Syzygium aqueum, consisting of various fruit colors, is one of the plants that have been used as traditional medicine. This study aims to evaluate and compare phytochemical, antioxidant, and cytotoxic activities and total phenolic content of leaves and stem bark extracts of S. aqueum with pink and red fruits, in order to identify the best extract that can be used as a natural antioxidant. Phytochemical constituents were evaluated qualitatively using chemicals, while cytotoxic activities were identified using the brine shrimp lethality test. Total phenolic content was determined via the Folin–Ciocalteu method. Leaves and stem bark of S. aqueum contained flavonoids, phenolics, and triterpenoids, but the stem bark also contained saponins and alkaloids. Methanol and ethyl acetate extracts of leaves and stem bark were categorized as very powerful antioxidants to DPPH (IC50 9.71–38.69 μg/mL) and hydrogen peroxide (IC50 16.44–44.02 μg/mL), while hexane extracts were inactive. Methanol, ethyl acetate, and hexane extracts of leaves and stem bark were categorized as moderately cytotoxic to A. salina larvae (LC50 104.04–440.65 μg/mL). Comparing leaves and stem barks, antioxidant and cytotoxic activities of stem bark extracts were higher than those of leaves extracts. Total phenolic content of leaves extracts was higher than that of stem bark extracts where the order of total phenolic content progressed from methanol extracts > ethyl acetate extracts > hexane extracts. Therefore, the stem bark of S. aqueum was identified as the better source of natural antioxidant compared with the leaves.

Abstract Cadaba farinosa is used in traditional medicine for treatment of cancer, diabetes, and rheumatism. The research was aimed at evaluating the phytochemical and antioxidant activity of the extracts. The powdered stem bark was extracted successively with the aid of Soxhlet extractor using n-hexane, ethyl acetate, and methanol. The resulting extracts were concentrated on rotary evaporator and the percentage yields were calculated. The phytochemical and TLC profiles of the extracts were studied. The antioxidant activity of the extracts and ascorbic acid (standard) were determined using 2, 2-diphenyl-1-picrylhydrazyl (DPPH) free radical assay. The total phenolic content of the extracts and tannic acid (standard) were evaluated using Folin–Ciocalteu reagent. The percentage yields of n-hexane, ethyl acetate, and methanol extracts were found to be 1.19, 1.37, and 13.93%, respectively. The phytochemical screening revealed the presence of tannins, flavonoids, saponins, cardiac glycosides, alkaloids, and triterpenoids. The TLC profiles of the extracts revealed the presence of compounds as evidenced from their R f values. The total phenolic content of ethyl acetate and methanol extracts were found to be 135 and 112 mg, respectively. The free radical scavenging activity demonstrated by the extracts was comparable to ascorbic acid. The ethyl acetate extract had higher phenolic content and demonstrated the highest free radical scavenging with IC50 31.07 mg/mL. The results of research have provided strong preliminary evidence of antioxidant activity which may be because of phenolic compounds in the extracts.

Abstract The bark of Chamaecyparis lawsoniana (A. Murray) Parl. was extracted with methanol and sequentially partitioned with n-hexane, ethyl acetate, n-butanol and deionized water. The antioxidant activities of the four extracts were evaluated using the DPPH• and ABTS+• methods. The total phenolic content of the extracts was determined using the Folin-Ciocalteu method and expressed as gallic acid equivalents (GAE). Butylated hydroxytoluene was used as a positive control in the radical-scavenging activity tests. All the bark extracts showed significant radical-scavenging activity. In the ABTS+• assay, n-butanol extracts exhibited the strongest radical-scavenging activity, followed by ethyl acetate, water, and n-hexane extracts. The greatest total phenolic content was 428.54 mg GAE per gram of dry extract and was detected in the n-butanol extract, followed by the ethyl acetate and n-hexane extracts. The antioxidant activities correlate with the amount of phenolics present in these extracts. The ethyl acetate and n-butanol extracts were rich in phenolics and may represent a good source of antioxidants.

One of the traditionally used herbs is the byproduct of the maize plant, the 10-20 cm long corn silk which from the female maize flowers. The aim of this study is to evaluate the total phenolic content, antioxidant activity, and cytotoxicity activity of corn silk. The corn silk was minced and was extracted with methanol-water (80 % v/v), methanol, ethanol, ethyl acetate, and hexane by using the maceration method. The total phenolic content (TPC) of corn silk was determined to assess the presence and level of phenolic compounds in each sample. The antioxidant activities of all corn silk extracts were determined via DPPH method, and MTT assay was used to study the viability of the cells after the cells were treated with corn silk extracts at different time intervals. The highest phenolic content was exhibited by the methanol extract. The EC50 value for methanol-water (80 % v/v), methanol, ethanol, ethyl acetate and hexane extracts were 251 μg/ ml, 300 μg/ ml, 330 μg/ ml, 550 μg/ ml and 1736 μg/ ml respectively. The MTT assay, the lowest IC50 values at 24 and 48 hours intervals, was exhibited by methanol-water extract (104 μg/ ml). In contrast, methanol (308 μg/ ml) was found with the highest IC50 value for all 24, 48, and 72 hours intervals. At 72 hours interval, ethyl acetate (88 μg/ ml) shown the lowest IC50 value. This study suggested that corn silk could be potentially used as a source of antioxidant and can further evaluate for cancer studies.

Current treatments for complex diseases have remarkable side effects that negatively impact patients’ quality of life. Thus, natural compounds with fewer side effects represent a promising source for safe drugs. The genus Senecio is widely used in folk medicine due to its various pharmacological properties. In the present study, the total phenolic content of Senecio glaucus, which is grown in Saudi Arabia, was assessed using the Folin-Ciocalteau colorimetric method. Scavenging DPPH and ABTS assays were utilized to determine the antioxidant properties of S. glaucus fractions, and MTT assay was used to screen the cytotoxic activity of S. glaucus against various cancer cells. In addition, HPLC-UV was utilized to detect the presence of two phenolic acids, namely, vanillic acid (VA) and gallic acid (GA). Among all fractions tested, S. glaucus chloroform fraction (SGCF) yielded the highest value (125.3 mg·GA/g) in terms of total phenolic content. SGCF also exhibited the highest scavenging activities (76.7 and 74.1%) on both DPPH and ABTS assays, respectively. Similarly, SGCF also possessed the most potent cytotoxic activity against the MCF-7 cell line, with an IC50 value of 41.8 μg/ml. The validated HPLC method confirmed the presence of VA (4.8 μg/mg DW) and GA (3.9 μg/mg DW) in SGCF. Overall, our data show that S. glaucus had antioxidant and cytotoxic properties. A developed validated HPLC method which could be helpful for quantifying phenolic compounds in S. glaucus was established.

AbstractIn vitro antimicrobial activity of 21 crude extracts obtained from seven taxa of the genus Teucrium (T. chamaedrys, T. montanum, T. arduini, T. polium, T. scordium subsp. scordium, T. scordium subsp. scordioides and T. botrys) was tested against bacterial and fungal species. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were determined using a microdilution analysis method. Total phenolic content and flavonoid concentrations were measured spectrophotometrically. Total phenols were determined using Folin-Ciocalteu reagent and their amounts ranged from 28.49 up to 159.84 mg CA/g of extract (chlorogenic acid equivalent). The amounts of flavonoids ranged from 38.17 up to 190.45 mg RU/g of extract (rutin equivalent). The plant extracts showed greater potential of antibacterial than antifungal activity. A relationship was found between total phenolics and biological activity. The highest level of total phenols was measured in the methanol extracts, which demonstrated higher antimicrobial activity than acetone and ethyl acetate extracts. Staphylococcus aureus ATCC 25923 appeared to be the most sensitive organism. Our results indicate that Teucrium spp extracts are rich sources of phenolic compounds and are promising candidates for further development as natural antimicrobial agents.
