The reduction in estrogen levels results in a decrease in bone density at menopause. Irisin is a myokine that modulates the benefits of exercise, which may include bone health. This study was planned to examine irisin’s impact in preventing osteoporosis after ovariectomy. 4 groups of female albino rats (10 rats/group): control, sham-operated, ovariectomized (OVX-control), and OVX-irisin-treated. Serum levels of bone markers [osteocalcin (OC), bone alkaline phosphatase (BALP), tartrate-resistant acid phosphatase (TRAP), calcium (Ca++), phosphorus (P)], glucose, and insulin were being measured. Body mass index, Homeostatic Model Assessment of Insulin Resistance (HOMA-IR), dry and ash femur weight, and bone contents of Ca++ and P were investigated. The femur was examined histopathologically. The OVX-control group showed an increase in serum levels of OC, BALP, TRAP, calcium, phosphorus, BMI, glucose, insulin, and HOMA-IR ( P < 0.05 ) and a reduction in dry and ash weight of the femur, the concentration of calcium and phosphorus content in bone ash ( P < 0.05 ). The OVX-irisin-treated group exhibited a decrease in serum levels of OC, BALP and TRAP, calcium, phosphorus, BMI, glucose, insulin, HOMA-IR ( P < 0.05 ), and a rise in dry and ash weight of the femur, the concentration of calcium and phosphorus in bone ash ( P < 0.05 ). Histological examination of the distal femur diaphysis of the OVX-irisin-treated group exhibited proper bone architecture and density compared with that of the OVX-control group. It is concluded that irisin treatment in the OVX rats safeguarded the regular bone architecture and normal levels of serum bone biomarkers. Irisin may be a possible novel target in the prohibition of postmenopausal osteoporosis.

Abstract Background Vitamin B12 (Vit B12) deficiency results in elevated homocysteine levels and interference with collagen cross-linking, which may affect tendon integrity. The purpose of this study was to investigate whether serum Vit B12 levels were correlated with degenerative rotator cuff (RC) tear. Methods Eighty-seven consecutive patients with or without degenerative RC tear were enrolled as study participants. Possible risk factors (age, sex, medical history, bone mineral density, and serum chemistries including glucose, magnesium, calcium, phosphorus, zinc, homocysteine, Vitamin D, Vit B12, homocysteine, and folate) were assessed. Significant variables were selected based on the results of univariate analyses, and a logistic regression model (backward elimination) was constructed to predict the presence of degenerative RC tear. Results In the univariate analysis, the group of patients with degenerative RC tear had a mean concentration of 528.4 pg/mL Vit B12, which was significantly lower than the healthy control group (627.1 pg/mL). Logistic regression analysis using Vit B12 as an independent variable revealed that Vit B12 concentrations were significantly correlated with degenerative RC tear (p = 0.044). However, Vit B12 levels were not associated with tear size. Conclusion Low serum levels of Vit B12 were independently related to degenerative RC tear. Further investigations are warranted to determine if Vit B12 supplementation can decrease the risk of this condition.

Abstract The aim of the present study was to examine the effect of prolonged use of finasteride on serum levels of dihydrotestosterone (DHT), estradiol (E2), progesterone, testosterone and androstenedione in women during the menstrual period. Further, to screen and compare the 5α-reductase activities through the expression of SRD5A1, SRD5A2 and AR gene and to determine the level of VEGF, VKOR and SAA gene expression and DNA damage. A total of 30 Saudi women aged between 25 and 35 years were enrolled in the study. The selected women were divided into two groups. The first group (n = 15) received 5 mg finasteride/day for prolonged period of one year and second group (n = 15) was taken as a healthy control. ELISA technique was used for measuring the serum levels of the targeted hormones, and Comet assay was used for checking the DNA integrity. Our findings revealed significant decrement of DHT, E2, progesterone and androstenedione levels and elevated levels of testosterone in group treated with daily oral doses of 5 mg finasteride/day compared with the control subjects. mRNA expression suggested that finasteride has concrete effects on the gene expression of the selected genes from the treated group in comparison with the control group. In addition, finasteride induced DNA damage, and heavy menstrual bleeding was noted in women treated with finasteride. In conclusion, the present findings revealed that finasteride has adverse health effects in women associated with gonadal sex steroids alterations, DNA damage and heavy menstrual bleeding with no consensus in the treatment of androgenetic alopecia in women.

Abstract Background Modulatory effects of soy extract and estradiol on the central nervous system (CNS) have been reported. The effect of soy on scopolamine-induced spatial learning and memory in comparison to the effect of estradiol was investigated. Materials and methods Ovariectomized rats were divided into the following groups: (1) control, (2) scopolamine (Sco), (3) scopolamine-soy 20 (Sco-S 20), (4) scopolamine-soy 60 (Sco-S 60), (5) scopolamine-estradiol 20 (Sco-E 20) and (6) scopolamine-estradiol 60 (Sco-E 60). Soy extract, estradiol and vehicle were administered daily for 6 weeks before training in the Morris water maze (MWM) test. Scopolamine (2 mg/kg) was injected 30 min before training in the MWM test. Results In the MWM, the escape latency and traveled path to find the platform in the Sco group was prolonged compared to the control group (p < 0.001). Treatment by higher doses of soy improved performances of the rats in the MWM (p < 0.05 – p < 0.001). However, treatment with both doses of estradiol (20 and 60 μg/kg) resulted in a statistically significant improvement in the MWM (p < 0.01 – p < 0.001). Cortical, hippocampal and serum levels of malondialdehyde (MDA), as an index of lipid peroxidation, were increased which was prevented by soy extract and estradiol (p < 0.001). Cortical, hippocampal as well as serum levels of the total thiol, superoxide dismutase (SOD) and catalase (CAT) in Sco group were lower than the control group (p < 0.001) while they were enhanced when the animals were treated by soy extract and estradiol (p < 0.01 – p < 0.001). Conclusions It was observed that both soy extract and estradiol prevented learning and memory impairments induced by scopolamine in ovariectomized rats. These effects can be attributed to their protective effects on oxidative damage of the brain tissue.

Objective. Sulphur, similar to phosphorus, is easily attached to organic compounds. The inadequate elimination of sulphate may cause high sulphate concentrations in hemodialysis (HD) patients because sulphate is low in free form in plasma. Although we are well aware of the accumulation of phosphorus in chronic dialysis patients, we do not have an adequate knowledge database about the sulphur compounds. This study was designed to determine the level of sulphate in hemodialysis patients. Materials and Methods. Ninety-four prevalent HD patients and 33 patients without renal failure were included in the study. The serum inorganic sulphate levels were measured by turbidimetric technique. Moreover, the serum level of urea, creatinine, albumin, calcium, phosphorus, and parathyroid hormone concentrations was simultaneously recorded. Results. Mean levels of plasma sulphate were significantly higher (0.56 ± 0.17 mM vs 0.31 ± 0.13 mM, p<0.001) in HD patients. Serum sulphate level correlated with patient’s age, serum albumin, serum BUN and creatinine, and serum phosphorus level in HD patients. Serum sulphate levels were not associated with serum parathyroid hormone levels. Conclusion. Serum sulphate levels were approximately twofold higher in HD patients than in the normal control group. Inorganic sulphate does not seem to accumulate in long-term dialysis patients, and mild increased serum levels of sulphate has no poor clinical outcome in these patients.

Abstract Background: Subjects with normal weight obesity (NWO) are supposed to cardiometabolic disorders. The aim of this study was to investigate the circulating levels of vaspin and leptin and their association with glycemic and lipid profiles in women with NWO compared to controlsMethods: Forty women with body mass index (BMI)=18.5-24.9 kg/m2 and fat mass (FM) ≥ 30% as NWO group and 30 age matched women with same BMI range and FM<30% as control group were enrolled in this study. Anthropometric measurement, fasting serum levels of blood sugar (FBS), insulin, Glycated hemoglobin (HbA1c), lipid profiles and also, leptin and vaspin were measured.Results: The mean ± standard deviation (SD) of age was 28.76 ± 4.76 years in NWO group and 29.23 ± 4.50 years in controls. Subjects in NWO group had higher serum levels of insulin (9.02 ± 4.75 vs.6.24 ± 2.51, p= 0.009), leptin (17.31 ± 8.10 vs. 9.94 ± 4.30, p<0.001) and homeostatic model assessment for insulin resistance (HOMA-IR) (33.77 ± 20.71 vs. 23.48 ± 10.03, p=0.009) as compared to normal weight non-obese (NWNO) as control group. Serum level of vaspin in NWO group (34.82 pg/ml) was higher than that in controls (27.72 pg/ml), (p=0.12). In NWO group, serum level of leptin was correlated positively with FBS (r=0.45, p=0.02), insulin (r=0.51, p=0.008), and HOMA-IR (r=0.46, p=0.02) and vaspin concentration was positively associated with insulin (r=0.36, p= 0.02) and HOMA-IR (r=0.30, p=0.06). Conclusion: We observed that concentration of insulin and HOMA-IR index were significantly increased in women with NWO compared to the controls. Higher levels of leptin and vaspin in NWO were associated with glycemic profiles in NWO group.

Background: Postmenopausal osteoporosis has become a global health issues recently. Several vitamins play a role in building and maintenance of bones. Objective: To evaluate the possible roles of vitamin B-complex supplementation alone or combined with estrogen in the improvement of osteoporosis in ovariectomized (OVX) rats. Materials and Methods: Fifty adult female albino rats were equally divided into 5 groups then 4 groups were exposed to ovariectomy: Control group, OVX group, OVX/ Ethinyl estradiol (EE) group, OVX/B-complex group and OVX/EE/B-complex only. All treatments were started 4 weeks after surgery and continued for 8 weeks. Blood samples were collected to measure serum levels of homocysteine (HCY), bone–specific alkaline phosphatase (BAP), collagen type 1 cross-linked C-telopeptide (CTX) and calcium (Ca++). Results: Ovariectomy increased serum levels of Ca++, bone turnover markers (BAP and CTX) and homocysteine (HCY). Serum Ca++ levels were increased, while serum levels of bone markers and HCY were decreased) in all treated groups but, only rats treated with EE and vitamin B-complex returned the serum levels of CTX and HCY to normal. Conclusion: Vitamin B-complex showed some antiosteoporotic effects in OVX rats.

Trigona honey (TH) is well known for its therapeutic characteristics. To date, the study of Trigona honey as a prophylactic or immune booster prior to the bacterial infection of the invivo model is not well covered. This study aims to investigate anti-inflammatory and immune activities in Wistar rats infected with respiratory infection following with Trigona honey. 25 Wistar rats were assigned to possitive groups, negative control group, positive control group was fed TH (5 g / kg body weight) orally, the untreated group was infected with Staphylococcus aureus to induce respiratory infection, the treated group has been infected with S. aureus followed by treatment with TH at a dose of 1.5 ×108 CFU / mL and the preventive group ingested TH one week before S. aureus infection. Blood was obtained for biochemical analysis. Lung tissues have been collected for molecular examination. The results showed a significant decrease in serum levels of ALT, AST, urea and creatinine in the preventive and treated groups, serum IgG increased significantly (P<0.05) in the preventive and treated groups, IFN-y increased in the preventive group while decreased in the treated group, and IL-8 increased in the treated group while decreased in the preventive group. The mRNA expression of AGP is up-regulated in the positive control, preventive and treated groups. The α2-MG, TNF-α , and mRNA expressions showed lower regulation after administration of TH in preventive and treated groups. The results show the ability of TH to counteract immune and inflammatory changes in serum levels and gene expressions.

Purpose: To determine the effect of Lactobacillus johnsonii Ncc533 (La1) on Th1/Th2 balance, the production of IL-4 and IFN-γ by splenocytes was evaluated following its administration to mice from newborn to adult. Changes in IL-4 and IFN-γ expression and serum levels of OVA-specific-IgE were then investigated in an asthma model. Methods: Using flow cytometry (FCM) and ELISA, the percentage of IL-4 and IFN-γ expressing splenocytes and serum levels of OVA-specific-IgE were measured in different groups of mice. Results: The percentages of IL-4 and IFN-γ expressing splenocytes in the offspring and in the adults of the La1-treated group were not significantly different when compared with the water-treated group. In the asthma model, the percentages of IL-4 expressing cells and the serum levels of OVA-specific-IgE in the La1-treated and water-treated group were significantly increased compared with those in the control group. The percentage of IFN-γ expressing cells was significantly lower in the La1-treated and water-treated groups. The percentage of IL-4 expressing cells and the serum levels of OVA-specific-IgE in the La1-treated group were significantly lower compared with those in the water-treated group, whereas the percentage of IFN-γ expressing cells was significantly higher. Conclusion: Administration of La1 had no effect on the immune system from the neonate to the adult in the normal mice. It did, however, significantly alter the percentages of IL-4 or IFN-γ expressing CD4+ T lymphocytes in the asthma model, suggesting that administration of La1 might regulate the immune response.

The mortality of individuals suffering from depression has been increasing, noticeably of postmenopausal women; consequently, their care and treatment are significant to retain a high quality of life. The aim of this study was to examine the effect of Camellia sinensis (CS) on repeated stress-induced changes of the depression related function on the tail suspension test (TST), forced swimming test (FST) in ovariectomized female rats. After behavioral test, we evaluated the changes in the neurotransmitter by measuring the level of dopamine in the nucleus accumbens (NaC) and the serum levels of estrogen and oxytocin. We used 18F-2-fluoro-deoxy-D-glucose positron emission tomography (18F-FDG-PET) to examine the effects of CS on glucose metabolism in ovariectomized rats. Female rats were randomly segregated into three groups. Nor group was considered as nonoperated and nonstressed group, while the control was the ovariectomized and stressed group (OVX+ST), and CS was the ovariectomized, stressed and CS treated group. The rats were exposed to immobilization stress (IMO) for 14 d (2 h/d), and CS (300 mg/kg, i.p.) was treated 30 min before IMO stress. Significant reduction of immobility in the TST and FST was indicated in rats treatment with CS compared to the control group (OVX+ST). The levels of estrogen in the serum of the Nor and CS groups were significantly elevated compared to the OVX+ST group. Also, CS activated brain glucose metabolism in the cortex. The present findings suggested that CS had antidepressant effectiveness in a menopausal depression animal model. These findings suggest evidence that CS plays a crucial role in stressful situation, providing that CS might be a dependable antidepressant medicine to treat menopausal depression.

The objective of the study was to find whether a single intratracheal inoculation with live Porphyromonas gingivalis ATCC 33277 influences local and systemic inflammatory and immune responses in mice.Twelve-week-old BALB/c mice were intratracheally inoculated with 2.9 × 109 CFU P. gingivalis ATCC 33277 diluted in 40 μl sterile phosphate buffer (treated group) or with sterile PBS (control group). The animals were sacrificed 2, 6, 24, 72 and 168 h after inoculation. TNFα, IL-1β, IL-6 and total protein concentrations were measured in the serum, lungs and kidneys. Six hours after P. gingivalis inoculation, TNFα concentration was significantly increased in serum (p = 0.02) and kidneys (p = 0.04), but in the lungs TNFα production was enhanced already 2 h (p < 0.0001) after inoculation, reaching the peak after 6 h (p < 0.0001). The IL-1β concentration was also significantly increased in serum after 2 h (p = 0.006), remaining significantly elevated up to 3 days (p ≤ 0.0001) after inoculation. In lungs IL-1β levels were significantly increased 6 and 24 h (p < 0.0001) and in kidneys 24 h (p < 0.0001) and 168 h (p = 0.01) after inoculation. The IL-6 concentration was significantly increased in serum after 72 and 168 h (p < 0.0001). However, IL-6 was significantly increased in lungs after 6 h (p < 0.0001), remaining elevated until 72 h and in kidneys 2 and 6 h (p < 0.0001) after inoculation. Significantly increased total protein concentration was detected in kidneys 6 and 24 h (p < 0.0001) after inoculation. These results suggest that a single intratracheal inoculation with P. gingivalis stimulates the local and systemic inflammatory and immune response, as shown by increased tissue and serum levels of proinflammatory cytokines.
