Methods: Participated in the study 50 men and 50 women; age range: 20-40 years old. The main outcome measures were: Body composition (bone densitometry), percentage of fat (% fat) tissue (g), fat (g), lean mass (g), bone mineral content (g) and bone mineral density (g/cm2); Anthropometry: body mass (kg), height (cm), length of the trunk-head (cm), length of lower limbs (cm). The following indices were calculated: body mass index (BMI) (kg/m2) and Postural balance test - center ofpressure displacement. Results: The correlation analysis showed low correlations between postural sway and anthropometric variables. The multiple linear regression model showed that the body composition and the anthropometry were able to explain only men's postural sway. Conclusion: The postural sway is sex type dependent. Men and women have different relations between body composition and postural sway. Only male's body composition affected the body sway.

Background. The most effective treatment of immune checkpoint inhibitors (ICIs) is restricted in microsatellite instability (MSI-H) subsets of advanced colorectal cancer, but MSI-H only accounts for 4-5% among them. ICIs are completely ineffective in advanced colorectal cancer patients with microsatellite stable (MSS), according to literatures published. Regorafenib is a novel tyrosine kinase inhibitor (TKIs) that could normalize tumor blood vessels by inhibiting vascular endothelial growth factor receptor and its downstream, thus improving cytotoxic T cell infiltration in tumor microenvironment, which has a synergistic effect with ICIs. Toripalimab is a type of anti-PD-1 monoclonal antibody produced by Junshi Biosciences in China. Herein, we aimed to explore the efficacy and safety of regorafenib combined with toripalimab in the third-line and beyond treatment of advanced colorectal cancer. Methods. We evaluated the outcomes of MSS patients with advanced colorectal cancer who received regorafenib combined with toripalimab in the Second Affiliated Hospital of Nanchang University from June 2019 to January 2021. These patients had previously received at least second-line treatment; the regimens were oxaliplatin and irinotecan-based chemotherapy and/or accompanied with bevacizumab or cetuximab. Thirty-three patients were treated orally with regorafenib 80 mg or 120 mg once daily for 21 days, 28 days as a cycle, combined with intravenous toripalimab until disease progression or intolerant to adverse reactions. We used the Kaplan–Meier method to estimate the rate of progression-free survival (PFS) and log-rank method to do a statistical test of the survival curve. The Cox regression model was used to analyze the influence of multiple factors on PFS. The primary endpoints were objective remission rate (ORR) and disease control rate (DCR). The secondary endpoints were the incidence of adverse reactions and median progression-free survival (mPFS). Results. The evaluation of treatment effects was assessed according to RECIST 1.1. Four patients (12.12%) got partial response, twelve patients (36.36%) experienced stable disease, and seventeen patients (51.52%) suffered progressive disease. ORR was 12.12% and DCR was 48.48%. mPFS was 113 days (95% CI: 0–272.1). In univariate analysis, patients who had previously received second-line treatment were significantly better than those who had received third-line or more treatment ( p = 0.005 ). Lung metastasis was a negative factor in combined therapy ( p = 0.032 ). Five patients without previous treatment of bevacizumab were effective. Previous treatment without bevacizumab showed a trend of effective when combination therapy ( p = 0.034 ). It was also a positive factor that the Eastern Cooperative Oncology Group performance status (ECOG) score was 0 ( p = 0.034 ). Multivariable Cox regression analysis showed the number of previous chemotherapy lines and excision of primary lesions were independent prognostic factors. The most common treatment-related adverse reactions were hand-foot syndrome (33.33%), liver dysfunction (27.27), hypothyroidism (24.24%), fever (24.24%), fatigue (21.21%), leukopenia (15.15%), hypertension (12.12%), platelet count decreased (6.06%), diarrhea (3.03%), and myocarditis (3.03%); one patient stopped treatment as myocarditis. The incidence of grade 3/4 adverse reactions was 9.09%. Conclusions. Regorafenib combined with toripalimab has a promising effect in the third-line and beyond treatment of advanced colorectal cancer. In the early use of combination therapy, excision of primary lesions can have a positive impact in regorafenib and toripalimab combination. This treatment-related adverse reactions are tolerant in combined therapy.

PurposeActivating mutation of the KRAS oncogene is an established predictive biomarker for resistance to anti–epidermal growth factor receptor (anti-EGFR) therapies in advanced colorectal cancer (aCRC). We wanted to determine whether KRAS and/or BRAF mutation is also a predictive biomarker for other aCRC therapies.Patients and MethodsThe Medical Research Council Fluorouracil, Oxaliplatin and Irinotecan: Use and Sequencing (MRC FOCUS) trial compared treatment sequences including first-line fluorouracil (FU), FU/irinotecan or FU/oxaliplatin in aCRC. Tumor blocks were obtained from 711 consenting patients. DNA was extracted and KRAS codons 12, 13, and 61 and BRAF codon 600 were assessed by pyrosequencing. Mutation (mut) status was assessed first as a prognostic factor and then as a predictive biomarker for the benefit of adding irinotecan or oxaliplatin to FU. The association of BRAF-mut with loss of MLH1 was assessed by immunohistochemistry.ResultsThree hundred eight (43.3%) of 711 patients had KRAS-mut and 56 (7.9%) of 711 had BRAF-mut. Mutation of KRAS, BRAF, or both was present in 360 (50.6%) of 711 patients. Mutation in either KRAS or BRAF was a poor prognostic factor for overall survival (OS; hazard ratio [HR], 1.40; 95% CI, 1.20 to 1.65; P < .0001) but had minimal impact on progression-free survival (PFS; HR, 1.16; 95% CI, 1.00 to 1.36; P = .05). Mutation status did not affect the impact of irinotecan or oxaliplatin on PFS or OS. BRAF-mut was weakly associated with loss of MLH1 staining (P = .012).ConclusionKRAS/BRAF mutation is associated with poor prognosis but is not a predictive biomarker for irinotecan or oxaliplatin. There is no evidence that patients with KRAS/BRAF mutated tumors are less likely to benefit from these standard chemotherapy agents.

Regular exercise during school hours is encouraged; however many children and adolescents fail to meet the recommendations during this time. Extracurricular activities may be a more appealing way for youth to achieve guidelines, and it is recommended that they attend two sessions each week. The aim of the study was to assess the influence of participation in a national physical activity program accompanied by nutritional education for trainers on the risk of obesity and body composition in a nationwide sample of boys and girls, after one year of intervention. The #goathletics Study was conducted in a group of 1014 adolescents aged 12–13: 507 individuals for the Athletics for All program (210 boys, 297 girls) and 507 pair-matched individuals not participating in any physical activity program (matching including: gender, age, city of residence). The body mass (kg), Body Mass Index (BMI) (kg/m2), waist circumference (WC) (cm), waist-to-height ratio (WHtR) (-) and body composition (%) (measured using bioelectrical impedance method) were compared in a gender-related sub-groups using t-Student test (for parametric distributions) or Mann-Whitney U test (for nonparametric distributions) and chi2 test (for the share of sub-groups). After one year of intervention, lower body mass percentile, BMI percentile, WC, WHtR and fat mass share, higher muscle mass share, as well as lower frequency of overweight/obesity and abdominal fat distribution were observed both for boys and girls participating in the physical activity intervention compared to the pair-matched controls. The after-school physical activity program accompanied by nutritional education for trainers may be a highly effective method for reducing the risk of obesity both for boys and girls, as regular participation is ensured.

Physical activity (PA) can bring incalculable benefits to people throughout the aging process. The objective of this study was to determine the relationships between the leg and arm strength, body composition and balance of elderly people. Twenty-one people (9 men and 12 women) over 60 years of age voluntarily participated in this study. Bioimpedance tests were carried out to obtain body mass data, manual dynamometer tests and the lower limbs test were conducted to obtain strength values, ​​and the stork balance stand test was conducted to assess static balance. As a result, medium-high correlation coefficients were obtained between the aforementioned dependent variables. The highest value corresponded to the relationship between the results of the dominant hand strength test (right hand in all participants) and the lean body mass (LBM) of the participant (R2 = 75%, p < 0.001). The results indicated that strength capacity, recorded by dynamometry, and balance ability have a direct relationship with the body composition of the subject, especially with their lean body mass. These data suggest that a greater lean body mass can provide greater strength and balance in the elderly, so it would improve or have greater duration the functionality and independence of the person, thereby justifying direct benefits in people and indirect benefits in public administrations that finance these social issues.
