This study was carried out to assess the in vitro antioxidant, anti-inflammatory and antidiabetic effects of Nauclea latifolia (Sm.) leaf extracts. Ethanolic (NLE) and aqueous (NLA) extract of N. latifolia leaves were prepared and assessed for their anti-inflammatory activity, antioxidant potential, α-amylase and α-glucosidase inhibitory activities, and the mechanism of enzyme inhibition in vitro using standard established methods. From the results, phytochemicals such as flavonoids, phenolics, glycosides, and tannins were detected in both extracts of N. latifolia with NLE having a significantly (p<0.05) higher phytochemical content. NLE displayed significantly (p<0.05) better total antioxidant capacity, reducing power, 2,2-diphenyl-2-picrylhydrazyl, and hydrogen peroxide radical scavenging activities. For anti-inflammatory activities, 70.54±2.45% albumin denaturation inhibition was observed for NLE while 68.05±1.03% was recorded for NLA. Likewise, 16.07±1.60 and 14.08±1.76% were obtained against hypotonic solution and heat-induced erythrocyte haemolysis, respectively, for NLE while 20.59±4.60 and 24.07±1.60% were respective NLA values. NLE (IC50: 4.20±0.18 and 1.19±0.11 mg/mL) and NLA (IC50: 11.21±0.35 and 2.64±0.48 mg/mL) α-glucosidase and α-amylase inhibitory activities were dose-dependent with uncompetitive and competitive inhibition elicited, respectively, by the extracts. A significant positive association (p<0.01 and 0.05) was identified between antioxidant activity and carbohydrate-metabolising enzyme inhibitory activity. The obtained result suggests N. latifolia leaf could serve as an alternative candidate for managing diabetes mellitus due to its antioxidant and anti-inflammatory association with diabetes-linked enzymes.

The flavonoid fraction was obtained from Elsholtiza bodinieri Vaniot (EBV) by ethanol-reflux and liquid-liquid extraction. The total content of flavonoid was 179.55 mg/g, and the purity was 64.6%. Then cynaroside with the purity of 94% was isolated from the fraction by preparative HPLC and characterized by the combined usage of HPLC, ESI-MS, and NMR. The antioxidant activity of cynaroside was determined using 2 complementary methods, namely, 2,2- diphenyl-1-picrylhydrazyl (DPPH) radical scavenging and reducing power assay. The anti-inflammatory effect of cynaroside was investigated based on in-vitro and in-vivo experiment. The results showed that cynaroside from EBV scavenged DPPH radical and reduced Fe3+ to Fe2+ effectively, inhibited NO and ROS production in LPS-stimulated RAW264.7 cells and attenuated the inflammation in the mouse model significantly ( p < 0.01), which showed it to be a nutraceutical product in the food industry.

The methanol, ethanol and chlorofom leaf extracts of Satkara, Citrus assamensis (family: Rutaceae), were subjected to in vitro anti-bacterial, thrombolytic, membrane stabilizing and in vivo anti-inflammatory and antitumor activity tests. The chloroform extract of C. assamensis showed the most important spectrum of activity against Bacillus subtilis, Bacillus cereus, Sarcina lutea among 6 gram positive and against 11 gram negative bacteria at the concentration of 1000 μg/disc, while the range of zones of inhibition were within 7-16 mm. Among the tested three extracts CHCl3 extract showed potent thrombolytic activity and hypotonic solution induced haemolytic activity where the percentages of inhibition were found to be 35% and 55% respectively. All the extracts established significant (p<0.05) anti-inflammatory effect by regulating biphasic inflammatory process induced by carrageenan. The leaf extract dose-dependently and significantly decreases the number of EAC cell count and inhibition of cell growth in comparison to the EAC control and standard. The results obtained in the present study indicate that, C. assamensis leaf can be a potential source of anti-bacterial, thrombolytic, membrane stabilizing, anti-inflammatory and antitumor agents.

Like other members of the Colchicum genus, C. szovitsii subsp. szovitsii is also of medicinal importance in Turkish traditional medicine. However, its biological properties have not been fully investigated. Herein, we focused on the evaluation of the in vitro antioxidant and enzyme inhibitory effects of flower, root and leaf extracts, obtained using different extraction methods. In addition, a comprehensive (poly)-phenolic and alkaloid profiling of the different extracts was undertaken. In this regard, ultra-high-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UHPLC-QTOF-MS) allowed us to putatively annotate 195 polyphenols and 87 alkaloids. The most abundant polyphenols were flavonoids (83 compounds), whilst colchicine and 2-demethylcolchicine were some of the most widespread alkaloids in each extract analyzed. However, our findings showed that C. szovitsii leaf extracts were a superior source of both total polyphenols and total alkaloids (being, on average 24.00 and 2.50 mg/g, respectively). Overall, methanolic leaf extracts showed the highest (p < 0.05) ferric reducing antioxidant power (FRAP) reducing power (on average 109.52 mgTE/g) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging (on average 90.98 mgTE/g). Interestingly, each C. szovitsii methanolic extract was more active than the water extracts when considering enzymatic inhibition such as against tyrosinase, glucosidase, and acetylcholinesterase (AChE). Strong correlations (p < 0.01) were also observed between polyphenols/alkaloids and the biological activities determined. Multivariate statistics based on supervised orthogonal projections to latent structures discriminant analysis (OPLS-DA) allowed for the detection of those compounds most affected by the different extraction methods. Therefore, this is the first detailed evidence showing that C. szovitsii subsp. szovitsii might provide beneficial effects against oxidative stress and the associated chronic diseases. Nevertheless, the detailed mechanisms of action need to be further investigated.

Objective: To evaluate the preliminary phytochemical content and antioxidant potential of the hydroalcoholic leaf extracts of Hemionitis arifolia. Methods: Total phenolic, flavonoid and alkaloid contents were evaluated using spectrophotometric methods. The free radical scavenging activity of the leaf hydroalcoholic extract were evaluated against DPPH+, ABTS+, Reducing power assay and nitric oxide assay were determined. Results: The hydroalcoholic concentrate of H. arifolia uncovered the most elevated polyphenol content when contrasted and the other phytoconstituents. Absolute phenol content of the hydroalcoholic separate was observed to be 31.78%, flavonoid content is 1.02% and Alkaloid content is 30.40% individually. The Solvent concentrates showed huge cell reinforcement movement, with hydroalcoholic extract. ABTS Assay, DPPH assay, Reducing power assay and Nitric oxide assay where the Inhibition concentration were 667.75µg/ml, 734.25 µg/ml, 791.58 µg/ml and 899.67 µg/ml. Conclusion: This study suggests that hydroalcoholic leaf extracts of H. arifolia could be a potential source of natural antioxidant and justifies its use in ethno-medicine.

Objective: The aim of the present study was to evaluate the alpha (α)-amylase and alpha (α)-glucosidase inhibitory activities and in vitro antioxidant activities of the 80 % aqueous ethanol extracts of Tinosporasinensis Lour (Merr.).Methods: The 80% aq. ethanol extract of the plant was prepared. The plant extract was examined for its antioxidant activity by using free radical 1,1-diphenyl-2-picryl hydrazyl (DPPH) scavenging method, ABTS radical scavenging ability, reducing power capacity, estimation of total phenolic content, flavonoid content and flavonol content. Different concentrations (2, 4, 8,10and 15 μg/ml) of the extract was subjected to α-amylase inhibitory and α-glucosidase inhibitory activities and IC50were calculated.Results: The study revealed that the different concentrations of the plant extract possessed a very good amount of total phenolics, flavonoid and flavonol and exhibited potent radical scavenging activity using DPPH and ABTS as a substrate. The ethanol extracts exhibited significant α-amylase and α-glucosidase inhibitory activities with an IC50 value1.093µg and 1.04µg dry extract respectively and well compared with standard acarbose drug.Conclusion: Thus, it could be concluded that due to the presence of antioxidant components the plant extracts could be used for the treatment of hyperglycemia, diabetes and the related condition of oxidative stress. This knowledge will be useful in finding more potent components from the natural resources for the clinical development of antidiabetic therapeutics.

The objectives of the study were to test the biological activities of peppermint and spearmint oils via (i) measuring in vitro anti-inflammatory effects with porcine alveolar macrophages (PAMs), (ii) determining the barrier integrity of IPEC-J2 by analyzing transepithelial electrical resistance (TEER), (iii) testing their antioxidant activities, and (iv) investigating the antimicrobial activity against enterotoxigenic Escherichia coli (ETEC) F18+. Briefly, (i) macrophages were seeded at 106 cells/mL and treated (24 h) with mint oils and lipopolysaccharide (LPS). The treatments were 2 (0 or 1 μg/mL of LPS) × 5 (0, 25, 50, 100, 200 µg/mL of mint oils). The supernatants were collected for TNF-α and IL-1β measurement by ELISA; (ii) IPEC-J2 cells were seeded at 5 × 105 cells/mL and treated with mint oils (0, 25, 50, 100, and 200 μg/mL). TEER (Ωcm2) was measured at 0, 24, 48, and 72 h; (iii) the antioxidant activity was assessed (0, 1, 50, 100, 200, 500, and 600 mg/mL) using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging and reducing power assays; (iv) overnight-grown ETEC F18+ were quantified (CFU/mL) after supplementing with peppermint and spearmint oils (0, 1.44, 2.87, 5.75, 11.50, and 23.00 mg/mL). All data were analyzed using the MIXED procedure. Both mint oils significantly inhibited (p < 0.05) IL-1β and TNF-α secretion from LPS-stimulated PAMs. Mint oil treatments did not affect TEER in IPEC-J2. Spearmint and peppermint oils exhibited (p < 0.05) strong antioxidant activities in DPPH and reducing power assays. Both mint oils also dose-dependently inhibited (p < 0.05) the growth of ETEC F18+ in vitro. The results of the study indicated that both mint oils are great candidate feed additives due to their in vitro anti-inflammatory, antioxidant, and antimicrobial effects. Further research is needed to evaluate their efficacy in vivo.

Considering the importance of diet in prevention of oxidative stress-related diseases including hypertension, this study was undertaken to evaluate thein vitroantioxidant and ACE inhibitory activities of selected culinary-medicinal mushrooms extracted by boiling in water for 30 min. Antioxidant capacity was measured using the following assays: DPPH free radical scavenging activity,β-carotene bleaching, inhibition of lipid peroxidation, reducing power ability, and cupric ion reducing antioxidant capacity (CUPRAC). Antioxidant potential of each mushroom species was calculated based on the average percentages relative to quercetin and summarized as Antioxidant Index (AI).Ganoderma lucidum(30.1%),Schizophyllum commune(27.6%), andHericium erinaceus(17.7%) showed relatively high AI. Total phenolics in these mushrooms varied between 6.19 to 63.51 mg GAE/g extract. In the ACE inhibitory assay,G. lucidumwas shown to be the most potent species (IC50= 50 μg/mL). Based on our findings, culinary-medicinal mushrooms can be considered as potential source of dietary antioxidant and ACE inhibitory agents.

Plants are adundantly and are very promisive to be used as source of drugs in many diseases or infections and also it is a main agents of antioxidants which prevents the oxidative stress that are caused by the free radicals. There are numerous studies based on the pharmaceutical and classification of medicinal plants throughout the world. Leaves, fruits, roots are most frequently plant parts used in many research and studies. Here the present study was aimed to evaluate the in vitro antioxidant activity of aqueous and ethanol leaf extracts of Andrographis paniculata Nees and Rhinacanthus nasutus Kurz. Antioxidant is a substance which is used to prevent some types of cell damage in the body. Determination of their in vitro antioxidant activity were carried out by using methods such as DPPH (2,2-diphenyl-1picrylhydrazyl assay), ABTS (2,2′-azinobis (3-ethylbenzothiazolin 6-sulfonic acid assay), FRAP (ferric reducing antioxidant power assay) and SOD (super oxide anion scavenging) assay, H2O2 (hydrogen peroxide radical scavenging) assay. Moreover the ethanolic leaf extracts showed best antioxidant activity than the aqueous leaf extracts. Experimental results reveals that the leaves of A. paniculata have potent antioxidant and free radical scavenging activity than R. nasutus. Further investigation must be done for these two medicinal plants for the discovery of the bioactive compounds.

Garcinia mangostana L. is medicinal plant. Its fruit, the mangosteen contains many bioactive xanthones. This study investigates the potential of organic leaf extracts of G. mangostana grown in Ivory Coast. We tested two organic leaf extracts: G. mangostana aqueous ethanolic leaf extract (ethanol: water, 80:20 v/v) (GMLE) and dichloromethane leaf extract (GMLD). We measured total phenolic and total Flavonoids. We analysed the in vitro anti-inflammatory, anti-radical and anti-proliferative activities of leaf extracts. Ethanol leaf extract showed a considerable amount of phenolic content (328.78±34.32 mg GAE/g) and moderate flavonoids content (43.60±1.48 mg QE/g), Dichloromethane extract had low values of phenolic (70.31±4.55 mg GAE/g) and flavonoids (8.49±0.69 mg QE/g). However, GMLD extract gave a significant anti-inflammatory activity (IC50=152.79±3.34 µg/mL), comparable to the standard drug diclofenac sodium (IC50=142.30±1.22 µg/mL), contrary to GMLE extract (IC50=652.33±12.23 µg/mL). The radical scavenging assay showed a very significant ability of ethanol leaf extract to reduce the DPPH radical (IC50=33.40±0.67 µg/mL) compared to references molecules such as Trolox (IC50=43.72±0.31 µg/mL) and acid ascorbic (IC50=27.20±0.17 µg/mL), dichloromethane extract results showed lowest activity (IC50 = 580.00±23.03 µg/mL). All the organic leaf extracts of G. mangostana had moderate anti-proliferative activity on L20B, RD and VS cell lines studied with IC50 values ranging from 110.89 ±4.82 µg/mL to 860.60±25.78 µg/mL). Our results prove the high potential of the G. mangostana leave extracts as anti-inflammatory and anti-oxidative stress drugs. However, further studies are to determine and validate all the medicinal properties of G. mangostana leaves extracts.

Plant nutrient management can influence the type and level of polyphenolic compounds within leafy vegetables. Therefore, we investigated the effects of fertilizer micro-dosing and harvest time on antioxidant activities of aqueous polyphenolic extracts from Amaranthus cruentus, Ocimum gratissimum and Solanum macrocarpon. Plants were cultivated using urea alone or combined with cattle manure for three staggered harvest periods. Polyphenolics profile (RP-HPLC), DPPH, hydroxyl and superoxide radical scavenging activities, ferric ion reducing power, ferrous ion chelation and inhibition of linoleic acid oxidation were determined. Polyphenolic contents of A. cruentus (caffeic acid, myricetin, quercetin and rutin) and O. gratissimum (catechin and gallic acid) as well as antioxidant activities of the vegetables extracts (except hydroxyl radical scavenging by A. cruentus) were fertilizer micro-dose and harvest time-dependent. Thus, combination of both factors highlighted the screening of optimal farming conditions for these vegetables in order to get leaf extracts possessing higher polyphenolic contents and antioxidant activities.
