The profound modification of lifestyle and food habits has led to an important increase in the prevalence of gout. Unfortunately, there are current unmet needs for the treatment of this disease, prompting the search for new alternatives. Paullinia pinnata is a plant used to treat various diseases including arthritis. The present work aimed to investigate the antigouty activities of the aqueous (AEPP) and methanolic (MEPP) extracts of P. pinnata as well as their in vivo antioxidant properties. The gouty arthritis was induced by injecting 50 μl of monosodium urate (MSU, 100 mg/ml) in the left hind ankle of rats. P. pinnata extracts were administered orally at the doses of 100 and 200 mg/kg/day for 6 days, starting 24 h after MSU injection. Allopurinol 5 mg/kg/day was used as reference drug. Inflammation and hyperalgesia were daily monitored from 24 hours after treatment initiation and for the 6 consecutive days. Myeloperoxidase (MPO) quantification was done in collected synovial fluid. Nitrite oxide (NO), malondialdehyde (MDA), and superoxide dismutase (SOD) were evaluated in the spinal cord and the brain. The serum content of SOD was additionally quantified. AEPP and MEPP significantly (p<0.001) reduce MSU-induced inflammation (22.41% to 93.65%) and hyperalgesia (33.33% to 64.44%) in both ankle and paw. AEPP and MEPP significantly (p<0.001) reduce synovial MPO production with the percentage ranging from 76.30% to 85.19%. AEPP and MEPP significantly (p < 0.001) reduce serum, spinal, left and right hemispheres NO, and MDA and increase the SOD activity (p<0.001). P. pinnata leaf extracts possess potent curative effects against MSU-induced gouty arthritis that combines analgesic, anti-inflammatory, and antioxidant activities. These findings support the use of P. pinnata leaves extracts in the treatment of gouty arthritis and further present the plant as a potent source of efficient antigouty medicine.

Background: Gout is a metabolic disease strictly related to hyperuricemia. The associated intense inflammation and pain are triggered by the deposited monosodium urate crystals (MSU) in joints. The principal therapeutic strategies of gout involve the control of hyperuricemia and anti-inflammatory medications. Objectives: This study aimed to investigate the possible beneficial effects of ozone therapy, a well-known antioxidant, and an immunomodulation, on gouty arthritis and the underlying mechanisms. Methods : Acute gouty arthritis was induced in male albino rats via MSU crystals intra-articular injection in the ankle joint. The gouty arthritic rats received pre-treatment with ozone, colchicine (as a reference drug), or combination. Results : The obtained results of ozone therapy showed obvious reduction in the degree of ankle edematous swelling, pro-inflammatory cytokines, lipid peroxidation, the nucleotide binding oligomerization domain like receptor containing pyrin domain 3 (NLRP3), procaspase-1, caspase-1, interleukin-1β synovial tissue levels with enhancement of antioxidant defense system. Additionally, ozone therapy significantly attenuated the histological derangements in gouty arthritic rats. Conclusion : This study suggests that ozone is able to treat gouty arthritis and reducing synovial injury through an anti-inflammatory effect as well as antioxidant activity.

Three Selaginella species, S. ornata, S. plana, and S. willdenowii, from Java Island, Indonesia, have been known to have antioxidant properties; however, in vivo antioxidant activities of these species have not been reported. This research aimed to evaluate the in vivo antioxidant activity of ethanolic extract of three Selaginella species. The 70% ethanol extract of three Selaginella species at four different doses was administered to mice one day before being treated with oxidative stress. The liver tissue of mice treated with or without oxidative stress was analyzed their lipid peroxidation by measuring MDA concentration and Superoxide Dismutase (SOD) activities. The results showed that there were variations in antioxidant activity among the three Selaginella species. In general, the dose of 0.3 g extract kg-1 BW has been able to reduce lipid peroxidation and increase SOD activity. The administration of S. ornata extract to the mice at 1.2 g extract kg-1 BW reduced the MDA concentration to the lowest level, but the same dose of two other Selaginella extracts caused toxic effects in mice. The antioxidant activities of S. ornata and S. plana were better than that of S. willdenowii extract, and among those species, S. ornata has the best antioxidant activity.

Piper crocatum and Persea americana Mill leaves are commonly used in traditional medicinal remedies, such as antidiabetes, antitumors and Alzheimer treatment. However, the extensive use of plant extracts over worldwide becomes significant concerns including its safety, efficacy, and quality. Therefore, the accurate scientific evaluation has become a precondition for acceptance of herbal health claims. The aim of this study was to determine the antioxidant activities of Piper crocatum Ruiz & Pav and Persea americana Mill leaf extracts. Isolation of antioxidant fractions were conducted using organic solvent extraction techniques. Antioxidant assays were conducted by using in vitro and in vivo methods involving DPPH and MDA methods. In this study, in vitro assays of Persea americana Mill and Piper crocatum Ruiz & Pav leaf extracts showed the best activity in water fraction. Moreover, in vivo assays of both plant leaf extracts showed the best dose at 8 mg. Persea americana Mill and Piper crocatum Ruiz & Pav leaf extracts have been successfully determined in antioxidant actions in vitro and in vivo. Persea americana Mill in water and ethanol solvents exhibit strong antioxidant properties. Meanwile, Piper crocatum Ruiz & Pav exhibit moderate activity in water and weak antioxidant activity in ethanol. Both plant leaves showed that 8 mg dose was better than the dose of 4 mg and 16 mg in vivo.

We have previously reported that the leaf extract of Albizia anthelmintica exhibited substantial antioxidant, anti-inflammatory, analgesic, and antipyretic properties in vivo. We also comprehensively characterized the active phytoconstituents and found several flavonoids and galloyl glucosides derivatives. In the current work, we explored the gastroprotective effects of the leaf extract in an indomethacin-induced ulcer model and the mechanisms involved. The rats being pretreated with the tested extract (100 and 200 mg kg−1) significantly prevented gastric lesions by 87.4% and 92.3%, respectively, and they had no structural derangements in the gastric mucosa. The extract significantly reduced the elevated levels of IKκB, NF-κB, TNF-α, IL-6, iNOS, and lipid peroxidation; increased the reduced level of glutathione peroxidase (GPx) activity; and reduced glutathione (GSH) in the indomethacin-induced ulcer model. The protective activities of the extract were similar in most aspects to those exerted by the known anti-ulcer drug famotidine. These activities might be attributed to the anti-inflammatory and antioxidant activities, and the reduction of iNOS levels. In conclusion, Albizia anthelmintica is a potential candidate for management of gastric ulcers with antioxidant properties.

This study was aimed to investigate the antioxidant potentials of methanol and petroleum ether leaf extracts of Asystasia vogeliana against paracetamol-induced liver injury in rats. For estimation of antioxidant potentials, in vitro radical scavenging assays were carried out using DPPH, FRAP, and ABTS. For in vivo study, twenty-five male Wistar rats weighing 100-120 g were randomized and assigned into 5 groups (I-V, n=5). Further, Paracetamol (PCM) at 2 g/kg was used to induce acute hepatotoxicity orally. Rats in group I received distilled water (10 ml/kg) only. While, the rats of groups II, III, and IV received MLEAV (200 mg/kg), PLEAV (200 mg/kg), and a standard hepatoprotective reference drug silymarin (25 mg/kg) respectively for 5 days before PCM induction. Rats in group V received distilled water for 5 days before PCM induction. Blood and liver samples were collected for hematology, serum biochemistry, and histopathology analyses using standard procedures. In vitro assays revealed that MLEAV showed significant (P < 0.05) increases in antioxidant activity compared with PLEAV. Further, significant (P < 0.05) reductions in the activities of ALT and ALP while a significant (P < 0.05) increases in the activity of antioxidant enzymes (CAT, SOD, and GPx) were reported in the group II and III compared with group V. There were also no observable lesions in their hepatocytes. Results of the study can be concluded that MLEAV elicited more in vitro and in vivo antioxidant activities than PLEAV, thus it protects the liver of rat from PCM-induced hepatotoxicity. Therefore, MLEAV could be used as a hepatoprotective agent for the clinical management of liver damage.

Introduction: Siphonochilus aethiopicus is a medicinal plant widely used in the treatment of many inflammatory conditions such as arthritis. The objective of this study was therefore to evaluate the antioxidant and anti-inflammatory properties of methanolic extract of S. aethiopicus rhizomes. Methods: The total phenolic compounds, flavonoid, and tannin content, as well as the in vitro antioxidant activity of the extract, were estimated. The in vivo anti-inflammatory activity was then evaluated in male mice aged 3 to 4 months using the arthritic mouse model induced by carrageenan (0.05 ml; 1%) and monosodium urate (MSU) crystals (26.6 mg/mL). Mice were treated with the methanolic extract of S. aethiopicus (75, 150, 300 mg/kg) and the reference drugs: indomethacin (3 mg/kg) and colchicine (1 mg/kg). The serum, splenic, and hepatic lysosomal enzymes were determined, and oxidative stress biomarkers were estimated. Paws were sectioned for histological analysis. Results: Results showed that S. aethiopicus extract had non-negligible concentrations of polyphenols, flavonoids, and tannins, which could confer it an antioxidant effect. Further, the methanolic extract of S. aethiopicus at different doses significantly (P < 0.05) reduced paw swelling, attenuated joint inflammation, limited the release of lysosomal enzymes, and improved antioxidant enzymes. Conclusion: The methanolic extract of S. aethiopicus has anti-inflammatory and antioxidant properties, and can be used to treat acute forms of gouty arthritis.

Five ethanol extracts of wild plants from the Casablanca region (Morocco) used in local traditional medicine for the treatment of inflammatory diseases were evaluated for their in vivo topical anti-inflammatory activity (inhibition of carrageenan-induced ear edema in mice) and in vitro antioxidant and antiradical properties (inhibition of linoleic acid oxidation, DPPH radical scavenging). All the extracts showed an anti-inflammatory effect: 300 μg/cm2 provoked edema reductions ranging from 22 to 28%. All the extracts also exerted radical scavenging and/or antioxidant properties, the most active plant being Mentha pulegium L. (Lamiaceae), which contained the highest amount of phenolics (339 mg/g), and flavonoids (16.7 mg/g).

Combretum fragrans (Combretaceae) is a Cameroonian medicinal plant containing various secondary metabolites and traditionally used for the treatment of several pathologies. Two cycloartane-type triterpenes, Combretin A and Combretin B, were isolated from this plant. This study was aimed at evaluating the anti-inflammatory, antioxidant, and anticolitis effects of these compounds. In vitro anti-inflammatory properties were evaluated by inhibition of cyclooxygenase, 5-lipoxygenase, and denaturation of the protein; antioxidant properties were assessed by using 1,1-diphenyl-2-picrylhydrazyl (DPPH), (2,2’-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid)) ABTS•+, capacity tests ferric reducing antioxidant (FRAP), and trapping nitric oxide. For in vivo analysis, we used the model of ulcerative colitis induced by Dextran Sulfate Sodium (DSS). Studies of the anti-inflammatory activity showed that Combretin A and Combretin B had maximal inhibitory activity on cyclooxygenase (71.92% and 89.59%), 5-lipoxygenase (76.68% and 91.21%), and protein denaturation (63.93% and 87.78%). Antioxidant activity on DPPH, ABTS•+, ferric reducing antioxidant capacity (FRAP), and nitric oxide scavenging showed that Combretin A and Combretin B showed good antioxidant activities. These compounds significantly reduced the signs of DSS-induced colitis in the treated animals by preventing the weight loss of the animals, by significantly reducing the disease activity index, improving the condition of the stool, preventing the reduction of the length of the colon, and preventing the degradation of the colon. This study revealed that Combretin A and Combretin B have anti-inflammatory, antioxidant, and curative properties against colitis experimentally induced by DSS in rats.

Bauhinia tomentosa L. belonging to the family Caesalpiniaceae is a traditional medicinal plant species used by the local healers for the treatment of inflammation and other ailments. The present study was addressed to isolate and characterize bioactive entities exists in the ethanolic extract of B. tomentosa flower. Moreover, the anti-inflammatory effects of crude and isolated compounds were assessed using carrageenan induced paw oedema model. Indomethacin, a renowned reference drug was used for comparison. In vivo antioxidant activities were achieved using standard procedures. In anti-inflammatory studies, the isolated compound, rutin (10mg/kgb.w.) manifested potent activity by inhibiting paw oedema (92.6%) than the crude extracts (88.7%). With reference to these findings, rutin also registered eminent therapeutic property in terms of in vivo antioxidant activity. Therefore, the current study exposed that the anti-inflammatory effects of isolated compound, rutin is more expected to depend on their ability to inhibit the production of proinflammatory mediators and proposes that the studied plant species can be deliberated as one of the finest, versatile and most frequently used household remedies for varied manifestations. Furthermore, the compound isolated may serve as a lead molecule for the formulation of active novel drugs without any adverse side effects.

Purpose: To evaluate the in vivo anti-inflammatory effect and in vitro antioxidant activity of the areal part of Sedum sediforme (Jacq.) extracts.Methods: The plant was extracted with solvents of varying polarity (Methanol, chloroform and ethyl acetate, respectively) allowed its separation into three sub-fractions: crude extract, chloroform extract and ethyl acetate extract (CrE, ChE and EaE, respectively). Total polyphenol contents of the extracts were determined. The phorbolmyristate acetate (PMA) induced mice ear edema method was using to evaluate the anti-inflammatory activity. The in vitro scavenging activity was evaluated using enzymatic and non-enzymatic methods. Chelating ability of extracts was assessed using Fe2+–ferrozine complex.Results: The highest content of phenolics compound was in EaE. The administration of CrE (12.5 and 25 mg/kg) reduced ear edema induced by PMA (%I = 35.81 ± 3.18 % and 38.57 ± 2.80 %, respectively), the effect was comparable with that of diclofenac used as a reference drug (%I = 38.84 ± 1.87 %). The in vitro scavenging activity of S. sediforme extracts confirmed that the CrE has the highest enzymatic and non-enzymatic activity with a half maximal inhibitory concentration (IC50) = 0.063 ± 0.005 mg/mL and 0.178 ± 0.006 mg/mL, respectively. However, the ChE present an excellent  chelating activity with a half-maximal effective concentration (EC50) of 0.397 ± 0.001 mg/mL.Conclusion: The results show that S. sediforme extracts have a strong antioxidant and antiinflammatory activities which lend some support their use in the traditional medicine.
