Antibiotic-resistant Staphylococcus aureus isolates pose a severe challenge to both veterinary and health professions and dairy cattle producers. Cross-sectional study was conducted from November 2014 to May 2015 to isolate and identify S. aureus from mastitic cows’ milk and estimate the occurrence of MRSA in the dairy farms of Shire. Physical examination and California mastitis test were performed on a total of 220 dairy cows. Bacteriological isolation and identification and antibiogram testing were performed. Furthermore, multiplex polymerase chain reaction (PCR) was done for the detection of mec A and fem A genes. Out of the 220 dairy cows, 64 (29.09%) were positive for bovine mastitis, and of these, 32.81% were coagulase-positive S. aureus (CoPS). Antibiogram test results showed 100% of the isolates were resistant to penicillin G, nalidixic acid, and ampicillin, and 33.33% of the CoPS showed resistance to oxacillin (phenotypically MRSA positive). But 38.09% of the CoPS were found to be resistant and susceptible to vancomycin. PCR amplification of the seven phenotypically MRSA isolates revealed that 42.9% and 71.4% of them were found to carry fem A and mec A genes, respectively. The current study revealed the existence of alarming level of CoPS and development of multidrug resistance.

A cross-sectional study was conducted from May 2016 to January 2017 in Rubavu and Nyabihu districts, Western Rwanda, aiming at estimating the prevalence of subclinical mastitis (SCM) and identifying its causative bacteria. Management practices and milking procedures were recorded through a questionnaire. 123 crossbreed milking cows from 13 dairy farms were randomly selected and screened for SCM using California Mastitis Test (CMT). Composite CMT positive milk samples were processed for bacterial isolation and identification. The overall SCM prevalence at cow level was 50.4%. 68 bacterial isolates were identified by morphological and biochemical characteristics. They included, Coagulase Negative Staphylococci (51.5%), Staphylococcus aureus (20.6%), Streptococcus species (10.3%), Bacillus species (10.3%), Streptococcus agalactiae (5.8%), and Escherichia coli (1.5%). About 67.1% of the farmers checked for mastitis; of these, 58.9% relied on clinical signs and only 6.8% screened with CMT. Only 5.5% and 2.7% of the farmers tried to control mastitis using dry cow therapy and teat dips, respectively. Thus, to reduce the prevalence of SCM, farmers in the study area need to be trained on good milking practices, including regular use of teat dips, application of dry cow therapy, and SCM screening. This will improve their sales and their financial status.

Background: Among Food-borne diseases, S. aureus is a leading cause of gastroenteritis resulting from the consumption of contaminated food. Objectives: The study aimed to isolate and characterize S. aureus from raw milk, yogurt and meat and determine its antibiotic sensitivity pattern. Materials and Methods: A cross-sectional study was conducted from December 2014 to June 2015 in Mekelle. A total of 284 samples were collected purposively. Enumeration of total viable bacteria count (TVBC), bacteriological isolation and identification, antimicrobial susceptibility testing, as well as PCR amplification of fem A and mec A genes were performed. Chi-square (χ2) and one way ANOVA tests were used for analysis. Results: Overall TVBC mean was found to be 1.29x108cfu/ml/g. The highest TVBC (1.38x108cfu/ml) was from the yogurt sample and the lowest (1.26x108cfu/g) was from meat. The overall prevalence of coagulase positive S. aureus (CoPS) was 39.1% (111) and of the smaples, 51(56.04%), 38(26.20%) and 22(45.83%) were isolated from meat, raw milk, and yogurt, respectively. There was a statistically significant difference (p<0.05) among the different sample types and sources in the prevalence of S. aureus. Almost half of the CoPS isolates were sensitive to Tetracycline, Gentamycin, and Kanamycin, but resistant to Amoxicillin (96.9%) and Penicillin G (93.8%). Moreover, 93.75% of the isolates developed multidrug resistance. All isolates carried the fem A gene and among these isolates, 12 (37.5%) carried mec A gene. Conclusion: The present study revealed that foods of bovine origin of the study area are found to be having less bacteriological quality, high prevalence of CoPS and development of drug resistance.

A cross-sectional study was carried out to determine the prevalence and risk factors of subclinical mastitis in dairy goats in Thika East Subcounty, Kenya. Further the bacterial pathogens and their antibiogram were investigated. Farm level data on risk factors were obtained from 41 farmers using questionnaires. Milk was obtained from 110 lactating dairy goats and tested for submastitis using California Mastitis Test (CMT). The prevalence of subclinical mastitis at goat level was estimated to be at 50.9% using CMT, out of which 86.5% yielded bacteria on culture. The significant risk factors associated with the occurrence of subclinical mastitis were cleaning schedule (p=0.022, OD=1.047) and parity of the goat (p=0048, OD=1.37). Higher prevalence of subclinical mastitis was observed for goats residing in houses cleaned at least once a fortnight. Does in the first parity were least affected. 169 bacterial isolates were obtained from culture, of which 52 isolates from major classes of isolated bacteria were tested for antibiotic sensitivity to six antibiotics. Fourteen different bacteria were isolated and identified from the milk samples. Coagulase-negative Staphylococci (20.7%), Serratia spp. (19.5%), Citrobacter spp. (16%), Klebsiella spp. (11%), Staphylococcus aureus (10.7%), Enterobacter spp. (6.5%), Escherichia coli (5.9%), Proteus spp. (3%), Corynebacterium spp. (1.8%), Morganella spp. (1.8%), Streptococcus spp. (1.2%), Providencia spp. (0.6%), Micrococcus spp. (0.6%), and Staphylococcus intermedius (0.6%) were isolated and identified from the samples. All the isolates were resistant to Penicillin G, while 98% of the isolates were sensitive to Streptomycin. In conclusion, the study showed that a large proportion of goats were affected by subclinical mastitis, with the main bacteria being Staphylococci spp. and coliforms, and that most of the tested antibiotics can be used in the treatment of mastitis. Farmers need to be trained on improved control of mastitis through adoption of good dairy husbandry and milking practices.

A cross-sectional study was conducted to estimate the prevalence of Staphylococcus aureus in raw milk and swab, to assess the antimicrobial susceptibility profile, milk handling practice, and its associated risk factors in selected dairy farms of Mukaturi and Sululta Town, Oromia Region, Ethiopia. A total of 247 samples collected from dairy farms were examined using standard microbiological techniques. The antimicrobial susceptibility profiles of the isolates were also investigated. The possible risk factors for Staphylococcus aureus contaminations in milk were evaluated through a structured questionnaire. Overall, 16.6% (n= 41) of the samples were positive for S. aureus. The prevalence of S. aureus was 15.3% from udder milk and 25%, 20%, and 10% from milkers’ hand, milking bucket, and drying towel swab, respectively. The prevalence of Staphylococcus aureus in milk showed statistically significant variation with respect to age (p≤ 0.001), parity (P≤ 0.001), drainage condition of milking area (P=0.035), study sites (P=0.035), and management system (P=0.035). Majority of the isolates were found resistant to penicillin G (97.6%) and amoxicillin (43.9%). According to this study, 12/28(42.9%) Staphylococcus aureus positive raw milk samples had 104-105cfu/ml S. aureus count, which is above the recommended level for human consumption. 47.1% of milking persons store milk at room temperature temporarily (between 6 and 12 hrs) till transport to collection center with no means of cooling aid. From consumers 25.6% had no health risk associated with raw milk consumption or aware of milk borne disease associated with drinking raw milk. Thus, 60.5% of milk users had habit of raw milk consumption. The study revealed a prevalence of S. aureus, poor milk handling practices, raw milk consumption behavior in study area. Proper handling and hygiene decrease milk contamination by S. aureus and make it safe for human consumption.

Mastitis is the most common disease of adult dairy cattle. The disease continues to be one of the most perplexing and costly diseases of dairy cattle. The present study was conducted to detect bovine mastitis cows and identify potential risk factors for the distribution of bovine mastitis of smallholder dairy farmers using the California mastitis test. A cross-sectional study was conducted from September 2013 to May 2015 in the southern zone of Tigray, Northern Ethiopia, on 146 head of randomly selected cattle, of which 99 (67.8%) were crossed zebu and 47 (32.2%) pure local zebu using the California mastitis test and questionnaire. The overall prevalence was 65 (45.5%), of which 72.3% of crossed local and 27.69% of local zebu were found positive for the CMT test. The investigated risk factors were the season of lactation (OR = 0.510, CI = 0.208–1.247, P = 0.140 ), washing hands between consecutive milking events (OR = 0.354, CI = 0.146–0.856, P = 0.021 ), and history of previous mastitis (OR = 0.441, CI = 0.226–0.858, P = 0.016 ), which had significant ( P < 0.05 ) effect on the present prevalence of bovine mastitis in the study area. A high prevalence of bovine mastitis was observed in the southern zone of Tigray. The summer season of lactation and not washing hands between consecutive milking of cows were significant mastitis causation risk factors in addition to the history of previous mastitis disease history.

Abstract Background Antibiotics must be safe, effective and have acceptable quality to use in both human and veterinary medicine. A cross-sectional study was conducted with the aim of evaluating in vitro efficacy of three brands of penicillin-streptomycin (penstrep) against Staphylococcus aureus isolated from dairy cow milk in Sebeta and Bishoftu, Ethiopia. A standardized questionnaire was also used to assess the knowledge, attitudes and practices (KAP) of veterinarians about brand antibiotics prescribing in dairy farms in the study area. Results A total of 43 S. aureus were isolated from 23 dairy farms. Thirty-three (81.4%) and 10 (23.3%) isolates were susceptible to brand A and intermediate susceptible to brand B, respectively, whereas only one isolate (2.3%) was intermediate susceptible to brand C. The isolates were also tested against the standard discs of penicillin and streptomycin, and the results showed that 42 (97.7%) and 27 (62.8%) of S. aureus were resistant, respectively. The KAP assessment showed penstrep was the most prescribed antibiotic in the dairy farms in the study area, followed by oxytetracycline and sulfa drugs. All veterinarians (100%, 30) agreed that antibiotics imported from Western countries perceived as having higher efficacy than those imported from eastern countries. Similarly, they preferred to use brand A, claiming better clinical improvements compared to the other two brands. The majority (86.7%) and more than half (53.3%) of the respondents agreed on the perception of overuse of antibiotics in veterinary clinics and dairy farms, respectively. Conclusions Penstrep Brand A has had better in vitro efficacy, while brands B and C have not been effective against S. aureus isolates. Moreover, this study revealed that a fixed combination dosage form of penicillin and streptomycin in brand A showed better efficacy towards S. aureus isolates compared to penicillin or streptomycin alone. The perception of the veterinarian about these brands agrees with the in-vitro antibacterial efficacy evaluation. The findings suggest that ineffective brands of penstrep are circulating in the local market, which constitute a potential danger to both human and animal health. Our findings warrant the need for stringent regulation and quality assessment criteria for imported veterinary antimicrobials.

Methicillin-resistant Staphylococcus aureus (MRSA) poses a serious problem in dairy animals suffering from mastitis. The study was carried out to evaluate the incidence of Methicillin resistant S. aureus from clinical mastitis milk samples and their antibiotic resistance profile and characterised with respect to the molecular features that contributed to the resistance in these pathogens. Isolation and identification of Methicillin resistant S. aureus were performed from acute clinical mastitis samples. The isolates were tested using agar disc diffusion method for their antimicrobial susceptibility and modified resazurin assay micro dilution technique for MIC to 8 different antimicrobial drugs. A total of 235 clinical mastitis milk samples from dairy cows were cultured for incidence of S. aureus. Methicillin resistant S. aureus was isolated from a total of 12 (44.25%) of the 116 S. aureus samples. Based on the antimicrobial sensitivity and MIC results, MRSA isolates were found sensitive to gentamicin, enrofloxcain, amoxicillin+sulbactam, ceftriaxone and resistant to amoxicillin, oxytetracycline, penicillin G and oxacillin. Most of MRSA isolates were found to be multi-drug resistant. MRSA alert kit test and mecA and blaZ target gene PCR were found to be useful in the confirmation of MRSA.

Background: Staphylococcus aureus is an opportunistic pathogen, which often causes asymptomatic diseases. This bacterium could cause several disorders, ranging from skin infections to life-threatening diseases. S. aureus could also develop resistance to a wide range of antibiotics. Objectives: The present study aimed to investigate the prevalence and pattern of antibiotic resistance in isolated S. aureus in Imam Reza Hospital of Kermanshah, Iran. Methods: This descriptive cross-sectional study was conducted on 2,228 patients with a positive S. aureus culture who were admitted to Imam Reza Hospital during 2016 - 2018. In accordance with the CLSI protocol, bacterial isolates were separated using laboratory tests, and antibiotic susceptibility was assessed using the standard disk-diffusion method. Data analysis was performed in SPSS version 24. Results: Most of the S. aureus-positive cases were isolated from the emergency ward (43.7%), blood samples (40.1%), and urine samples (23.9%). The highest antibiotic resistance was observed against erythromycin, penicillin G, ofloxacin, cefoxitin, clindamycin, and piperacillin. With 93.3% and 81.8% sensitivity, vancomycin and teicoplanin were respectively the most effective antibiotics against S. aureus. Conclusions: According to the results, the prevalence of resistant S. aureus strains could be due to the long-term hospitalization of patients and the overuse of antibiotics in infection treatment. Therefore, proper monitoring and development of effective infection control methods are essential in these healthcare settings.

Udder inflammation in dairy cows known as mastitis is still a major problem in dairy industry. Economic loss caused by mastitis burden on farmers due to decrease on milk yield and decrease on the milk quality. Most of traditional farmers recognize clinical mastitis as the symptoms was obvious, while subclinical mastitis were not easy to detect and can be potential source of infection for the herd. Detection on subclinical mastitis were using indirect test california mastitis test (CMT) and direct test with Somatic Cell Count (SCC). This research was a cross sectional study using udder quarters milk sample from traditional dairy farms in Bakir, Sukomulyo village, Batu regency. Field test conduct uding CMT and SCC counting in laboratorium. Data collection using Questionnaire were conduct to asses predisposing variables on farmers. Result shows prevalence of subclinical mastitis based on SCC was 60,22% and based on CMT 18,20% of quarter sampels. Predisposing factors that have effect on mastitis were the sanitation of the cow and the farm before and after the milking process.

Background: Staphylococcus aureus is a leading cause of food poisoning resulting from the consumption of contaminated food with staphylococcal enterotoxins. Raw meat is a good medium for the survival and spread of drug-resistant S. aureus. Objective: To look for the prevalence of drug-resistant S. aureus in Addis Ababa abattoir enterprise. Material & Methods: A cross-sectional study was conducted from November 2013 to April 2014 in Addis Ababa abattoir enterprise. A total of 185 swab samples were collected from the carcasses of sheep, goat and slaughtering materials such as workers’ clothes, vehicles, knives and hands. Isolation and identification of S. aureus were conducted using the conventional culture methods and signatory tests. Antimicrobial sensitivity was conducted using standard methods. Results: The overall prevalence rate for S. aureus in the present study was 33%. The higher prevalence rates of S. aureus were recorded from sheep carcasses 36%, followed by 30% from the environment and 16% from goat carcasses. The variation in the prevalence of S. aureus between the carcasses and environment was not statically significant (p > 0.05). More than 90% of S. aureus strains were sensitive to vancomycin, chloramphenicol, gentamicin and kanamycin. While 86.9% S. aureus strains were resistant to penicillin G 80.3% resistant to ampicillin, 63.9% resistant to ceftriaxone, 62.3% resistant to oxacilin, and 62.3% resistant to cefoxitin respectively. Conclusion: The present study indicated that the quality of slaughtered sheep carcasses was more contaminated by S. aureus as compared to goat carcasses, during slaughtering, processing, handling and transportation. The presence of MDR strain in the carcasses demonstrates that there is a growing need to control antimicrobial resistance in sheep and goat carcasses.
