In this study, optimization of enzyme-assisted extraction, purification, characterization, and its bioactivities of polysaccharides from Hedyotis corymbosa (HCP) was investigated. It was found that the optimum extraction conditions were 3% of enzyme concentration (X1), 30 of liquid-to-solid ratio (X2), 56°C of extraction temperature (X3), 200W of ultrasonic power (X4), 10 min of extraction time (X5), and 5 of pH value (X6). Under optimum conditions, the experimental yield (4.10 ± 0.16%) was closed to the predicted value (4.02%). The crude HCP was further purified using DEAE-52 and Sephadex G-150 gel column, and a major polysaccharide fraction from HCP, designed as HCP-1a with molecular weight of 33.9 kDa, was obtained. The HCP and HCP-1a were characterized by chemical analysis, FT-IR, and HPLC. For antioxidant activities in vitro, HCP possessed strong hydroxyl radical scavenging, DPPH radical scavenging, and Fe2+ chelating activities. In subsequent immunostimulatory studies, significantly decreased NO, IL-1β, and TNF-α concentrations were observed in both of HCP and HCP-1a treated RAW264.7 cells. Therefore, this study may indicate some insights into the application of polysaccharides from Hedyotis corymbosa as potential natural antioxidants and immunostimulants.

Ultrasonic-assisted extraction (UAE) method was used to polysaccharides from Radix Astragali (RAP), and the orthogonal test was applied to optimize extraction conditions. Furthermore, the antioxidant activities of RAP were evaluated by DPPH and superoxide anion radical scavenging activities assay. The results indicated that the optimal conditions for RAP extraction as followings: solid/liquid ratios of 1:20, extraction time of 10 min, ultrasonic power of 65 W and extraction temperature of 60°C. Meanwhile, RAP had relatively higher scavenging activities on DPPH and superoxide anion radicals.

Abstract Background Secoisolariciresinol di glucoside (SDG) is a natural antioxidant generally extracted from flaxseed, which is one of the most important oil crops in China, the by-product of the flaxseed oil, i.e., flaxseed meal, still contains a lot of lignans. However, flaxseed meal is generally treated as waste, resulting in a huge waste of resources. Objective To establish an efficient and convenient method for extraction and purification of lignans from flaxseed meal. Methods First, we used response surface methodology (RSM) to optimize the extraction conditions of the ultrasonic-assisted aqueous two-phase system (UAATPS), and we obtained the purified extracts by macroporous resin purification (MRP). Second, the antioxidant ability of the extracts was studied in vitro. Results The best extraction conditions obtained were as follows: 9.0 wt% sodium hydroxide, 30.0 wt% isopropanol, extraction time 39 min, liquid-to-solid ratio of 52.0 mL/g, ultrasonic wave 560 W, and extraction temperature 40 °C. Under the optimal conditions, the purity of crude extracts (SDG-APTS-C) reached 21.5%. The desorption conditions of MRP were as follows: eluting 3 BV with ultrapure water, and then eluting with 25% ethanol at 2 BV/h to collect eluents. The purified extracts (SDG-ATPS-P) had a purity quotient of 73.9%, 52.4% higher than that of SDG-ATPS-C. Additionally, experiments conducted in this paper revealed that SDG-ATPS-C and SDG-ATPS-P could effectively remove DPPH, ABTS, and hydroxyl free radicals in vitro. Conclusions The method was validated for extracting SDG from flaxseed meal, thus achieving the reuse of flaxseed meal. Highlights This research provides some references for the application of UAATPS combined with MRP in natural products.

Free radicals, reactive nitrogen species (RNS) and reactive oxygen species (ROS) have been known to contribute several degenerative diseases such as cardiovascular diseases, aging, certain types of cancers, rheumatoid arthritis, neurodegenerative, and diabetes mellitus. In order to overcome the negative effects of these radicals, some scientists have explored some natural antioxidants from plants and it's by-products. The antioxidant can be defined as any substances or samples capable of inhibiting free radical reactions in the oxidation reaction. Due to curcuminoids contained, Curcuma species such as Curcuma longa, Curcuma heyneana, Curcuma mangga, and Curcuma xanthorriza were commonly used for herbal components in some traditional medicine. Several in vitro tests been introduced and used to measure antioxidant activities, namely radical scavenging assay using 2,2’-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azinobis-(3-ethylbenzothiazoline-6 -sulfonic acid) (ABTS•+), ferric reducing antioxidant power (FRAP), ferric-thiocyanate, phosphomolybdenum method, cupric ion reducing antioxidant capacity, metal chelating power, beta-carotene bleaching linoleic-ferric-thiocyanate, and thiobarbituric acid methods. This review highlighted the antioxidant activities in vitro of C. longa, C. heyneana, C. mangga, and C. xanthorriza through several tests. To perform this review, several repute databases were analyzed and used. From this review, it can be stated that Curcuma species have powerful antioxidant activities, therefore they could be potential sources of natural antioxidants and can be used as food supplements.

The seed coat of red sword bean (Canavalia gladiata (Jacq.) DC.) is rich in antioxidant polyphenols. It is often discarded as a byproduct with the consumption of red sword bean, since it is very thick and not consumed by people. The aim of this study was to develop an ultrasound-assisted extraction method to extract natural antioxidants from the seed coats. The extraction process was optimized by using response surface methodology. After the single-factor experiments, three key factors, including ethanol concentration, liquid/solid ratio, and extraction time, were selected and their interactions were studied using a central composite design. The optimal extraction condition was 60.2% hydroethanol, a liquid/solid ratio of 29.3 mL/g, an extraction time of 18.4 min, an extraction temperature of 50 °C, and ultrasound power of 400 W. Under the optimal conditions, antioxidant activity of the extract was 755.98 ± 10.23 μmol Trolox/g dry weight (DW), much higher than that from maceration (558.77 ± 14.42 μmol Trolox/g DW) or Soxhlet extraction (479.81 ± 12.75 μmol Trolox/g DW). In addition, the main antioxidant compounds in the extract were identified and quantified by high-performance liquid chromatography–diode array detection–tandem mass spectrometry (HPLC–DAD–MS/MS). The concentrations of digalloyl hexoside, methyl gallate, gallic acid, trigalloyl hexoside, and digallic acid were 15.30 ± 0.98, 8.85 ± 0.51, 8.76 ± 0.36, 4.27 ± 0.21, and 2.89 ± 0.13 mg/g DW. This study provides an efficient and green extraction method for the extraction of natural antioxidants from the bean coat of red sword bean. The extract of antioxidants might be added into functional foods or nutraceuticals with potential beneficial functions.

Crossostephium chinensis (L.) (CC) Makino is a common traditional Chinese medicinal plant used to dehumidify and cure rheumatism and arthralgia. The water and methanol extracts of C. chinensis (CCW and CCM) were evaluated for their antioxidant and antiproliferative activities. The antioxidant activities of CC were evaluated by using ABTS radical scavenging, DPPH radical scavenging, nitric oxide scavenging and superoxide scavenging methods. Iron chelating activity, lipid peroxidation, total polyphenol contents, total flavonoid contents and total flavonol contents were also detected. In all the tested models, both CCW and CCM showed their ability to scavenge the free radicals in a does-dependent manner. CCW had higher antioxidant and antiproliferative activities than CCM. In LC-MS-MS analysis, the chromatograms of CCW with good antioxidant activities were established. Rutin might be an important bioactive compound in CCW. The antiproliferative activities of CCW and CCM were also studied in vitro by using human hepatoma HepG2 cells. CCW exhibited good antiproliferative activity. These results indicated that CCW might be used as a potential source of natural antioxidants and as an anti-tumor agent.

Background Amaranthus hybridus L. is an annual, erect or less commonly ascending herb that is a member of the Amaranthaceae family. Polysaccharides extracted from traditional Chinese medicines may be effective substances with antioxidant activity. Methods In this study, we isolated crude polysaccharides from A. hybridus (AHP-M) using microwave-assisted extraction. Then, the AHP-M was purified by chromatography with DEAE-32 cellulose, and two fractions, AHP-M-1 and AHP-M-2, were obtained. The structural characteristics of AHP-M-1 and AHP-M-2 were investigated, and their antioxidant activities were analyzed in vitro. Results We found that the monosaccharide composition of AHP-M-1 was different from that of AHP-M-2. The molecular weights of AHP-M-1 and AHP-M-2 were 77.625 kDa and 93.325 kDa, respectively. The results showed that the antioxidant activity of AHP-M-2 was better than that of AHP-M-1. For AHP-M-2, the DPPH radical scavenging rate at a concentration of 2 mg/mL was 78.87%, the hydroxyl radical scavenging rate was 39.34%, the superoxide anion radical scavenging rate was 80.2%, and the reduction ability of Fe3+ was approximately 0.90. The total antioxidant capacity per milligram of AHP-M-2 was 6.42, which was higher than that of Vitamin C (Vc). Conclusion The in vitro test indicated that AHP-M-1 and AHP-M-2 have good antioxidant activity, demonstrating that A. hybridus L. polysaccharide has immense potential as a natural antioxidants.

Flavonoids were extracted from Phyllostachys heterocycla leaves by adopting microwave-assisted extraction technology. Based on the single factor experiment and Plackett-Burman design results, the extraction process of flavonoids was further optimized using the response surface methodology. The optimum conditions were as follows: an ethanol concentration of 78.1%, an extraction time of 24.9 min, and a microwave power of 559 W. Under these conditions, the extraction yield of flavonoids was 4.67%, which was in close proximity to the predicted value (4.70%) and higher than the extraction yield from traditional Soxhlet extraction (3.35%). Moreover, the possible extraction mechanisms of these two extraction methods were further derived to explain why the microwave-assisted extraction of flavonoids was more efficient compared with traditional Soxhlet extraction. Ultimately, the antioxidant activities in vitro of flavonoids from Phyllostachys heterocycla leaves were evaluated via DPPH and ABTS radical scavenging assay. The flavonoids from Phyllostachys heterocycla leaves exhibited excellent antioxidant activities in vitro and Phyllostachys heterocycla leaves could be a new natural source for developing antioxidants. Overall, the findings of this research could provide a theoretical reference for the further comprehensive development and utilization of bamboo resources.

Dietary polyphenols exist in two forms; extractable polyphenols (EPP) or compounds solubilised by aqueous/organic solvents, and non-extractable polyphenols (NEPP) or compounds remain in the corresponding residues after the extraction. At present, most researchers focus on EEP fractions, while NEPP is neglected. Thus, this study aimed to release NEPP from the remaining powder residue of Barhi date palm kernels (BDPK) with acid hydrolysis. The related extraction conditions were determined and optimised using response surface methodology (RSM) for maximisation of NEPP with highest cytotoxic and antioxidant activities. The face-centred central composite design (FCCCD) was used to establish treatments based on three independent variables, namely; extraction temperature, time, and solvent/sample ratio. Under the optimal conditions, the experimental values for DPPH radical-scavenging capacity of NEPP (IC50=57.52µg/mL), and cytotoxicity of NEPP against A549 and HT29 cells were IC50=17.4 µg/mL and 31.4µg/mL, respectively. The experimental values were in agreement with those predicted by RSM models, confirming the suitability of the model employed and the success of RSM for optimisation of the extraction conditions for NEPP from BDPK. These results indicate that NEPP from industrial date fruit waste could be a promising candidate as natural antioxidants with significant antiproliferation effect against A549 and HT29 cancer cells in-vitro.

Rich in phenolic compounds, Moringa oleifera leaf extract (ME) exhibits significant antioxidant activity both in vitro and in vivo. ME has already been widely used in fields of medicine, functional food, and cosmetics. Ultrasonic extraction (UE) method has been improved to be one of the most effective ways to extract phenols from M. oleifera leaves. The purpose of this study was to optimize ultrasonic extraction of phenols by response surface methodology (RSM). Four parameters were discussed, such as ethanol concentration, solvent-sample ratio, extraction temperature, and extraction time. Also, purification methods of the crude ME by organic solvent extraction and column chromatography were examined. Antioxidant activities of ME and each fraction were evaluated by DPPH, ABTS, and hydroxy radical-scavenging activities and reducing power. The phenol content of the purified ME reached up to 962.6 mg RE/g, extremely higher than the crude extract 107.22 ± 1.93 mg RE/g. The antioxidant activity of the purified ME was also significantly improved. Furthermore, phenols were identified by using the HPLC-MS method, and the results showed that there were 6 phenolic acids and derivatives and 7 flavonoids in ME. Quercetin-3-O-β-D-glucoside isolated from ME showed excellent DPPH and ABTS radical-scavenging abilities, which were comparable to VC.

In this study, the ultra-high pressure extraction (UHPE) conditions for obtaining the maximum flavonoid yield from Xinjiang jujube (Ziziphus jujuba Mill.) leaves (XJL) were optimized by response surface methodology (RSM). Box–Behnken design (BBD) was applied to evaluate the effects of four variables (extraction temperature, pressure, time and liquid-to-solid ratio) on flavonoid yield. The results showed that the optimal flavonoid yield (25.45 ± 0.21 mg/g) was derived at 50.0 °C, 342.39 MPa, 11.56 min, and a liquid-to-solid ratio of 43.95 mL/g. Eight compounds were tentatively identified and quantified as kaempferol and quercetin glycosides with UPLC-ESI-MS. Compared to ultrasound-assisted extraction (UAE), UHPE can obtain higher concentrations of total flavonoids and stronger DPPH and ABTS radical-scavenging activities in a much shorter time. Therefore, UHPE is an alternative to UAE for obtaining flavonoids from XJL, which may be an optional method for large-scale industrial flavonoid extraction from XJL.
