Oxidative stress is the result of the disparity between pro-oxidants and antioxidants in an organism, and it is important in the pathogenesis of several degenerative disorders, such as arthritis, Alzheimer’s, cancer, and cardiovascular diseases. Free radicals can damage biomolecules, such as nucleic acids, lipids, proteins, polyunsaturated fatty acids, and carbohydrates, and the DNA leading to mutations. The use of antioxidants is effective in delaying the oxidation of biomolecules. Antioxidants are complexes found in the food that can retard or deter oxidation by preventing the initiation and propagation of oxidizing chain reactions. Medicinal plants have been used for centuries by man to manage diseases and have a host of antioxidant complexes. Traditionally, Caesalpinia volkensii, Vernonia lasiopus, and Acacia hockii have folkloric remedies against associated oxidative stress-mediated complications. However, the upsurge in its use has not been accompanied by scientific validations to support these claims. In this study, in vitro antioxidant activity of Caesalpinia volkensii, Vernonia lasiopus, and Acacia hockii collected from Embu County (Kenya) were determined by radical scavenging activities of 1,1-diphenyl-2-picrylhydrazyl (DPPH) and hydroxyl radical in addition to ferric reducing antioxidant power analyzed against that of L-ascorbic acid as the standard. The obtained results revealed remarkable antioxidant activities of the studied plant extracts as evidenced by the low IC50 and EC50 values. These antioxidant activities could be due to the presence of antioxidants phytochemicals such as flavonoids, phenols, terpenoids, and saponins among others. Therefore, the therapeutic potential of this plant could be due to their antioxidant properties. This study recommends bioassay of the extracts against oxidative stress-related disorders for development of phytomedicine with antioxidant properties.

AbstractThe distribution of phytochemicals and their contribution to antioxidant potentials in whole and dehulled Bambara groundnut (BGN) seeds was evaluated. Whole BGN seeds were sorted using the testa and hilium colour and further grouped into whole and dehulled BGN seeds. Extractions of both whole and dehulled BGN seeds was done using methanol and the extracts assayed for total phenolics (TPC), flavanol, flavonol, anthocyanin content, oxygen radical absorbance capacity (ORAC) and ferric reducing antioxidant power (FRAP). Methanolic extract of whole BGN seed exhibited higher flavanol and flavonol content as well as significantly higher in-vitro antioxidant activities than dehulled BGN seeds. The TPC of whole BGN seed extract ranged from 3.6 to 11.0 GAE/g, while that of dehulled BGN ranged from 2.7 to 3.2 GAE/g. Identification of phenolics in whole and dehulled BGN seed extract using UPLC-qTOF-MS, revealed the presence of monoterpenoids (iridoids), phenolic acids, flavonoids and lignans. Bivariate correlations showed anthocyanin demonstrated weak positive correlation between flavanol, flavonol and ORAC for whole BGN seed extract; and negative correlation between flavanol, TPC, FRAP and ORAC for dehulled BGN. Aside the effect of dehulling, whole BGN seeds exhibited the presence of phytochemicals with beneficial properties for food and industrial application.

This study aimed to isolate, prepare and identify the main flavonoids from a standardized Smilax glabra flavonoids extract (SGF) using preparative HPLC, MS, 1H NMR and 13C NMR, determine the contents of these flavonoids using UPLC, then compare their pharmacological activities in vitro. We obtained six flavonoids from SGF: astilbin (18.10%), neoastilbin (11.04%), isoastilbin (5.03%), neoisoastilbin (4.09%), engeletin (2.58%) and (−)-epicatechin (1.77%). The antioxidant activity of six flavonoids were evaluated by determining the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical and 2,2′-Azinobis (3-ethylbenzothiazoline-6-sulphonic acid) diammonium salt (ABTS+) radical scavenging activity and ferric reducing antioxidant power (FRAP). In addition, the anti-inflammatory activity of six flavonoids were evaluated by determining the production of cytokines (IL-1β, IL-6), nitric oxide (NO) using enzyme linked immunosorbent assay and the NF-κB p65 expression using Western blotting in lipopolysaccharide (LPS)-stimulated RAW264.7 cells. The results showed that (−)-epicatechin, astilbin, neoastilbin, isoastilbin and neoisoastilbin had strong antioxidant activities, not only in DPPH and ABTS+ radicals scavenging capacities, but in FRAP system. Furthermore, all the six flavonoids could significantly inhibit the secretion of IL-1β, IL-6, NO (p < 0.01) and the protein expression of NF-κB p-p65 (p < 0.01) in LPS-stimulated RAW264.7 cells. This study preliminarily verified the antioxidant and anti-inflammatory activities of six flavonoids in S. glabra.

Doxorubicin (DOX) is an anthracycline antibiotic widely used as a chemotherapeutic agent in the treatment of several tumours. However, its cardiac toxicity limits its use at maximum therapeutic doses. Most studies implicated increased oxidative stress as the major determinant of DOX cardiotoxicity. The local Saudi flora is very rich in a variety of plants of quite known folkloric or traditional medicinal uses. Tribulus macropterus Boiss., Olea europaea L. subsp. africana (Mill.) P. S. Green, Tamarix aphylla (L.) H. Karst., Cynomorium coccineum L., Cordia myxa L., Calligonum comosum L’ Hér, and Withania somnifera (L.) Dunal are Saudi plants known to have antioxidant activities. The aim of the current study was to explore the potential protective effects of methanolic extracts of these seven Saudi plants against DOX-induced cardiotoxicity in rats. Two plants showed promising cardioprotective potential in the order Calligonum comosum > Cordia myxa. The two plant extracts showed potent in vitro radical scavenging and antioxidant properties. They significantly protected against DOX-induced alterations in cardiac oxidative stress markers (GSH and MDA) and cardiac serum markers (CK-MB and LDH activities). Additionally, histopathological examination indicated a protection against DOX-induced cardiotoxicity. In conclusion, C. comosum and C. myxa exerted protective activity against DOX-induced cardiotoxicity, which is, at least partly, due to their antioxidant effect

The antioxidant activities of the methanolic extracts of 9 Salvia species and 15 other Lamiaceae plants growing in Iran were evaluated using ferric reducing antioxidant power (FRAP) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging assays. FRAP values ranged form 8.5 to 79.0 μM quercetin equivalents/g dry weight, and IC50 values in the DPPH assay from 115.7 to 1350.2 μg dry weight/mL. Salvia species showed the highest antioxidant activities. S. santolinifolia, S. eremophila and S. palestina, which have not been studied before, were the most active plants. These were more active than the previously studied species from this family, such as S. multicaulis and Marrubium vulgare. S. hydrangea and Gontscharovia popovii also showed high antioxidant activities. FRAP and DPPH assay results showed good correlations with the total phenolic contents of the plants, measured by the Folin-Ciocalteau assay (r2 = 0.925 and 0.799, respectively, p < 0.0001). In conclusion, our study shows that some Lamiaceae plants growing in Iran represent good potential sources of natural antioxidants useful for either prevention or treatment of oxidative stress-related diseases.

Aims: This study was intended to investigate and quantify the phytochemicals and the antioxidant activities of fresh and powdered wheatgrass. Study Design:  The experiment was performed by solvent extraction technique for determining the phytochemicals of wheatgrass samples. Place and Duration of Study: The study was conducted at the Food engineering laboratory in Hajee Mohammad Danesh Science and Technology University, Dinajpur, Bangladesh between June 2018 and December 2018. Methodology: Fresh and powdered wheatgrass were used for the experiment. Methanol and ethanol solutions were separately utilized, in an attempt to assume the efficient solvent for extraction. UV-Vis spectrophotometric methods were employed in general throughout the study. Total phenolic contents (TPC), total flavonoid contents (TFC), DPPH scavenging activity, ferric reducing antioxidant power (FRAP) and the total chlorophyll contents of both solvent extracts were analyzed. Results: Phytochemicals and antioxidant activity were found lower in both solvent extracts of powdered wheatgrass than fresh leaves. Phenolic content and Chlorophyll were significantly (P < 0.05) higher in fresh samples (extracts). Due to heating, a significant decrease was observed in the DPPH radical scavenging activity of powdered extracts. Ferric Reducing Antioxidant Power (FRAP) was significantly (P < 0.05) higher in methanolic extracts than in ethanolic extracts. No significant (P > 0.05) difference was observed in total flavonoids contents. Conclusion: These findings endorsed that both solvents demonstrate competitive potentials to extract bioactive compounds from wheatgrass and its derivatives and hence can be chosen specifically according to the components.

Leaves of Olax subscorpioidea, Hoslundia opposita, Cleistopholis patens, Plumbago zeylanica and Dioscoreophyllum cumminsii that are used as anti-diabetics were evaluated for hyperglycaemic-lowering and antioxidant activities to justify their Nigerian ethnomedicinal usage. Leaf methanolic extracts (100, 200 and 400 mg/kg, p.o.) were assayed in normal, glucose- and alloxan-induced diabetic rats, while 1,1,-diphenyl-2-picrylhydrazyl, total antioxidant capacity, ferric reducing antioxidant power and hydroxyl radical scavenging tests were used for their antioxidant activity. Effects of n-hexane, dichloromethane, ethylacetate and aqueous partition fractions of the three most active anti-hyperglycaemic extracts were also tested in glucose-loaded rats. In normoglycaemic rats, all extracts generally lacked activity, while with glucose-loaded rats, only O. subscorpioidea (200mg/kg) and C. patens (400 mg/kg) at 1 h and O. subscorpioidea (200, 400 mg/kg) and C. patens (400 mg/kg) at 2-4 hours gave lowered (p < 0.05) blood glucose levels than glibenclamide (5 mg/kg), while activity of H. opposita and P. zeylanica (200 mg/kg) were only comparable to glibenclamide. When daily fed for fourteen days to alloxan-induced diabetic rats, all extracts, at their most active doses, gave significantly higher activity than glibenclamide. Olax subscorpioidea leaf extract had the highest hyperglycaemic-lowering and least antioxidant activities. Highest antioxidant activity of H. opposita may suggest some contribution of antioxidant property to its hyperglycaemic-lowering activity. The glucose-lowering and insulinotropic constituents of O. subscorpioidea, H. opposita and C. patens should be concentrated in their aqueous, aqueous and dichloromethane fractions, respectively. Anti-hyperglycaemic ethnomedicinal use of these plants was justified and H. opposita has additional antioxidant property. Keywords: Antioxidant activity; Diabetes mellitus; Nigerian anti-diabetic plants

Phytochemical investigations of Taxus baccata L. by successive chromatographic methods resulted in the isolation of the lignans lariciresinol (1), taxiresinol (2), 3´-demethylisolariciresinol- 9´-hydroxyisopropylether (3), isolariciresinol (4), and 3-demethylisolariciresinol (5) as well as taxoids. Compounds 1 - 5 were evaluated for their acetylcholinesterase (AChE), butyrylcholinesterase (BChE), and lipoxygenase (LOX) inhibitory activities, which play a role in the pathogenesis of Alzheimer’s disease (AD), by in vitro spectrophotometric methods, while they were also screened for their antioxidant capacity in 2,2-diphenyl-1- picrylhydrazyl (DPPH) radical scavenging, ferrous ion-chelating effect, and ferric-reducing antioxidant power (FRAP) at 125, 250, 500, and 1000 μg ml-1. All compounds exhibited a moderate inhibition against both BChE and LOX, whereas they were inactive towards AChE. The compounds displayed a great scavenging activity against DPPH especially at 500 and 1000 μg ml-1. Besides, they were found to exert noteworthy reducing antioxidant power on ferric ions. In particular, the FRAP of compounds 2 (3.552 ± 0.02), 4 (3.021 ± 0.71), and 5 (3.533 ± 0.01) were as high as that of the reference chlorogenic acid (3.618 ± 0.01) at 1000 μg ml-1. None of the compounds exhibited chelating ability against ferrous ions.

The decoction of Tessaria absinthioides is used in traditional medicine of South America as hypocholesterolemic, balsamic, and expectorant; but it is also useful for the prevention of hepatitis, renal insufficiency, and diabetes, and is used as digestive. A lyophilized decoction from the aerial parts of this plant (TLD) collected in San Juan (TLDSJ) and Mendoza (TLDM) provinces (Argentina) and one collection from Antofagasta, Chile (TLDCH) were characterized regarding antioxidant and antibacterial activities, phenolics and flavonoids content, and ultrahigh resolution liquid chromatography Orbitrap MS analysis UHPLC–PDA–OT-MS/MS metabolite profiling. The antioxidant properties were carried out "in vitro" using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and trolox equivalent antioxidant activity (TEAC) methods, ferric-reducing antioxidant power (FRAP), and lipoperoxidation in erythrocytes (LP). The antibacterial activity was evaluated following the Clinical and Laboratory Standards Institute (CLSI) rules. TLDSJ, TLDM, and TLDCH displayed a strong DPPH scavenging activity (EC50 = 42, 41.6, and 43 µg/mL, respectively) and inhibition of lipoperoxidation in erythrocytes (86–88% at 250 µg TLD/mL), while a less effect in the FRAP and TEACantioxidant assays was found. Additionally, the decoctions showed a content of phenolics compounds of 94 mg gallic acid equivalents (GAE)/g, 185 GAE/g, and 64 GAE/g, for TLDSJ, TLDM, and TLDCH samples, respectively. Regarding the flavonoid content, the Chilean sample was highlighted with 19 mg quercetin equivalents (QE)/g. In this work, several phenolic compounds, including sesquiterpenes, flavonoids, and phenolic acids, were rapidly identified in TLDSJ, TLDM, and TLDCH extracts by means UHPLC–PDA–OT-MS/MS for the first time, which gave a first scientific support to consider this medicinal decoction from both countries as a valuable source of metabolites with antioxidant effects, some with outstanding potential to improve human health.

In this work, some new 2-[(3,4-dichlorophenyl)methyl]-1H-benzimidazole derivatives containing different five-membered heterocycles like 1,3,4-oxadiazole, 1,3,4-triazole, and 1,3,4- thiadiazole moieties were designed and efficiently synthesized starting from 2-[(3,4-dichlorophenyl) methyl]-1H-benzimidazole. The products were screened for their in vitro antioxidant and α-glucosidase inhibitory activities. Among the synthesized compounds, some of them showed efficient α-glucosidase inhibition with IC50 values ranging between 16.05 ± 0.94 and 77.02 ± 1.12 μg/mL when they were compared with the standard α-glucosidase inhibitor acarbose having IC50 value 12.04 ± 0.68 μg/mL. The antioxidant activity of all products was screened by using various in vitro antioxidant assays, including CUPric Reducing Antioxidant Capacity (CUPRAC) and Ferric Reducing Antioxidant Power (FRAP) assays. Also, the radical scavenging activities of the products were assayed by using the ABTS method. method.

A new dihydroisocoumarin glucoside, vacillanoside (3), and two new anthrone C-glycosides microdantin derivatives; vacillantin A (10) and B (11), together with nine known compounds belonging to the anthraquinone, anthrone and isocoumarin groups were isolated from the leaves of Aloe vacillans. The structures were determined based on spectroscopic evidence including 1D and 2D nuclear magnetic resonance (NMR) spectroscopy and high resolution mass spectrometry (HRESIMS) data, along with comparisons to reported data. The leaves were used to extract compounds with different solvents. The extracts were tested for antioxidant activity with a variety of in vitro tests including 2,2-diphenyl-1-picrylhydrazyl (DPPH•), 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonate (ABTS•+), ferric reducing antioxidant power assay (FRAP), superoxide, and nitric oxide radical scavenging assays. The dichloromethane fraction was most active, displaying significant free radical scavenging activity. The n-butanol fraction also showed notable activity in all assays. Therefore, these findings support the potential use of A. vacillans leaves as an antioxidant medication due to the presence of polyphenolic compounds.
